Establishment of the model of KBv200 nude mice xenograft and studies on its characterization of multidrug resistance

Abstract:AIM Multidrug resistance (MDR) is a main obstacle for cancer chemotherapy. A promising method for circumventing MDR is to find the modulators with potent MDR reversing activity and low toxicity. It is important to establish a model of the nude mice xenograft with human MDR tumor cells for MDR study in vivo . The purpose of this research is to determine whether the MDR characterization persisted in KBv200 cells in vivo . METHODS KBv200 cells were grown as subcutaneous xenografts in athymic nude mice. The comparisons of the possibility of resistant difference between KBv200 (KBv200.MDR) and nude mice xenografts (KBv200/nude.MDR) were investigated. Cytotoxicity was determined with tetrazolium (MTT) assay. Pgp was examined by flow cytometry. RESULTS Tumor incidence was 100% at 5 days after implantation subcutaneous by 10 7 of cells. IC 50 of vincristine (VCR) to KB (WT) and KBv200 (MDR), KB/nude, KBv200/nude(MDR) were 0 018, 1 479, 0 018, 1 472 μmol·L -1 , respectively. The expressions of Pgp in KBv200 (MDR) ware similar to those in KBv200 /nude(MDR). CONCLUSION Like the MDR cells in culture, KBv200 nude xenografts are extremely resistant to VCR and retain the characteristics of MDR phenotype.
