Distribution and Immunotherapeutic Effects of IL-2 Administered Via the Splenic Artery in the Murine Model of Liver Cancer

Abstract:To investigate the distribution of IL 2 administered via the splenic artery, and its influence on the immune functions in the rat model of liver cancer. Methods: The rat model of orthotopic liver cancer was established as reported previously. 131 I IL 2 labeled by the Iodogen method, was administered intraperitoneally (IP group), intravenously (IV group) and via the splenic artery (SA group) respectively. The distribution and the accumulation rate of the 131 I IL 2 were compared among the groups. The level of endogenic IL 2, NK and LAK activity, changes in T cell subgroups, and level of immunosuppressive factors sIL 2R, TGF βand IL 10 were investigated. Results: After IP, IV and SA injection, 131 I IL 2 preferentially accumulated in the liver and the spleen. The elevation of serum IL 2 and its accumulation in the liver and spleen was faster and lasted longer after SA injection than the other group. In the IP and IV groups, the serum IL 2 level reached its peak on Day 4, then subsided gradually and reached the level of the control group on Day 8. Whereas in the SA group, the serum IL 2 peak level was reached on Day 6, and still maintained a high level on Day 8. The serum levels of sIL 2R, TGF β and IL 10 were lower in all groups than that of the control. The decrement was sustained for the longest duration in the SA group. The NK and LAK activity, CD + 4 and CD + 4/CD + 8 ratio were increased in the IP and IV groups, but, as compared to the control, there was no statistical significance. The above mentioned parameters were significantly increased in the SA group. Conclusions: In comparison to the other 2 approaches, the administration of IL 2 via the splenic artery is more effective in maintaining a high and durable concentration in the blood, liver and spleen.
