Abstract:An ultra-high pressure liquid chromatography-tandem mass spectrometry method has been developed and validated for identification and quantification of five major bioactive components in rat plasma after oral administration of Qihuotongqiao tablets. The analysis was performed on an Acquity, UPLC HSS T3 column (100 mm x 2.1 mm, 1.8 mu m; Waters, USA) utilizing a gradient elution profile and a mobile phase consisting of(A) water containing 0.5 mM ammonium chloride and (B) acetonitrile. Electrospray ionization (ESI) tandem interface was employed prior to mass spectrometric detection. The calibration curve was linear over the range of 4.2-416.0 ng/mL for notoginsenoside RI, 38.4-3840.0 ng/mL for ginsenoside Rg(1), 3.7-368.0 ng/mL for ginsenoside Re, 37.6-5640.0 ng/mL ginsenoside Rb-1 and 4.5-448.0ng/mL for icariin. respectively. The average accuracies ranged from 87.2 to 109.3% with RSD <= 13.7%. The results indicated that ultra-performance liquid chromatography-tandem mass spectrometry (UPLC-MS) provided improved chromatographic parameters resulting in significantly increased sample throughput including lower solvent consumption and lower limits of quantitation (LLOQ) for most of target analytes compared to previous method employing conventional high-performance liquid chromatography (HPLC) separation. Abstract:To support pharmacokinetic studies of ginsenosides, a novel method to quantitatively analyze ginsenoside Rg3 (Rg3), its prosapogenin ginsenoside Rh2 (Rh2) and aglycone 20(S)-protopanaxadiol (ppd) in rat plasma was developed and validated. The method was based on gradient separation of ginsenosides present in rat plasma using high performance liquid chromatography (HPLC), followed by detection with electrospray ionization(ESI) mass spectrometry (MS) in negative ion mode with the mobile phase additive, ammonium chloride (500 microM). Differentiation of ginsenosides was achieved through simultaneous detection of the [M(+)Cl(-)] adduct of ginsenoside Rg3 and [M(+)Cl(-)] adducts of its deglycosylated metabolites Rh2 and ppd, and other ions after solid phase extraction (SPE). The /specific ions monitored were m/z 819.50 for Rg3, m/z 657.35 for Rh2, m/z 495.40 for ppd and m/z 799.55 for the internal standard (digitoxin). The mean recoveries for Rg3, Rh2 and ppd were 77.85, 82.65 and 98.33%, respectively using 0.1 ml plasma for extraction. The lower limits of quantification were 10.0, 2.0 and 8.0 ng/ml (equivalent to 0.1, 0.02 and 0.08 ng in each 10 microl injection onto the HPLC column) for Rg3, Rh2 and ppd, respectively. The method has been demonstrated to be highly sensitive and accurate for the determination of Rg3 and its metabolites in rat plasma.

Abstract:A rapid and sensitive ultra high performance liquid chromatography with tandem mass spectrometry method was established and validated for simultaneous determination of thirteen bioactive components (gallic acid, protocatechuic acid, puerarin, p-hydroxycinnamic acid, daidzin, ononin, daidzein, naringenin, genistein, apigenin, formononetin, biochanin A, and beta-sitosterol) of Radix Puerariae extract in rat plasma and tissues. The plasma and tissues samples were pretreated by protein precipitation extraction, and umbelliferone and rutin were used as internal standards. Sample separation was performed on a ZORBAX RRHD Eclipse plus C-18 column (2.1 mm x 50 mm, 1.8 mu m, Agilent) with a mobile phase consisting of methanol-water (containing 0.1% formic acid). The mass spectrometry analysis was conducted in positive and negative ionization modes with multiple reaction monitoring. The lower limit of quantitation range for the 13 analytes was 0.2-35 ng/mL. The intra- and inter-day precision of all the analytes were less than 10.92%, with an accuracy ranging from -13.10 to 11.96%. Both the recovery and matrix effect were within acceptable limits. This method was successfully applied to pharmacokinetic and tissue distribution study of the 13 bioactive components in rats after oral administration of R. Puerariae extract.

