Spatiotemporal model of focal inflammatory and immune cellular response in ischemic rat brains

Abstract:Objective: To investigate spatial location and time-course of inflammatory and immune cells, and to inquire into pathological relationship of inflammatory and immune factors with cerebral ischemic injury. Methods: Spatial distribution and reactive time-course of monocyte/macrophages, microglia, T-lymphocytes and immunomolecules were immunohistochemically explored with focal ischemic model. Results: ED1 positive cells infiltrated into ischemic penumbra at 3 day postischemia, and progressively increased in number, and invaded ischemic core at 1-2 week postischemia. Hypertrophic and proliferative changes of OX42 positive cells occurred in the ischemic penumbra at 3 day postischemia, and persisted up to 2 week postischemia. OX6 positive ramified cells and round cells appeared in ischemic penumbra (ramified) and core (round) at 3 day postischemia, and increased and peaked in number at 1 week postischemia, and then declined gradually. Numerous CD3 positive cells were located at the ischemic penumbra at 3 day postischemia, and increased further in number and invaded the infarct core at 1-2week postischemia. Conclusion: Accumulation and activation of monocyte/macrophages, microglial cells and T-lymphocytes, and immunomolecules were induced following cerebral focal ischemia, which indicates that cerebral focal ischemia initiates inflammatory and immune response which may be involved in pathological process of cerebral ischemic injury.
