Abstract:To develop novel fluorescent probes for detecting endogenous histidine (His) plays significant roles in physio-logical and pathological fields. Herein, a non-emissive Ir(III) complex [Ir(C-N)(2)(CH3CN)(2)]+NO3- (IrCN, C-N represents 2-phenylpyridine derivative) has been demonstrated to be a reaction-based fluorescent probe for the recognition of His in HEPES buffer. It is found that IrCN can selectively detect His with a distinct "turn-on" luminescence and low limit of detection (LOD = 0.62 mu M), relying on which, IrCN can light-up the cytoplasm and nuclear region of the fixed cells, exhibiting negligible cytotoxicity and suitable solubility under test concentration.

Abstract:A new zinc(II) porphyrin conjugate with an appended pyrene subunit has been synthesized and shown to exhibit significant and analytical usefulness for fluorescence sensing toward imidazole derivatives. The molecular recognition was based on the bridging interaction of the imidazole ring of analyte with the zinc(II) center of the porphyrin, while the transduction signal for the recognition process was the pyrene excimer fluorescence. The sensor was constructed and applied for fluorescence assay of histidine in aqueous solution by immobilizing the sensing material in a plasticized PVC membrane. When the membrane was bathed in an alkaline solution void of histidine, zinc(II) porphyrin was present in the monomer form, and pyrene emitted monomer fluorescence at 378 and 397 nm. With the presence of histidine in the sample solution, histidine was extracted into the membrane phase and bridged with the Zn(II) center of the porphyrin, causing the monomer porphyrin to be converted to its dimeric species. Since the formation of porphyrin dimer was accompanied by the enhancement of pyrene excimer emission at 454 nm, the chemical recognition process could be directly translated into a fluorescent signal. With the optode membrane M1 described, histidine in sample solution from 6.76 x 10(-7) to 5.01 x 10(-3) M can be determined. The limit of detection was 1.34 x 10(-7) M. The optical selectivity coefficient obtained for histidine over biologically relevant amino acids and anions met the selectivity requirements for the determination of histidine in biological samples. Serum histidine values obtained by the optode membrane fell in the normal range of the content reported in the literature and were in good agreement with those obtained by HPLC.

Abstract:A plasticized polymer film, poly(vinyl chloride) (PVC) incorporated with a specific porphyrin dimer, is shown to exhibit significant and analytical usefulness for optical response toward histidine. The porphyrin dimer containing a free-base porphyrin and a covalently linked metalloporphyrin is shown to be weakly fluorescent as a result of the photoinduced intramolecular electron transfer from the inner free-base porphyrin in singlet excited state to a low-spin cobalt(II). The fluorescence enhancement of the membrane by histidine is based on favorable extraction of histidine into the bulk organic membrane and complexation with the inner metallopophyrin moiety and inhibiting the electron transfer process. With the optode membrane described, histidine in a sample solution from 0.0045 to 1.53 mM can be determined. The calibration curve of the optode membrane for histidine shows a good correlation with the mathematically derived formalism and, thus, confirms the theoretically expected behavior. The sensor presented exhibits high selectivity toward histidine over several amino acids and common inorganic anions. The optical selectivity coefficients obtained for histidine over other biologically relevant amino acids and anions are shown to meet the selectivity requirements for the monitoring concentration levels of histidine in biological samples. The selective characteristic of the sensor has been discussed in the view of the coordination chemistry of metalloporphyrin.