Abstract:In this study, a rapid and highly sensitive ultra-high performance liquid chromatography-tandem mass spectrometry method was established for the quantification of paeoniflorin in rat plasma. The analyte and the internal standard (IS), hyperoside, were extracted from rat plasma via liquid-liquid extraction with ethyl acetate. LC separation was carried on a Phenomenex Luna C18 column within 2.5 min. Quantitation was performed on a triple quadrupole mass spectrometer employing electrospray ionization and operated in the multiple reaction monitoring and negative ion modes. The precursor to product ion transitions monitored for paeoniflorin and the IS were m/z 524.8→449.0 and 463.1→300.0, respectively. The assay was validated with a linear range of 0.02~25.6 μg/mL for paeoniflorin. The intra- and inter-day precisions (relative standard deviation%) were within 9.7%, and the recoveries were greater than 65.2%. Paeoniflorin was proven to be stable during all sample storage, preparation and analytical procedures. The method was successfully applied to a pharmacokinetic study of the Baixiangdan Capsule in eight female rats.

Abstract:A reversed-phase high-performance liquid chromatographic (RPHPLC) method has been developed for the determination of polydatin (PD) in rat plasma, bile, urine, feces and tissue homogenates using 2,3,5,4′-tetrahydroxychrysophenine-β-d-glucoside as an internal standard. The sample pretreatment included deproteinization for plasma samples and a liquid–liquid extraction for bile, urine, feces and tissue homogenates. Separation was obtained on a C18 reversed-phase column with the mobile phase consisting of methanol and water (35:65v/v). The flow rate was 1ml/min and the effluent was monitored at 310nm. The method showed good linearity over the concentration ranges employed for various matrices (r>0.998). The quantification limits of PD in rat plasma, bile, urine, feces and tissue homogenates were 0.0251, 0.126, 0.025μg/ml, 0.189 and 0.0378μg/g, respectively. The accuracy and precision of the method were less than 12.0% for the various matrices. No interferences from endogenous substances were found. The method was successfully applied to study the pharmacokinetics of PD in rats after intravenous administration.

Abstract:An ultra-high pressure liquid chromatography-tandem mass spectrometry (UPLC-MS/MS) method was developed for the quantification of three major ingredients in Chinese lantern preparations (CLP) in rat plasma. Following extraction by ethyl acetate, the analytes were separated on an Acquity UPLC BEH Shield RP C(18) column using a gradient mobile phase system of acetonitrile-water. Electrospray ionization (ESI) tandem interface was employed prior to mass spectrometric detection. The calibration curves were linear over the range of 5.0-500.0 ng/ml for physalin D, 2.3-230.0 ng/ml for physalin G and 0.71-71.0 ng/ml for 4,7-didehydroneophysalin B. The average extraction recoveries, examined at four concentration levels, carried from 57.1% to 76.9%, and the accuracies ranged from 94.0% to 113.3% with precision (RSD) <15%. The validated method was successfully applied to the determination of the three physalins in rat plasma after intragastric administration of CLP suspension.

Abstract:A sensitive HPLC method based on post-column modification and fluorescence detection has been developed for determination of puerarin and daidzein in rat plasma. Chromatographic separation was performed on a C8 column with a linear gradient prepared from 0.5% aqueous acetic acid and 0.5% acetic acid in acetonitrile, delivered at a flow rate of 0.8 mL min−1. Naringin was used as the internal standard. It was necessary to use acetic acid in the mobile phase to achieve good separation, but this led to fluorescence signal suppression, because puerarin and daidzein have native fluorescence at pH 8.0–9.0. To enhance the sensitivity, post-column modification with alkaline buffer was adopted. After this modification, detection sensitivity for puerarin and daizein increased more than 500-fold and 600-fold, respectively, compared with direct fluorescence detection. Signal-to-noise ratios for detection for puerarin were more than 150 times better than for UV detection after use of the same method of sample preparation. This sensitive analytical method was successfully used to determine pharmacokinetic data for puerarin and daidzein in rat plasma after oral administration of a single dose of Puerariae radix extract containing puerarin (approx. 8.4 mg) and daizein (approx. 5.9 mg) to male SD rats.

