Abstract:Objective: To set up a three-parameter method for cell cycle analysis by two-laser flow cytometer, which can detect two types of cyclin plus DNA content in one measurement, and that analyze unscheduled expression of cyclins. Methods: Three-color fluorescence was used for analysis of two types of cyclins and DNA content simultaneously in individual cells by two-laser flow cytometry. MOLT-4 cells were used to study the expression of major cyclins in mammalian cells. A Triton-X100 permeabilization procedure was optimized for detection of two types of cyclins. One cyclin was stained directly with a FITCconjugated monoclonal antibody (mAb), and the other, indirectly with RPE-Cy5-conjugated secondary antibody, while DNA was stained with the fluorochrome DAPI. mAMSA and mimosine treated MOLT-4cells were used to test this three-parameter method. Results: Permeabilization with 0.5% Triton-X100 in PBS containing 1% BSA for 5 min on ice provided optimal conditions for the simultaneous labelling of two cyclins plus DNA in single cells. It was found that the emission spectrum of the three dyes (DAPI, FITC and RPE-Cy5) could be measured with no compensation. Based on cyclinA/cyclinE/DNA flow cytometric analysis, asynchronously growing MOLT-4 cells could be dividedinto 6 compartments (G1o, G1e, G1l,S, G2, and M) simultaneously, allowing for analysis of cell cycle phase specific perturbations without the necessity of cell synchronization. Unscheduled cyclin B1 expression was observed in G1 cells treated with mimosine and cyclin E in G2 cells treated with mAMSA. We found that unscheduled cyclin expression paralleled expected cyclin expression. Conclusion: Thus, three-color FCM analysis of cells may not only be applied to measure unscheduled vs. expected cyclin expression but may also be used to estimate the fraction of cycling cells in up to 6 cell populations.
A Miniature Modular Fluorescence Flow Cytometry System

Abstract:Fluorescence flow cytometry is a powerful instrument to distinguish cells or particles labelled with high-specificity fluorophores. However, traditional flow cytometry is complex, bulky, and inconvenient for users to adjust fluorescence channels. In this paper, we present a modular fluorescence flow cytometry (M-FCM) system in which fluorescence channels can be flexibly arranged. Modules for particle focusing and fluorescence detection were developed. After hydrodynamical focusing, the cells were measured in the detection modules, which were integrated with in situ illumination and fluorescence detection. The signal-to-noise ratio of the detection reached to 33.2 dB. The crosstalk among the fluorescence channels was eliminated. The M-FCM system was applied to evaluate cell viability in drug screening, agreeing well with the commercial cytometry. The modular cytometry presents several outstanding features: flexibility in setting fluorescence channels, cost efficiency, compact construction, ease of operation, and the potential to upgrade for multifunctional measurements. The modular cytometry provides a multifunctional platform for various biophysical measurements, e.g., electrical impedance and refractive-index detection. The proposed work paves an innovative avenue for the multivariate analysis of cellular characteristics.
Microfluidic Quantitative Flow Cytometer with Light Modulation

Abstract:Quantitative detections of proteins for single cells have made great contributions to tumor heterogeneity and immune variance while effective tools of measuring single-cell intracellular proteins are under developments. This study reported a flow cytometer based on constriction microchannel and light modulation, which was capable of quantifying intracellular proteins for single cells with high resolution. Cells tagged with fluorescein labelled antibodies were squeezed into the constriction microchannel as either the calibration or detection structure where a light source with a carrier wave was used for fluorescent modulation, which was further demodulated into the low-frequency domain and translated into numbers of single-cell proteins. In platform characterization, the inclusion of light modulation was shown to effectively improve the detection resolution from ~1000 intracellular proteins per cell without modulation to ~100 intracellular proteins per cells with modulation. As demonstrations, expressions of mutant p53 or phosphorylated p53 from tens of thousands of single T47D and/or A431 cells were quantitatively measured by the microfluidic quantitative flow cytometer, which were 1.78 ± $0.96 \times10$ 5 mutant p53 per cell for T47D cells ( ${N}_{c} =10\,315$ ), 1.99 ± $1.49\times10$ 5 mutant p53 per cell for A431 cells ( ${N}_{c} =15\,665$ ), and 6.34 ± $7.50\times10$ 3 phosphorylated p53 per cell for A431 cells treated with doxorubicin ( ${N}_{c} =12\,812$ ). In conclusion, the inclusion of light modulation can effectively decrease electrical and optical noises and therefore improve detection resolution in microfluidic quantitative flow cytometry.

