Abstract:A new limonoid named rohituka-15 1, with a known limonoid dregcana-1 2, was isolated from the seed of Aphanamixis polystachya (Meliaccac). The C-13 NMR data assignment of dregoana-1 2 and the Structural elucidation of the new compound 1 were based on spectral analysis including H-1-H-1 COSY, HMQC and HMBC experiments.

Abstract:Five compounds, huyouyisu (1), vomifoliol (2), isoferulic acid (3), 1,2,3-trihydroxy-phenol (4) and p-hydroxy-phenol (5), were isolated from the peels of Citrus changshan-huyou Y.B.Chang for the first time by utilizing repeated column chromatography on silica gel. Among them, huyouyisu (1) is a new compound. The structure was elucidated by using 1D and 2D spectra.

Abstract:A new ring-intact limonoid, ciliatonoid D (1), five new 7,8-seco limonoids, ciliatonoids E–I (2–6), three new 7,8-seco-29-nor limonoids, ciliatonoids J–L (7–9), and a new 7,8:9,11-diseco limonoid, ciliatonoid M (10), together with eleven known limonoids were isolated from the twigs of Toona ciliata. Their structures were established by comprehensive analyses of spectroscopic, electronic circular dichroism, single-crystal X-ray diffraction data. Compound 6 bears an uncommon 1,30-oxygen bridge. Compound 10 represents a rare example of a structure with 9,11-seco limonoid from Meliaceae. The orientation of the 14,15-epoxy moiety in a known limonoid, toonayunnanae C (12), was revised as in a β-orientation by quantum NMR calculations with ML-J-DP4 probability analysis. Compound 9 and a known analogue, toonanoronoid C (11), were found to show modest in vitro cytotoxicity against HL-60 (human premyelocytic leukemia) and P-388 (murine leukemia) cell lines.

Abstract:Chemical investigation of the EtOAc extract of an insect-associated fungus Letendraea sp. 5XNZ4-2 cultured in Potato Dextrose Broth (1/2 PDB) medium lead to the isolation of three new polyketides, named letendronol D (1), phomopsiketones H-I (2-3). The structures of new compounds were elucidated by the analysis of HRESIMS and NMR spectroscopic data, and the absolute configurations were determined by modified Mosher’s method, ECD calculation and single-crystal X-ray diffraction. Cytotoxicity and antibacterial activities of 1 were assayed and regrettably 1 didn’t display any cytotoxicity and antibacterial activity. 3 was the first phomopsiketone derivative obtaining the lactone.

Abstract:Limonoids are bioactive plant specialized metabolites found in the Meliaceae family. The basic limonoids, i.e., azadiradione, epoxyazadiradione, and gedunin have been exploited for various bioactivities and therefore are the potential drug leads for tomorrow. However, their low abundance, structural similarity, and lack of adequate mass fragmentation data have hampered their accurate identification and quantification from various sources. In the present study, basic limonoids such as azadirone, azadiradione, epoxyazadiradione, and gedunin isolated from Neem were utilized for the synthesis of their derivatives and isotopologs. A total of 30 one compounds were used in this study among which five were isolated, two were biotransformed, and 24 were synthesized. Among the synthesized compounds nine are novel compounds including six deuterated analogs/isotopologs which are (1,3-2H)-1,2-dihydro-3β-hydroxyazadiradione (9), (1,3,16-2H)-1,2-dihydro-3β-16β-dihydroxyazadiradione (10), 3β-hydroxyazadiradione (11), 3β-16β-dihydroxyazadiradione (12), (3-2H)-3β-hydroxyazadiradione (13), (3,16-2H)-3β-16β-dihydroxyazadiradione (14), (1,3,7-2H)-1,2-dihydro-3β-hydroxy-7-deacetylazadiradione (15), 1,2,20,21,22,23-hexahydroazadiradione (17), and (1,3-2H)-1,2-dihydro-3β-hydroxygedunin (29). These limonoids along with their semisynthesized derivatives were subjected to ultra high performance liquid chromatography mass spectrometry (UHPLC-MS/MS) and the fragmentation pathway was established based on structure-fragment relationships (SFRs), utilizing high resolution MS/MS data. We have developed a most reliable and easily reproducible protocol describing in depth analysis of SFRs based on the structural modifications and synthesis of isotopologs. Also, the MS/MS fragment library of these basic limonoids generated in this study acts as a fingerprint for accurate identification and quantification of limonoids by MS/MS analysis in various plant tissue extracts, phytopharmaceutical formulations and biological samples.

