Abstract:Objective To analyze the single nucleotide polymorphisms(SNPs) in promoter region,5' untranslated region,exon 1 and haplotypes of mannose binding-lectin(MBL) gene in Han nationality and Hui etheic children in China.Methods Sixty nine Hui ethnic children from Ningxia Huizu Auto.Reg.and 105 Han children from Zhejiang Prov.were enrolled in the present study.Whole blood samples(1.5 ml) were collected into potassium-EDTA tubes.SNPs in promoter region,5' untranslated region and exon 1 of MBL gene were determined by sequence analysis using BigDye Mix 3730 genetic analyzer,and genetic analysis was performed using SHEsis software.Results The variant allele frequencies at-221 sites in Han and Hui objects were 0.091 and 0.123,respectively,with no difference between the two groups(χ2=0.684,P=0.408).No mutation was found at sites +223 and +239 in exon 1 of MBL gene in the study.The variant frequency at +230 site in Hui children(0.268) was significantly higher than that in Han objects(0.167,χ2=5.223,P=0.022).The most common haplotype was YA,and the frequencies of YA haplotype in Han and Hui ethnic were 0.770 and 0.669,respectively,with significant difference between the two groups(χ2=4.312,P=0.038).Conclusion The variant allele frequency at +230 sites in exon 1 in Hui ethnic children is higher than that in Han subjects.The most common haplotype is YA,and the frequency of YA haplotype is higher in Han children than that in Hui subjects.
Analysis on the Polymorphism in Exons 2,3,4 of the MICA Genes in Han Nationality Population from Anhui Province

Abstract:Objective In order to know genetic polymorphism in the exons 2 to 4 of the MICA gene in Han nationality population from Anhui province and play a basis on the study of pathogeny of diseases associated with MICA allele.Method DNA samples from 112 random healthy individuals,unrelated Han nationality population were genotyped by PCR-SSOP to analyse the frequencies of MICA alleles. Results MICA*008 was dominant in MICA allele,accounted for 69.8%,the others were MICA*010(16.96%)*002(13.84%)*009(12.05%) respectively. The haplotype analysis revealed that linkage disequilibrium were lied in MICA*008-B40,B07,B13,MICA*010-B46,B15,MICA*002-B35,B15 and MICA*009-B52.Conclusion The distribution of MICA alleles has basic characteristics in Han nationality population from Anhui province.

Abstract:Major histocompatibility complex (MHC) class I chain-related gene A (MICA) is located 46 kb centromeric to HLA-B and encodes a stress-inducible protein. MICA allelic variation is thought to be associated with disease susceptibility and immune response to transplants. This study was aimed to investigate the haplotypic diversity and linkage disequilibrium between human leukocyte antigen (HLA)-B and (GCT)(n) short tandem repeat in exon 5 of MICA gene (MICA-STR) in a southern Chinese Han population. Fifty-eight randomly selected nuclear families with 183 members including 85 unrelated parental samples were collected in Hunan province, southern China. HLA-B generic typing was performed by polymerase chain reaction-sequence-specific priming (PCR-SSP), and samples showing novel HLA-B-MICA-STR linkage were further typed for HLA-B allelic variation by high-resolution PCR-SSP. MICA-STR allelic variation and MICA gene deletion (MICA*Del) were detected by fluorescent PCR-size sequencing and PCR-SSP. Haplotype was determined through family segregation analysis. Statistical analysis was applied to the data of the 85 unrelated parental samples. Nineteen HLA-B specificities and seven MICA-STR allelic variants were observed in 85 unrelated parental samples, the most predominant of which were HLA-B*46, -B60, -B*13, and -B*15, and MICA*A5, MICA*A5.1 and MICA*A4, respectively. Genotype distributions of HLA-B, MICA-STR loci were consistent with Hardy-Weinberg proportions. The HLA-B-MICA-STR haplotypic phases of all 85 unrelated parental samples were unambiguously assigned, which contained 30 kinds of HLA-B, MICA-STR haplotypic combinations, nine of them have not been reported in the literature. Significant positive linkage disequilibria between certain HLA-B and MICA-STR alleles, including HLA-B*13 and MICA*A4, HLA-B*38 and MICA*A9, HLA-B*58 and MICA*A9, HLA-B*46 and MICA*A5, HLA-B*51 and MICA*A6, HLA-B*52 and MICA*A6, and HLA-B60 and MICA*A5.1, were observed. HLA-B*48 was linked to MICA*A5, MICA*A5.1 and MICA*Del. HLA-B*5801-MICA*A10 linkage was found in a family. Our data indicated a high degree of haplotypic diversity and strong linkage disequilibrium between MICA-STR and HLA-B in a southern Chinese Han population, the data will inform future studies on anthropology, donor-recipient HLA matching in clinical transplantation and HLA-linked disease association.
Studies on the Polymorphism of MICA Gene in Four Chinese Populations

