Abstract:A set of autotrophic mixed ammoxidation bacteria with high efficient was screened from active sludge of a sewage purification-factory.Batch tests were performed.The results showed that the optimum temperature and pH were 30℃ and 7.0～7.5,respectively.DO had little effect on ammoxidation. Ammonia oxidation rate (AOR) has no apparent change under 0～ 1.0 % salinity stress but decreased rapidly when the salinity was over 1.0%.AOR changed little on PC of 20～900 mg/L.These bacteria grew well on ANC (0～1000 mg/L).The maximum AOR could reach 150 mg·L~(-1)·d.The end products of oxidation mainly were nitrite and the residual ANC was below 3 mg/L.Five strains of ammoniaoxidizing bacteria were identified and primarily classified as Nitrosornonas sp.by using PCR-DGGE and sequencing.In addition,some other bacteria related to nitrogen transformation were found by PCR with specific primers.

Abstract:Anaerobic ammonium oxidation(anammox) is one of important discoveries in both environmental and microbiological fields.It supplements biogeochemistry of nitrogen cycle in nature,extends microbial diversity and urges development of novel nitrogen removal biotechnology.Recently,plenty breakthroughs have been made in the research of anammox bacteria.Their properties in bacterial morphology,cytochemistry,enrichment and isolation,physiology and biochemistry,ecology were revealed,and probable metabolic models were proposed,and the important enzymes were purified and characterized.A family of Anammoxaceae including five genus,nine species has been established.This paper is a systematic review in the features of anammox bacteria and the nine identified species.

Abstract:Anaerobic ammonium oxidation (anammox) is not only an important part of the global nitrogen cycle,but also a vital foundation of new biological nitrogen removal technologies for wastewater treatment and environmental bioremediation.As the biocatalyst of anammox process,anaerobic ammonium oxidation bacteria (AnAOB) attract increasing attention in the fields of microbiology,environmental engineering,and geoscience with extensive research in recent years.In this study,we reviewed the newest research advance on taxonomy,characteristics,and biotechniques used for the analysis of AnAOB.In all,6 genera containing 21 AnAOB species have been identified based on the polyphasic taxonomy by using the genotypic group-dividing method;this includes the newly discovered genus Candidatus Anammoximicrobium.Some studied species were further compared to identify the similarities and differences between their morphological,cellular components,physiological,and ecological characteristics;they were found to have different sensitivities to environmental factors such as temperature,salinity,and organic matter,which caused niche divergence among them.These species were favorable for application to environmental engineering.In addition,a series of advanced molecular biotechniques such as polymerase chain reaction denaturing gradient gel electrophoresis,fluorescence in situ hybridization,and quantitative polymerase chain reaction have been successfully applied to assess species clarification,community distribution,microbial activity,and cell numbers of AnAOB.Moreover,many specific primers for AnAOB 16S rRNA or functional genes (hzo,nirS) have been developed.Furthermore,some important topics and obstacles for further research,i.e.,obtaining purified AnAOB,elucidating metabolic pathways and genetic features of AnAOB,clarifying the ecological contribution of AnAOB in the environment,were proposed.The findings of this study might provide valuable information for both anammox process and AnAOB researches.
Characterization of Oligotrophic AnAOB Culture: Morphological, Physiological, and Ecological Features

Abstract:Anaerobic ammonium oxidation (anammox) process is regarded as a promising nitrogen removal technology to treat ammonium wastewaters in a wide concentration range. Oligotrophic anaerobic ammonia oxidation bacteria (O-AnAOB) culture has been successfully achieved from a new anammox system to treat superlow ammonium concentration wastewaters. In this work, the O-AnAOB culture was compared with the eutrophic AnAOB (E-AnAOB) culture to reveal its physiological, morphological, and ecological features. Results showed that the specific anammox activity (SAA) of O-AnAOB culture was 0.07 kgN/(kgVSS·d) with the nitrogen removal rate (NRR) of 0.20 kgN/ (m3 d) in the reactor, while the SAA of E-AnAOB culture was 2.11 kgN/(kgVSS·d) with the NRR of 11.10 kgN/(m3 d). The hzs gene transcription levels (hzs-mRNA) of O-AnAOB and E-AnAOB cultures were 1.32 × 109 copies/gVSS and 1.51 × 1010 copies/gVSS, respectively. Morphologically, the O-AnAOB culture took on the unique brown color rather than the typical red color of E-AnAOB. The O-AnAOB cells lived in a disperse pattern in the culture. The cells were seriously deformed with deep craters on the cell wall. The size of anammoxsome and paryphoplasm compartments inside the O-AnAOB cells was smaller than that inside the E-AnAOB cells. Ecologically, the O-AnAOB culture had special microbial community with a higher bacterial diversity than the E-AnAOB. The most dominant genera in O-AnAOB were Anaerolineaceae (33.7%, fermentative bacteria), Candidatus Kuenenia (17.4%, anammox bacteria), and Nitrospira (7.3%, nitrite oxidizing bacteria). This study provided an insight into the new anammox process for deep nitrogen removal from wastewaters.

