Abstract:A review of p-terephthalic acid (PTA) process development carried out in Zhejiang University during recent years was made. The R & D works cover oxidation and crystallization kinetics, gas-liquid mass transfer, reactor modeling, database development, novel reactor design, process modeling, simulation and optimization. A viewpoint was emphasized through this case study, which chemical reaction engineering should be developed to process system engineering to extend its scope, and particular attention should be paid on reactor and process unit modeling.
Multivariate Statistical Process Monitoring and Control: Recent Developments and Applications to Chemical Industry

Abstract:Multivariate statistical process monitoring and control (MSPMC) methods for chemical process monitoring with statistical projection techniques such as principal component analysis (PCA) and partial least squares (PLS) are surveyed in this paper. The four-step procedure of performing MSPMC for chemical process, modeling of processes, detecting abnormal events or faults, identifying the variable(s) responsible for the faults and diagnosing the source cause for the abnormal behavior, is analyzed. Several main research directions of MSPMC reported in the literature are discussed, such as multi-way principal component analysis (MPCA) for batch process, statistical monitoring and control for nonlinear process, dynamic PCA and dynamic PLS, and on-line quality control by inferential models. Industrial applications of MSPMC to several typical chemical processes, such as chemical reactor, distillation column, polymerization process, petroleum refinery units, are summarized. Finally, some concluding remarks and future considerations are made.

Abstract:Process design and control are closely related to each other in chemical engineering activities. Traditionally, process design and control system design are carried out in sequence. However, the integration of process design and control (IPDC) can bring greater economic benefits and process dynamic performance than traditional sequential design methods. This is true, particularly for modern chemical processes, in which various process units become more interacting and compact owing to the widespread use of heat integration and recycled streams, and the resulted impacts between process design and control begin to significantly influence both the capital and operational costs. Recently, considerable studies about the IPDC for chemical processes have been reported in published literature. The purpose of the paper is to survey the applications of optimization-based integrating process design and control for chemical processes. Firstly, attention has been focused on the applications of IPDC to different process units, for example, chemical reactors and separation columns. Then, the survey is extended to the applications of IPDC to plant-wide chemical processes. Finally, the future research challenges in the application of IPDC to chemical processes have been briefly discussed.

Abstract:A review of progress in PTA process RD in China was made in this paper. It summarized the works contributed by Zhejiang University group. The works cover chemical engineering aspects: reaction kinetics, flow and transport phenomena, reactor modeling, the database development, the novel reactor and novel process flowsheet design. It was shown that the domestic development of PTA process now is proceeding to the stage of industrialization. Both market and technique environment are best at the moment, and we must do our best to promote industrial application of the novel domestic techniques.

Abstract:A light-addressable potentiometric sensor (LAPS) is a semiconductor electrochemical sensor based on the field-effect which detects the variation of the Nernst potential on the sensor surface, and the measurement area is defined by illumination. Thanks to its light-addressability feature, an LAPS-based chemical imaging sensor system can be developed, which can visualize the two-dimensional distribution of chemical species on the sensor surface. This sensor system has been used for the analysis of reactions and diffusions in various biochemical samples. In this review, the LAPS system set-up, including the sensor construction, sensing and substrate materials, modulated light and various measurement modes of the sensor systems are described. The recently developed technologies and the affecting factors, especially regarding the spatial resolution and temporal resolution are discussed and summarized, and the advantages and limitations of these technologies are illustrated. Finally, the further applications of LAPS-based chemical imaging sensors are discussed, where the combination with microfluidic devices is promising.

Abstract:Multivariate statistical process control (MSPC) has been successfully applied to performance monitoring and fault diagnosis for chemical processes. However, classical methods of MSPC are based on the premise that the separated latent variable must be subjected to normal distribution, which sometimes can't be satisfied. In this paper, a new method based on independent component analysis OCA) whose goal is to find a line representation of nomgaussian data to depict the chemical process and improve the monitoring performance of the system is presented. Due to the uncertainty of the probability distribution of the independent component, the paper devises a kind of classifier with Parzen density estimation for classifjring the normal data and fault data. Then the nonisothemal CSTR is monitored and diagnosed by the present method, the simulation result verifies the effectiveness of ICA-based monitoring method.

Abstract:Principal component analysis (PCA) can be effectively used to eliminate system noise and correlation between process variables but to reserve enough original data information. Based on principal component model, performance monitoring and analysis was carried out on control system with multivariate statistical index, such as Q residuals, Hotelling T2 and principal scores. This method with multivariable statistical technologies gives explicit knowledge about control system: whether the system is in-control or out-control, how disturbance and system variables influence the performance and how the control strategy work. Abstract:The concept of quality by design (QbD) is widely applied in the process development of pharmaceuticals. However, the additional cost and time have caused some resistance about QbD implementation. To show a possible solution, this work proposed a rapid process development method, which used direct analysis in real time mass spectrometry (DART-MS) as a process analytical technology (PAT) tool for studying the chromatographic process of Ginkgo biloba L., as an example. The breakthrough curves were fast determined by DART-MS at-line. A high correlation coefficient of 0.9520 was found between the concentrations of ginkgolide A determined by DART-MS and HPLC. Based on the PAT tool, the impacts of process parameters on the adsorption capacity were discovered rapidly, which showed a decreased adsorption capacity with the increase of the flow rate. This work has shown the feasibility and advantages of integrating PAT into QbD implementation for rapid process development.

