Abstract:Oxidative stress plays an important role in the pathological process of neurodegenerative diseases. The effects of four major flavonoids present in Scutellaria baicalensis Georgi on hydrogen peroxide-induced neuronal cell damage are studied in this paper. When human neuroblastoma SH-SY5Y cells were incubated in Hanks’ solution with the addition of 400 μM hydrogen peroxide for 2 h, the viability of cells was decreased remarkably, while the cell lipid peroxidation and the percentage of lactose dehydrogenase released into the culture medium was significantly increased. Addition of 10 μM of baicalein and baicalin significantly attenuated the cellular injury induced by hydrogen peroxide, while the effect of wogonin was marginal and wogonoside showed no effect at the tested concentration. In a separate experiment, 10μM of baicalein and baicalin also antagonized the intracellular free-calcium concentration ( [Ca2+ ]i) increase caused by 1 mM hydrogen peroxide. The effects of baicalein in both experiments were similar to those of quercetin, a well-studied antioxidant flavonoid. These results demonstrated the protective effects of flavonoids originating from Scutellaria baicalensis Georgi on the oxidative injury of neuronal cells.
Protective effects of flavonoids extracted from Scutellaria baicalensis Georgi on free radical induced rat cortex mitochondria injury

Abstract:AIM: To evaluate the protective effects of four main flavonoids in Scutellaria baicalensis Georgi on oxidative injury of rat cortex mitochondria. METHODS: Lipid peroxidation was induced by three different system, e.g. ascorbic acid-Fe2+, 2, 2-azobis (2-amidinopropane) hydrochloride and NADPH; the swelling of mitochondria and the decreasing of membrane fluidity were induced by ascorbic acid-Fe2+ system. RESULTS: Baicalein, baicalin, wogonin and wogonoside at 10 μmol · L-1 can protect mitochondria from NADPH induced lipid peroxidation, while only baicalein and baicalin showed remarkable effects on ascorbic acid-Fe2+ and AAPH induced lipid peroxidation. The swelling of mitochondria and the decreasing of membrane fluidity induced by ascorbic acid-Fe2+ system can be retarded more or less by these four flavoids. CONCLUSION: Flavonoids in Scutellaria baicalensis Georgi may protect mitochondria from oxidative injury induced in different ways. Baicalein is the most effective flavonoids among the tested agents in different systems.

Abstract:The protective effect of flavonoids extracted from Scutellaria baicalensis Georgi on cerebral ischemia injury has been explored in experimental animals. Scutellaria flavonoid (SF) could significantly prolong gasping time (prolonged ratio, 23.79%) and survival time after carotid artery occlusion, and decrease attenuate malondialdehyde (MDA) content in damaged brain tissues from 118.56+/-47.95 nmol/g in untreated to 199.29+/-24.24 nmol/g. SF could also increase the content of superoxide dismutase (SOD) in brain tissues after ischemic mice from 1486+/-94 NU/g in untreated to 1168+/-76 NU/g, and showed significant protective effect on cerebral hypoxia and reperfusion brain tissues in middle cerebral artery occlusion (MCAO) procedure. Additionally, SF has inhibitory effect on platelet aggregation, with the average inhibition rate of 45.52%, while the aspirin group was 54.96%. These results suggest that SF has a significant protective effect on cerebral ischemia and ischemia-reperfusion induced brain injury.
Protective Effects of the Aqueous Extract Ofscutellaria Baicalensisagainst Acrolein-Induced Oxidative Stress in Cultured Human Umbilical Vein Endothelial Cells

Abstract:Objective: aEuro,This study was designed to determine the protective effects of the extract of SbG against the acrolein-induced oxidative stress in cultured human umbilical vein endothelial cells (HUVEC).Materials and methods: aEuro,The MTT reduction assay was employed to determine cell viability. The total cellular glutathione (GSH) level was detected using a colorimetric GSH assay kit. Cellular GSH production was conducted by detecting the mRNA expression levels of gamma gamma-glutamylcysteine ligase catalytic subunit and modifier subunit.Results: aEuro,Concentration-dependent cytotoxic effects of acrolein were observed while SbG could effectively protect the acrolein-induced oxidative damage. The protective mechanism was investigated, showing that the increased GSH content in the SbG-incubated HUVE cells was associated with the protective effects of SbG-treated cells. Further RT-PCR data confirmed the elevated mRNA expressions of GSH synthesis enzymes.Discussion and conclusion: aEuro,The current study strongly indicated that SbG could be a potential antioxidant against oxidative stress in treating cardiovascular diseases.