Abstract:We report a novel turn-on two-photon fluorescent probe (TPNF) for detecting formaldehyde based on aza-Cope rearrangement and intramolecular charge transfer (ICT). The probe, containing a 1,8-naphthalimide dye with a homoallylamino group, has was shown with high selectivity, sensitivity and low cytotoxicity and it was capable of detecting formaldehyde in live cells. Moreover, a time-dependent density functional theory (TD-DFT) calculation was performed to demonstrate the turn on mechanism of the probe via ICT effect. Finally, a zebrafish larvae imaging experiment reveals that TPNF displayed a significant signal change when treated with FA in vivo. Abstract:We described a new ensemble-approach-based chemosensor, NCH-Cu2+, for highly selective and reversible detection of histidine (His) in aqueous solution and live cells. The ligand NCH exhibited specific binding with Cu2+ ions over other metal ions, accompanied with a 92.2% fluorescence quenching. The decomplexation of NCH-Cu2+ ensemble by His led to the liberation of the fluorophore, NCH, and thus the fluorescence was recovered. The specific fluorescence enhancement of NCH-Cu2+ towards His showed a good linearity with a detection of limit at 70nm. Quantification of intracellular His at the single cell level was achieved by microscopy and flow cytometry. Besides the UV/Vis and emission titration, reversibility of the NCH-Cu2+ towards His was further confirmed by imaging and cytometry analysis. In addition, microscopy studies revealed that NCH-Cu2+ was distributed in the lysosome of live cells, where it could be employed as a fluorescent biosensor for imaging of His at subcellular level.

Abstract:A naphthalimide-based fluorescent probe CP has been synthesized with simple steps. It can selectively and sensitively recognize copper ions (Cu2+) in HEPES buffer (50mM, pH 7.2). The fluorescence intensity of CP is linearly proportional to the concentration of Cu2+ ranging from 0–8.3μM (correlation coefficient R2=0.9808). The resulted complex CP@Cu can serve as a turn-on fluorescent probe for the detection of histidine and histidine rich proteins in broad pH application range. Upon the addition of histidine, the fluorescence intensity of CP@Cu exhibits a linear correlation with the concentration of histidine ranging from 0–200μM (correlation coefficient R2=0.9912). Moreover, CP@Cu has potential for imaging histidine in vitro experiments and has promise in real sample applications with great validity.

Abstract:A novel fluorescence sensor based on porphyrinic zirconium-based metal-organic frameworks, L-cysteine modified PCN-222 (L-Cys/PCN-222), was developed to selectively recognize histidine enantiomers and sensitively detect Hg2+. The dual-functional sensor was successfully prepared by solvent-assisted ligand incorporation method and characterized by X-ray diffractometer (XRD), scanning electron microscopy (SEM), 1H nuclear magnetic resonance spectroscopy (1H NMR), Fourier transform infrared (FT-IR), circular dichroism (CD), X-ray photoelectron spectroscopy (XPS), and nitrogen adsorption-desorption. L-Cys/PCN-222 not only showed a higher quenching response of L-histidine than that of D-histidine with a fast fluorescent response rate under < 40 s but also exhibited the low detection limits of L- and D-histidine (2.48 μmol L-1and 3.85 μmol L-1). Moreover, L-Cys/PCN-222 performed as a fluorescent and visual sensor for highly sensitive detection of Hg2+ at the line range of 10~500 μmol L-1, and the detection limit was calculated to be 2.79 μmol L-1 in the surface water. The specific and selective recognition of our probe towards chiral compound and metal ion made it suitable for real field applications.

Abstract:Introduction: Hematoporphyrin (Hp) and hematoporphyrin derivatives (HpDs) have been widely used as photosensitizers in photodynamic therapy (PDT). Radiolabeling of HpDs is helpful for preclinical and clinical studies of PDT.Methods: The histidine-coupled hematoporphyrin (His-Hp) was synthesized and radiolabeled with [Tc-99m(CO)(3)(H2O)(3)](+). Biodistribution of the radioligand and fluorescent imaging of His-Hp in mice bearing S180 tumor were investigated.Results: [Tc-99m(CO)(3)](+)-labeled His-Hp was electrically neutral, hydrophilic and stable. The biodistribution of the radioligand in S180 tumor-bearing mice was similar with that of nonlabeled HpD in the literature. The uptake of His-Hp in tumors and livers was confirmed by fluorescent imaging.Conclusions: The complex [Tc-99m(CO)(3)](+)-His-Hp might be suitable for in vivo dose evaluation of HpD in PDT. Abstract:Two new ditopic metalloporphyrin receptors constructed by combining metalloporphyrin with crown ethers have been prepared and characterized. 1H NMR and MS spectra confirmed the complexation of receptor with peptide driven by coordination interaction and hydrogen bonding. UV/vis experiments revealed that the receptors exhibited high binding affinity to histidine-containing peptides. These receptors could differentiate short peptides of C-terminal histidine and N-terminal histidine and formed the most stable complexes with tripeptide.

