Abstract:OBJECTIVE:To investigate the secondary metabolites from Chinese marine Sponge Cinachyrella australiensis.METHODS:Column chromatography techniques including HPLC were used for the separation and purification of the compounds, and extensive spectral analyses including various 2D NMR spectra were employed for structure elucidation.RESULTS:Nineteen compounds were obtained,including 2-methyoxy-6,12,15-trien-8-yne-octadecanoic acid (1), 2-benzenedicarboxylic acid dibutyl ester(2), 1,2-benzenedicarboxylic acid bis(2-ethylhexyl)ester(3), (-) (3S)-1,2,3,4-tetrahydro-beta-carboline-3-carboxylic acid(4), L-Tryptophan (5), p-hydroxylbenzaldehyde (6), p-hydroxyl-benzylethanol(7), p-hydroxyl-benzyl-propanol(8), cholesta-4-en-3-ol(9), 2-methyl-6-amino-9-(2-deoxy-beta-D- ribofuranosyl-purine(10), 2'-Deoxyadenosine (11), 6-amino-9-beta-D-ribofuranosyl-9H-purine (12),uracil(13), thymine(14), thymidine(15), 1-(2-deoxy-beta-D-Ribofuranosyl) uracil(16), 1-ethyl-alpha-(2-deoxy)-beta-D-ribofuranos(17),isolumichrome(18),and zarzissine(19).CONCLUSION:Compounds 1 and 18 are new natural products,and compounds 2 to 17 as well as 19 are isolated from this species for the first time.

Abstract:Objective:To study the chemical constituents in the fresh marine sponge Spongilla Wagner. Methods:Compounds were separated and purified through various chromatographic methods and their structures were identified by spectroscopic data. Results:Seven compounds were isolated and identified as 4-hydroxybenzoic acid(Ⅰ) , p-hydroxycinnamic acid(Ⅱ) , Pyrimidine-2, 4(1H,3H)-dione(Ⅲ), Ferulic acid methyl ester(Ⅳ) , Dodecyl ethers of glycerol(Ⅴ) , Tetracosane(Ⅵ). Conclusion: All compounds are isolated from this sponge for the first time.

Abstract:Marine sponges are a family of important natural sources producing natural products with rich chemical diversity and significant bioactivities.In this paper,we summarizes the research progress of our group on the structural diversity and bioactivity of metabolites from the sponges collected in South China Sea to provide useful data for further discovery of lead compounds derived from marine organisms.

Abstract:Three indole compounds(namely 1H-Indole-3-carboxaldehyde(1),1H-Indole-3-carboxylic acid(2),and ethyl 1H-Indole-3-carboxylate(3)) and 3-β-5-en-cholest(4) were isolated from the south China sea sponge,Xetospongia testudinaria,by repeatedly silica gel and Sephedex LH-20 chromatography.Their structures were identified by extensive spectroscopic analysis together with NMR、IR and EI(ESI)-MS spectrum,and further determined by comparing their physical and chemical data with the literature.All the identified compounds are isolated from the sponge,Xetospongia testudinaria,at the first time.

Methods The chemical constituents were isolated by various methods of isolation and their structures were elucidated by physicochemical properties and the analysis of spectral data compared with those reported in literatures. Results Six steroids including cholesterol (1), cholest-4-en-3-one (2), cholest-3,6-dione (3), 3β-hydroxy-cholest-5 -en-7-one ( 4 ), 6β-hydroxy-cholest-4 -en-3-one ( 5 ), cholest-3β, 5α,6β-triol (6)were isolated from the marine sponge Spheciospongia sp. collected from the South China Sea.Conclusions The compounds 2 -6 are isolated form the marine sponge for the first time.

Abstract:OBJECTIVE:To study the chemical constituents of the marine sponge Rhabdastrella aff. distincta.METHOD:Compounds were obtained by normal phase silica gel, Sephadex LH-20, reverse phase chromatogrophy and methylation method. These structures were determined on the basis of IR, MS, NMR spectral analysis.RESULT:Eight compounds were isolated from bioactive CH3Cl fraction and identified as geoditin A (1), geoditin B (2), stellettin A (5), stellettin B (4), stellettin C (3), stellettinE (7), rhabdastrellic acid A (8) and the methyl ester of stellettin E (6).CONCLUSION:All these compounds were obtained from this species for the first time.