Abstract:The progress in the research of antioxidative mechanism of flavonoids extracted from Scutellaria baicalensis Georgi was summarized,which provides the theory basis to development new antioxidative traditional Chinese drugs.It was demonstrated by pharmacological experiments that Scutellaria baicalensis Georgi and its extracts exhibited significant antioxidative and free-radical scavenging activities in tissue injury-including cardiomyocytes injury,liver injury,brain injury and neurocytes injury.Scutellaria baicalensis will be exploited to new antioxidative traditional Chinese drugs,however,the detailed antioxidative mechanism in vivo would study in future.

Abstract:Flavonoids from the stems and leaves of Scutellaria baicalensis Georgi, an antioxidant, markedly improve memory impairments and neuronal injuries. In the present study, primary cortical neurons of rats were exposed to potassium cyanide to establish a model of in vitro neural cell apoptosis. Inhibition of apoptosis by flavonoids from the stems and leaves of Scutellaria baicalensis Georgi at concentrations of 18.98, 37.36, and 75.92 μg/mL was detected using this model. These flavonoids dramatically increased cell survival, inhibited cell apoptosis and excessive production of malondialdehyde, and increased the activities of superoxide dismutase, glutathione peroxidase, and Na(+)-K(+)-ATPase in primary cortical neurons exposed to potassium cyanide. The flavonoids from the stems and leaves of Scutellaria baicalensis Georgi were originally found to have a polyhydric structure and to protect against cerebral hypoxia in in vitro and in vivo models, including hypoxia induced by potassium cyanide or cerebral ischemia. The present study suggests that flavonoids from the stems and leaves of Scutellaria baicalensis Georgi exert neuroprotective effects via modulation of oxidative stress, such as malondialdehyde, superoxide dismutase, glutathione peroxidase and Na(+)-K(+)-ATPase disorders induced by potassium cyanide.

Abstract:OBJECTIVE:To study the protective effects of flavonoids from Cuscuta chinensis (CF) on oxidative stress in cultured PC12 cells and investigate the mechanism of the effects.METHODS:The cell viability was analyzed by MTT method and the radical scavenging activity of CF was examined by DPPH (1, 1-diphenyl-2-picrylhydrazyl). The morphological changes were observed by Hoechst 33258 staining assay, and the apoptosis rate of PC12 cells was detected by propidium iodide stain flow cytometry (FCM).RESULTS:Application with 0.3-0.5 mM H2O2 induced a dose and time dependent viability loss in PC12 cells; Treatment with 0.5 mM H2O2 for 24 h was shown to cause nearly 50% viabliity loss and apoptosis in PC12 cells; Pretreatment with different concentrations of CF for 0.5 h increased the survival rate of PC12 cells, inhibited apoptosis induced by H2O2; CF had the activity of scavenging free radicals generated by DPPH in a dose-dependent manner.CONCLUSION:CF can protect PC12 cells against oxidative stress. The mechanism of it may be the ability of scavenging ROS and increasing the activity of antioxidant enzyme.

Abstract:Purpose To investigate the neuroprotective effect and its mechanism of total flavonoids of Hippophae Rhamnoide(TFH).Methods Models of neuronal impairment by PC12 cells with NaCN,glucose deprivation,Na2S2O4 and KCl were used to induce hypoxia and Ca2+ overload injury and to assay the neuroprotective effect of the drug in cultured PC12 cells.Results The cells were protected by using TFH through MTT activity in a dose-dependent manner.Conclusion TFH has protection effects on neurons.
Effects of Flavonoids Extracted from Scutellaria Baicalensis Georgi on Hemin–nitrite–h2o2 Induced Liver Injury

