Abstract:A series of chiral tridentate ligands derived from readily available enantiopure l-proline were designed and synthesized. The ligands together with Cu(OAc)2 were successfully used in asymmetric Henry reactions. Various structurally divergent aldehydes and nitromethane were converted into versatile β-nitro alcohols in MeOH at room temperature with very good yields (up to 85%) and enantioselectivities (up to 86%).

Abstract:A novel protocol for the facile synthesis of chiral 3-substituted 1,3,4,5-tetrahydrobenzo[e] [1,4]diazepin-2-ones via a domino reaction has been developed, which involves a cascade of S(N)2 nucleophilic substitution and copper-catalyzed C-N coupling reactions of a-amino acid amides and orthohalobenzyl halides. This protocol has some merits, such as one-pot process, easy operation, a wide scope of substrates. Furthermore, no heavy racemization occurs during the reaction.

Abstract:A series of chiral C-2-symmetric bis(oxazoline) ligands containing semicrown ether unit were synthesized. The copper complexes prepared in situ from copper(I)-triflate, and the new chiral oxazoline ligands were assessed as chiral catalysts in the enantioselective cyclopropanation of styrene with diazoacetates. Enantioselectivities up to 84% and up to 65%, respectively, for trans- and cis-2-phenylcyclopropanecarboxylate were observed.

Abstract:The asymmetric B & uuml;chner reaction and related arene cyclopropanations represent one type of the powerful methods for enantioselective dearomatization. However, examples of asymmetric B & uuml;chner reactions via a non-diazo approach are quite scarce, and the related arene cyclopropanation based on alkynes has not been reported. Herein, we disclose an asymmetric B & uuml;chner reaction and the related arene cyclopropanation by copper-catalyzed controllable cyclization of N-propargyl ynamides via vinyl cation intermediates, leading to chiral tricycle-fused cycloheptatrienes and benzonorcaradienes in high yields and enantioselectivities. Importantly, this protocol represents an asymmetric arene cyclopropanation reaction of alkynes and an asymmetric B & uuml;chner reaction based on vinyl cations. Asymmetric B & uuml;chner reactions are an effective methodology to bring about synthetic complexity from the common benzene motif, but effective, general catalytic methods are scarce. Here, the authors present divergent asymmetric arene dearomatization from ynamides using copper catalysis, wherein the catalytic cycle proceeds via a putative vinyl cation intermediate.

Cu ( 11) complexes of 1a similar to 1c were applied to the asymmetric cyclopropanation of 1, 1-diphenyl ethylene with alkyl diazoacetate.

Abstract:A new kind of copper(I)-chiral Schiff base complex, formed in situ by Cu (CH 3CN) 4ClO 4 and the Schiff bases derived from (-)-(1R,2S)-2-amino-1,2-diphenyl ethanol and substituted salicylaldehyde, was employed in the catalytic asymmetric cyclopropanation of 2,5-dimethyl-2,4-hexadiene with L-menthyl diazo acetate, and 90% of yield and 30% of e.e. (cis) were obtained. Compared with copper(II) complex catalyst, this catalysis system showed better enantiomeric excess and higher activity. And in the system, Cu(CH 3CN) 4ClO 4 gave far higher activity than Cu(I)OTf and was believed to be a good reagent for the title reaction.

Abstract:A direct, efficient, and practical protocol to install a chiral oxazoline unit onto aryl/heteroaryl rings via palladium-catalyzed C-H functionalization of 2-positions of oxazolines with a variety of halides using dppe as the ligand has been developed. Various chiral oxazoline ligands could be synthesized, even in a 10-g scale process. This protocol is a good supplement to traditional methods and for diverse synthesis of chiral oxazoline ligands.

Abstract:Bipyridines represent a class of ligands renowned for their versatility and efficacy in numerous transition metal-catalyzed reactions. Chiral bipyridine ligands are noted for their distinctive reactivity and stereoselectivity. In this work, we have designed and synthesized a class of bipyridine ligands endowed with an axially chiral scaffold. These ligands feature: (a) a precisely aligned coplanar bipyridine framework that is beneficial for metal chelation; (b) a spacious environment around the metal-bipyridine complexes, capable of hosting substrates with substantial steric bulk; (c) a chiral pocket induced by the axially chiral architecture. These atropochiral planar bipyridine ligands were successfully applied in copper-catalyzed ring-opening reactions of cyclic diaryliodoniums with bulky secondary amines, achieving high efficiency and stereoselectivity that were unsuccessful in our previous efforts.