Abstract:Chemical examination of the Chinese marine sponge Xestospongia testudinaria led to the isolation of 39 new brominated polyunsaturated compounds, which were designated with the trivial names xestospongienes A–Z and Z1–Z13. The structures of these compounds were elucidated by analyzing extensive spectroscopic (IR, MS, 1D, and 2D NMR) data and the results from CD spectral assignment, chemical conversion, and Mosher reaction.

Abstract:Objective To investigate the chemical constituents and bioactivity of the South China Sea sponge Luffariellasp..Methods The compounds were isolated and purified by using silica gel column chromatography, reversed-phase C18 ODS chromatography and semi-preparative HPLC.Their structures were identified by 1 Dand 2 DNMR、MS、UV、IR and comparison with the data of literature.They were performed an NF-κB inhibitory activity evaluation test.Results Six sesterterpenoids withα、β-unsaturated-γ-lactone ring were isolated from the sponge.Their structures were identified as Hippolide H (1) 、Hippolide F (2) 、Hippolide G (3) 、Hippolide E (4) 、6 Z-neomanoalide (5) 、6 E-neomanoalide (6).In the evaluation of NF-κB inhibitory activity, compounds 1, 4 and6 showed good NF-κB inhibitory activity.Conclusion The South China Sea sponge Luffariella sp.could produce sesterterpenoids, some of which have good NF-κB inhibitory activity.

Abstract:Marine sponges are one of the prolific producers of bioactive natural products with therapeutic potential. As an important subgenus of Haliclona, Reniera sponges are mainly distributed in the Mediterranean Sea and Atlantic area, and had been chemically investigated for over four decades. By an extensive literature search, this review first makes a comprehensive summary of all natural products from Reniera sponges and their endozoic microbes, as well as biological properties. Perspectives on strengthening the chemical study of Reniera sponges for new drug-lead discovery are provided in this work.

Abstract:Four new sesterterpenes, namely, (+)-20-formylhyrtiosal (1), (+)-16-O-acetyl-20-formylhyrtiosal (2), 12alpha-O-acetylhyrtiolide (3), and 5,10-dihydroxyfurospinulosine-1 (4), together with seven known sesterterpenes (5-11) were isolated from the marine sponge Hyrtios erectus collected at Hainan, China. The structures were elucidated on the basis of extensive spectroscopic analysis involving mainly one- and two-dimensional NMR as well as mass spectroscopy.

Abstract:Five sterols with a nucleus skeleton of 6-hydroxy-4-en-3-one, namely cholesta-6beta-hydroxy-4-en-3-one (1), ergosta-6beta-hydroxy-4,24(28)-dien-3-one (2), ergosta-6alpha-hydroxy-4,24(28)-dien-3-one (3), ergosta-6alpha-hydroxy-4,22-E-dien-3-one (4), and ergosta-28-methyl-6beta-hydroxy-4,24(28)-dien-3-one (5) have been isolated from the marine sponge Iotrochoto birotulata, collected from the southern China sea. Sterols 2-4 are new compounds, and 1 has been isolated from marine organisms for the first time. The structures have been elucidated on the basis of extensive spectroscopic and chemical properties.

Abstract:A bioassay-guided chemical investigation of the South China Sea sponge Acanthella cavernosa resulted in the isolation of eight new diterpenoids, kalihinols M–T (1–8), together with seven known analogues (9–15). These compounds featured a trans-decalin ring bearing a tetrahydrofuran or a tetrahydropyran ring at C-7. Compounds 1 and 2, with a formamide functionality beared at C-4, extended the structure breadth of this diterpenoid family. The absolute stereostructures of 1–14 were determined by a combination of 2D NMR and CD spectroscopic analysis and single crystal X-ray diffraction. Compounds 1 and 2 were confirmed to have the configurations of 4S, 5S, whereas 3–14 were determined as 4R, 5R. Compounds 3–14 displayed significant antifouling activity against the barnacle Balanus amphitrite larvae, and the cytotoxic activities of 3–14 were evaluated against the H1299, A549, PC3, CT-26, and HCT-116 cancer cell lines.