Abstract:One strain of autotrophic Ammonia-Oxidizing Bacteria (AOB) CM-NRO14 was isolated from the aerobic activated sludge of a domestic wastewater treatment plant using enrichment, isolation and purification. This study was aimed to determine phylogenetic position and ammonia oxidation characteristics of the strain. Strain CM-NRO14 was identified as Nitrosomonas eutropha based on morphological and biochemical/biophysical characters and 16S rDNA sequence. The maximal rate of ammonia oxidation was 24.54 mg/(L·d) under the optimal ammonia concentration 331.2 mg/L. Km and Ki for ammonia were 45.66 mg/L and 2 401 mg/L, respectively. Ki for nitrite was 1 524 mg/L. The optimal pH for ammonia oxidation was 7.79. The optimal temperature was 34℃. When cultured in the conditions of ammonia nitrogen 180 mg/L, pH 7.79 and 34℃, strain CM-NRO14 removed 91.6% of ammonia in 4 days. Doubling time of the strain was 2.94 d.

Abstract:Anaerobic ammonium oxidation (anammox) is the microbial conversion of ammonium and nitrite to dinitrogen gas. The functional microbes of anammox reaction are anammox bacteria, which were discovered in a wastewater treatment system for nitrogen removal. Anammox bacteria are prevalent in anoxic ecosystems and play an important role in both biological nitrogen cycle and nitrogen pollution control. In this paper, we reviewed the investigation on ecological characteristics of anammox bacteria, and tried to figure out their complicated intraspecies and interspecies relationships. As for intraspecies relationship, we focused on the quorum sensing system, a cell density-dependent phenomenon. As for interspecies relationship, we focused on the synergism and competition of anammox bacteria with other microorganisms for substrate and space. Finally, we discussed the great influence of environmental factors (e.g., dissolved oxygen, organic matters) on the constitution, structure and function of anammox bacteria community.

Abstract:Sulfate-dependent anaerobic ammonium oxidation is a novel biological reaction, in which ammonium is oxidized with sulfate as the electron acceptor under anoxic conditions. Ammonium and sulfate are cosmopolitan chemical species which are an integral part of the global nitrogen and sulfur cycles. A detailed exploration of sulfate-dependent anaerobic ammonium oxidation is quite practical. In this work, a bacterial strain named ASR has been isolated from an anaerobic ammonia and sulfate removing reactor working under steady-state. On the basis of electron microscopy, physiological tests and 16S rDNA phylogenetic sequence analysis, the strain ASR is found to be related to Bacillus benzoevorans. According to the biological carbon source utilization test, the strain ASR could use many carbon sources. Its optimum pH value and temperature were 8.5 and 30 °C, respectively. The test proves that the strain ASR is able to use sulfate to oxidize ammonia anaerobically. The maximum ammonia and sulfate removal rates were 44.4% and 40.0%, respectively. The present study provided biological evidence for the confirmation and development of sulfate-dependent anaerobic ammonium oxidation and brought new insights into the global nitrogen and sulfur cycles.

Abstract:Various studies have revealed anaerobic ammonium oxidation (anammox) as a very attractive alternative process suitable for nitrogen removal from wastewater. Here we investigated anammox bacteria in eight different nitrogen removal reactors. The diversity and abundance of anammox bacteria were determined by the 16S rRNA gene analysis, fluorescence in situ hybridization with specific probes and real-time quantitative PCR (qPCR). In these reactors, at least eight unique near full length anammox 16S rRNA gene sequences were detected, which were distributed over two genera; Candidati Brocadia and Kuenenia. FISH results confirmed that only one anammox bacterium dominated the community in each of the eight reactors investigated in this study. qPCR analysis revealed that anammox bacteria were present in seven of the reactors in the order of 10(9) cells/ml and 10(7) cells/ml in reactor A1. The dominant and divergent Brocadia-like anammox phylotype in one reactor represented a novel species for which we propose the name Candidatus Brocadia sinica. Taken together, these results indicated that a single seeding source could be used to seed anammox reactors designed to treat different types of wastewater, which could lead to a faster start-up of bioreactors.

