Abstract:We investigated the effect of tension strain on proliferation and reconstruction of vascular smooth muscle cells (VSMCs). We designed a device in order to load cyclically a silicon membrane and the deformation of the membrane was transferred to the VSMCs cultured on the membrane. VSMCs therefore were exposed to a cyclic load by two dimension tension strain. Then microscopic observation, computer image processing, 3H TdR incorporation were applied to investigate the morphology and proliferation of VSMCs. Result: first, the spread area of VSMCs raised during 30′~6h stretch, and reduced during loading of 6~24 hrs. Second, in non confluent case, the arrangement of cells changed with stretch and proliferation. Third, in confluent case, the cells were observed as oriental arrangement and the relaxation and constriction of cells were observed after loading of ten hours. Forth, the incorporation rate of 3H TdR of stretch group was notably higher than control group after loading of twenty four hours. Conclusion: the mainly response of cells to strain is proliferation in non confluent case in contrast with reconstruction in confluent case.
