Abstract:Polyacrylic acid is a biocompatible and biodegradable polymer that has been widely investigated for use in drug delivery. It has a number of properties that make polyacrylic acid a promising material for various applications, including ability to form hydrogels, mucoadhesive properties, and stimuli-responsive behavior. Polyacrylic acid hydrogels can be used to deliver drugs to a variety of tissues and organs. They can be prepared in a variety of forms, including injectable gels, films, and tablets. These hydrogels can be loaded with a variety of drugs, including proteins, peptides, and small molecules. The mucoadhesive properties of polyacrylic acid make it a good choice for delivering drugs to the eye, nose, and skin. Polyacrylic acid can adhere to these mucosal surfaces, which can help to prolong the drug's release and improve its bioavailability. The stimuli-responsive behavior of polyacrylic acid can be used to develop drug delivery systems that release drugs in response to specific stimuli, such as changes in pH, temperature, or ionic strength. This can be used to control the release of drugs in a way that is tailored to the specific needs of the patient. The research on the use of polyacrylic acid in drug delivery is ongoing. Abstract:Poly(α-amino acid)s, containing amino acid repeat units, are remarkable biomaterials with similar structures to protein, which are mainly synthesized by solid phase peptide synthesis, ring opening polymerization of N-carboxyanhydrides, polymerization of activated urethane derivatives of α-amino acids and so on. α-Amino acid N-thiocarboxyanhydride (NTA) and N-substituted glycine N-thiocarboxyanhydride (NNTA) have recently emerged as a subject of scientific interest due to their easy synthesis and purification in open air, long shelf-life and well controlled polymerization to produce poly(α-amino acid)s. In this feature article, we review the history and the recent development on the design of NTA monomers, and summarize the recent advances in poly(α-amino acid)s synthesis through ring-opening polymerization of NTA and NNTA by using different initiators and reaction mediums. We also highlight the applications of poly(α-amino acid) materials resulting from NTA polymerization.

Abstract:Poly(lactic acid) (PLA) is the most promising biodegradable alternative to replace conventionalpetrochemical-based polymers in manufacturing high performance materials. Here we review the main methods to obtain polylactic acid and briefly discuss its functionalization and application in the field of controlled drug release. We conducted a bibliographic search of scientific databases and summarized the results of the research carried out by our group. We show that the most commonly used PLA modifications in drug delivery systems are functionalization with glycolic acid (GA) and polyethylene glycol (PEG) through copolymerization or blending, where the use of compatibilizers is essential for good adhesion. Active vectorization is discussed as its choice depends on the size of the nanoparticle and the type of disease to be treated.

Abstract:Poly-L-lysine (PLL) is a water-soluble biopolymer consist of repeating unit of L-lysine. It displays intrinsic non-antigenicity, antibacterial property, biocompatibility, and biodegradability, and facile synthesis procedure, therefore has attracted particular attention in various biomedical and pharmaceutical applications. Following a brief introduction to PLL synthesis, this review focuses on the significant advances of PLL and PLL-derived complexes utilized in cargo delivery, tissue adhesives, and antibacterial materials. Meanwhile, this review summarized the modification and optimization of PLL complexes for further clinical translational studies. The final section discusses the challenges and opportunities of PLL-based complexes in the biomedical areas.

Abstract:Nanoparticles are appealing drug delivery systems since they promise prolonged circulation time and predictable release behaviors. The current work reports a novel hyperbranched amphiphilic block copolymer synthesized using deactivation-enhanced atom transfer radical polymerization (DE-ATRP) for smart drug delivery. PEG2000-Br was applied as a macroinitiator to initiate acid-cleavable divinyl (ACD) monomers linked by acetal groups, formulating the hydrophilic (PEG) and hydrophobic (ACD) segments. The polymer appeared to self-assemble into micelles with diameters in the range of 70-100 nm, and was examined for the controlled release of doxorubicin (DOX). Results showed that DOX-loaded micelles (similar to 90 nm) could achieve drug loading as high as 8.2 wt%, with interesting pH-dependent release behaviors. Studies with flow cytometry (FCM) and confocal laser scanning microscopy (CLSM) showed that DOX-loaded micelles exhibited a high cellular uptake performance by HeLa cells, which indicated promising antitumor efficacy for such a drug delivery system. Additionally, such DOX-loaded micelles exhibited remarkable cytotoxicity against HeLa cells in a dose-and time-dependent manner due to the enhanced cell uptake behavior of micelles. These results indicated that the polymeric micelles might be used as a promising candidate for a pH-responsive drug delivery for cancer therapy.