Abstract:Objective To understand with greater clearness the genetic polymorphism of (GCT)n repeat of MHC class I chain-related gene A (MICA)in some Chinese populations and provide preliminary genetic evidence for the independent origin of Chinese Baima Tibetan (BMT). Methods ACD-blood or saliva specimens of 411 unrelated individuals from four Chinese populations were collected. A primer pair spanning exon 5 of MICA gene was used to amplify the GCT region. Alleles were detected by PCR and denaturing PAGE. Comparison of the allelic distributions among the four populations was carried out. Results Five previously reported alleles have been observed in all the four populations, but the allelic distributions are different from one another. The most frequent allele is the A5 in all the four populations (0.325 in BMT, 0.345 in Tibetans, 0.390 in Chengdu Hans and 0.319 in Qiangs). A5.1 allele is the second most frequent allele in Chengdu Hans (0.230) and in Qiangs (0.293), while the second most frequent alleles for BMT and Tibetans are A4 (0.254) and A9 (0.272) respectively. The distribution of alleles in BMT is significantly different from that in the other three populations. Conclusion Alleles of MICA gene exon 5 are conservative in all populations studied so far. The results suggest that genetically BMT might be an independent ethnic population.
MICA polymorphism in a northern Chinese Han population: The identification of a new MICA allele, MICA*059

Abstract:The major histocompatibility complex class I chain-related gene A (MICA) in humans, located 46 kb centromeric to human leukocyte antigen (HLA)-B, is highly polymorphic. In addition to its primary role in immune surveillance, recent data highlight the importance of MICA in organ transplantation and in susceptibility to some diseases. In this study, 104 healthy, unrelated Han subjects recruited from central Inner Mongolia Autonomous Region, northern China, were investigated by sequence-based typing and fragment analysis for MICA allelic variation, MICA-HLA-B linkage disequilibrium, and HLA-A-Cw-B-MICA haplotypic diversity. Nineteen MICA alleles were observed, the most frequent of which were MICA*00801, MICA*010, MICA*00201, MICA*00901, and MICA*045, with gene frequencies of 23.08%, 18.75%, 12.02%, 12.02%, and 8.17%, respectively. The peculiarity in HLA-B-MICA haplotypic configurations was also uncovered. In particular, there was a clear-cut dichotomy between MICA*00801 and MICA*045 in their linkage to members of HLA-B*13 lineage, which was frequently represented in this population. A new MICA allele, MICA*059, was identified, which appeared to be evolutionarily linked to MICA*045. Haplotype HLA-A*30-Cw*06-B*1302-MICA*00801, previously not reported in other populations, was found with a frequency of 8.65% in this population. Our results provide new data about MICA genetic polymorphism in Chinese Han populations, which will form the basis for future studies of the potential role of MICA in allogeneic organ transplantation and disease susceptibility in related ethnic groups.