Abstract:Hemin–nitrite–H2O2 system may play a role in liver oxidative injury in some pathological events. In this paper, the effects of the three active components of the root of Scutellaria baicalensis Georgi, i.e. baicalin, baicalein and wogonin, on hemin–nitrite–H2O2 induced liver injury were studied in liver homogenate, liver microsome and human hepatoblastoma cell line HepG2 cells. It was found that hemin–nitrite–H2O2 could induce liver homogenate protein nitration, lipid peroxidation and liver microsome protein oxidation; it also caused a decrease of HepG2 cells viability. Baicalein, baicalin and wogonin could inhibit protein nitration and lipid peroxidation in liver homogenate as well as in HepG2 cells in a dose-dependent manner, the inhibition order was baicalein>baicalin≫wogonin. These three flavonoids also inhibited the oxidation of protein in liver microsome, the decrease of cell viability and the content of GSH in HepG2 cells, among which baicalin represented the most inhibitory effect. Besides, hemin–H2O2 induced cell injury could be augmented with the existence of nitrite, indicating protein nitration involved in hemin–nitrite–H2O2 induced liver injury. These results demonstrated hemin–nitrite–H2O2 could induce liver injury through oxidizing or nitrating different biomolecules. Baicalein, baicalin and wogonin could inhibit hemin–nitrite–H2O2 induced liver injury in dose-dependent manners by inhibiting oxidation and nitration.

Abstract:We have investigated the protective effects of echinacoside (ECH), one of the phenylethanoid glycosides, on H2O2-induced cytotoxicity in the rat pheochromocytoma cell line (PC12 cells). Our data show that application of ECH to H2O2-injured PC12 cells (HIPCs) increased cell viability and decreased the apoptotic ratio. Flow cytometry (FCM) and laser scanning confocal microscopy (LSCM) analysis suggested that ECH exerted its inhibitory effects on the formation of reactive oxygen species (ROS) and the accumulation of intracellular free Ca2+ ([Ca2+]i). In addition, ECH elevated the mitochondrial membrane potential (MMP) in HIPCs. Furthermore, Western blot analysis revealed that ECH prevented an H2O2-induced increase of the Bax/Bcl-2 ratio by down-regulating Bax protein expression and upregulating Bcl-2 protein expression. In summary, ECH showed significant neuroprotective effects on HIPCs through the mitochondrial apoptotic pathway, and could be a potential candidate for intervention in neurodegenerative diseases such as Alzheimer's and Parkinson's disease.
Protective Effect of SSTF against H2O2 Induced Cardiomyocyte Injury in Rats

Abstract:Objective:To investigate the protective effects of scutellaria baicalensis stem-leaf tatal flavonoid(SSTF)on freshly isolate cardiomyocytes injured by hydrogen peroxide(H2O2).Methods:Neonatal rat cardiomyocytes were cultured in DEME medium and the cellular injury was induced by H2O2.The viability of cardiomyocytes,activity of dehydrogenase(LDH),content of malondialdehyde(MDA),activity of superoxide dimutase(SOD) in the medium were measured.The apopto-sis rate was detected.Results:The SSTF(50～200 mg·L-1) significantly increased the viability of cardiomyocytes(P < 0.01),inhibited activity of LDH(P < 0.01),decreased the content of MDA and increased the activity of SOD(P < 0.01).The apop-tosis rate of cardiomyocytes injured by H2O2 was significantly decreased by SSTF(P < 0.01).Conclusion:The SSTF can ease the injury of cardiomyocytes,and decrease the apoptosis of cardiomyocytes injured by H2O2.