Abstract:Ginseng saponins isolated from ginseng, have been regarded as the principal constituents responsible for the biological activities. The aim of this study was to set up a liquid chromatography–mass spectrometry (LC–MS) method for simultaneously determine the concentration of Ginsenoside Rg1 and its secondary glycoside Rh1 and aglycone protopanaxatriol (PPT) in rat plasma so as to study the pharmacokinetics of Rg1 after intraveneous (i.v.) and intragastric gavage (i.g.) administration. One hundred microliters or 1.0ml of rat plasma samples from i.v. or i.g. treated rats were used respectively for analysis. After solid-phase extraction (SPE) and high performance liquid chromatography (HPLC) separation, the chloride adduct anions [M+Cl]− of Rg1, Rh1 and PPT were analyzed by LC–MS in selected ions monitoring (SIM) mode. Rg1 could be determined by this LC–MS method over the ranges of 1.56–250ng/ml and 250–20,000ng/ml with the correlation coefficients of 0.999 and 0.9998, respectively. The detection limits (LOD) of this method was 20pg (S/N>3) for Rg1, 100pg for Rh1 and 10pg for PPT. Chromatographic separation was achieved in less than 8mins. The method has been used for the pharmacokinetic study of Rg1 in rats.
A Simple HPLC Assay for Ginsenoside-Rh2 in Plasma and Its Application for Pharmacokinetic Study in Rats

Abstract:Ginsenoside-Rh2 (G-Rh2) is an important constituent in ginseng and has significant anti-tumor property.A simple HPLC assay was developed to study pharmacokinetics of G-Rh2 in rat plasma.The HPLC consisted of a C 18 reversed phase analytical column, a variable wavelength ultraviolet (UV) spectrophotometric detector, and a mobile phase made up of 0.005 M KH 2 PO 4 (pH 7.2): acetonitrile: methanol (23:7:70).The system was operated at ambient temperature isocratically at a flow rate of 0.5 mL/min and wavelength at 203 nm.Extraction of G-Rh2 from plasma was achieved by solid phase extraction (SPE) using 100 mg/mL C 18 SPE columns.The results showed that standard curves using 50 mL of plasma sample were linear from 0.25 to 100 mg/mL, with regression coefficient (r 2 )>0.99.The intra-and inter-assay variations over a 3-month study period were <10% and <20%, respectively.After 10 mg/kg twice daily for 4 doses by subcutaneous (sc) injection, the mean maximum plasma concentration (C max ), and time to C max (T max ) of G-Rh2 was 0.79 mg/mL and <0.5 hour, respectively.The described HPLC is readily performed in most laboratories and should have adequate sensitivity and specificity to study pharmacokinetics of G-Rh2 in rats following multiple doses.

Abstract:A sensitive rapid resolution liquid chromatography–tandem mass spectrometry method was developed to determine the pharmacokinetics of ginsenoside Rb1, Rb2, and Rb3 in rats, after oral administration (50mg/kg) and intravenous administration (10mg/kg) of Rb1, Rb2, and Rb3, respectively. The plasma samples were extracted by saturated N-butanol with Rg2 as internal standard. Chromatographic separation was performed on a Zorbax SB-C18 column (50mm×4.6mm, 1.8μm) with a mobile phase consisting of methanol and 1mM ammonium formate (74:26, v/v). Multiple reaction monitoring mode was performed using the fragmentation transitions of m/z 1107.7→m/z 178.9, m/z 1077.7→m/z 148.6, and m/z 1077.7→m/z 783.4 for Rb1, Rb2, and Rb3, respectively. Calibration curves were recovered over a concentration range of 20–1000ng/ml for Rb1 and Rb2, and 50–2500ng/ml for Rb3. The limits of detection were 3.0ng/ml, 4.0ng/ml, and 6.5ng/ml. Both intra-day and inter-day variances were less than 15% and the accuracy was within 86–114% for the three ginsenosides. All three ginsenosides had poor oral bioavailability (0.78%, 0.08%, and 0.52% for Rb1, Rb2, and Rb3, respectively). The value of Rb1 is higher than that of Rb2 or Rb3, indicating that ginsenosides with hexose and hydroxyl groups (Rb1) could present better pharmacokinetic behaviors than those with pentose groups in the same glycosylation site by oral administration.

