Abstract:Two new chiral calix[4]arenes bearing chiral pendants, which were from by-product of the antibiotic industry, were synthesized and characterized by H-1 NMR, MS-FAB and elemental analysis. Studies of H-1 NMR of the two calix [4] arene derivatives indicate that they exist in cone conformation in solution. Results of chiral recognition of the two chiral ligands 2a mid 2b towards the tartaric add derivative 3 show that ligand 2a exhibited good chiral recognition abilities compared to ligand 2b.

Abstract:An efficient synthetic route to inherently chiral calix[6]arenes with an ABC substitution pattern at the wide rim and a mesitylenyl unit at the narrow rim in the cone conformation was developed for the first time. Based on the selective formylation and bromination of 1,3,5-bridged calix[6]arene 1, two new inherently chiral calix[6]arene derivatives 4 and 5 have been prepared with moderate yields. The 1H NMR spectra indicate that these inherently chiral compounds adopt a cone conformation and their three different groups at the wide rim can create a chiral environment inside the cavity that maybe sensed by some chiral guests, which has also been confirmed by X-ray crystal structure of 4 and molecular modelling of 5. Complexation studies show that 5 displays exceptional properties for the binding of [image omitted].

Efficient Synthesis and Resolution of Meta-Substituted Inherently Chiral Aminocalix[4]arene Derivatives

Abstract:Efficient synthesis and resolution of a series of meta-substituted inherently chiral aminocalix[4]arene derivatives have been described. Consequently, the meta-nitro, bromo and chloro substituted inherently chiral calix[4]arenes could be directly synthesized by the nitration, bromination, and chlorination of the acylating product of aminocalix[4]arene. meta-Cyano and phenyl substituted inherently chiral aminocalix[4]arenes were readily obtained by the nucleophilic substitution reaction or Suzuki coupling reaction from the meta-bromo substituted one under the Pd-catalyzed conditions. For kinetic resolution of the racemic inherently chiral aminocalix[4]arenes, it was found that the electron-withdrawing ability of substituent was helpful to improving the resolution efficiency of the acylation process, and the kinetic resolution could be efficiently applied to the resolution of meta-nitro substituted inherently chiral aminocalix[4]arene, providing (cS)- or (cR)-isomer in up to 95% or 99.9% ee value, respectively, with the corresponding chiral acylating reagent. Moreover, by introduction of the chiral auxiliary, enantiopure antipodes of meta-cyano and phenyl substituted inherently chiral aminocalix[4]arenes could also be readily obtained. These enantiopure aminocalix[4]arenes are potential building blocks for constructing novel chiral receptors and asymmetric catalysts.

Abstract:A series of p-tetra-tert-butyl calix[4]arene derivatives with alkyl guanidinium or chiral bicyclic guanidinium functionalities were synthesized as the receptors of organic anions. Mono- and di- alkyl guanidinium derivatives were prepared by the reaction of 5,11,17,23-tetra-tert-butyl-25,27-(2-aminoethoxy)-26,28-dihydroxy calix[4]arene (1) with methyl isothiourea sulphate. While mono- and di- chiral bicyclic guanicinium derivatives were all obtained via an O-alkylation reaction of p-tetra-tert-butyl calix[4]arene with chloromethyl chiral bicyclic guanidinium (6 or 7) in the presence of anhydrous K2CO3 in acetonitrile, good yields were only observed for mono- substituted derivatives.

Abstract:The p-tetra-tert-butyl calix[4] arene derivatives (3 and 4) with(S, S) chiral bicyclic guanidinium, as the receptors of amino acid zwitterions, have been synthesized via a O-alkylation reaction of p-tetra-a-tert-butyl calix [4] arene with chloromethyl chiral bicyclic guanidinium 2 in the presence of anhydrous K2CO3 in acetonitrile. The results obtained from liquid-liquid competitive extraction experiments indicate that the two receptors may selectively recognize L-aromatic amino acids, and that the enantioselective recognizability of the receptor 4 with two chiral bicyclic guanidinium units reachs up to about 90% for L-Phe.