Abstract:The MICA gene encodes nonclassical major histocompatibility complex class I molecules, centromeric to HLA-B and telomeric to HLA-DRB1. The MICA genes are polymorphic. The immune response against MICA may correlate with a decrease in graft survival after transplantation. However, data on the frequency of MICA polymorphisms in different populations are limited. In this study, we determined MICA allelic frequencies in a Han population living in Guangdong Province in south China. A total of 15 MICA alleles were identified using sequence-based typing. The most frequent allele was MICA*010 (22.22%), followed by MICA*002:01(18.56%), MICA*008:01(16.32%), and MICA*019(14.93%). The MICA null gene (MICA*Del) exhibited a frequency of 1.743% in this population. MICA and HLA, MICA-HLA-B, and MICA-HLA-A/HLA-B/HLA-DRB1 haplotype frequencies were estimated. The most common 2-, 3- and 4-locus haplotypes were HLA-B*40:01-MICA*008:01 (13.70%), HLA-A*11:01-B*40:01-MICA*008:01(8.25%), and HLA-A*33:03-B*58:01-DRB1*03:01-MICA*002:01(5.22%). A new MICA allele, MICA*061, was identified and appears to be evolutionarily related to MICA*012:01. This study provides high-resolution information on the distribution of haplotypes with MICA, HLA-A, HLA-B, and HLA-DRB1 in China. This information should help determine the mechanisms underlying diseases and allotransplant rejection associated with MICA polymorphisms in the southern Chinese Han population.

Abstract:In this study, polymorphisms of major histocompatibility complex class I chain-related genes A and B (MICA and MICB) and human leucocyte antigen (HLA)-B gene were investigated for 158 unrelated Chinese Mongolian subjects recruited from central Inner Mongolia Autonomous Region, northern China, by polymerase chain reaction-sequence-based typing (PCR-SBT) and cloning. Collectively, 79 alleles, including 20 MICA, 12 MICB and 47 HLA-B alleles, were identified. MICA*008:01 (21.2%), MICB*005:02 (48.1%) and HLA-B*51:01 (7.91%) were the most common alleles. Significant global linkage disequilibrium (LD) was detected between HLA-B and MICA, HLA-B and MICB, and MICA and MICB loci (all P < 0.000001). The most frequent haplotypes were HLA-B*51:01-MICA*009:01 (7.28%), HLA-B*58:01-MICB*008 (6.96%), MICA*010-MICB*005:02 (13.92%) and HLA-B*58:01-MICA*002:01-MICB*008 (6.96%). HLA-B-MICA haplotypes such as HLA-B*50:01-MICA*009:02 were associated with single MICB allele. Some HLA-B-MICA haplotypes were associated with multiple MICB alleles, including HLA-B*51:01-MICA*009:01. One novel MICB allele, MICB*031, was identified, which has possibly arisen from MICB*002:01 through single mutation event. We also confirmed the existence of a recently recognized MICA allele, MICA*073, whose ethnic origin has not been previously described. Genotype distributions at MICA, MICB and HLA-B were consistent with a neutrality model. Our results provide new insight into MIC genetic polymorphisms in Chinese ethnic groups. Findings shown here are important from an anthropologic perspective and will inform future studies of the potential role of MIC genes in allogeneic organ transplantation and HLA-linked disease association in populations of related ancestry.
Association of MICA-STR with leukemia in Hunan Han population

