Abstract:A simple and new synthetic approach to isoflavones 1, 2 and 3 was described. This method was started from 1,3-dimethoxybenzene or 1,3,5-trimethoxybenzene via a synthetic sequence of Hoesch reaction or Friedel-Crafts acylation, demethylation and cyclization to obtain common key intermediate methoxyisoflavones. Selective hydrolysis of the methoxyisoflavones gave isoflavones 1, 2 and 3, respectively with one or more hydroxyl groups.

Traditionally there are two main methods available to synthesize flavones : the chalcone route and Baker-Venkataraman method. Recently many new methods have been discovered. Here, the modified Baker-Venkataraman methods, the oxidative cyclization of 2'-hydroxychalcones, oxidation of flavanones and other new methods for constructing flavones are introduced.

Abstract:An efficient synthesis of novel phenoxyimine ligands containing a difluorovinylbiphenyl group was based on the Suzuki coupling coupling of 4-bromo-2,6-difluoroaniline with 4-styrylboronic acid followed by a condensation reaction with various arylaldehydes. The structures were established by IR, 1H NMR, 13C NMR spectra and HRMS. The advantages of this synthetic route were its simple operation, mild reaction conditions and good yields.

Abstract:Farrerol (Ⅴ) has been isolated from the leaves of Rhododendron dauricurn L. (Man-Shan-Hong) of Heilongjiang province. Its structure is ascertained s 6, 8-dimethyl-5,7,4′-trihydroxyflavanone, which is a phlegm-reducing principle.We report here two routes of syntheses for farrerol (Ⅴ). The first is condensation of compound Ⅰ with compound Ⅳ to give compound Ⅵ, followed by a Fries reaction and cyclization. The second route makes use of the condensation of compound (ⅩⅣ) with p-hydroxybenzaldehyde (Ⅻ) in the presence of boric acid using ethylene glycol as a diluent.Synthetic (±)-farrerol was found to be identical with the natural specimen by IR and NMR spectra. They were also comparable pharmacologically, toxicologically and clinically.

Abstract:Two novel flavones (±)-Anastatins A and B, each containing a benzofuran moiety, were synthesized for the first time by using bromination, Suzuki coupling reaction, and an oxidation/Oxa-Michael reaction cascade as the key steps. The syntheses of (±)-Anastatins A and B were achieved in eight steps from the commercially available phloroglucinol with 9% and 10% overall yield, respectively.

Abstract:Starting from readily available vanillin and α-D-acetobromo glucose, two natural acylated phenolic glycosides vitexnegheteroin A and ovatoside D were synthesized for the first time in 4 steps with overall yields of 54% and 65%, respectively. The key steps involve the directly regioselective O-6 acylation of vanillin β-D-glucopyranoside with acyl chlorides, and simultaneous removal of the allyl protecting groups on the phenolic acid moiety and reduction of the aldehyde in the aglycon moiety by using Pd(PPh)3-NaBH4 system in one pot.

Abstract:Since flavonoids exhibit a remarkable number of pharmacological properties like antioxidant, anti-inflammatory, and antimicrobial activities, a great deal of work has been done on the medicinal and therapeutical application of these compounds. Despite the central role of flavan-3,4-diols in the synthesis of many other classes of flavonoids, only a few methods for the preparation of these compounds have been reported. In this paper, the results on the preparation of methoxy-substituted flavan-3,4-diols, displaying natural substitution patterns, from readily available starting materials through a 3-step process is disclosed. It is also indicated that the 4-aryl-substituted analogues can become available from the corresponding flav-3-enes if an aromatic nucleophile is added during the dimethyldioxirane (DMDO) epoxidation process. Characterization and purification of extracellular proteases of Trichomonas vaginalis.

Abstract:Extracellular protease activity was detected in serum-free culture filtrates of Trichomonas vaginalis. The activity was demonstrated by hydrolysis of hide powder azure and possessed the characteristics of cysteine type proteases: inhibition by N-ethyl maleimide, Cu2+, antipain, N-tosyl-L-phenylalanine chloromethyl ketone, N-tosyl-L-lysine chloromethyl ketone, leupeptin, chymostatin, and iodoacetamide, and enhancement by cysteine, EDTA, and dithiothreitol. The activity was optimal at acid pH and the protease was also active on peptide nitroanilides with arginine derivatives. Purification of this activity by ethanol precipitation, ammonium sulfate fractionation, ion exchange chromatography, and gel filtration resulted in the isolation of two proteases estimated by sodium dodecyl sulfate - polyacrylamide gel electrophoresis to have molecular masses of 60 and 30 kilodaltons (kDa), respectively. The larger molecular mass protease broke down during purifications to two subunits of approximately 23 and 43 kDa, as determined by gel electrophoresis. Rabbit sera derived by immunization with the 23-kDa subunit cross-reacted by immunoblot with the 60- and 43-kDa subunits, but not with the 30-kDa protease. These soluble products of T. vaginalis growth could be important pathogenically in establishing T. vaginalis infection in the normally acid (pH less than or equal to 4.5) environment of the vagina.