Abstract:Polymeric-based particulate systems have been intensively developed to increase the short biological half-life and prevent enzymatic degradation of therapeutic proteins. These techniques demonstrate the useful characteristics for the delivery of therapeutic proteins and peptides to the targeted site of application and prevent the interaction of encapsulated drug with the normal cells. In this article, we have described the in depth of different pharmaceutical-based techniques that are currently being practiced for efficient delivery of therapeutic proteins and peptides. A comprehensive English literature was searched using different electronic search databases including PubMed, Science Direct, Web of Science, google scholar and library search. Different search terms and advanced search were made by combining all the search fields in abstract, keywords and/or titles. Findings of various studies that have been discussed in this article clearly indicate that polymeric-based techniques can significantly increase the therapeutic potentials of incorporated proteins with no known toxic effects. These techniques have shown to maintain the stability and retain biological activity of protein therapeutics. Hence it can be suggested that pharmaceutical-based techniques are promising drug carriers for efficient delivery of therapeutic proteins.

Abstract:Surface engineering of particles based on a polymeric coating is of great interest in materials design and applications. Due to the disadvantages of non-biodegradability and undesirable biocompatibility, the application of petroleum-based synthetic polymers coating in the biomedical field has been greatly limited. In addition, there is lack of a universal surface modification method to functionalize particles of different compositions, sizes, shapes, and structures. Thus, it is imperative to develop a versatile biopolymeric coating with good biocompatibility and tunable biodegradability for the preparation of functional particle materials regardless of their surface chemical and physical structures. Recently, the natural polysaccharide polymers (e.g. chitosan and cellulose), polyphenol-based biopolymers (e.g. polydopamine and tannic acid), and proteins (e.g. amyloid-like aggregates) have been utilized in surface modification of particles, and applications of these modified particles in the field of biomedicine have been also intensively exploited. In this review, the preparation of the above three coatings on particles surface are summarized, and the applications of these materials in drug loading/release, biomineralization, cell immobilization/protection, enzyme immobilization/protection, and antibacterial/antiviral are exemplified. Finally, the challenges and the future research directions on biopolymer coating for particles surface engineering are prospected.

Abstract:Highlights • Synthesis of polyacrylic acid (PAA) by different methods such as free radical polymerization, ATRP, RAFT and plasma polymerization have been discussed. • Design of PAA-based stimuli-responsive formulations by different crosslinking methods and the fabrication of PAA nano-systems is addressed. • Delivery of therapeutics from PAA-based systems through oral, buccal, nasal, ophthalmic, topical, vaginal and colonic administration routes is reviewed. • Efficacy of PAA in the delivery of genes, vitamins, proteins and growth factors is elucidated with a special focus on PAA-based injectable drug delivery systems. Polyacrylic acid (PAA)-based drug carrier systems have garnered much significance towards the delivery of various therapeutic agents via different administration routes for the accomplishment of a controlled and sustained release profile. PAA renders a safe and effectual system for drug delivery owing to its nontoxicity and mucoadhesiveness. Furthermore, the versatility of PAA is paramount towards the fabrication of varied architectures namely hydrogels (including injectable hydrogels), microparticles, nanoparticles and the like. The possibility of delivery of therapeutics using various routes such as oral, buccal, transdermal, nasal, vaginal and gene/vitamin/growth factors delivery further diversify the prospects of PAA in the biomedical field. This review presents an elaboration on the recent advances on PAA-based drug carrier systems along with their delivery routes. Abstract:AbstractPolypeptoids, with similar structure to polypeptides, are nowadays rising stars due to their excellent solubility, processibility, enzymatic stability, and biocompatibility. Attractive properties of polypeptoid‐containing copolymers endow them with various applications in surface antifouling, biosensing, microreactor, drug delivery, and stimuli–response. The syntheses of block copolymers containing polypeptoids have attracted a great amount of interest so far. The review summarizes the synthetic strategies of polypeptoid‐containing copolymers, that is, polypeptoid‐b‐polypeptoid, −polypeptide, −polyester, −polyether and ‐polyolefin developed recently, as well as their phase‐separation, assembly and stimuli–response properties.