Abstract:A series of novel chiral C2-symmetric bis (oxazoline) ligands have been synthesized.The copper and magnesium complexes,prepared in situ from copper(Ⅱ)-triflate of magnesium triflate with the new enantiopure oxazoline ligands,were evaluated as chiral catalysts in the enantioselective Diels-Alder reaction of cyclopentadiene with N-crotenoyl-oxazolidin-2-one.Primary results showed that diastereoselectivity up to 94% and enantioselectivity up to 68% ee for endo products were observed respectively with these ligands.

Abstract:As a structural analog of oxazoline, imidazoline (4,5-dihydroimidazole) has received much attention in the rational design of chiral ligands. The additional N-substituent provides broader space for fine-tuning of electronic and steric effects, and it offers a good handle for immobilizing onto solid supports. In the past decades, imidazoline ring has emerged as a powerful candidate for the design of chiral nitrogen-containing ligands, as well as a significant alternative for oxazoline ring. Various chiral imidazoline ligands have been designed and utilized in asymmetric organic reactions. These new catalysts can not only be applied in classical reactions, but also be employed to develop new organic reactions with high enantioselectivities. This review provides an overview of chiral imidazoline ligands. Their applications in asymmetric synthesis are also summarized.

Abstract:A palladium-catalyzed C-H functionalization/intramolecular asymmetric allylation cascade of N-sulfonyl benzamides with 1,3-dienes has been developed. In the presence of a chiral pyridine-oxazoline ligand, this protocol enables the synthesis of chiral 3,4-dihydroisoquinolones in yields of up to 83% with enantioselectivities of up to 96%, using environmentally friendly air as the terminal oxidant.

Abstract:A wide range of isoquinoline ring-modified Quinap oxides with different steric and electronic variations have been constructed through a palladium-catalyzed imidoylative cyclization of arylethenyl isocyanides with 2-diphenylphosphinyl-1-naphthyl bromides. The Pd-catalysis plays dual roles in the formation of both axial C–C bond and isoquinoline ring. This de novo synthetic strategy features good functional group tolerance, high yields and easy scale-up, providing Quinap derivatives with substitution patterns that could not be obtained using coupling reactions. Chiral ligands 7 and 12 can be readily prepared by transformation of the resulting Quinap oxide to their BINOL esters, and have been proven to be superb chiral ligands for the copper-catalyzed enantioselective A3-coupling and alkynylation of quinoline reactions. In general, the enantioselectivies obtained using ligands 7 and 12 are excellent, and the ee values are higher than those using Quinap as ligand, even three times higher in some cases. Mechanism studies revealed that a monomeric copper(I) complex bearing a single chiral ligand was formed and served as the catalytically active species.

Abstract:Two N,N'-bichiral lariat crown ethers, (S,S)-1,7-bis(4-benzyl-5-hydroxy-2-oxo-3-azapentyl)-1,7-diaza-12-crown-4 (1a) and (S,S)-1,7-bis(4-benzyloxazolin-2-yl-methyl)-1,7-diaza-12-crown-4 (1b), were obtained by alkylation of 1,7-diaza-12-crown-4 with (S)-(1-hydroxy-3-phenylprop-2-yl)-chloroacetamide or (S)-2-chloromethyl-4-benzyloxazoline, which were obtained by the reaction of (S)-2-amino-3-phenylpropanol with triethyl orthochloroacetate. la was used successfully in the chiral separations of D/L-carnitine by capillary zone electrophoresis. Cu(II) complex of 1b was applied to the asymmetric cyclopropanation of 1,1-diphenylethylene with alkyl diazoacetate.

Abstract:A series of chiral C2-symmetric bis(oxazoline) ligands containing dibenzo[a,c]cycloheptadiene units were synthesized. The copper complexes, prepared in situ from copper (I)-triflate and the new enantiopure oxazoline ligands, were assessed as chiral catalysts in the enantioselective cyclopropanation of styrene with diazoacetate. Abstract:A new C 2 symmetric chiral Schiff base compound A with relatively flexible chiral environment was synthesized from L (+) tartaric acid. Its structure was determined by NMR, IR, MS etc, and its preponderant conformation was modeled by CS Chem 3D Program. The results showed that its structure is not in a plane and has a big chiral cavity. It was showed that the complex B has different catalytic performance in asymmetric cyclopropanation of styrene and DMHD (2,5 dimethyl 2,4 hexadiene) with diazoacetate. The catalytic mechanism was also discussed.

Abstract:The efficient asymmetric preparation of the C-2-symmetric chiral 1,4-diamine, (1R,2S,4R,5S)-1,4-diamino-2,5-dimethylcyclohexane (5), and its salen ligands is described. With the Mitsunobu reaction as the key step, the overall yield of the four-step synthesis of 5 is 45%. The enantioselective alkynylation of meso-epoxides catalyzed by chiral gallium complexes is also achieved.