Abstract:Objective:To explore the association between the short tandem repeat polymorphism of exon 5 of MICA gene(MICA-STR)and leukemia in Hunan Han population.Methods:62 patients with leukemia and 112 randomly selected normal controls residing in Hunan province were analyzed for MICA-STR allelic variation and MICA gene deletion by fluorescent polymerase chain reaction-genescanning and polymerase chain reaction-sequence specific priming.Results:The overall comparison showed no significant difference of MICA-STR frequency between patient group and control group(P>0.05).Subsequent stratification revealed that MICA*A5 was present at significantly lower frequency in patient group with chronic granulocytic leukemia(CML)(RR=0.635,P=0.038 0);MICA*A4 at significantly lower frequency in patient group with lyphololastic leukemia(ALL)(RR=0.120,P=0.029 7);MICA*A5 at significantly higher frequency in patient group with acute non-lyphololastic leukemia(ANLL)(RR=2.229,P=0.021 8),all compared with control group.Conclusion:MICA-STR allelic variation is associated with leukemia in Hunan Han population;The data also suggest the heterogeneity of MICA-STR allele associated with different clinical types of leukemia.

Abstract:In this study, 201 healthy, unrelated Han subjects in Hunan province, southern China, were investigated by sequence-based typing (SBT) for the allelic variation of the human major histocompatibility complex (MHC) class I chain-related gene A (MICA). Nineteen MICA alleles were observed, among which MICA*008:01 predominated with gene frequency of 30.35%. There was significant linkage disequilibrium (LD) of MICA*012:01 with HLA-B*54 and HLA-B*55, which was not observed in a northern Chinese Han population. Haplotype HLA-A*11-C*07-B60-MICA*008:01 (9.16%) was highly specific to this southern Chinese Han population. The most common five-locus haplotype in this population was HLA-A*02-C*01-B*46-MICA*010-DRB1*09 (8.73%). A new MICA allele, MICA*060, was identified on an HLA-A*02-C*01-B*55:02-DRB1*14 haplotype through extended family analysis. MICA*060 has probably arisen from MICA*012:01. Another new MICA allele, MICA*062, was identified by screening 1432 subjects using polymerase chain reaction–sequence-specific priming technology. MICA*062 has probably derived from MICA*010. Of particular interest is that MICA*062 was carried on an HLA-C*08-B*48:01-DRB1*14 haplotypic segment, as HLA-B*48 has been consistently shown to be primarily linked to MICA gene deletion in east Asian populations. Our results provide new insight into MICA genetic polymorphism in human populations. The findings reported here are of importance for future studies on the potential role of MICA in allogeneic organ transplantation and disease association in populations of Chinese ancestry.

Abstract:Objective To study on the characterization of MICA/B genetic polymorphism in a northern Chinese Hunan Han population.Methods Ninty-five unrelated individuals were involved in this study and MICA/B genotypes were determined by two methods:PCR-sequence-specific primers (PCR-SSP) and PCR-sequence-based typing (PCR-SBT).Results In northern Hunan Han population,eleven MICA alleles were found,among which MICA * 010 (28.95％),MICA * 008 ∶ 01 (20.53％) and MICA * 002 ∶ 01 (15.79％) were the common alleles.Five MICA-STR(short tandem repeat) alleles were found,among which MICA * A5 (37.89％) and MICA * A5.1 (21.05％) predominated.In this population,ten MICB alleles were found.The common alleles were MICB * 005 ∶ 02/* 010 (58.42％),MICB * 002 ∶ 01 (10.00％),and MICB * 008 (7.89％).Two kinds of MICA-MICB haplotypes were MICA * 004-MICB * 004 ∶ 01 and MICA * 010-MICB * 005 ∶ 02/010 in significant linkage disequilibrium.This study also showed MICA/B gene with high polymorphism in different populations.Conclusion MICA/B alleles distribution in northern Hunan Han population with its unique characteristics.