Abstract:A series of new flavanone derivatives of farrerol was designed and synthesized as a potent inhibitor of vascular smooth muscle cells(VSMCs) vegetation according to a convenient method. The structures of all the synthesized compounds were confirmed by H-1 NMR, C-13 NMR and EIHR-MS. The biological activities of these compounds against VSMCs in vitro were evaluated. The assay results indicate that two compounds, 5,7-dihydroxy-6,8-dimethyl-2-(2-nitrophenyl)chroman-4-one(7f) and 2,3-dibromo-4,5-dihydroxydiphenylmethanone(7j) exhibited high activity against VSMCs in vitro with IC50 values of 9.9 and 6.7 mu mol/L, respectively, and the preliminary structure-activity relationship(SAR) was described.

Abstract:We succeeded in a first total synthesis of flavocommelin (1), a component of the blue supramolecular pigment, commelinin (2), from Commelina communis, by direct 6-C-glycosylation of the flavan 4 using perbenzylglucosyl fluoride 8 in the presence of MS 5 angstroms in CH2Cl2 and a catalytic amount of BF3 x Et2O. After 6-C-glycosylation of 4, oxidation with CAN to flavanone 18 and subsequent 4'-O-glycosylation, promoted with a combination of BF3 x Et2O and DTBMP, afforded diglucosylflavanone 20. DDQ oxidation of 20 and deprotection successively gave 1.
Study on the Total Synthesis of Resveratrol Dimers Quadrangularin A and Pallidol

Abstract:This paper has focused on developing a novel methodology for the total synthesis of quadrangularin A (3) and pallidol (4). These two resveratrol dimers have been obtained from the inexpensive materials of 3,5-dimethoxybenzaldehyde (5) and 3,5-dimethoxybenzaldehyde (6) within 9 steps and 11 steps with total yields of 2.6% and 1.3%, respectively. The stilbene skeleton constructed by two symmetric or asymmetric aromatic rings (A and B) has been synthesized simply through Knoevenagel reaction under mild conditions with yield of more than 98%, which expanded the diversity of stilbene skeleton (7), and addressed the problems of limited expansion of asymmetric structure, low yield and complex operation. In addition, the key intermediate of 4-enaldehy de skeleton structure (9) could be used to construct aromatic rings of C and D efficiently, which has thrown light on the expansion of resveratrol dimers with aromatic ring A, B, C and D in diversity structure. All the intermediates and natural products have been characterized by (HNMR)-H-1, C-13 NMR, and HRMS.

Abstract:The efficient total syntheses of Nigrasin I and Kuwanon C, two biologically interesting natural flavonoids with two regioisomeric isoprenyl side chains, were realized for the first time starting from commercially available 1-(2,4,6-trihydroxyphenyl)ethanone via a linear reaction sequence of 11 steps with the overall yields of 22% and 21%, respectively, in which the selection of protective groups and the Claisen rearrangement of the allylated 16 featured the synthetic strategy. The use of acetic anhydride as the solvent and the employment of microwave irradiation are disclosed to be critically important in the efficient and selective Claisen rearrangement.
Effects of temperature on cycle length dependent changes and restitution of action potential duration in guinea pig ventricular muscle.