Abstract:Flow cytometry has been proven to be a very versatile tool for cell biology research and clinical applications. A drawback of conventional technique lies in the fundamental limit of extracting morphological characteristics of the interrogated cells. In this paper, we present recent results of instrumentation design of flow cytometer systems with a new diffraction imaging method for the of 3D cell features measurement consisting of a jet-in-fluid flow chamber, a dual-polarization imaging system and a friendly human-computer interaction interface. A precise fluid control unit has been developed for producing stable laminar flow that is critical for acquisition of dual-polarization diffraction images and cell classification. Virtual simulation and experiment test results of the flow control unit and the new system with polystyrene microspheres and cells indicated the strong correlations between the diffraction image patterns and the 3D morphology of the particles.

Abstract:This paper presented a microfluidic flow cytometry with a uniform optical field, which is capable of translating fluorescent signals of antibodies bound with targeted proteins of single cells into protein numbers regardless of distribution patterns due to volume equivalence. In this study, the uniform optical field for fluorescent excitation and detection was modeled with numerical simulations. Furthermore, the microfluidic flow cytometry with the uniform optical field was used to quantify two key transcriptional factors of OCT4 and c-Myc without specific expression locations within cells from three well-established tumor cell lines of HeLa, A549 and Hep G2, validating the capability of this platform in analyzing single-cell proteins with arbitrary distributions.

Abstract:Abstract Imaging flow cytometry (IFC) combines flow cytometry and fluorescence microscopy to enable high-throughput, multiparametric single-cell analysis with rich spatial details. However, current IFC techniques remain limited in their ability to reveal subcellular information with a high 3D resolution, throughput, sensitivity, and instrumental simplicity. In this study, we introduce a light-field flow cytometer (LFC), an IFC system capable of high-content, single-shot, and multi-color acquisition of up to 5,750 cells per second with a near-diffraction-limited resolution of 400-600 nm in all three dimensions. The LFC system integrates optical, microfluidic, and computational strategies to facilitate the volumetric visualization of various 3D subcellular characteristics through convenient access to commonly used epi-fluorescence platforms. We demonstrate the effectiveness of LFC in assaying, analyzing, and enumerating intricate subcellular morphology, function, and heterogeneity using various phantoms and biological specimens. The advancement offered by the LFC system presents a promising methodological pathway for broad cell biological and translational discoveries, with the potential for widespread adoption in biomedical research.

Abstract:Flow cytometry is among the most powerful tools for single-cell analysis, while the high cost and mechanical complexity of the commercial instrumentation limit the applications in personalized single-cell analysis. For this issue, we hereby construct an open and low-cost flow cytometer. It is highly compact to integrate the functions of (1) single cell aligning by a lab-made modularized 3D hydrodynamic focusing device, and (2) fluorescence detection of the single cells by a confocal laser-induced fluorescence (LIF) detector. The ceiling cost of the entire hardware for the LIF detection unit and 3D focusing device is 400 respectively. A sheath flow velocity of 150 μL/min produces a focused sample stream of 17.6 μm × 14.6 μm at sample flow of 2 μL/min, based on the LIF response frequency and the laser beam spot diameter. The assay performance of the flow cytometer was evaluated by characterizing fluorescent microparticles and acridine orange (AO) stained HepG2 cells, producing throughputs of 40.5/s and 6.2/s respectively. Favorable assay precision and accuracy were demonstrated by the agreement of frequency histogram with imaging analysis, and good Gaussian-like distributions of fluorescent microparticles and AO-stained HepG2 cells. Practically, the flow cytometer was successfully applied for the evaluation of ROS generation in single HepG2 cells.

Abstract:Molecular interactions in live cells play an important role in both cellular functions and drug discovery. Current methods for measuring binding kinetics involve extracting the membrane protein and labeling, while the in situ quantification of molecular interaction with surface plasmon resonance (SPR) imaging mainly worked with fixed cells due to the micro-motion related noises of live cells. Here, an optical imaging method is presented to measure the molecular interaction with live red blood cells by tracking the nanometer membrane fluctuations. The membrane fluctuation dynamics are measured by tracking the membrane displacement during glycoprotein interaction. The data are analyzed with a thermodynamic model to determine the elastic properties of the cell observing reduced membrane fluctuations under fixatives, indicating cell fixations affect membrane mechanical properties. The binding kinetics of glycoprotein to several lectins are obtained by tracking the membrane fluctuation amplitude changes on single live cells. The binding kinetics and strength of different lectins are quite different, indicating the glycoproteins expression heterogeneity in single cells. It is anticipated that the method will contribute to the understanding of mechanisms of cell interaction and communication, and have potential applications in the mechanical assessment of cancer or other diseases at the single-cell level, and screening of membrane protein targeting drugs.

