Determination of surface properties and cell uptake in vitro of stearic acid solid lipid nanoparticles

Abstract:OBJECTIVE: To investigate the relationship between the surface properties and the cell uptake of stearic acid solid lipid nanoparticles (SLN) modified with different surfactants. METHODS: The stearic acid solid lipid nanoparticles were prepared by the method of emulsion-evaperation-solidfied at low temperature with different surfactants. Then the contact angle and the surface tension of each kind of nanoparticle colliodal suspension were determined. The mouse peritoneal macrophages (MPM) were separated as a cell model to investigate the uptake of nanoparticles by cells in vitro. RESULTS: The diameter of each kind of nanoparticles was (162.0 ± 67.4) nm, (50.2 ± 28.9) nm, (326.8 ± 195.2) nm and (204.5 ± 91.3) nm when its surface was modified with Brij 78, Myrj 53, Myrj 59 or Pluronic F68, respectively. Compared with the (9,10)3 H-stearic acid solution, obvious increases of cpm value in MPM were observed by binding (9,10)3 H-stearic acid with SLN. The lowest cpm value in MPM was observed in SLN surface-modified by Myrj 59. A direct relationship was observed between the hydrophilicity of the surface of the nanoparticles and the uptake by cells. CONCLUSION: The surface properties of nanoparticles plays a major role in targeting to phagocytes.
