Relationship between survival effect of caffeine and levels of intercellular calcium in cerebellar granule neurons

Abstract:To study the relationship between the protective effect of caffeine and intercellular calcium concentration ([Ca 2+](i)) in cultured cerebellar granule neurons of rats, agarose gel eletrophoresis and fura-2 were used to measure [Ca 2+](i). Cultured cerebellar granule neurons died by apoptosis when transferred from a medium containing 25 mmol·L -1 KCl to one with 5 mmol·L -1 KCl. Death resulting from low [K +], was prevented by caffeine (5-20 mmol·L -1) in a concentration-dependent manner. Inhibiting ryanodine-sensitive Ca 2+ release by ryanodine and dantrolene did not affect the survival action, neither did L-type Ca 2+ channel blockers (nifedipine, nimodipine, verapamil) nor NMDA receptor blocker dizocilpine (MK801). The results indicate that increasing [Ca 2+](i) is not necessary for the protective action of caffeine on cultured cerebellar granule cells.