Abstract:Nine strains of ammonia nitrogen degradation strains from C1 to C9 were isolated from industrial wastewater to study their degradation and conversion of ammonia nitrogen. The results showed that C2 strain with a high degradation activiity of ammonia nitrogen, and the ammonia nitrogen degradation rate of the activated C2 strain was 93% within 24 h when the initial concentration of ammonia nitrogen was 200 mg/L under the conditions of inoculation 10%, temperature 35?C, pH 7.0, rotation 200 r/min. And C2 was identified as Bacillus amyloliquefaciens.

Abstract:The ammonium-oxidizing microbial community was investigated in a granular sludge anaerobic ammonium-oxidizing (anammox) reactor that was operated for about 1 year with high anaerobic ammonium oxidation activity (up to 0.8 kg NH4+-N m(-3) day(-1)). A Planctomycetales-specific 16S rRNA gene library was constructed to analyse the diversity of the anaerobic ammonium-oxidizing bacteria (AnAOB). Most of the specifically amplified sequences (15/16) were similar to each other (> 99%) but were distantly related to all of the previously recognized sequences (< 94%), with the exception of an unclassified anammox-related clone, KSU-1 (98%). An ammonia monooxygenase (amoA) gene library was also analysed to investigate the diversity of 'aerobic' ammonium-oxidizing bacteria (AAOB) from the beta-Proteobacteria. Most of the amoA gene fragments (53/55) clustered in the Nitrosomonas europaea-Nitrosococcus mobilis group which has been reported to prevail under oxygen-limiting conditions. The quantitative results from real-time polymerase chain reaction (PCR) amplification showed that the dominant AnAOB comprised approximately 50% of the total bacterial 16S rRNA genes in the reactor, whereas the AAOB of beta-Proteobacteria represented only about 3%. A large fragment (4008 bp) of the rRNA gene cluster of the dominant AnAOB (AS-1) in this reactor sludge was sequenced and compared with sequences of other Planctomycetales including four anammox-related candidate genera. The partial sequence of hydrazine-oxidizing enzyme (hzo) of dominant AnAOB was also identified using new designed primers. Based on this analysis, we propose to tentatively name this new AnAOB Candidatus 'Jettenia asiatica'.

Abstract:Anaerobic ammonium oxidation (anammox) is an important discovery in microbiology and environmental sciences, which can simultaneously remove NH4(+) -N and NO3(-) -N, being valuable in environmental engineering. However, anaerobic ammonium oxidizers are extremely slow-growing, and their population's doubling time is longer than 11 days, which seriously restricts the application of anammox process. Therefore, the study of anammox bacteria is of significance. It has been proved that besides planctomycetes, the first recognized anammox bacteria, both nitrifying bacteria and denitrifying bacteria are also capable of anaerobic ammonium oxidation. These anammox bacteria have wide-spread habitats, which offered a chance to exploit new bacterial resources for anammox. Nitrifying bacteria and denitrifying bacteria have the function of anammox, and their metabolic diversity provides a basis to speed up the start-up of anammox reactor. It was revealed that anaerobic digestion sludge can present anammox activity, with sulphate as electron acceptor. The new bioreaction lays a foundation for the development of novel N-removal biotechnology, being conducive to the development and application of anammox to get more bacterial resources for anammox and to make clear the ecological distribution of anammox bacteria.