Abstract:This review highlights the advantages of incorporating Process Analytical Technologies into continuous flow processes, especially in the context of telescoped multistep flow sequences. Use of FlowIR, in-line UV-Vis, online HPLC, online MS and Flow NMR are discussed within this review, with multiple PAT techniques used in conjunction with one another in some instances. PAT is key in ensuring as much information as possible can be gathered during a chemical transformation, with real-time analysis allowing for rapid reaction optimization.

Abstract:A promising process analytical technology (PAT) tool has been introduced for batch processes monitoring. Direct analysis in real time mass spectrometry (DART-MS), a means of rapid fingerprint analysis, was applied to a percolation process with multi-constituent substances for an anti-cancer botanical preparation. Fifteen batches were carried out, including ten normal operations and five abnormal batches with artificial variations. The obtained multivariate data were analyzed by a multi-way partial least squares (MPLS) model. Control trajectories were derived from eight normal batches, and the qualification was tested by R(2) and Q(2). Accuracy and diagnosis capability of the batch model were then validated by the remaining batches. Assisted with high performance liquid chromatography (HPLC) determination, process faults were explained by corresponding variable contributions. Furthermore, a batch level model was developed to compare and assess the model performance. The present study has demonstrated that DART-MS is very promising in process monitoring in botanical manufacturing. Compared with general PAT tools, DART-MS offers a particular account on effective compositions and can be potentially used to improve batch quality and process consistency of samples in complex matrices.

Abstract:Process analytical technology (PAT) has been defined as a mechanism to design, analyze and control pharmaceutical manufacturing processes through measurement of critical process parameters which affect critical quality attributes. PAT checks the quality of raw material attributes both physically and chemically (i.e. at off-line, on-line, in-line). PAT involves a shift from testing the quality of building to the quality of products by testing at several intermediate steps. PAT saves a huge amount of time and money required for sampling and analysis of products. The main goal of PAT is to provide successful tools such as multivariate data analysis and acquisition tools, modern process analyzers or analytical chemistry, endpoint process monitoring, controlling tools and continuous improvement and knowledge improvement tools. In this review attempt has been carried out to explore the concept of PAT, different tools of PAT, goals of PAT, How it Works and Its benefits. Abstract:The field of Process system engineering (PSE) is constantly developing with the availability of advanced computational resources. Since 1960s, process simulation has been one of the most successful story in using computer aided methods and tools in the field of chemical engineering. Nowadays, several software tools are available for process simulation, process optimization, process synthesis and design. However, the current software in PSE is mainly focussed on processes without consideration of controller design or validation. The gap between process design and controller design seems to be increasing, which may lead to suboptimal solutions especially for closed-loop systems. Therefore, there is always a demand for integrating the process design with consideration of the controller design; even though the development of such software tool has not been well addressed so far, or are industry specific. In this paper, a model-based integrated process and controller design software tool called ProCACD (computer aided controller design) is presented along with a case study to design controllers and optimize controller parameters for a system of three-tanks-in-series with recycle.