Abstract:Two compounds (1 and 2) have been isolated from the stem bark of Dysoxylum binectariferum (Roxb.) Hook. f. ex Bedd. Compound 1 is rohitukine [5,7-dihydroxy-2-methyl-8-[4-(3-hydroxy-1- methyl)-piperidinyl]-4H-1-benzopyran-4-one], a known compound that has anti-inflammatory and immunomodulatory activities. Compound 2 is a new alkaloid, named rohitukine N-oxide, the structure of which was elucidated on the basis of spectral evidences and comparison with the 1H NMR data of rohitukine.

Abstract:Alokicenones A-H (1-8), eight new tetrahydroanthracenes and one known okicenone (9) were identified from the secondary metabolites of mangrove-derived Streptomyces sp. HN-A101. Their structures were elucidated by HRESIMS and NMR spectroscopic data. The absolute configurations of them were determined by the calculated and experimental ECD curves. Compounds 1-2 and 9 showed moderate cytotoxicity against HCT116 and SW620 cancer cell lines with IC50 values from 0.63 to 7.73 mu M. Abstract:Aphanamgrandiol A (1), a novel triterpenoid with a bicyclo[3,2,1]octane ring skeleton produced by 2,3-ring opening and 2,6-ring closure, was isolated from the stems of Aphanamixis grandifolia. The structures were established on the basis of extensive spectroscopic methods, including 1D and 2D NMR techniques, and determined unambiguously by X-ray crystal diffraction. Aphanamgrandiol A showed moderate cytotoxicities against MCG-803, SKOV-3, HCT116 and HepG2 cell lines.

Abstract:Two new lappaconitine‐type C18‐diterpenoid alkaloids, named as leucostosines C (1) and D (2), together with six known compounds (3‐8), were isolated from the roots of Aconitum leucostomum Worosch. Their structures were elucidated by various spectroscopic analyses, including IR, HR‐ESI‐MS, NMR spectra and X‐ray experiments. Leucostosine C is the first diterpenoid alkaloid bearing the 7‐amino group. The isolated compounds were tested for the acetylcholinesterase (AChE) inhibitory effect and neuroprotective activity, none of them showed significant activities.

Abstract:Six undescribed 1,8,9-phragmalin limonoid orthoesters (chukorthoesters A-F, 1–6) and two undescribed mexicanolide-type limonoids (ivorenoids H and I, 7 and 8), together with ten known limonoids, were isolated from the seeds of common wood-source and greening plants Chukrasia tabularis A. Juss. Their chemical structures were elucidated using HR-ESI-MS, 1D and 2D NMR spectroscopic analysis, and that of 1 along with absolute configuration was confirmed by X-ray diffraction experiment with Cu Kα radiation. Bioactivities screening indicated the limonoid orthoesters 1 and 2 shown reversing resistance in MCF-7/DOX cells at 10 μM (RF:4–5 folds).

Abstract:This study reports simultaneous isolation of three new limonoids (1-3), six known regio isomers (6, 7, 9-12), and three more known limonoids (4, 5, 8) from Swietenia macrophylla (S. macrophylla) seeds. Structures of these compounds were determined via extensive study of their 1D/2D-NMR and mass spectral data. Known limonoids (4-12) were identified by comparing their physical and spectroscopic data with literature values. A novel environmentally friendly supercritical fluid chromatography (SFC) technique facilitated simultaneous and rapid separation of these compounds. The pharmacological activities of the new limonoids were investigated.

