Abstract:For the purpose of producing pyruvate from dl-lactate by enzymatic methods, four microorganism strains that produce lactate oxidase (LOD) were screened and isolated from many soil samples. Among them, strain SM-6, which showed high potential for pyruvate production, was chosen for further research. Physiological studies and 16S rDNA relationship reveal that SM-6 belongs to Pseudomonas putida. The optimized pH and temperature of the enzyme-catalyzed reaction were pH 7.2, and 39°C, respectively. Low-concentration EDTA (1mM) could improve the stability of pyruvate and conversion ratio of lactate oxidase. Vmax and Km value for dl-lactate were 2.46μmol/(minmg) protein and 9.53mM, respectively. On preparation scale, cell-free extract from SM-6, containing 300mg/l of crude enzyme (4037U/ml lactate oxidase), could convert 66% of 116mM of dl-lactate into 76.6mM pyruvate in 18h, and 82% of substrate was transformed after 48h, giving 95.0mM (10.5mg/ml) of pyruvate. The ratio of product to biocatalyst was 34.8:1 (g/g).

Abstract:A bacterial strain of Acinetobacter sp., which was capable of enzymatic production of pyruvate from lactate, was cultured in a 5-l reactor with a basal salt medium. After 14 h of fed-batch fermentation, 9.56 g l(-1) cell concentration in the broth was obtained with 20 g l(-1) (178 mM) sodium lactate and 4 g l(-1) NH(4)Cl in the medium; and the biotransformation ability was 2.51 units ml(-1). The cells were harvested from one reactor and then used for pyruvate production from lactate in the same reactor. l-lactate at a concentration about 527 mM was almost stoichiometrically converted to pyruvate in 28 h. After a total 42 h of cell culture and biotransformation, the transformative yield was about 0.72 g g(-1) pyruvate from lactate and the rate of pyruvate production was calculated as 1.33 g l(-1) h(-1) during the process. The results suggested this simple enzymatic production of pyruvate from lactate should be a promising process and may bring a yield higher than that by microbial fermentation. By this process, the recovery of pyruvate from such a simple reaction liquid is relatively easy and inexpensive to perform.

Abstract:Pseudomonas stutzeri SDM oxidized DL-lactic acid (25.5 g l(-1)) into pyruvic acid (22.6 g l(-1)) over 24 h. Both NAD(+)-independent D-lactate dehydrogenase and NAD(+)-independent L-lactate dehydrogenase were found for the first time in the bioconversion of lactate to pyruvate based on the enzyme activity assay and proteomic analysis.

Abstract:Background: The platform chemical lactate is currently produced mainly through the fermentation of sugars presented in biomass. Besides the synthesis of biodegradable polylactate, lactate is also viewed as a feedstock for the green chemistry of the future. Pyruvate, another important platform chemical, can be produced from lactate through biocatalysis.Methodology/Principal Findings: It was established that whole cells of Pseudomonas stutzeri SDM catalyze lactate oxidation with lactate-induced NAD-independent lactate dehydrogenases (iLDHs) through the inherent electron transfer chain. Unlike the lactate oxidation processes observed in previous reports, the mechanism underlying lactate oxidation described in the present study excluded the costliness of the cofactor regeneration step and production of the byproduct hydrogen peroxide.Conclusions/Significance: Biocatalysis conditions were optimized by using the cheap DL-lactate as the substrate and whole cells of the lactate-utilizing P. stutzeri SDM as catalyst. Under optimal conditions, the biocatalytic process produced pyruvate at a high concentration (48.4 g l(-1)) and a high yield (98%). The bioconversion system provides a promising alternative for the green production of pyruvate.

Abstract:d-Lactic acid and pyruvic acid are two important building block intermediates. Production of d-lactic acid and pyruvic acid from racemic lactic acid by biotransformation is economically interesting. Biocatalyst prepared from 9g dry cell wtl−1 of Pseudomonas stutzeri SDM could catalyze 45.00gl−1 dl-lactic acid into 25.23gl−1 d-lactic acid and 19.70gl−1 pyruvic acid in 10h. Using a simple ion exchange process, d-lactic acid and pyruvic acid were effectively separated from the biotransformation system. Co-production of d-lactic acid and pyruvic acid by enantioselective oxidation of racemic lactic acid is technically feasible.