Abstract:Anaerobic ammonium oxidation is considered to be the most economical and low-energy biological nitrogen removal process. So far, anammox bacteria have not yet been purified from cultures. Some nitrogen-removing microorganisms cooperate to perform the anammox process. The objective of this research was to analyze the abundance and diversity of nitrogen-removing microorganisms in an anammox reactor started up with bulking sludge at room temperature. In this study, the ammonia-oxidizing archaea phylum Crenarchaeota was enriched from 9.2 to 53.0%. Nitrosomonas, Nitrosococcus, and Nitrosospira, which are ammonia-oxidizing bacteria, increased from 3.2, 1.7, and 0.1% to 12.8, 20.4, and 3.3%, respectively. Ca. Brocadia, Ca. Kuenenia, and Ca. Scalindua, which are anammox bacteria, were detected in the seeding sludge, accounting for 77.1, 11.5, and 10.6%. After cultivation, the dominant genus changed to Ca. Kuenenia, accounting for 82.0%. Nitrospirae, nitrite oxidation bacteria, decreased from 2.2 to 0.1%, while denitrifying genera decreased from 12.9 to 2.1%. The results of this study contribute to the understanding of nitrogen-removing microorganisms in an anammox reactor, thereby facilitating the improvement of such reactors. However, the physiological and metabolic functions of the ammonia-oxidizing archaea community in the anammox reactor need to be investigated in further studies.

Abstract:Nitrate production during anammox can decrease total nitrogen removal efficiency, which will negatively impact its usefulness for the removal of nitrogen from waste streams. However, neither the performance characteristics nor physiological shifts associated with nitrate accumulation in anammox reactors under different nitrogen loading rates (NLRs) is well understood. Consequently, these parameters were studied in a lower NLR anammox reactor, termed R1, producing higher than expected levels of nitrate and compared with a higher NLR reactor, termed R2, showing no excess nitrate production. While both reactors showed high NH4+-N removal efficiencies (>90%), the total nitrogen removal efficiency (<60%) was much lower in R1 due to higher nitrate production. Metagenomic analysis found that the number of reads derived from anammox bacteria were significantly higher in R2. Another notable trend in reads occurrence was the relatively higher levels of reads from genes predicted to be nitrite oxidoreductases (nxr) in R1. Binning yielded 27 high quality draft genomes from the two reactors. Analysis of bin occurrence found that R1 showing both a decrease in anammox bacteria and an unexpected increase in nxr. In-situ assays confirmed that R1 had higher rates of nitrite oxidation to nitrate and suggested that it was not solely due to obligate NOB, but other nxr-containing bacteria are important contributors as well. Our results demonstrate that nitrate accumulation can be a serious operational concern for the application of anammox technology to low-strength wastewater treatment and provide insight into the community changes leading to this outcome.
The Anammox Process at Typical Feast-Famine States: Reactor Performance, Sludge Activity and Microbial Community

Abstract:Anaerobic ammonium oxidation (Anammox) is a chemolithotrophic bioprocess which has been widely applied in the treatment of different concentrations of ammonium-containing wastewaters. However, there is less attention on the problem that the instantaneous growth rate (or metabolic rate) and equilibrium growth rate were un-synchronous for anammox bacteria due to their long generation time and self-immobilization of the granular sludge which could lead to the inaccurate estimation. In this study, the anammox process was firstly divided to four typical feast-famine (starvation, satiation, tolerance and poison) states based on the combination of both off-site and in-situ anammox reaction kinetics. Then, four respective lab-scale bioreactors were operated at each state for over a year to achieve stable anammox performance. The results showed that the nitrogen removal rates of bioreactors were 0.53, 2.24, 9.30 and 12.96 kg N/(m(3).d); and the specific anammox activities of granular sludge were 188.94 (48%), 313.29 (79%), 397.50 (100%) and 198.60 (50%) mg N/(g VSS.d) which could reflect the reactivity of each feast-famine state. The stable microbial communities of bioreactors were cultured and analyzed, whose species diversity went down with the decrease of Shannon and ACE index. The relative abundance of anammox bacteria increased from 11% to 57% from starvation to poison state. Candidatus Brocadia/Nitrospira, Candidatus Kuenenia and Brocadiaceae unclassified were revealed to be the distinctive functional bacteria, which could serve as the indicator of each state. The setting up of the typical feast-famine states could be regarded as the landmark to help the design, control and optimization of anammox process.