Abstract:A number of powerful tools have been developed for process simulation, optimization, and design. However, the current software in Process Systems Engineering is mainly focused on processes without consideration of controller design or validation. Therefore, there is always a demand for integrated platforms for solving various process design and control problems. In this paper, an integrated software, ProCACD, developed for solution and analysis of control related problems is presented. Embedded within the software architecture are process design methods, analysis methods, integrated tools, and a library of process models that allow users to study a wide range of problems. With ProCACD, users can solve problems involving generation of process models, steady-state and/or dynamic simulations, analysis of process dynamics in open-loop and/or closed-loop, design of the controller structure, and solving integrated process-control design problems. Abstract:<p>Real time monitoring of bioprocesses by integration of analytics at critical unit operations is one of the paramount necessities for quality by design manufacturing and real time release of biopharmaceuticals. A well‐defined process analytical technology (PAT) roadmap enables the monitoring of critical process parameters and quality attributes at appropriate unit operations to develop an analytical paradigm which is capable of providing real time data. We believe a comprehensive PAT roadmap should entail not only integration of analytical tools into the bioprocess but also should address automated‐data piping, analysis, aggregation, visualization and smart utility of data for advanced‐data analytics such as machine and deep learning for holistic process understanding. In this review, we discuss a broad spectrum of PAT technologies spanning from vibrational spectroscopy, multivariate data analysis, multi‐attribute chromatography, mass spectrometry, sensors and automated‐sampling technologies. We also provide insights, based on our experience in clinical and commercial manufacturing, into data automation, data visualization and smart utility of data for advanced‐analytics in PAT. This review is catered for a broad audience, including those new to the field to those well versed in applying these technologies. The article is also intended to give some insight into the strategies we have undertaken to implement PAT tools in biologics process development with the vision of realizing real time release testing (RTR) in biomanufacturing and to meet regulatory expectations.</p><p>This article is protected by copyright. Abstract:Owing to the advancement in pharmaceutical technology, traditional Chinese medicine industry has seen rapid development. Preferring conventional manufacturing mode, pharmaceutical enterprises of traditional Chinese medicine have no effective process detection tools and process control methods. As a result, the quality of the final products mainly depends on testing and the quality is inconsistent in the same batch. Process analytical technology(PAT) for traditional Chinese medicine manufacturing, as one of the key advanced manufacturing techniques, can break through the bottleneck in quality control of medicine manufacturing, thus improving the production efficiency and product quality and reducing the material and energy consumption. It is applicable to the process control and real-time release of advanced manufacturing modes such as intelligent manufacturing and continuous manufacturing. This paper summarized the general idea of PAT for traditional Chinese medicine manufacturing. Through the analysis of the characteristics and status quo of the technology, we summed up the methodology for the continuous application and improvement of PAT during the whole life-cycle of traditional Chinese medicine. The five key procedures(process understanding, process detection, process modeling, process control, and continuous improvement) were summarized, and the application was reviewed. Finally, we proposed suggestions for the technical and regulatory challenges in implementing PAT in traditional Chinese medicine industry. This paper aims to provide a reference for development and application of PAT in advanced manufacturing, intelligent manufacturing, and continuous manufacturing of traditional Chinese medicine industry.

Abstract:Background: Pharmaceutical industry is witnessing increased pressure to introduce innovative and efficient processes for manufacturing Active Pharmaceutical Ingredients (APIs) in order to be competitive as well as to meet the stringent product quality requirements set by regulatory authorities. Crystallization with its ability to engineer the final product to the desired qualities such as purity, polymorphic form, particle size and shape is one of the most important steps involved in the manufacturing of APIs. Therefore, development of crystallization processes with better understanding of process parameters and their impact on quality of APIs and subsequently the drug products assume great significance for the pharmaceutical industry. Methods: This review paper focuses on the application of PAT tools, an integral part of Quality by Design (QbD) approach, for better understanding, control, and design of crystallization processes in the manufacturing of APIs. Results: Firstly, various steps involved in the drug development process are introduced briefly with emphasis on crystallization as one of the most important steps in manufacturing of drug products. Secondly, Critical Quality Attributes (CQAs) of drug products, their dependence on material attributes of APIs and role of crystallization in manipulating material attributes of APIs has been discussed. Finally, application of PAT tools such as advanced process analyzers for continuous monitoring, chemometric methods for multivariate data analysis, and control strategy for APIs crystallization processes has been reviewed along with some examples. Conclusion: Application of PAT in crystallization of APIs facilitates development of robust processes that works within the design space to produce the drug products of consistent quality. Furthermore, it opens up the opportunities for continuous improvement of the process by generating knowledge base of existing processes.

Abstract:Sensitive principal component analysis (SPCA) is proposed to improve the principal component analysis (PCA) based chemical process monitoring performance, by solving the information loss problem and reducing nondetection rates of the T-2 statistic. Generally, principal components (PCs) selection in the PCA-based process monitoring is subjective, which can lead to information loss and poor monitoring performance. The SPCA method is to subsequently build a conventional PCA model based on normal samples, index PCs which reflect the dominant variation of abnormal observations, and use these sensitive PCs (SPCs) to monitor the process. Moreover, a novel fault diagnosis approach based on SPCA is also proposed due to SPCs' ability to represent the main characteristic of the fault. The case studies on the Tennessee Eastman process demonstrate the effect of SPCA on online monitoring, showing its performance is significantly better than that of the classical PCA methods.

Abstract:The pharmaceutical industry is now paying increased attention to continuous manufacturing. While the revolution to continuous and automated manufacturing is deepening in most of the top pharma companies in the world, the advancement of automated pharmaceutical continuous manufacturing in China is relatively slow due to some key challenges including the lack of knowledge on the related technologies and shortage of qualified personnels. In this review, emphasis is given to two of the crucial technologies in automated pharmaceutical continuous manufacturing, i.e., process analytical technology (PAT) and self-optimizing algorithm. Research work published in recent 5 years employing advanced PAT tools and self-optimization algorithms is introduced, which represents the great progress that has been made in automated pharmaceutical continuous manufacturing.