Abstract:Polymers occupy a major portion of materials used for controlled release formulations and drug-targeting systems because this class of materials presents seemingly endless diversity in topology and chemistry. This is a crucial advantage over other classes of materials to meet the ever-increasing requirements of new designs of drug delivery formulations. The polymer architecture (topology) describes the shape of a single polymer molecule. Every natural, seminatural, and synthetic polymer falls into one of categorized architectures: linear, graft, branched, cross-linked, block, star-shaped, and dendron/dendrimer topology. Although this topic spans a truly broad area in polymer science, this review introduces polymer architectures along with brief synthetic approaches for pharmaceutical scientists who are not familiar with polymer science, summarizes the characteristic properties of each architecture useful for drug delivery applications, and covers recent advances in drug delivery relevant to polymer architecture.

Abstract:Due to the specific physiological pH throughout the human body, pH-responsive polymers have been considered for aiding drug delivery systems. Depending on the surrounding pH conditions, the polymers can undergo swelling or contraction behaviors, and a degradation mechanism can release incorporated substances. Additionally, polyurethane, a highly versatile polymer, has been reported for its biocompatibility properties, in which it demonstrates good biological response and sustainability in biomedical applications. In this review, we focus on summarizing the applications of pH-responsive polyurethane in the biomedical and drug delivery fields in recent years. In recent studies, there have been great developments in pH-responsive polyurethanes used as controlled drug delivery systems for oral administration, intravaginal administration, and targeted drug delivery systems for chemotherapy treatment. Other applications such as surface biomaterials, sensors, and optical imaging probes are also discussed in this review.

Abstract:Controlled drug release is an important aspect of biomedical applications of polymer materials. Polylactic acid (PLA)-based materials have been thoroughly researched for biomedical applications, owing to their attractive properties. Here, the advancements in the preparation and application of PLA-based polymers for drug delivery are reviewed. In addition to discussing the potentials of the PLA-based materials for drug delivery, the production techniques and various geometries of the drug carriers are also covered. The review has established that several drugs, including ibuprofen, amphotericin B, paclitaxel, 5-fluorouracil, doxorubicin, ketoprofen, rifampicin, salinomycin, ciprofloxacin, and ornidazole, have been successfully loaded into and released from PLA-based materials. Due to the hydrophobic nature of PLA, its modification to accommodate both the hydrophobic and hydrophilic drugs is emphasized in the literature.

Abstract:Polylysine-based composites have emerged as promising materials in biomedical applications due to their versatility, biocompatibility, and tunable properties. In drug delivery, polylysine-based composites furnish a novel platform for targeted and controlled release of therapeutic agents. Their high loading capacity and capability to encapsulate diverse drugs make them ideal candidates for addressing challenges such as drug stability and controlled release kinetics. Additionally, their biocompatibility ensures minimal cytotoxicity, vital for biomedical applications. They also hold substantial potential in tissue engineering by providing a scaffold with tunable mechanical characteristics and surface properties, and can support cell adhesion, proliferation, and differentiation. Furthermore, their bioactive nature facilitates cellular interactions, promoting tissue regeneration and integration. Wound healing is another area where polylysine-based composites show promise. Their antimicrobial properties help prevent infections, while their ability to foster cell migration and proliferation accelerates the wound healing procedure. Incorporation of growth factors or other bioactive molecules further enhances their therapeutic effectiveness. In biosensing applications, they serve as robust substrates for immobilizing biomolecules and sensing elements. Their high surface area-to-volume ratio and excellent biocompatibility improve sensor sensitivity and selectivity, enabling accurate detection of biomarkers or analytes in biological samples. Abstract:Background: Biocompatible polymers are gaining great interest in the field of biomedical applications. The term biocompatibility refers to the suitability of a polymer to body and body fluids exposure. Biocompatible polymers are both synthetic (man-made) and natural and aid in the close vicinity of a living system or work in intimacy with living cells. These are used to gauge, treat, boost, or substitute any tissue, organ or function of the body. A biocompatible polymer improves body functions without altering its normal functioning and triggering allergies or other side effects. It encompasses advances in tissue culture, tissue scaffolds, implantation, artificial grafts, wound fabrication, controlled drug delivery, bone filler material, etc. Objectives: This review provides an insight into the remarkable contribution made by some well-known biopolymers such as polylactic-co-glycolic acid, poly(ε-caprolactone) (PCL), polyLactic Acid, poly(3- hydroxybutyrate-co-3-hydroxyvalerate) (PHBV), Chitosan and Cellulose in the therapeutic measure for many biomedical applications. Methods: Various techniques and methods have made biopolymers more significant in the biomedical fields such as augmentation (replaced petroleum based polymers), film processing, injection modeling, blow molding techniques, controlled / implantable drug delivery devices, biological grafting, nano technology, tissue engineering etc. Results: The fore mentioned techniques and other advanced techniques have resulted in improved biocompatibility, nontoxicity, renewability, mild processing conditions, health condition, reduced immunological reactions and minimized side effects that would occur if synthetic polymers are used in a host cell. Conclusion: Biopolymers have brought effective and attainable targets in pharmaceutics and therapeutics. There are huge numbers of biopolymers reported in the literature that has been used effectively and extensively.