Abstract:To track the pharmacokinetic features of red ginseng (RG), a rapid and sensitive ultra fast liquid chromatographic coupled with electrospray ionization triple quadrupole tandem mass spectrometry (UFLC-MS/MS) method was developed for simultaneous quantification of twenty-one ginsenosides and their three aglycones, including 18 prototype compounds (ginsenosides Rb1, Rb2, Rc, Rd, Re, Rg1, Rg5, Rh4, Rk1, Rk3, 20(S)-Rf, 20(S)-Rg2, 20(R)-Rg2, 20(S)-Rg3, 20(R)-Rg3, 20(S)-Rh1, 20(R)-Rh1, 20(S)-NG-R2), and 6 metabolites (ginsenosides 20(S)-Rh2 and Rh3, 20(S)-protopanaxadiol (PPD), 20(S)-protopanaxatriol (PPT), 20(R)-PPT, ginseng saponin compound K) of RG in rat plasma after oral administration of RG water extract at a single dose of 4g/kg body weight to rats. All analytes with internal standard (digoxin) were detected by multiple reaction monitoring in negative ionization mode and separated on an ACQUITY UPLC® BEH RP-C18 column (1.7μm, 100×2.1mm). This established method was well validated in terms of linearity, sensitivity, intra- and inter-day precisions, accuracy, recovery, matrix effect, stability, and had a lower limit of quantification at the concentration range of 0.12-8.12ng/mL for all of analytes. This UFLC-MS/MS approach was successfully applied to the pharmacokinetic study for RG water extract in rats. We firstly proposed that Rb1, Rb2, Rc, Rd, Rg1, Rg5, 20(S)-Rg3, 20(S)-Rh2, and 20(S)-PPD measured in rat plasma were suitable pharmacokinetic markers of RG extract in rats due to their high systemic exposure levels. Thus, this specific and reliable method will be useful for future applications to pharmacokinetic studies for various sources of ginsenoside samples and Panax herbs in vivo.
Gradient High-Performance Liquid Chromatography for the Simultaneous Determination of Chlorogenic Acid and Baicalin in Plasma and Its Application in the Study of Pharmacokinetics in Rats.

Abstract:A novel HPLC–UV method was developed for the simultaneous determination of two major active components in Yinhuang injection, chlorogenic acid and baicalin, in rat plasma. Extracted from the plasma samples with methanol–acetonitrile (3:1, v/v), the two compounds were successfully separated using a C18 column with a gradient elution composed of 15 and 54% methanol–acetonitrile (1:1, v/v) in 0.2% (v/v) phosphoric acid water solution (pH 2.0). The flow-rate was set at 1mlmin−1 and the eluent was detected at 327nm for chlorogenic acid, 278nm for baicalin. Puerarin and rutin were used as the internal standards for chlorogenic acid and baicalin, respectively. The method was linear over the range of 0.388–12.4μgml−1, 0.485–124μgml−1 for chlorogenic acid and baicalin, respectively. The correlation coefficient for each analyte was above 0.998. The intra-day and inter-day precisions were better than 7 and 9%, with the relative error ranging from −9.5 to 7.3% and from −4.2 to 1.8%. The limit of detection (LOD) and the limit of quantification (LOQ) for chlorogenic acid and baicalin in plasma were 0.194, 0.122, 0.388 and 0.485μgml−1, respectively. This assay has been successfully applied in the pharmacokinetic study of chlorogenic acid and baicalin in vivo through intravenous administration of Yinhuang injection to rats.
Simultaneous Determination and Pharmacokinetic Study of Water-Soluble and Lipid-Soluble Components of Danshen in Rat Plasma Using Hplc-Uv Method

