Study on Tea Polyphenols Regulating Mitochondrial Autophagy and Inhibiting Senescence of Vascular Endothelial Cells

Abstract:OBJECTIVE: To study the effects of tea polyphenols on mitochondrial autophagy in the senescence of vascular endothelial cells, specifically inhibiting mitochondrial autophagy, and to further explore the possible mechanism of tea polyphenols in inhibiting cell senescence. STUDY DESIGN: A cell senescence model was established. CCK-8 was applied to detect cell proliferation viability. GFP-LC3 and MitoTracker fluorescent co-localization test was conducted to evaluate the formation of mitochondrial autophagosomes. Parkin and MitoTracker fluorescent co-localization test were performed to evaluate Parkin protein translocation. Western blot was employed to detect the expression levels of mitochondrial autophagy-related proteins PINK1, Parkin, LC3B-II, and p62, as well as senescence-related proteins pl6 and p21. SA-p-gal staining was used to detect the degree of cell senescence, and immunofluorescence was applied to detect the Y-H2A.X (Serl39) expression, mtROS generation, and mitochondrial membrane potential (MMP) changes. RESULTS: Compared with the blank control group, the co-localization degree of GFP-LC3 and MitoTracker in the human aortic endothelial cells (HAECs) of the model group decreased, the expression of Parkin protein and the degree of translocation decreased, and the expression of p62 increased. Compared with the model group, the GFP-LC3, the co-localization degree of Parkin and MitoTracker in the administration group increased, the expression levels of Parkin and LC3B-II were upregulated, while the expression level of p62 was downregulated. Mdivi-1 was given to inhibit mitochondrial autophagy. Compared with the model group, the co-localization degree of GFP-LC3 and MitoTracker and the expression and translocation of Parkin did not change in the administration group. The expression levels of LC3B-II and p62 were upregulated, and mtROS was generated. There were no statistically significant differences in the levels of MMP, the number of SA-p-gal blue-stained cells, pl6, p21, and Y-H2A.X (Serl39) protein expressions. CONCLUSION: High-sugar and high-fat stimulation can inhibit mitochondrial autophagy in HAEC cells, while tea polyphenols can restore mitochondrial autophagy in HAEC cells. This may be one of the important mechanisms for improving mitochondrial function, reducing mtROS production, and delaying cell senescence.