Abstract:The present study investigated the protective actions of the antioxidant scutellarin against the cytotoxicity produced by exposure to H2O2 in PC12 cells. This was done by assaying for MTT (3,(4,5-dimethylthiazole-2-yl)2,5-diphenyl-tetrazolium bromide) reduction and lactate dehydrogenase (LDH) release. Reactive oxygen species (ROS) and Ca2+ in cells were evaluated by fluorescent microplate reader using DCFH and Fura 2-AM, respectively, as probes. Lipid peroxidation was quantified using thiobarbituric acid-reactive substances (TBARS). Mitochondrial membrane potential (MMP) was assessed by the retention of rhodamine123 (Rh123), a specific fluorescent cationic dye that is readily sequestered by active mitochondria, depending on their transmembrane potential. The DNA content and percentage of apoptosis were monitored with flow cytometry. Vitamin E, a potent antioxidant, was employed as a comparative agent. Preincubation of PC12 cells with scutellarin prevented cytotoxicity induced by H2O2. Intracellular accumulation of ROS, Ca2+ and products of lipid peroxidation, resulting from H2O2 were significantly reduced by scutellarin. Incubation of cells with H2O2 caused a marked decrease in MMP, which was significantly inhibited by scutellarin. PC12 cells treated with H2O2 underwent apoptotic death as determined by flow cytometric assay. The percentage of this H2O2-induced apoptosis in the cells was decreased in the presence of different concentrations of scutellarin. Scutellarin exhibited significantly higher potency compared to the antioxidant vitamin E. The present findings showed that scutellarin attenuated H2O2-induced cytotoxicity, intracellular accumulation of ROS and Ca2+, lipid peroxidation, and loss of MMP and DNA, which may represent the cellular mechanisms for its neuroprotective action.
NMDA Receptor-Mediated Neuroprotective Effect of the Scutellaria Baicalensis Georgi Extract on the Excitotoxic Neuronal Cell Death in Primary Rat Cortical Cell Cultures

Abstract:The objective of the current research work was to evaluate the neuroprotective effect of the ethanol extract of Scutellaria baicalensis (S.B.) on the excitotoxic neuronal cell death in primary rat cortical cell cultures. The inhibitory effects of the extract were qualitatively and quantitatively estimated by phase-contrast microscopy and lactate dehydrogenase (LDH) assays. The extract exhibited a potent and dose-dependent inhibition of the glutamate-induced excitotoxicity in the culture media. Further, using radioligand binding assays, it was observed that the inhibitory effect of the extract was more potent and selective for the N-methyl-D-aspartate (NMDA) receptor-mediated toxicity. The S.B. ethanol extract competed with [3H] MDL 105,519 for the specific binding to the NMDA receptor glycine site with 50% inhibition occurring at 35.1 μg/mL. Further, NMDA receptor inactivation by the S.B. ethanol extract was concluded from the decreasing binding capability of [3H]MK-801 in the presence of the extract. Thus, S.B. extract exhibited neuroprotection against excitotoxic cell death, and this neuroprotection was mediated through the inhibition of NMDA receptor function by interacting with the glycine binding site of the NMDA receptor. Phytochemical analysis of the bioactive extract revealed the presence of six phytochemical constituents including baicalein, baicalin, wogonin, wogonoside, scutellarin, and Oroxylin A.
Protective Effects of Red Clover Flavonoids Against H2O2-induced Injury in Vascular Endothelial Cells

Abstract:Aim To investigate the protective effects of red clover flavonoids against H2O2-induced injury in vascular endothelial cells and to probe into its possible mechanisms.Methods All the experiments were carried out with human umbilical vein vascular endothelial cells ECV-304 in vitro,which were divided into 6 groups: control,model,quercetin(30 mg·L-1) and red clover flavonoids(12.5,25,50 mg·L-1) preincubation.Cell viability was determined colorimetrically by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium(MTT) assay;Hoechst33342 and PI staining were used for the qualitative and quantitative analyses of the apoptotic endothelial cells.JC-1 staining was used for the detection of mitochondrial membrane potential of endothelial cells injured by H2O2.Then ROS production of endothelial cells in different groups was detected.The supernates and the cells were collected respectively after different treatments for measuring LDH and MDA levels,as well as SOD,CAT and GSH-Px with the corresponding detection kit according to the manufacturers instructions.Fluorescence spectroscopy was used to detect the activity of caspase-3.Results Red clover flavonoids restrained the apoptotic of endothelial cells;the activity of SOD,CAT and GSH-Px increased and caspase-3 decreased;the contents of LDH,MDA and ROS decreased and mitochondrial membrane potential increased.Conclusion Red clover flavonoids exert protective effects against H2O2-induced injury in endothelial cells,which may act through its anti-oxidative effects and the decrease of caspase-3 activity and the stabilization of mitochondrial membrane potential.