Abstract:A sensitive and specific HPLC-UV method was developed for the simultaneous determination of major active components of danshen in rat plasma. Both water-soluble and lipid-soluble compounds were included, i.e. danshensu, salvianolic acid B and tanshinone IIA. Protocatechuic aldehyde and diazepam were used as internal standards. The chromatographic separation was achieved on a reversed-phase C(18) column by gradient elution using acetonitrile and 0.025% (v/v) phosphoric acid solution as mobile phase, at a flow rate of 1.0 mL/min. Salvianolic acid B, danshensu and internal standards were detected at 281 nm, while the detection of tanshinone IIA was carried out at 272 nm. All calibration curves showed good linearity (r(2) > 0.999) within test ranges. The limit of detection and the limit of quantification for danshensu, salvianolic acid B and tanshinone IIA in plasma were 0.065, 0.043, 0.022, 0.131, 0.085 and 0.044 microg/mL, respectively. This is the first report on the determination and pharmacokinetic study of danshensu, salvianolic acid B and tanshinone IIA in rat plasma and the results indicated that this method was reliable for the determination of the major active components of danshen in rat plasma.
Simultaneous Determination of Ferulic Acid, Paeoniflorin, and Albiflorin in Rat Plasma by Ultra-High Performance Liquid Chromatography with Tandem Mass Spectrometry: Application to a Pharmacokinetic Study of Danggui-Shaoyao-San

Abstract:A rapid, selective, and sensitive ultra-high performance liquid chromatography-tandem mass spectrometry method was developed for simultaneous determination of ferulic acid, paeoniflorin, and albiflorin, the major active constituents of Danggui-Shaoyao-San, in rat plasma using geniposide as the internal standard. The plasma samples were processed by protein precipitation with acetonitrile, and then separated on a Shim-Pack XR-ODS C-18 column (75 mm x 3.0 mm, 2.2 mu m) using gradient elution program with a mobile phase consisting of 0.1% aqueous formic acid and acetonitrile at a flow rate of 0.4 mL/min. The detection was achieved on a 3200 QTRAP mass spectrometer equipped with electrospray ionization source in negative ionization mode. Quantification was performed using multiple reaction monitoring mode by monitoring the fragmentation of m/z 192.9 -> 134.0 for ferulic acid, m/z 525.0 -> 120.9 for paeoniflorin, m/z 525.2 -> 121.0 for albiflorin, and m/z 433.1 -> 225.1 for the internal standard, respectively. The calibration curve was linear in the range of 5-2500 ng/mL for all the three analytes (r >= 0.9972) with the lower limit of quantitation of 5 ng/mL. The intraday and interday precisions were below 12.1% for all the analytes in terms of relative standard deviation, and the accuracy was within +/- 11.5% in terms of relative error. The extraction recovery, matrix effect and stability were satisfactory in rat plasma. The validated method was successfully applied to a pharmacokinetic study of ferulic acid, paeoniflorin, and albiflorin after oral administration of Danggui-Shaoyao-San to rats.