Abstract:As an important method for building blocks synthesis, whole cell biocatalysis is hindered by some shortcomings such as unpredictability of reactions, utilization of opportunistic pathogen and side reactions. Due to its biological and extensively studied genetic background, Pseudomonas putida KT2440 is viewed as a promising host for construction of efficient biocatalysts. After analysis and reconstruction of the lactate utilization system in the P. putida strain, a novel biocatalyst that only exhibited NAD-independent d-lactate dehydrogenase activity was prepared and used in l-2-hydroxy-carboxylates production. Since the side reaction catalyzed by the NAD-independent l-lactate dehydrogenase was eliminated in whole cells of recombinant P. putida KT2440, two important l-2-hydroxy-carboxylates (l-lactate and l-2-hydroxybutyrate) were produced in high yield and high optical purity by kinetic resolution of racemic 2-hydroxy carboxylic acids. The results highlight the promise in biocatalysis by the biotechnologically important organism P. putida KT2440 through genomic analysis and recombination.

Abstract:Three strains that can produce lactate oxidase were screened and isolated from many soil samples. Strain SM-10#, which showed the highest activity among the three strains, was chosen as the enzyme source for further research and preliminarily identified as Staphylococcus saprophyticus . Cell disruption, growth curve, stability of crude enzyme and the rate of transforming lactate into pyruvate were studied, respectively. The best transforming rate could reach 80% in 4 h. Abstract:Background: Phenyllactic acid (PLA), a novel antimicrobial compound with broad and effective antimicrobial activity against both bacteria and fungi, can be produced by many microorganisms, especially lactic acid bacteria. However, the concentration and productivity of PLA have been low in previous studies. The enzymes responsible for conversion of phenylpyruvic acid (PPA) into PLA are equivocal.Methodology/Principal Findings: A novel thermophilic strain, Bacillus coagulans SDM, was isolated for production of PLA. When the solubility and dissolution rate of PPA were enhanced at a high temperature, whole cells of B. coagulans SDM could effectively convert PPA into PLA at a high concentration (37.3 g l(-1)) and high productivity (2.3 g l(-1) h(-1)) under optimal conditions. Enzyme activity staining and kinetic studies identified NAD-dependent lactate dehydrogenases as the key enzymes that reduced PPA to PLA.Conclusions/Significance: Taking advantage of the thermophilic character of B. coagulans SDM, a high yield and productivity of PLA were obtained. The enzymes involved in PLA production were identified and characterized, which makes possible the rational design and construction of microorganisms suitable for PLA production with metabolic engineering.

Abstract:Production of pyruvate from lactate through biocatalysis is a valuable process for its simple composition of reaction system and convenience of recovery. Biocatalyst with lactate-induced NAD-independent lactate dehydrogenases (iLDHs) can effectively catalyze lactate into pyruvate. To reduce the cost of biocatalyst preparation caused by indispensable lactate addition, the mutants with constitutive iLDH of Pseudomonas sp. XP-M2 were screened. Mutant XP-LM exhibited high iLDHs activities in minimal salt medium with cheap substrate glucose as the carbon source. The biocatalyst (8.2gdry cellweightl−1) containing 169.8Ul−1 l-iLDH was prepared with 20g1−1 glucose. The cost-effective biocatalyst prepared from the mutant XP-LM could efficiently catalyze lactate into pyruvate with high yield (0.961molmol−1). Based on the different thermostability of d-iLDH and l-iLDH in the biocatalyst, whole cells of the strain might also have the potential in production of pyruvate and d-lactate from racemic lactate.