Abstract:A microfluidic chip is designed and fabricated, and coupled with flow cytometry. By using Na2HPO4 solution added with hydroxypropyl methyl cellulose (HPMC) as buffer and sheathing fluid, the problems met with the flowing of microspheres in the microchannel were solved, and the fluidic conditions were under more effective control. By using a home-made laser induced fluorescence system and electrokinetic focusing, the counting of the microspheres and real time observation of their actual fluidic state through an inverted fluorescence microscope is made. The whole system is compact and easy to be operated. The consumption of reagent and sample is little and the speed of the assay is quick.

A low-cost flow cell for flow cytometry

Abstract:Flow cytometry is an essential analytical technique used in biomedical diagnostics to measure properties of cells, micro-organisms, and particles. Laser light is scattered from particles focused in a flow cell and collected by light sensors, where the intensity of the scattered light is a function of the scattering angle, the refractive index of the particle and surrounding medium, the wavelength of light, and the size and the shape of the particle. One of the critical parts of the cytometer is the flow cell where the particle stream is constrained into a tight region within 10-30 μm using hydrodynamic focusing. The conventional flow cells use thick quartz flow cells, which are expensive and therefore not suitable for instruments targeted for resource-constrained settings. We demonstrate a compact, economical, bio-compatible flow cell assembly design that incorporates inexpensive and easily available capillaries attached to sturdy polymer fixtures in a simple manner that performs the focusing of a sample stream of particles. The flow cell has been tested by studying the relation between sample core diameter, and sample and sheath flow rates. Small-angle scattering (forward scatter) and wide-angle scattering (side scatter) have been captured for the enumeration and characterization of particles. We show excellent agreement between the size distribution obtained via direct imaging and that obtained from light scattering. The flow cell was also used to successfully size white blood cells in human blood samples.

Abstract:Illumination spot in a flow cytometer is a crucial factor determining the measurement accuracy and stability. The traditional mechanism is to precisely calibrate multiple optical components to convert circular Gaussian beams into elliptical Gaussian beams, making it difficult to shape multiwavelength lasers simultaneously. A diffractive beam shaper for multicolor lasers with high simplicity, only containing one diffractive optical element and one focusing lens is created in this work. It can produce rectangular spots, of which the number, the sizes, and the positions are accurately determined by the incident wavelengths. Demonstrated in the customized microflow cytometer, the coefficient of variations (CV) of the optical signals by the beam shaper are 3.6–6.5%, comparable to those derived from the commercial instrument with 3.3–6.3% CVs. Benefiting from the narrow rectangular spots and the flexibility of diffractively shaped lasers, the measurement of bead sizes with 4–15 μm diameters and the real‐time detection of flow velocity from 0.79 to 9.50 m/s with the CV of <5% are achieved. © 2020 International Society for Advancement of Cytometry

Abstract:Flow cytometry is a widespread and powerful technique whose resolution is determined by its capacity to accurately distinguish fluorescently positive populations from negative ones. However, most informative results are discarded while performing the measurements of conventional flow cytometry, e.g., the cell size, shape, morphology, and distribution or location of labeled exosomes within the unpurified biological samples. Herein, we propose a novel approach using an anti-diffraction light sheet with anisotroic feature to excite fluorescent tags. Constituted by an anti-diffraction Bessel–Gaussian beam array, the light sheet is 12 μm wide, 12 μm high, and has a thickness of ~0.8 μm. The intensity profile of the excited fluorescent signal can, therefore, reflect the size and allow samples in the range from O (100 nm) to 10 μm (e.g., blood cells) to be transported via hydrodynamic focusing in a microfluidic chip. The sampling rate is 500 kHz, which provides a capability of high throughput without sacrificing the spatial resolution. Consequently, the proposed anti-diffraction light sheet flow cytometry (ADLSFC) can obtain more informative results than the conventional methodologies, and is able to provide multiple characteristics (e.g., the size and distribution of fluorescent signal) helping to distinguish the target samples from the complex backgrounds.

A high-throughput all-optical laser-scanning imaging flow cytometer with biomolecular specificity and subcellular resolution

Abstract:Image-based cellular assay advances approaches to dissect complex cellular characteristics through direct visualization of cellular functional structures. However, available technologies face a common challenge, especially when it comes to the unmet need for unraveling population heterogeneity at single-cell precision: higher imaging resolution (and thus content) comes at the expense of lower throughput, or vice versa. To overcome this challenge, a new type of imaging flow cytometer based upon an all-optical ultrafast laser-scanning imaging technique, called free-space angular-chirp-enhanced delay (FACED) is reported. It enables an imaging throughput (>20 000 cells s-1 ) 1 to 2 orders of magnitude higher than the camera-based imaging flow cytometers. It also has 2 critical advantages over optical time-stretch imaging flow cytometry, which achieves a similar throughput: (1) it is widely compatible to the repertoire of biochemical contrast agents, favoring biomolecular-specific cellular assay and (2) it enables high-throughput visualization of functional morphology of individual cells with subcellular resolution. These capabilities enable multiparametric single-cell image analysis which reveals cellular heterogeneity, for example, in the cell-death processes demonstrated in this work-the information generally masked in non-imaging flow cytometry. Therefore, this platform empowers not only efficient large-scale single-cell measurements, but also detailed mechanistic analysis of complex cellular processes.
High-Fidelity Single Molecule Quantification in a Flow Cytometer Using Multiparametric Optical Analysis