Abstract:OBJECTIVE The aim was to investigate the effects of temperature on cycle length dependent changes of action potential duration and on restitution of action potential duration. METHODS Guinea pig papillary muscle action potentials were recorded using conventional microelectrode techniques. Action potential duration was measured at cycle lengths ranging from 500 to 2000 ms at both 27 degrees C and 37 degrees C. Restitution of action potential duration was determined by introducing an extra stimulus at progressively longer diastolic intervals from 40 to 9000 ms at pacing cycle lengths of 500, 1000, and 2000 ms. RESULTS At 37 degrees C, action potential duration measured at 90% of repolarisation (APD90) during continuous pacing and the maximum value of APD90 achieved during restitution (APD90pl) decreased by 18(SEM 6) ms (n = 7) and 24(7) ms (n = 6), respectively, when pacing cycle length was reduced from 2000 to 500 ms. At 27 degrees C, the magnitude of the shortening of APD90 and APD90pl observed when pacing cycle length was similarly reduced was greater than at 37 degrees C, ie, 143(21) ms (n = 6) and 115(11) ms (n = 6), respectively. Thus the relation for restitution of action potential duration shifted downwards with reduction in pacing cycle length, and the magnitude of this shift was greater at 27 degrees C than at 37 degrees C. The difference between APD90 at the shortest diastolic interval (40 ms) and at diastolic interval of 100 ms (range of premature action potential durations) was much greater at 27 degrees C than at 37 degrees C at all three pacing cycle lengths. CONCLUSIONS Reduction in temperature magnifies the cycle length dependent changes in action potential duration both during abrupt changes in cycle length, as with an extra stimulus, and during changes of steady state cycle length. This may indicate a greater dispersion of premature action potential durations during hypothermia, and hence predispose to hypothermia induced arrhythmias.

Abstract:Flavanols are privileged heterocyclic compounds in medicinal chemistry. It is notable to develop an efficient and straightforward protocol for accessing chiral flavanols with precise control of the stereocenters. Here, a highly efficient kinetic resolution of chromenes was reported via Cu-catalyzed asymmetric hydroboration. This previously unidentified approach features a one-step synthesis of chiral flavan-3-ols containing two vicinal stereogenic centers via a highly efficient kinetic resolution ( s factor up to 1060, >99% ee for most products). In addition, the anti-inflammation effects of these diversified flavan-3-ols were studied by the in vitro experiments and RNA sequencing analysis. These flavan-3-ols showed inhibitory effects on the secretion of pro-inflammation cytokines including interleukin-1β (IL-1β), IL-6, and tumor necrosis factor–α (TNF-α), as well as inhibiting the inflammation responses through down-regulating the gene transcriptions closely related to PI3K-Akt signaling pathway and TNF signaling pathway. The results suggested that these newly synthesized flavan-3-ols have the potential to be lead compounds for anti-inflammatory drugs.

The developed chemistry was applied to the synthesis of the naturally occurring and biologically active flavonoids wogonin (2), oroxylin A (3), and their glycosylated derivatives (4 and 5) as potential carnitine palmitoyltransferase 1 activators.

Abstract:The first enantioselective total syntheses of (-)-kuwanon X, (+)-kuwanon Y, and (+)-kuwanol A have been accomplished by using asymmetric Diels-Alder cyclo-addition promoted by chiral VANOL or VAPOL/boron Lewis acid. The biosynthesis-inspired asymmetric Diels-Alder cyclo-addition shows high exo selectivity (exo/endo = 13/1), which was unprecedented in the previous total syntheses of related prenylflavonoid Diels-Alder natural products. An acid catalyzed intramolecular ketalization process enabled a biomimetic transformation to construct the polycyclic skeleton of kuwanol A efficiently.

Abstract:Azaflavanones are synthetic aza analogues of natural flavanones that have received attention in medicine. More recently, some alpha-ylidene azaflavanones have emerged as promising bioactive compounds. In the last decades, several approaches to synthesizing azaflavanones and alpha-ylidene azaflavanones have been described. These include intramolecular cyclizations of 2'-aminochalcones and analogues, coupling of 2-aminoacetophenones with aromatic aldehydes, metal-catalyzed coupling reactions of ortho -iodoanilines and the condensations of N -protected 2-aminoacetophenones and N -protected 2'-aminochalcones with aromatic aldehydes. This review discusses the different approaches in the racemic synthesis of azaflavanones and alpha-ylidene azaflavanones.

Abstract:A series of novel alkyl substituted fructose-based oxadiazoles were synthesized and their cytotoxic activities toward tumor cells were investigated. We studied the reaction mechanism and the stereochemistry of the reaction. Tautomerization between isomers 2 and 3 was observed in solution. The tautomerization was accelerated by heating or in the presence of acetic acid. An intermediate 6 during the heterocyclization was isolated, and two different pathways for the heterocyclization were suggested. On the basis of these findings, an efficient method, with the assistance of microwave irradiation was developed for the synthesis of 4 and 5. The yields were satisfactory and no by-products were found. We also proposed that the (R/S)-configurations of oxadiazoles were determined by the E/Z configurations of hydrazones.