Abstract:Free radical scavenging and antioxidant activities of baicalein, baicalin, wogonin and wogonoside, the four major flavonoids in the radix of Scutellaria baicalensis Georgi, were examined in different systems. ESR results showed that baicalein and baicalin scavenged hydroxyl radical, DPPH radical and alkyl radical in a dose-dependent manner, while wogonin and wogonoside showed subtle or no effect on these radicals. Ten micromol/l of baicalein and baicalin effectively inhibited lipid peroxidation of rat brain cortex mitochondria induced by Fe(2+)-ascorbic acid, AAPH or NADPH, while wogonin and wogonoside showed significant effects only on NADPH-induced lipid peroxidation. In a study on cultured human neuroblastoma SH-SY5Y cells system, it was found that 10 micromol/l of baicalein and baicalin significantly protected cells against H(2)O(2)-induced injury. Baicalein was the most effective antioxidant among the four tested compounds in every system due to its o-tri-hydroxyl structure in the A ring. Compared with a well-known flavonoid, quercetin, the antioxidant activity of baicalein was lower in DPPH or AAPH system, but a little higher in those systems which might associate with iron ion. These results suggest that flavonoids in the radix of Scutellaria baicalensis with o-di-hydroxyl group in A the ring, such as baicalein and baicalin, could be good free radical scavengers and might be used to cure head injury associated with free radical assault.
Effects of Luteolin on Endothelial Cell Injury Induced by H2O2

Abstract:AIM:To study the effect of luteolin on endothelial cell injuries induced by H2O2.METHODS:Endothelial cells were cultured in vitro and divided into seven groups:control;solvent;H2O2;quercetin+H2O2;luteolin-L+H2O2;luteolin-M+H2O2;luteolin-H+H2O2 group,respectively.Endothelial cells were incubated with 750 μmol/L H2O2 for 18 h or 14 h in the absence or presence of various concentrations of luteolin and quercetin.The expression of caspase-3,the pro-apoptotic factors,was detected by immunohistochemical technique,and the apoptotic index was detected by flow cytometry.Meanwhile,the amount of malondialdehyde(MDA),nitric oxide(NO),lacate dehydrogenase(LDH) in serum,and cell viability were measured.RESULTS:Incubation of endothelial cells with H2O2 for 18 h elicited the increase in the extent of immunostaining for caspase-3,the rate of apoptosis,the release of LDH,and the number of dead cells accompanied by the decrease in the content of NO in the medium,which were inhibited by pretreatment of luteolin at concentrations of 0.5-50 μmol/L in a concentration-dependent manner(P0.01).CONCLUSION:Luteolin possesses a protective effect against endothelial cell injuries induced by H2O2,which may be related with anti-oxidation,increasing the concentration of NO and inhibiting the activation of caspase-3.
Antioxidant Effect of Scutellarin on PC12 Cell Injury Induced by Hydrogen Peroxide in Culture

Abstract:PurposeTo investigate the antioxidant effect of scutellarin on PC12 cell injury induced by hydrogen peroxide in culture.MethodsThe PC12 cells injury was induced by H_2O_2 in culture.Morphological changes of PC12 cells were visualized under light microscope.Lactate dehydrogenase(LDH),malondialdelyde(MDA) and superoxide dismutase(SOD) in the culture medium and MDA,SOD in the cell were measured by spectroscopy respectively.ResultsAfter being exposed to 100 μmol/L H_2O_2 for 4 hours,typical morphological characteristics of injury were observed.The level of LDH and MDA was increased significantly while SOD level was decreased significantly in H_2O_2 induced group compared with that in the sham group.Scutellarin can decrease LDH,MDA content and increase SOD activity significantly.ConclusionScutellarin has protective effect on PC12 cell injury induced by hydrogen peroxide in culture.The protective effect may be related to protecting cell membrane,to increasing the activity of clearance enzyme of free radical and to inhibiting lipid peroxidation.
Baicalin Ameliorates H2O2 Induced Cytotoxicity in HK-2 Cells Through the Inhibition of ER Stress and the Activation of Nrf2 Signaling.