Abstract:A new homocysteine-selective sensor based on the iridium(III) complex Ir(pba)(2)(acac) (Hpba = 4-(2-pyridyl)benzaldehyde; acac = acetylacetone) was synthesized, and its' photophysical properties were studied. Upon the addition of homocysteine (Hcy) to a semi-aqueous solution of Ir(pba)(2)(acac), a color change from orange to yellow and a luminescent variation from deep red to green were evident to the naked eye. The blue-shift of the absorption spectrum and enhancement of the phosphorescence emission upon the addition of Hcy can be attributed to the formation of a thiazinane group by selective reaction of the aldehyde group of Ir(pba)(2)(acac) with Hcy, which was confirmed by H-1 NMR studies. Importantly, lr(pba)(2)(acac) shows uniquely luminescent recognition of Hcy over other amino acids (including cysteine) and thiol-related peptides (reduced glutathione), in agreement with the higher luminescent quantum yield of the adduct of Ir(pba)(2)(acac) with Hcy (0.038) compared with that of the adduct with Cys (-0.002). Both surface charge analysis and the electrochemical measurement indicated that a photoincluced electron-transfer process for lr(pba)(2)(acac)-Cys might be responsible for the high specificity of lr(pba)(2)(acac) toward Hcy over Cys.

Abstract:By using histamine as the template molecule, methacrylic acid as the functional monomer, ethylene glycol dimethacrylate as the crosslinking agent, and zinc (II)-protoporphyrin as the functional monomer, a new kind of molecularly imprinted polymer which may emit fluorescence was synthesized. Its binding property was evaluated with a batch adsorption and a column method. The results of both methods indicate that the recognition of histamine with the MIP is superior. to that of the NMIP.

Abstract:The design and development of new pyrene-based fluorescent probes, P-Hcy-1 and P-Hcy-2, which display selective fluorescence enhancements in response to homocysteine (Hcy), are described. The distinctly different fluorescence responses of P-Hcy-1 and P-Hcy-2 to Hcy vs. Cys are explained by theoretical calculations. Finally, the results of cell experiments show that these probes can be used to selectively detect Hcy in mammalian cells.

Abstract:We present a highly sensitive and selective fluorescence "turn‐on" sensor for L‐histidine (His) detection in aqueous solutions utilizing a 1‐Cu2+ complex. This sensing platform employs a fluorescence‐based ligand displacement approach, featuring a meso‐(N'‐acetyl‐hydrizide)‐based BODIPY derivative (1) complexed with Cu2+. Initially highly fluorescent, the emission of 1 is selectively quenched by Cu2+ ions, forming the 1‐Cu2+ complex. The high affinity between His and Cu2+ effectively displaces 1 from the complex, restoring fluorescence. The system exhibits rapid response (within 5 minutes), excellent sensitivity (detection limit of 78 nM), operational simplicity, and a large fluorescence "turn‐on" signal. It demonstrates remarkable selectivity for His over other amino acids, with maleimide masking cysteine interference. Notably stable in complex biological matrices, the sensor has successfully quantified His in artificial urine samples. Its practical applicability extends to paper‐based test strips, offering portability and potential for real‐time His monitoring in clinical diagnostics and biological systems.