Abstract:Objective To analyze the association between MHC class I chain-related genes A(MICA)polymorphisms and rheumatoid arthritis(RA)in ethnic groups of Qinghai Tu and Tibetan nationalities.Methods The florescence microsatellite typing,MICA*Del detection and HLA-B PCR/SSP typing method were used to analyze the association between MICA polymorphisms and RA in ethnic groups of Qinghai Tu and Tibetan nationalities.Results The results showed that the relatively higher gene frequency of MICA in Tu nationality is MICA*00801,MICA*00201,MICA*018 in turn and MICA*016,MICA*00801,MICA*00201,MICA*015 in turn in Tibetan;the relatively lower gene frequency of MICA in Tu nationality is MICA*00701,MICA*035,MICA*00902,MICA*017 in turn and MICA*035,MICA*004,MICA*046,MICA*019,MICA*00701 in turn in Tibetan.The results of association analysis involving MICA and RA showed that there was no significant relation concerning OR rates,95% C.I and significant detection between two groups.Conclusions It is considered that at least in this study there is no remarkable association between MICA and RA susceptibility in Tu and Tibetan nationalities of Qinghai province.

Abstract:The majority of Miaos, Bouyeis, and Shuis are distributed in the Guizhou province of southwest China. They live within vast areas, while each ethnicity lives in individual concentrated communities in small areas. Their origin, migration, and relationship have long been interesting to anthropologists. In the present study, polymorphism of HLA-A, -B, and -C genes was investigated using the sequencing-based typing method in the Miao, Bouyei, and Shui from Guizhou, southwest China. Generally, Miao, Bouyei, and Shui share the most high-frequency alleles, suggesting that these three ethnic groups might be subject to intensive gene exchange because of their close location. However, it appears that some alleles distribute ethnic-specifically for each ethnicity. The dendrogram constructed according to the neighbor-joining method demonstrates that Miao, Bouyei, and Shui cluster together and form a branch with other southern Chinese ethnic groups, indicating that Miao, Bouyei, and Shui are three genetically close ethnic groups and inherit more characteristics of southern Chinese populations. The genetic architecture of 3'untranslated region of the MICA gene: polymorphisms and haplotypes.

Abstract:In this study, the 3′untranslated region (3′UTR) of MHC class I chain-related gene A (MICA) were investigated in 104 healthy, unrelated Han individuals recruited from northern China, using PCR-sequencing method. Nine polymorphic sites were detected, which were in very strong linkage disequilibrium with each other .Seven different MICA 3′UTR alleles were identified, among which UTR1 predominated (0.6971),followed by UTR2 (0.2356). Twenty-one extended haplotypes incorporating the 3′UTR and MICA exons 2–5 were observed in this population. Phylogenetic analysis revealed the existence of two MICA lineages, each with multiple subsets. The 2 lineages were primarily linked to UTR1 and UTR2 in the 3′UTR, respectively. Ewens–Watterson homozygosity statistics at MICA coding and 3′UTR regions were consistent with neutral expectations. Our data provided for the first time the data of genetic variation in the 3′UTR of MICA gene in human populations. The findings are valuable for future studies of the mechanisms underlying MICA post-transcriptional regulation, and will inform studies of evolution of the MHC gene complex.
Distribution of MICA Microsatellite in 13Population Groups of China

Abstract:The genetic data of MICA microsatellite were obtained by genotyping 577 samples in 13 population groups of China, which are Han-YN, Han-GD, Han-SD, Bai, Dai, Lahu, Li, Naxi, Sala, She,Tu, Wa and Zang-YN, with genescan. Five alleles have been observed in the population groups,which are A4, A5, A5. 1, A6 and A9. A5 allele is the most frequent in all population groups except Lahu and Li, while the most frequent allele for Lahu and Li is A5. 1 and A4 respectively. The second most frequent allele is the A5. 1 in Han-YN, Han-SD, Dai, Naxi, Sala, She, and Wa. The lest frequent allele for Han-YN, Han-GD, Lahu, Naxi, She, Wa is the A6 which is not observed in Li. A4 allele is the lest frequent in Han-SD, Bai, Dai, Sala, Tu, Zang-YN. The results show that the distribution of MICA microsatellite is different in these population groups, and the polymorphism information contents (PIC) of this microsatellite is high. It is a potential useful marker in the study of human origin and migration, personal identification, gene mapping and location, and disease diagnosis.

