Studies on Pharmacokinetics of Ketoprofen Injection in Rats

Abstract:The aim of this paper is to establish a RP-HPLC method to measure ketoprofen concentrations in plasma of rats and compare the intramuscular and intravenous pharmacokinetics of ketoprofen injection. The separation was performed on a Shim-pack VP-ODS C-18 (250 x 4.6 mm, 5 mu m) column. The mobile phases were composed of phosphate buffer (pH 3.5): acetonitrile: water = 2:43:55 (v/v/v) with a flow rate of 12 mL/min. The detection was carried out at 255 nm under the column temperature of 25 degrees C and the injection volume was 20 mu L. Healthy male SD rats were randomly divided into-two groups, a single intramuscular, caudal intravenous injection were conducted respectively, and administered dose was 10 mg/kg, blood samples were collected from the orbital venous plexus. After the protein was precipitated by hydrochloric acid, extracted and separated in ether, blow-dried in nitrogen, dissolved in mobile phase, purified in pre-column, the content of ketoprofen in rats' plasma was determinated by RP-HPLC, and the data of each group was analyzed by DAS 2.0 pharmacokinetic software. Ketoprofen was separated from impurities with no other interference. The linear range was determined from 0.02 to 40 mu g/mL with the correlation coefficient of 0.9998 for plasma concentrations of ketoprofen; the lower limit of quantification (LLOQ) was 0.02 mu g/mL at S/N >= 5. Average extraction recoveries were 81.47, 8232, and 79.88%, inter-day precisions were 2.98, 3.19, and 3.43%, intra-day precisions were 6.03, 4.82, and 5.27% for high, middle and low concentrations of ketoprofen, respectively. Pharmacokinetics parameters of ketoprofen in rats after single intramuscular and intravenous administration: T-1/2z were 5.19 +/- 0.08 and 2.95 +/- 0.14 h, T-max were 0.33 +/- 0.01 and 0.02 +/- 0.01 h, C-max were 1832 +/- 0.02 and 19.02 +/- 0.01 mg/L, AUC(0-24) were 36.46 +/- 0.17 and 37.30 +/- 0.23 mg/L.h, respectively. In conclusion, the method is simple, accurate, with strong specificity and good reproducibility and can be used for detecting plasma concentration of ketoprofen; ketoprofen by intramuscular injection was absorbed quickly and completely, and with high bioavailability, which could provide basic data for clinical intramuscular injection administration.