Abstract:Renal ischemia-reperfusion injury plays a key role in renal transplantation and greatly affects the outcome of allograft. Our previous study proved that Baicalin, a flavonoid glycoside isolated from Scutellaria baicalensis, protects kidney from ischemia-reperfusion injury. This study aimed to study the underlying mechanism in vitro. Human renal proximal tubular epithelial cell line HK-2 cells were stimulated by H2O2 with and without Baicalin pretreatment. The cell viability, apoptosis and oxidative stress level were measured. The expression of endoplasmic reticulum (ER) stress hallmarks, such as binding immunoglobulin protein (BiP) and C/EBP homologous protein (CHOP), were analyzed by western blot and real-time PCR. NF-E2-related factor 2 (Nrf2) expression was also measured. In the H2O2 group, cell viability decreased and cell apoptosis increased. Reactive Oxygen Species (ROS) and Glutathione/Oxidized Glutathione (GSH/GSSG) analysis revealed increased oxidative stress. ER stress and Nrf2 signaling also increased. Baicalin pretreatment ameliorated H2O2-induced cytotoxicity, reduced oxidative stress and ER stress and further activated the anti-oxidative Nrf2 signaling pathway. The inducer of ER stress and the inhibitor of Nrf2 abrogated the protective effects, while the inhibitor of ER stress and the inducer of Nrf2 did not improve the outcome. This study revealed that Baicalin pretreatment serves a protective role against H2O2-induced cytotoxicity in HK-2 cells, where the inhibition of ER stress and the activation of downstream Nrf2 signaling are involved.
Effects of Flavonoids from Hypericum Attemuatum on Oxidative Stress Injury of PC12 Cell DNA

Abstract:Objective To observe the effect of flavonoids from hypericum attemuatum on relieving oxidative stress injury of PC12 cell DNA. Method PC12 cell was exposed to H2O2 in vitro to establish oxidative injury model. MTT staining was applied to analyze the survival rate of cell and DNA injury was detected by single cell gel electrophorisi. Results The cell survival rates of all the four flavonoids large-dose groups were higher than H2O2 group (P <0.01,P <0.05, quercetin>quercetin-3’ -O-L-rhamnoside>quercetin-4’ -O -β-D-glucoside>quercetin- 3’ -O- β-D-glucoside), while the incidence of comets was lower than H2O2 group (P <0.01). Conclusion All of these four kinds of flavonoid had protective effects on oxidative injury cell induced by H2O2.
Protective Effect of 18β‑glycyrrhetinic Acid Against H2O2‑induced Injury in Schwann Cells Based on Network Pharmacology and Experimental Validation

Abstract:The aim of the present study was to assess the protective effects of 18 beta-GA against hydrogen peroxide (H2O2)-induced injury. First, the SMILES annotation for 18 beta-GA was used to search PubChem and for reverse molecular docking in Swiss Target Prediction, the Similarity Ensemble Approach Search Server and the TargetNet database to obtain potential targets. Injury-related molecules were obtained from the GeneCards database and the predicted targets of 18 beta-GA for injury treatment were selected by Wayne diagram analysis. Subsequently, Kyoto Encyclopedia of Genes and Genomes analysis was performed by WebGestalt. The experimental cells were assorted into control, model, 10 mu M SB203580-treated, 5 mu M 18 beta-GA-treated and 10 mu M 18 beta-GA-treated groups. Hoechst 33258 staining was performed and intracellular reactive oxygen species (ROS) levels, cell apoptosis, Bcl-xl, Bcl-2, Bad, Bax, cleaved-caspase 3, cleaved-caspase 7, transient receptor potential ankyrin 1 (TRPA1) and transient receptor potential vanilloid 1 (TRPV1) levels, as well as p38 MAPK phosphorylation were measured. The 'Inflammatory mediator regulation of TRP channels' pathway was selected for experimental verification. The results indicated that 10 mu M 18 beta-GA significantly increased cell viability as compared with the H2O2-treated model group. As suggested by the difference in intracellular ROS fluorescence intensity, 18 beta-GA inhibited H2O2-induced ROS production in Schwann cells. Hoechst 33258 staining indicated that 18 beta-GA reversed chromatin condensation and the increase in apoptotic nuclei following H2O2 treatment. Furthermore, flow cytometry suggested that 18 beta-GA substantially inhibited H2O2-induced apoptosis. Pre-treatment with 18 beta-GA obviously reduced Bad, Bax, cleaved-caspase3, cleaved-caspase 7, TRPA1 and TRPV1 levels and p38 MAPK phosphorylation after H2O2 treatment and increased Bcl-2 and Bcl-xl levels. In conclusion, 18 beta-GA inhibited Schwann cell injury and apoptosis induced by H2O2 and may be a potential drug to prevent peripheral nerve injury.
