Effects of Tai lake bloom microcystins on immune function of mice

Abstract:AIM: In order to investigate the latent risk of Tai lake cyanobacteria blooms on mammals. METHODS: The mice were administered intraperitoneally with the microcystins extract from cyanobacteria blooms collected from the Tai lake in dose of 5 mL·kg -1 of body weight (equal to 20 mg·kg -1 of body weight), over a period of 7 d. On d 8, the immune function of mice was measured with macrophagocyte phagocytosis assay, the splenocyte proliferation assay, hemolytic spectrophotometry, reverse transcription-polymerase chain reaction (RT-PCR) and enzyme labeled immunosorbent assay(ELISA). RESULTS: Toxicity was displayed by a drop in body weight, increase in spleen index, reduction of phagocytic activity of phagocyte. The mitogenic response of splenic T cell to concanavalin A and the primary antibody response to sheep erythrocyte were significantly decreased in the BMC-treated mice. The results of PCR and ELISA elucidated that BMC down-regulated the expression level of tumor necrosis factor(TNF)-α mRNA and decreased the content of TNF-α in mouse serum. CONCLUSION: The findings indicate that BMC obviously mediated the immunosuppression on mice.