Abstract:On an industrial scale, the production of pyruvate at a high concentration from the cheaper lactate substrate is a valuable process. To produce pyruvate from lactate by whole cells, various lactate-utilizing microorganisms were isolated from soil samples. Among them, strain WLIS, identified as Acinetobacter sp., was screened as a pyruvate producer. For the pyruvate preparation from lactate, the preparative conditions were optimized with whole cells of the strain. The cells cultivated in the medium containing 100 mM of l-lactate showed the highest biotransformation efficiency from lactate to pyruvate. The optimized dry-cell concentration, pH, and temperature of reaction were 6 g/L, pH 7.0-7.5, and 30 degrees C, respectively. The influences of ethylenediaminetetraacetic acid (EDTA) and aeration on a biotransformation reaction were carried out under the test conditions. Under the optimized reaction conditions, l-lactate at concentrations of 200 and 500 mM were almost totally stoichiometrically converted into pyruvate in 8 and 12 h, respectively. About 60% of 800 mM of l-lactate was transformed into pyruvate in 24 h. This reduced conversion rate is probably due to the high substrate inhibition in biotransformation.

Abstract:Pseudomonas sp. F12 isolated from soil could transform DL-2-amino-Δ(2)-thiazoline-4-carboxylic acid (DL-ATC) to L-cysteine. It could grow in minimal medium containing DL-ATC as the sole carbon and nitrogen source, and the apparent activity of L-cysteine synthesis (CS) achieved 122 U/mL in a 5-L bioreactor. Pseudomonas sp. F12 could utilize glucose as carbon source and ammonia as nitrogen source for growth, but no CS activity was formed. To reduce the cost of DL-ATC, the cultivation process was divided into a growth stage on glucose and ammonia and a production stage induced by DL-ATC. The excessive glucose led to the production of byproduct(s) which seriously inhibited cell growth and CS production. Ammonium was accumulated when DL-ATC was consumed, and ammonium did not inhibit CS activity formation until 60 mM. Based on the above features, fed-batch cultivation of the growth stage was developed by supplying glucose restrictively. The volumetric CS activity was enhanced more than two times that obtained under the initial conditions.

Abstract:Efficient enzymatic synthetic pathways for biomanufacturing chemicals often require addressing bottlenecks by enhancing the catalytic efficiency or thermostability of unsatisfied enzymes. Laminaribiose, a valuable commodity disaccharide, can be biosynthesized from starch via an in vitro four-enzyme cascade previously described, but a thermolabile laminaribiose phosphorylase from Paenibacillus sp. YM1 (PsLBP) is a potential bottleneck. Here, through several computationally guided approaches, the thermostability of PsLBP was successfully enhanced, with the best mutant, M8, exhibiting a 15.3 °C higher T m value, a 71.1-fold longer half-life, and a 2.4-fold increased specific activity at 60 °C than those of the wild-type enzyme. Structure analysis and molecular dynamics simulations revealed that the formation of new disulfide bonds and salt bridges and the changes to hydrogen bonds optimized the overall structure and improved the thermostability of M8. Subsequently, by employing an industrial procedure of heat-permeabilized whole cells, the in vitro biosystem incorporating PsLBP-M8 in a volume of 30 L converted 100.0 g/L starch to ∼77.0 g/L laminaribiose with a molar yield of ∼75% at 60 °C. These findings highlight the utility of hybrid computationally guided strategies for engineering large proteins such as laminaribiose phosphorylase and demonstrate the power of adapting incompatible enzymes in in vitro biosystems to enhance industrial performance.

Pyruvate can be produced by both chemical and biological systems. Novel biotechnological systems that can yield pyruvate have been the focus of process development in pyruvate production. In this review, we summarize recent developments related to pyruvate production by biotechnological systems, with emphasis on the enzymatic synthesis of pyruvate from the cheaper substrate lactate.

Abstract:The modification of intracellular metabolic pathways by metabolic engineering has generated many engineered strains with relatively high yields of various target products in the past few decades. However, the unpredictable accumulation of toxic products, the cell membrane barrier, and competition between the carbon flux of cell growth and product synthesis have severely retarded progress toward the industrial-scale production of many essential chemicals. On the basis of an in-depth understanding of intracellular metabolic pathways, scientists intend to explore more sustainable methods and construct a cell-free biosynthesis system in vitro. In this review, the synthesis and application of pyruvate as a platform compound is used as an example to introduce cell-free biosynthesis systems. We systematically summarize a proposed methodology workflow of cell-free biosynthesis systems, including pathway design, enzyme mining, enzyme modification, multienzyme assembly, and pathway optimization. Some new methods, such as machine learning, are also mentioned in this review.

