Experimental study on the effects of recombinate human BDNF in the cultured neurons of Alzheimer's disease model

Abstract:Objective: To investigate the effects of recombinate human BDNF (rhBDNF) on normal prime cultured hippocampal neurons and neurons of Alzheimer's disease (AD) model. Methods: 20 μmol·L-1 Aβ 25-35 to cultured hippocampal neurons before it was treated with rhBDNF. The results were obtained by morphological observation, ACHE histochemistry, confocal microscopy observation and MTT methods. Results: The neuron survival rate in control group decreased to 59.5%, which showed significant difference compared with that of blank group(P<0.01). The neuron survival rate in rhBDNF experimental group rose to 65.7%-72.6%, and there appeared a significant difference between experimental group and control group (P<0.05). The optimum concentration of rhBDNF was 50 ng·mL-1. The quantity of cholinergic neuron in rhBDNF experimental group increased, diameter of cell body and length of processes augmented, and [Ca2+]i comparatively stable. Conclusion: rhBDNF has neurotrophic effects on normal cultured hippocampal neurons and can prolong the living time of the cell. In cultured hippocampal neurons of AD model, rhBDNF has the protective function, and it can decrease cell mortality rate greatly, weaken the toxicity caused by the Aβ and help prevent and cure AD.
