Effects of estrogen on the activation of hepatic stellate cells and collagen synthesis in liver fibrotic rats

Abstract:Objective: To study the effects of estrogen on the activation of hepatic stellate cells and collagen synthesis in liver fibrotic rats, and then to investigate the suppressive effects of estrogen on liver fibrosis. Methods: Liver fibrosis was induced in rats by CCl 4 administration, and was treated with benzoic estradiol (1 mg ·kg -1) twice weekly. The expression of hepatic stellate cells positive for a-smooth muscle actin (α- SMA), the collagen synthesis and other indicators were studied at the end of the experiment. Results: Estradiol treatment reduced AST, ALT, hyaluronic acid (HA) and type IV collagen (CIV) in serums, suppressed hepatic collagen content, decreased areas of hepatic stellate cells positive for α- SMA, and lowered the synthesis of hepatic type I collagen significantly in the fibrotic rats induced by CCl 4 administration. Conclusion: Estradiol suppresses the activation of hepatic stellate cells and collagen synthesis, thus reducing CCl 4-induced hepatic fibrosis in rats.