Abstract:An anaerobic ammonium-oxidation (anammox) reactor was operated for more than 500 days and the anammox activity of the biomass in the reactor reached 0.58kg Ntotal/kg VSSd. The removal ratios of NO2−-N to NH4+-N in both reactor and activity tests were nearly 1.1. The bacterial diversity in the reactor was investigated by analysis of 16S rRNA gene clone libraries and quantitative real-time PCR (qPCR). The analysis showed that more than half of the clones in the library were affiliated to recognized filamentous bacteria. The previously recognized anammox bacterium (AnAOB) Candidatus Kuenenia stuttgartiensis was only detected by using a Planctomycetes-specific 16S rRNA gene primer set. However, at least two different types of AnAOB were detected by the primer set targeting the hydrazine-oxidizing enzyme gene (hzo). The aerobic ammonium-oxidizing bacteria (AAOB) Nitrosomonas europaea–eutropha group, which is widely detected in oxygen-limited environments, was also found in this reactor. The result of qPCR indicated that AnAOB comprised 16% of the community population while AAOB comprised less than 1% in the reactor.

Abstract:A total of 8 pure strains of ammonia-oxidizing bacteria (AOB) were collected from the rhizosphere of wetland plants including Arundo donax, Herb of rice galingale and willow. By microscopic observation and color reaction of Griess reagent, two strains of YL-1 and YS-2 were selected as the dominant genus to conduct performance investigation. Bath experiment results demonstrated that YL-1 shows the similar properties with YS-2 on the degradation of ammonia under the conditions of different pH value, ammonia loading and oxygen concentration. Both the two strains all showed excellent adaptability to the conditions of strong alkaline (pH=8.5) and high free ammonia (FA) concentration (18.2 mg/L). In spite of it, the ammonia degradation performance of the two strains to the high ammonia loading was all inhibited by the low DO condition.

These strains were identified as Acinetobacter junii YB, Pseudomonas putida YH and Pseudomonas aeruginosa YL, respectively. All three strains exhibited efficient heterotrophic nitrification-aerobic denitrification capabilities, with a low accumulation of nitrification products over a wide range of ammonium loads, and were able to utilize nitrite and nitrate as a sole nitrogen source. About one-half of the consumed total nitrogen was converted into intracellular nitrogen, and the remainder was believed to be denitrified. Strain YH was found to possess higher nitrogen removal capacity than the other two strains. Single-factor experiments indicated that the optimal conditions for all three heterotrophic ammonium oxidation strains were as follows: succinate as the carbon source, C/N ratio of 10, pH of 7.0, temperature of 30 °C, and shaking speed of 160 rpm. The presence of hydroxylamine oxidase, nitrite reductase and nitrate reductase provided additional evidence of heterotrophic nitrification and aerobic denitrification of the isolates. Higher enzyme activity observed in strain YH further demonstrated its superior nitrogen removal ability.

Abstract:In order to solve nitrogen pollution in environmental water, two heterotrophic nitrifying and aerobic denitrifying strains isolated from acid paddy soil were identified as Achromobacter sp. strain HNDS-1 and Enterobacter sp. strain HNDS-6 respectively. Strain HNDS-1 and strain HNDS-6 exhibited amazing ability to nitrogen removal. When (NH4)(2)SO4, KNO3, NaNO2 were used as nitrogen resource respectively, the NH4+-N, NO3- -N, NO2- -N removal efficiencies of strain HNDS-1 were 93.31%, 89.47%, and 100% respectively, while those of strain HNDS6 were 82.39%, 96.92%, and 100%. And both of them could remove mixed nitrogen effectively in low C/N (C/ N = 5). Strain HNDS-1 could remove 76.86% NH4+-N and 75.13% NO3- -N. And strain HNDS-6 can remove 65.07% NH4+-N and 78.21% NO3(-) -N. A putative ammonia monooxygenase, nitrite reductase, nitrate reductase, assimilatory nitrate reductase, nitrate/nitrite transport protein and nitric oxide reductase of strain HNDS-1, while hydroxylamine reductase, nitrite reductase, nitrate reductase, assimilatory nitrate reductase, nitrate/nitrite transport protein, and nitric oxide reductase of strain HNDS-6 were identified by genomic analysis. DNA-SIP analysis showed that genes Nxr, narG, nirK, norB, nosZ were involved in nitrogen removal pathway, which indicates that the denitrification pathway of strain HNDS-1 and strain HNDS-6 was NO3- -> NO2- -> NO -> N2O -> N-2 during NH4+ -> N removal process. And the nitrification pathway of strain HNDS-1 and strain HNDS-6 was NO2- -> NO3- , but the nitrification pathway of NH4+ -> NO2- needs further studies.