Abstract:Nine new limonoids, named thaixylomolins S–Z (1–8) and 2-O-acetylthaixylomolin Z (9), were isolated from seeds of the mangrove, Xylocarpus moluccensis, collected in the mangrove swamp of Trang Province, Thailand. Thaixylomolin S (1) is the fourth member of the khayalactone class of limonoids containing a hexahydro-2H-2,5- propanocyclopenta[b]furan motif. Thaixylomolins T–Y (2–7) are structurally diverse mexicanolides; whereas thaixylomolin Z (8) and 2-O-acetylthaixylomolin Z (9) are phragmalin 8,9,30-orthoesters. The structures of these compounds were established by HRESIMS and extensive 1D and 2D NMR investigations. The absolute configurations of thaixylomolins S (1), U (3), and Z (8) were unambiguously established by single-crystal X-ray diffraction analyses, conducted with Cu Kα radiation; whereas that of 2-O-acetylthaixylomolin Z (9) was determined to be the same as that of thaixylomolin Z (8) by the accurate fit of their experimental electronic circular dichroism spectra. Thaixylomolin S (1), featuring the presence of a 30-(2′-methyl)butyryloxy group, is the first limonoid of the khayalactone class, whose constitution and absolute configuration are unequivocally determined by X-ray crystallography. The inhibitory activities of all the compounds, except for the epimers 4, were assayed against human carboxylesterase 2. All the tested compounds exhibited inhibition rates in the range of 16–65% at the concentration of 100.0 μM.

Abstract:One new sterol, (3 β ,21 α ,22a)-stigmast-5-ene-3,21,22-triol ( 1 ), together with 36 compounds ( 2 – 37 ), were isolated from the stems of Aphanamixis grandifolia . The structure of 1 was elucidated by physical, chemical, and spectroscopic methods, including 1D and 2D NMR and HR-ESI-MS. Besides, 33 compounds ( 2 , 4 , 5 , 7 – 10 , 12 – 30 , and 32 – 37 ) were isolated from the Aphanamixis genus for the first time, and 15 was isolated from the plant for the first time.

Abstract:Toosendanone A (1), a new euphane (tirucallane)-type triterpene bearing a five-membered ring in the side chain and the first cyclopentanyl protolimonoid, was isolated from the bark of Melia toosendan, along with two new tirucallanes, toosendanic acids A (2) and B (3). The structure and absolute configuration of compound 1 was elucidated by spectroscopic data interpretation and X-ray diffraction analysis. Compounds 1-3 were evaluated for cytotoxicity against a small panel of cancer cell lines.

Abstract:A new acorane sesquiterpene glucoside, 1 R,3 S,4 R,5 R,10 R-3,10-dihydroxyacoronene-3- O-β-D-glucoside (1), was isolated from the EtOAc-soluble partition of the ethanol extract of Lysionotus paucflorus, together with six known compounds, namely p-hydroxybenzoic acid (2), vanillic acid (3), caffeic acid (4), β-ydroxypropiosyringone (5), α,β-dihydroxypropiosyringone (6), and lyoniresinol (7). The structure of the new compound was elucidated on the basis of spectroscopic methods, including D and 2D NMR, and high-resolution MS analysis. The absolute configuration of 1 was determined from CD spectra. When evaluated against several bacterial and fungal strains, and human cancer cell lines, compound 1 and its aglycone were inactive.

Abstract:A structurally novel 8,9-seco-eudesma-9,4:8,12-dilactone sesquiterpenoid, chlorajapodiolide (1), a new eudesmane sesquiterpenoid, chlorajapotriol (2), a new lindenane-type sesquiterpenoid dimer, chlorajaponilide I (3), and four known lindenane-type sesquiterpenoid dimers were isolated from the whole plant of Chloranthus japonicus. Abstract:Nine new limonoids, meliazedarines A–I (1–9), seven known analogues (10–16), and five known triterpenoids (17–21) were isolated from the fruits of Melia azedarach. Abstract:A new C20‐denudatine‐type diterpenoid alkaloid (DA) 11S‐aconicarnine D (1) and fifteen known DAs were isolated from the lateral roots of Aconitum brachypodum Diels. Their structures were identified on the basis of extensive spectroscopic analyses, NMR calculations and DP4+ analysis. The antimicrobial activities of compound 1 was evaluated, compound 1 exhibited potent antimicrobial activity against Alternaria panax and Fusarium solani with MICs of 2.00 and 16.00 μg/mL (Nystatin, 1.00 μg/mL), respectively.

Abstract:Fifteen new labdane-type diterpenoids, sublyratins A-O (1-15), along with four known analogues (16-19) were isolated from the aerial parts of Croton sublyratus. Their structural assignments were challenging due to the stereoisomeric features evident and were achieved by analyzing comprehensively the spectroscopic data and electronic circular dichroism spectra and using X-ray crystallographic analysis. Compounds 9 and 16-18 displayed cytotoxic activity against the HL-60 cell line with IC50 values of 1.5-2.8 μM.