Abstract:L‐Pipecolic Acid (L‐PA) is a valuable building block for the synthesis of pharmaceuticals such as anesthetics and immunosuppressants. Thus, more efficient and greener strategies are desired for its production. Herein, we have applied a previously engineered variant of the Lysine Cyclodeaminase from Streptomyces pristinaespiralis (e‐SpLCD) for the bioconversion of L‐Lysine into L‐PA. The reaction can be performed by the free e‐SpLCD reaching full conversion to 50 mM L‐PA. From a biotechnological perspective, the process scale‐up has been trialed in a SpinChem® reactor, albeit with lower conversion yields. To further enhance the biocatalyst stability, we present a detailed study of the e‐SpLCD immobilization on microparticles. This enabled the integration of the immobilized biocatalyst into a packed‐bed reactor for the continuous flow synthesis of L‐PA. The full conversion was achieved in 90 min, maintaining also high operational stability. Remarkably, the addition of exogenous cofactor was not needed for the flow reaction, although the long‐term operational stability was improved by the addition of NAD+.

Abstract:As a valuable versatile building block, L-phenyllactic acid (L-PLA) has numerous applications in the fields of agriculture, pharmaceuticals, and biodegradable plastics. However, both normally chemically synthesized and naturally occurring PLA are racemic, and the production titer of L-PLA is not satisfactory. To improve L-PLA production and reduce the high cost of NADH, an in vitro coenzyme regeneration system of NADH was achieved using the glucose dehydrogenase variant Ls GDH D255C and introduced into the L-PLA production process. Here an NADH-dependent L-lactate dehydrogenase-encoding variant gene (L- Lcldh 1 Q88A/I229A ) was expressed in Pichia pastoris GS115. The specific activity of L- Lc LDH1 Q88A/I229A (Pp) was as high as 447.6 U/mg at the optimum temperature and pH of 40°C and 5.0, which was 38.26-fold higher than that of wild-type L- Lc LDH1 (Pp). The catalytic efficiency ( k cat / K m ) of L- Lc LDH1 Q88A/I229A (Pp) was 94.3 mM −1 s −1 , which was 67.4- and 25.5-fold higher than that of L- Lc LDH1(Pp) and L- Lc LDH1 Q88A/I229A (Ec) expressed in Escherichia coli , respectively . Optimum reactions of L-PLA production by dual-enzyme catalysis were at 40°C and pH 5.0 with 10.0 U/ml L- Lc LDH1 Q88A/I229A (Pp) and 4.0 U/ml Ls GDH D255C . Using 0.1 mM NAD + , 400 mM (65.66 g/L) phenylpyruvic acid was completely hydrolyzed by fed-batch process within 6 h, affording L-PLA with 90.0% yield and over 99.9% ee p . This work would be a promising technical strategy for the preparation of L-PLA at an industrial scale.

Abstract:Phenyllactic acid (PLA), a novel biological antiseptic agent with broad and effective antimicrobial activity, finds wide applications in the food processing field. In this work, a new approach for biocatalytic synthesis of PLA from l-phenylalanine (Phe) was constructed. First, selection of l-lactate dehydrogenase (LDH) was done to achieve the efficient synthesis of PLA from phenylpyruvic acid (PPA); Second, l-amino acid deaminase (l-AAD) and LDH were co-expressed to achieve the synthesis of PLA from Phe; Finally, formate dehydrogenase (FDH) was overexpressed to accelerate the regeneration of cofactor NADH from NAD, which is necessary for the biocatalysis of LDH. By this biocatalytic system, 30 gL(-1) Phe was transformed into PLA with 100% conversion ratio in 12 hr. This work developed a new and efficient biocatalytic approach for PLA production with Phe as a cheap substrate, which provides an alternative for PLA production in a green manner. PLA is a novel antimicrobial compound with broad inhibition activity and has less side-effect on the health of human being. It is an ideal antiseptic that has received much attention in the food industry, for the control of microbial contaminants. In addition, PLA can be used as feed additives to replace antibiotics in livestock feeds. The newly established biosynthetic system in this work may be a promising technique for the industrial production of PLA due to high production and high efficiency.