Abstract:Hydrogen sulfide (H2S) as small molecular signal messenger plays key functions in numerous biological processes. The imaging detection of intracellular hydrogen sulfide is of great significance. In this work, a ratiometric fluorescent probe BH based on an asymmetric BODIPY dye for detection of H2S was designed and synthesized. After the interaction with hydrogen sulfide, probe display colorimetric and ratiometric fluorescence response, with its maximum emission fluorescence wavelength red-shifted from 542 nm to 594 nm, which is attributed to the sequential nucleophilic reaction of H2S leading to enhanced molecular conjugation after ring formation of the BODIPY skeleton. A special response mechanism has been fully investigated by NMR titration and MS, so that the probe has excellent detection selectivity. Furthermore, probe BH has low cytotoxicity and fluorescence imaging experiments indicate that it can be used to monitor hydrogen sulfide in living cells.

Abstract:The chiral Schiff base N-(2-hydroxy-1-naphthyl)methylidene-(S)-α-phenylethylamine (1) was designed and synthesized as a fluorescent chemosensor based on the photoinduced electron transfer (PET) and CN isomerization mechanisms. The receptor exhibited a highly selective switch-on fluorescence response toward Zn2+ and especially avoided the interference of Cd2+ when the detection was conducted in unbuffered methanol solution. Furthermore, the in situ generated 1·ZnII-complex could be employed for the specific recognition of H2PO4− by fluorescence turn-off signaling through a ligand displacement process.

Abstract:A molecular chemodosimeter PHSN, with a naphthalimide fluorophore connected to a newly designed phenazine energy acceptor, for the selective detection of cysteine was effectively synthesized. PHSN featured efficient intramolecular fluorescence resonance energy transfer (FRET) based on spectral overlap between the emission of naphthalimide and the absorption of phenazine. A cystamine chain with a S-S bond was applied to play the role of recognition moiety and the linker part. The specific reaction between the biological thiols and PHSN gave rise to an obvious fluorescence intensity enhancement at 540 nm, which is induced by cleavage of the disulfide bond followed by elimination of the FRET effect. High sensitivity and selectivity for cysteine over homocysteine and glutathione were also achieved. In addition, upon excitation at 400 nm, a relatively weak NIR emission provided an internal standard making PHSN a promising ratiometric sensor for cellular detection of cysteine.

Abstract:A highly selective colorimetric and fluorescence enhanced probe S1 (M2@Cu) for histidine and histidine-rich proteins has been developed. In neutral aqueous ethanol solution, probe S1 can selectively detect histidine out of twenty DNA encoded amino acids by showing a color change from brownish red to light green, and with a fluorescence enhancement up to 99-fold at 537 nm, simultaneously.

Abstract:A new 4-hydroxy-1,8-naphthalimide-based ratiometric fluorescent probe for hydrogen sulfide (H2S) detection was designed and synthesized. The probes showed fast response, high sensitivity and selectivity for hydrogen sulfide over other reactive sulfur species. The mechanism is based on H2S-mediated nucleophilic addition followed by an intramolecular cyclization to give the product, 4-hydroxy-1,8naphthalimide, which showed outstanding intramolecular charge transfer (ICT). Therefore, ratiometric signal was observed and with a H2S detection of 1-10 mu M. Abstract:We designed and synthesized a series of BODIPY based probes with fast and distinct ratiometric responsiveness for discriminative detection of GSH from Cys and Hcy. The discriminative detection is based on the different products obtained by the SNAr between probes and thiol-containing amino acids. The amino group of the obtained thioether from the reaction with Cys or Hcy but not GSH would trigger an intramolecular nucleophilic substitution through five- or six-membered cyclic transition state, finally yielding an amino substituted derivative. To achieve highly discriminative detection and fast response, a series of structure modifications and improvements have been made by elongating the pi-conjugation and introducing electron withdrawing groups, finally affording probe BOD-DBNPF with optimized responsiveness and selectivity. Importantly, BOD-DBNPF was successfully used for the selective detection of GSH from Cys with distinct fluorescent ratiometric responses in living HeLa cells. (C) 2019 Elsevier Ltd.