Abstract:Microfluidic techniques are widely used for high-throughput quantification and discrete analysis of micron-scale objects but are difficult to apply to molecular-scale targets. Instead, single-molecule methods primarily rely on low-throughput microscopic imaging of immobilized molecules. Here we report that commercial-grade flow cytometers can detect single nucleic acid targets following enzymatic extension and dense labeling with multiple distinct fluorophores. We focus on microRNAs, short nucleic acids that can be extended by rolling circle amplification (RCA). We labeled RCA-extended microRNAs with multicolor fluorophores to generate repetitive nucleic acid products with submicron sizes and tunable multispectral profiles. By cross-correlating the multiparametric optical features, signal-to-background ratios were amplified 1600-fold to allow single-molecule detection across 4 orders of magnitude of concentration. The limit of detection was measured to be 47 fM, which is 100-fold better than gold-standard methods based on polymerase chain reaction. Furthermore, multiparametric analysis allowed discrimination of different microRNA sequences in the same solution using distinguishable optical barcodes. Barcodes can apply both ratiometric and colorimetric signatures, which could facilitate high-dimensional multiplexing. Because of the wide availability of flow cytometers, we anticipate that this technology can provide immediate access to high-throughput multiparametric single-molecule measurements and can further be adapted to the diverse range of molecular amplification methods that are continually emerging.

Abstract:Raman flow cytometry (RFC) uniquely integrates the "label-free" capability of Raman spectroscopy with the "high-throughput" attribute of traditional flow cytometry (FCM), offering exceptional performance in cell characterization and sorting. Unlike conventional FCM, RFC stands out for its elimination of the dependency on fluorescent labels, thereby reducing interference with the natural state of cells. Furthermore, it significantly enhances the detection information, providing a more comprehensive chemical fingerprint of cells. This review thoroughly discusses the fundamental principles and technological advantages of RFC and elaborates on its various applications in the biomedical field, from identifying and characterizing cancer cells for in vivo cancer detection and surveillance to sorting stem cells, paving the way for cell therapy, and identifying metabolic products of microbial cells, enabling the differentiation of microbial subgroups. Moreover, we delve into the current challenges and future directions regarding the improvement in sensitivity and throughput. This holds significant implications for the field of cell analysis, especially for the advancement of metabolomics.

Abstract:Flow cytometer (FCM) can be used to obtain the muti - parameters of the light scattering and fluorescence of individual cell rapidly and conveniently. It has been more and more applied in oceanographic and limnological studies. The work principles of FCM and its applications in the ecological studies of nano- and pico- phytoplankton were introduced and reviewed based on some recent publications abroad and at home in this paper. By means of FCM, sub- populations of phytoplankton can easily be classified, counted and sorted, and then further studies of the adaptive ability and ecological responses of the sub - populations can be implemented. It was concluded that FCM would have a prosperous prospect in the studies of the primary production and the microbial food web in marine ecosystem.
A Quantitative Method for Measurement of HL-60 Cell Apoptosis Based on Diffraction Imaging Flow Cytometry Technique.

Abstract:A quantitative method for measurement of apoptosis in HL-60 cells based on polarization diffraction imaging flow cytometry technique is presented in this paper. Through comparative study with existing methods and the analysis of diffraction images by a gray level co-occurrence matrix algorithm (GLCM), we found 4 GLCM parameters of contrast (CON), cluster shade (CLS), correlation (COR) and dissimilarity (DIS) exhibit high sensitivities as the apoptotic rates. It was further demonstrated that the CLS parameter correlates significantly (R(2) = 0.899) with the degree of nuclear fragmentation and other three parameters showed a very good correlations (R(2) ranges from 0.69 to 0.90). These results demonstrated that the new method has the capability for rapid and accurate extraction of morphological features to quantify cellular apoptosis without the need for cell staining.

Abstract:We report a method of coupled CE-LIF detection with flow cytometry for high-throughput determination and quantitation of fluorophores in single intact K562/S (KS) cells. The membrane properties of KS cell including fluophore transport rate and apparent permeability coefficient were further quantitatively characterized. The method has advantages for accurate quantitation and unique capacity of high-throughput analysis. The strategy will be useful for the quantitation of fluorophores in the intact cells, such as measurement of multidrug resistance, quantitation of specific protein expression, and quantitative characterization of protein and enzyme functions.