Abstract:A sensitive and specific reversed-phase high-performance liquid chromatography with ultraviolet detection (RP-UV-HPLC) method has been developed and validated for the identification and quantification of pinocembrin in rat plasma using chrysin as the internal standard. Following protein precipitation with acetonitrile, the analytes were separated by the mobile phase 0.01M ammonium acetate (pH 4.0)–methanol (35:65, v/v) with an Agilent TC-C18 column (5μm, 4.6mm×150mm) at a flow rate of 1ml/min, column temperature 40°C and detection wavelength 290nm. A good linear relationship was obtained in the concentration range studied (0.07–133.33μg/ml, r=0.9995). The lowest limit of quantification (LLOQ) was 66.7ng/ml and the lowest limit of detection (LLOD) was 25ng/ml. Average recoveries ranged from 93.9 to 97.8% in plasma at the concentrations of 0.33 and 33.33μg/ml. Intra- and inter-batch relative standard deviations were 0.15–2.03 and 1.18–9.96%, respectively. This method was successfully applied to the pharmacokinetic studies in rats after intravenous administration of pinocembrin.
Determination of Ginsenoside Rg2 in Rat Plasma by High‐Performance Liquid Chromatography–Mass Spectrometry after Solid‐Phase Extraction

Abstract:A sensitive liquid chromatography-mass spectrometric (LC/MS) method for the quantification of ginsenoside Rg2 (Rg2) in rat plasma was developed after solid-phase extraction (SPE). Chromatographic separation was achieved on a reversed-phase Kromasil C 18 column with the mobile phase of acetonitrile-ammonium chloride (500 mu M/L) and step gradient elution resulted in a total run time of about 9 min. The analytes were detected using electrospray negative ionization mass spectrometry in the selected ion monitoring (SIM) mode. A good linear relationship was obtained in the concentration range (5-2500 ng/mL) (r=0.9999). Limit of quantification (LOQ) was 5 ng/mL and the limit of detection (LOD) was 2 ng/mL using 100 mu L plasma sample. Average recoveries ranged from 72.43-84.73% in plasma at the concentrations of 20, 200, and 2000 ng/mL. Intra- and interday coefficients of variation for the assay were 4.93-10.87% and 4.06-7.84%, respectively. The method was successfully applied to the analysis of ginsenoside Rg2 in rat plasma. The applicability of this assay was examined in a preliminary pharmacokinetic study of ginsenoside Rg2 in rats.
[Quantitative determination of ginsenoside Re and Rg1 in human plasma by ultra performance liquid chromatography-tandem mass spectrometry].

Abstract:OBJECTIVE To develop a rapid and sensitive method for the pharmacokinetic study of ginsenoside Re and Rg1 in SHEN-FU injective powder in human plasma by Ultra Performance Liquid Chromatography-Tandem Mass Spectrometry (UPLC/ESI/MS/MS). METHODS A Waters C18 column (1.7 microm, 2.1 x 100 mm) was used in this study. The detection of Re and Rg1 was performed on a triple-quadruple mass spectrometer with multiple-reaction monitoring (MRM) mode using the following transitions: m/z 969.6-->789.3 for Re; m/z 823.5-->643.2 for Rg1 and m/z 803.5-->387.1 for digoxin. A total of 324 plasma samples from 18 healthy volunteers were tested. RESULTS A total run could be accomplished in 4 minutes. Only 50 microL plasma sample was needed to detect Re and Rg1. The lowest detectable concentration for both Re and Rg1 was 0.025 ng/mL. Good linearity appeared from 0.1 to 200 ng/mL (r2>0.999). The decline of Re and Rg1 in plasma could be described by a triple-compartment model. CONCLUSION The proposed method provides a rapid and sensitive method for the quantification of Re and Rg1 in human plasma, which has been successfully applied to the pharmacokinetic study on intravenous infusion of SHEN-FU powder.