Poly (amino acid)s as new co-formers in amorphous solid dispersion

Abstract:The drug-amino acid co-amorphous systems and amorphous solid dispersions (ASDs) are promising methods to address the poor water solubility of poorly water-soluble drugs. However, some amino acids might not be perfect co-formers for co-amorphous systems, and the relatively low drug-loading of many ASDs is one of the main disadvantages of ASDs. Thus, poly-l-lysine and polyglutamic acid were selected as the co-formers, ball milled with basic mebendazole, neutral tadalafil and acidic valsartan at different weight ratios (from 3:1 to 1:3) to prepare poly (amino acid)-based ASDs, aiming to combine the advantages of co-amorphous systems (high drug-loading) and ASDs (relatively high Tg and high physical stability). All the mixtures were converted into amorphous after milling. The powder dissolution studies showed that drug-poly (amino acid) ASDs improved the dissolution rate of the drug in different ways and to different degrees. Moreover, the two poly (amino acid)s enhanced the physical stability of amorphous drugs. It is worthy to mention that the salt formation between the drug and the poly (amino acid) does not necessarily mean better performance compared to non-salt forming systems, and salt formation is also not a prerequisite for the formation of promising drug-poly (amino acid) ASDs.
Efficient Synthesis and Application of Protein-Poly(alpha-amino Acid) Conjugates

Abstract:Protein-polymer conjugates are important therapeutics for various diseases.There are currently two major challenges in this field:one is the search of new biodegradable polymers beyond traditional PEGylation,and the other is to develop highly efficient and site-specific conjugation strategy.Poly(α-amino acid)s (PαAAs) are biodegradable and biocompatible polymers with tunable properties and numerous functions,making them promising candidates for protein modification.In this review,we summarize our recent progresses in protein-PαAAs conjugates.Specifically,we discuss our developments in:(1) Recent developments in the controlled ring-opening polymerization (ROP) of α-amino acid N-carboxyanhydrides (NCAs),including amine-based initiators,organometallic initiators,organosilicon amines initiators and sulfide-based initiators.For instance,trimethylsilyl phenylsulfide (PhSTMS) is a novel initiator for controlled ROP of NCAs.It exhibits higher nucleophilicity than conventional amine-based initiator,and thus affords considerably higher chain initiation rate to ensure a more controlled polymerization.Moreover,this initiator is well-tolerated to various functional groups.(2) In situ functionalization of PαAAs for site-specific protein conjugation,and construction of various topological structures.Using PhSTMS initiator,it in situ generates a reactive phenyl thioester group at one end of the PαAAs,which can be used for protein N-terminus conjugation via native chemical ligation (NCL);moreover,ROP of glycine NCA yields oligoglycine at the other end of PαAAs,which can be used for C-terminus protein conjugation via sortase-A mediated ligation (SML).More interestingly,combinatory use of the two methods can construct various topological protein-PαAA conjugates including the head-to-tail circular conjugates.(3) Development of functional PαAAs for potential protein conjugation.Various functional PαAAs have been developed as delivery materials or hydrogels.To further expand the arsenal of PαAAs for potential modulation of protein functions,PαAAs that mimic protein post-translational modifications (PTM) are synthesized;On the other hand,a series of multiple stimuli-responsive PαAAs are also produced.These PαAAs show interesting enzyme,light,and/or thermal responsiveness,which could be potentially harnessed for modulation of protein functions in the future.
