Abstract:Objective To evaluate the effect on the osteoge nic phenotype of fibroblasts by usin g rhBMP-2and bFGF togeth-er.Method Add rhBMP-2and bFGF with different c oncentration into the cultured NIH3T3cells.Observe the morphologic ch ange of the cells and the variety of ALP activ ity and the expression of osteocalcin.The staining of ALP in the cells and the proliferation cycle were also under analysis.Result Under the effect of bFGF and rhBMP-2,fibroblast became lager with more pr ocesses and granules.The ALP activity and the expression of osteocalcin increased.The ALP granul es in the cells stained thicker and th e cells' proliferation cycle shortened.Conclusion Using bFGF and rhBMP-2together coul d augment the expression of osteogen ic phenotype and promote prolifer-ation of fibroblasts.