Results From the butanol fraction of the 95 % EtOH extract of the starfish Asterias rollestoni, a new compound N7-2'-deoxypseudoxanthosine (1), along with sixteen known compounds, 2'-O-methyl-inosine (2), 2'-deoxyinesine (3), 2'-O-methylguanosine (4), inosine (5); thymine (6), uracil (7), thymidine (8), deoxyuridine (9), 2'-O-methyluridine (10), (-)-( 1S, 3S)-l-methyl-1, 2, 3, 4-tetrahydro-β-carboline-3-carboxyl-ic acid (11), ( - )-(1R, 3S)-l-methyl-1, 2, 3, 4-tetrahydro-β-cacboline-3-carboxylic acid (12), ( - )-(3S)- 1, 2, 3,4-tetrahydro-β-carboline-3-carboxylic acid (13), L-tryptophan (14), L-phenylalanine (15), 3-carboxy indole (16), and p-hydroxybenzoic acid (17), have been isolated. Conclusion Compound 1 is a new natural product, and compounds 8, 9 and 10 are isolated from natural sources for the first time, and the known compounds except 14 and 15 are first reported from starfish Asterias rollestoni.

Abstract:In this study, a marine medicinal brown alga Sargassum cristaefolium-derived fungal strain Scytalidium lignicola SC228 has been isolated and identified. Column chromatography of the extracts from liquid-fermented products of the fungal strain was carried out, and led to the purification of eight compounds. Their structures were characterized by spectroscopic analysis, and the absolute configurations were further established by single X-ray diffraction analysis and modified Mosher's method as four previously undescribed compounds, namely scytabenzofurans A-C (1-3), and (3S,4S)-5-chloro-3,4-dihydro-4,6,8-trihydroxy-3-methyl-1H-2-benzopyran-1-one (4), along with four known compounds 5-8. All the isolates were subjected to anti-inflammatory and anti-angiogenic assays. Compounds 1-4, 7, and 8 showed moderate nitric oxide production inhibitory activities in lipopolysaccharide-activated BV-2 microglial cells with IC50 in the range of 19.6 ± 0.1 to 49.0 ± 1.2 μM in comparison with that of curcumin (IC50 = 2.7 ± 0.3 μM). Compounds 5-7 exhibited moderate to potent inhibitory effects on EPCs growth with IC50 in the range of 0.5 ± 0.1 to 42.7 ± 0.9 μM as compared to sorafenib (IC50 = 5.50 ± 1.50 μM).

Abstract:Three new spiro-sesquiterpenoids, myrmekiones A-C (1-3), were isolated from the marine sponge Myrmekioderma sp. collected from the South China Sea. The structures of 1-3 were experimentally illuminated though comprehensive NMR spectra, X-ray diffraction analysis and calculated ECD. These three compounds possessed a special spiro skeleton. Compound 1 was characterized by a chamigrane-type structure, it is the first time to obtain the single-crystal of this type of oil compounds. 2 and 3 were a pair of diastereoisomers that possessed an acorane skeleton. This study expands the chemical diversity of marine origin spiro-metabolites.

Abstract:Two cembranoid diterpenes, sinulariol D (Compound 1) and mayolide A (Compound 2) were firstly isolated from the soft coral Sinularia sp. by repeatedly silica gel chromatography. Their structures were identified by extensive spectroscopic analysis together with IR、EI(ESI)-MS spectrum, and further determined by comparing their physical and chemical data with the literatures. Compound 1 showed a moderate cytotoxicity against the selected tumor lines at the concentration 50 μg·mL -1 in vitro.

Abstract:OBJECTIVE: To study the chemical constituents of Sargassum thunbergii. METHODS: The compounds were isolated and purified upon extensive column chromatography using silica gel, ODS, Sephadex LH-20, and prepative HPLC, while their structures were identified on the basis of physicochemical properties and spectroscopic data analysis. RESULTS: Fifteen compounds were isolated from the EtOH extract. The structures were identified as(+)-isololiolide(1), (-)-loliolide(2), loliolide acetate(3) grasshopper ketone(4), apo-9β-fucoxanthinone(5), 3α-hydroxy-5, 6-epoxy-7-megastigmen-9-one(6), deacetylate-apo-13'-fucoxanthinone(7), apo-13'-fucoxanthinone(8),(R)-(-)-3-hydroxy-β-ionone(9) and 6 other compounds sargassum ketone(10), 2-heptenoicacid, 4,4-dihydroxy-2,3-dimethyl-7-lactone(11), (R)-2-hydroxy-3-phenylpropanamide(12), thymidine(13), fucosterol(14) and hexade-canoic acid-2β, 3β-dihydroxypropyl ester(15). CONCLUSION: Deacetylate-apo-13'-fucoxanthinone is a new compound, and compounds 3,5,6,8,9,15 are obtained from the genus Sargassum for the first time.