Abstract:Five chiral calix[4]proline derivatives were designed and synthesized from aminocalix[4]arenes, and their structures were characterized by H-1 NMR, C-13 NMR, IR, ESI-MS and elemental analysis. The chiral recognition ability for enantiomers of mandelic acid was investigated by NMR techniques, and the results showed that 5,11-diprolinamides substituted calix[4]arene 2b could well and selectively recognize the two enantiomers of mandelic acid (K-L/K-D approximate to 94). A possible recognition mechanism was proposed, which was constructed by cooperative multi-supramolecular interactions including intermolecular acid-base, hydrogen bond and pi center dot center dot center dot pi stacking interactions.

Abstract:Chiral calix[4]arene amino alcohol 2 was synthesized by the reaction of 25,27-di(2-bromoethoxy)-26,28-dihydroxy-5,11,17,23-t-butylcalix[4]arene with optical pure (1S,2R)-(+)-2-amino-1,2-diphenylethanol. 2 can form 1:1 dimer by self-assembly in d-methanol and the association constant of the dimer is 34.7 M -1 determined by 1H NMR titration.

Abstract:We report herein an efficient approach for the enantioselective synthesis of inherently chiral calix[4]arenes via palladium-catalyzed asymmetric intramolecular C-H arylations. Using a chiral bifunctional phosphine-carboxylate ligand, the inherent chirality on macrocyclic scaffolds was induced successfully, from which a wide range of calix[4]arenes with fluorenone motifs were obtained with good yields and excellent enantioselectivities (up to >99% ee). The synthetic utility of this method was demonstrated by diverse transformations of the products, thus substantially expanding the chemical space of chiral calix[4]arenes. Further investigations of photophysical and chiroptical properties revealed that calix[4]arenes bearing two fluorenone moieties displayed remarkable glum values (up to 0.019), highlighting the great potential of inherent chirality in the development of organic optoelectronic materials.

Among them lots of host compounds and have wide applications in chiral calix[4]arene is investigated most widely due to its stable works reported the chiral calix[4]arenes can be used in enantiomeric recognition and dectection. Fluorescence, UV-vis and NMR are the most common methods in the research of chiral recognition. According to these methods, the latest research progress in the synthesis of chiral receptors based on calix[4]arenes and their applications in the chiral recognition are summarized. The prospect of chiral calix[4) arene is also discussed.

Abstract:Inherently chiral calix[4]arenes are an excellent structural scaffold for asymmetric synthesis, chiral recognition, sensing, and circularly polarized luminescence. However, their catalytic asymmetric synthesis remains challenging. Herein, we report an efficient synthesis of inherently chiral calix[4]arene derivatives via cascade asymmetric cyclization and oxidation reactions. The three-component reaction features a broad substrate scope (33 examples), high efficiency (up to 90% yield), and excellent enantioselectivity (>95% ee on average). The potential applications of calix[4]arene derivatives are highlighted by their synthetic transformation and a detailed investigation of their photophysical and chiroptical properties.

Abstract:Three novel types of chiral calixarene derivatives 5, 8, and 10 were designed and synthesized by introducing chiral units to parent calixarenes. Their chiralities were confirmed by rotational analysis. Chiral recognition properties of these host compounds towards L- and D-threonine were studied by UV-Vis spectroscopy. The results indicated that calixarene derivatives 5 and 8 exhibited good chiral recognition capabilities toward L- or D-threonine. Although calixarene derivative 10 had no evident chiral recognition ability, the supramolecules of calixarene derivative 10 with L- or D-threonine showed a hypochromic effect or hyperchromic effect respectively. Therefore, calixarene derivative 10 might serve as a good chiral UV-indicator.

Abstract:Inherently chiral calixarenes hold great potential for applications in chiral recognition, sensing, and asymmetric catalysis due to their unique structures. However, due to their special structures and relatively large sizes, the catalytic asymmetric synthesis of inherently chiral calixarenes is challenging with very limited examples available. Here, we present an efficient method for the enantioselective synthesis of inherently chiral sulfur-containing calix[4]arenes through the desymmetrizing electrophilic sulfenylation of calix[4]arenes. This catalytic asymmetric reaction is enabled by a chiral 1,1'-binaphthyl-2,2'-diamine-derived sulfide catalyst and hexafluoroisopropanol. Various inherently chiral sulfur-containing calix[4]arenes are obtained in moderate to excellent yields with high enantioselectivities. Control experiments indicate that the thermodynamically favored C-SAr product is formed from the kinetically favored N-SAr product and the combination of the chiral sulfide catalyst and hexafluoroisopropanol is crucially important for both enantioselectivity and reactivity.