Abstract:A platform for the pharmacokinetic study of multiple constituent traditional Chinese medicine was developed and validated. An HPLC/ESI/MS method was employed for the simultaneous determination of panax notoginsenoside R1, ginsenoside Rg1, Rd, Re and Rb1 in rat plasma. After the addition of digoxin as an internal standard (IS), rat plasmas were extracted with n-butanol saturated with pure water and all analytes were separated on a reversed-phased C-18 column with a mobile phase of acetonitrile-water (0.5 mM ammonium chloride) and pumped at a flow rate of 0.2 mL/min. Analytes were determined in a single quadrupole mass spectrometer using an electrospray ionization source. HPLC/ESI/MS was performed in the selected-ion monitoring mode with the chlorinated adducts of molecular ions [M + Cl](-) at m/z 967.75, 835.80, 981.80, 981.80, 1143.65 and 815.40 for R1, Rg1, Rd, Re, Rb1 and digoxin, respectively. The method showed excellent linearity over the concentration range 3.03-775.00 ng/mL (r(2) = 0.9994) for R1, 4.00-1025.00 ng/mL (r(2) = 0.9991, 0.9988, 0.9991) for Rg1, Rd and Re, respectively, and 2.77-710.00 ng/mL for Rb1 (r(2) = 0.9990). The low limit of quantification was 3.03, 4.00, 4.(X), 4.00 and 2.77 ng/mL for R1, Rg1, Rd, Re and Rb1, respectively, with S/N > 10. The intra- and inter-day precisions were below 12.00% and the accuracy was between -2.31 and +4.43% for all analytes. The extract recoveries of analytes were from 67.47 to 94.18%. All analytes were stable in rat plasma after storage for 12 h at ambient temperature, at 4 degrees C for 12 h in the sample pool, at -20 degrees C for 4 weeks and at -20 degrees C for three thaw-freeze cycles. The HPLC/ESI/MS technique provided an excellent method for the simultaneous quantification of R1, Rg1, Rd, Re and Rb1 in rat plasma and was successfully applied to the pharmacokinetic study of a multiple-constituent traditional Chinese medicine, total panax notoginsenoside (Xuesaitong injection). Abstract:AIM: To develop an accurate,sensitive and reliable HPLC-MS method for the determination of ginsenoside Rg1 in rat plasma,and to study the absorption characteristics of Rg1 at different intestinal sections(duodenum,ileumand,jejunum and colon) of rats.METHODS: Ginsenoside Rg1 or total ginsenoside which containing equal amount of Rg1 was injected into different intestinal sections(duodenum,ileum,jejunum and colon) of each rat.At the same time,Ginsenoside Rg1 or total ginsenoside was administrated to rats by intragastric gavage.Blood samples were collected according to preset timetable and Rg1 concentrations were determined.RESULTS: The bioavailability of Rg1 in rats was increased significantly when total ginsenoside was administrated compared with that of Rg1 given alone.The absorption profile of Rg1 was different in duodenum and ileum but similar in jejunum and colon.CONCLUSION: There might be some specific substance in total ginsenoside,which could improve the absorption of Rg1 in duodenum and ileum.

Abstract:A specific ultra-performance liquid chromatography-mass spectrometry (UPLC-MS) method was developed for the simultaneous determination of puerarin, daidzein, baicalin, wogonoside and liquiritin in rat plasma. Chromatographic separation was performed on a C(18) column packed with 1.7 microm particles by a linear gradient elution. The analytes and carbamazepine (internal standard, I.S.) were monitored in a selected-ion reaction (SIR) mode with a positive electrospray ionization (ESI) interface by the following ions: m/z 417.2 for puerarin, m/z 255.2 for daidzein, m/z 271.0 for baicalin, m/z 461.0 for wogonoside, m/z 441.0 for liquiritin and m/z 237.2 for carbamazepine (I.S.), respectively. The calibration curves of these analytes were linear over the concentration ranges from 0.00254-1.02 microg mL(-1) to 0.0102-10.2 microg mL(-1). Within-batch and between-batch precisions (RSD%) were all within 15% and accuracy (RE%) ranged from -10% to 10%. The extraction recoveries were on average 79.8% for puerarin, 90.8% for daidzein, 74.4% for baicalin, 70.2% for wogonoside and 84.7% for liquiritin. The validated method was successfully applied to investigate the pharmacokinetics of five bioactive compounds of GegenQinlian decoction (GQD) in rats.
