Identification of T Cell Receptor Expression in Rheumatoid Arthritis by Genescan

Abstract:Objectives To explore the role of T cells in the pathogenesis of rheumatoid arthritis in local cases(China) and provide some database to potential new therapy. Methods TCR Vβ gene expression and T cell clonality from synovial tissue and peripheral blood of rheumatoid arthritis (RA), osteoarthritis and accidently amputed patients were analyzed. The complementary determining region 3 (CDR3) of 25 TCR Vβ subfamily genes in unselected T cell populations was amplified semi-quantitatively by RT-PCR, the products were further studied by genescan for frequency of Vβ usage and identification of T cell clonality. Results The usage of most TCR Vβ subfamilies was found in all these patients but the expressions were skewed. Vβ 6, Vβ 17 and Vβ 22 were predominant subfamilies in RA synovial tissue. It was noteworthy that the expression of Vβ 17 in RA synovium was significantly increased. Most of the Vβ subfamily T cells (including predominant subfamily T cells) were found to be polyclonal. Conclusions Our results indicate that the usage of Vβ subfamilies expressed by RA peripheral blood and synovial T cells is not significantly restricted but biased. The responses of synovial T cells are polyclonal in RA patients with long duration and greater severity. Our data are consistent with the hypothesis that antigen or superantigen-driven processes may be involved in the pathogenesis of rheumatoid arthritis.
