Effects of facial nerve axotomy and repair on vesicular acetylcholine transporter in facial nucleus of rats

Abstract:Aim. Vesicular acetylcholine transporter (VAChT) is a molecule that in responsible for the transportation of acetylcholine from the cytoplasm of cholinergic axon terminals into the synaptic vesicles. The biosynthesis of acetylcholine is greatly reduced following facial nerve injury, however, the effects of nerve injury on VAChT is unclear. In present study, the effects of facial nerve axotomy and repair on VAChT were examined in facial nucleus in rats. Methods. Sixty adult rats with 200 - 250 g were divided into two groups of 30 subjects: facial axotomy group and end-to-end anastomosis group. Immunohistochemical methods were conducted to observe the changes of VAChT-positive neurons two groups. Results. Axotomy of facial nerve resulted in a strikingly reduction in the number of VAChT-positive neurons and the intensity of immuno-staining in facial nucleus with the peak of the reduction reaching 7 day after operation. The patterns of reduction of VAChT immunostaining following an immediate end-to-end anastomosis 7 days after operation were similar to those of axotomy group. The redomination of facial nerve was accompanied by the recover of VAChT-positive neurons in facial nucleus. The t-test showed significant differences between two group for the intensity of immunostaining of VAChT 14, 21 and 35 d post-operation respectively (t14 = 10.382, t21 = 13.892, t35 = 16.245, P < 0.05). Conclusion. The facial nerve injury resulted in a striking reduction in VAChT synthesis in facial nucleus, while VAChT recovered accompanied by the regeneration of facial nere.