Abstract:Here, we report for the first time the polymorphisms of MICA and MICB in a healthy Li population of 344 unrelated individuals. By using polymerase chain reaction-sequence specific priming (PCR-SSP) and sequence-based typing (PCR-SBT), 17 MICA-sequence alleles and 5 MICA-STR (short tandem repeats, STR) alleles, as well as 17 MICB alleles were detected, among which MICA*010, MICA*A4 and MICB*005:02 were the most frequent alleles. In addition, linkage disequilibrium was investigated and the most common two-locus haplotypes were MICB*005:02-MICA*010 and MICB*008-MICA*002:01. These results present informative genetic markers for the investigation of possible origins and the evolution of MHC class I haplotypes in the Li population.

Abstract:The human major histocompatibility complex (MHC) class I chain-related gene A ( MICA) is located 46 kb upstream of HLA-B and encodes a stress-inducible protein which displays a restricted pattern of tissue expression. MICA molecules interact with NKG2D, augmenting the activation of natural killer cells, CD8(+) alpha beta T cells, and gamma delta T cells. MICA allelic variation is thought to be associated with disease susceptibility and immune response to transplants. We investigated MICA allelic variations and linkage disequilibrium with HLA-A, B, and DRB1 loci on 110 parental haplotypes from 29 African-American families. PCR/sequence-specific oligonucleotide probing (SSOP) was used to define MICA polymorphisms in exons 2, 3, and 4. Ambiguous allelic combinations were resolved by sequencing exons 2, 3, and 4. Exon 5 polymorphisms were analyzed by size sequencing. For HLA-A, B and DRB1 typing, low-resolution PCR/SSOP and allelic PCR/sequence-specific priming techniques were used. Twelve MICA alleles were observed, the most frequent of which were MICA*008, MICA*004, and MICA*002, with gene frequencies of 28.2, 26.4, and 25.5%, respectively. Thirty-eight HLA-B- MICA haplotypic combinations were uncovered, 22 of which have not been reported in the HLA homozygous typing cell lines from the 10th International Histocompatibility Workshop. Significant positive linkage disequilibria were found in 8 HLA-B- MICA haplotypes. Furthermore, haplotypes bearing HLA-B*1503, *1801, *4901, *5201, *5301, and *5703 were found to segregate with at least two different MICA alleles. Our results provide new data about MICA genetic polymorphisms in African-Americans, which will form the basis for future studies of MICA alleles in allogeneic stem cell transplantation outcome.

Abstract:Objective To study the correlation between polymorphism of MICA gene and lung cancer in Han nationality of Hainan.Methods PCR-SSP and PCR-SBT were used to analyze the gene polymorphism of MICA.Results 11 allelic genes were found in lung cancer patients.The frequency of MICA * A5/ 010 was significantly higher in lung cancer group than that of control group (MICA * A5:OR =1.62,95％ CI:1.18-2.23,Pc＜0.05;MICA *010:OR=6.13,95％CI:3.5～10.75,Pc＜0.000).Conclusions MICA * A5/010 allelic gene may be associated with the susceptibility to lung cancer.

Abstract:To assess the potential contribution of major histocompatibility complex class I chain-related gene A (MICA) polymorphisms toward the pathogenesis of leukemia, 107 leukemia patients and 162 ethnically matched controls from Hunan province, Southern China, were genotyped for the MICA polymorphism using polymerase chain reaction-sequence-specific priming (PCR-SSP) and sequence-based typing (PCR-SBT). The relevance between these genotypes and risk of leukemia was assessed by means of odds ratio (OR) with 95% confidence intervals (95% CIs). Allele frequencies of MICA-sequence and MICA-STR were different in leukemia patients in comparison with normal controls (both P < 0.05). MICA A5 was directly associated with leukemia (OR = 2.3257, Pc < 0.0005), whereas MICA A5.1 and MICA*008 were inversely associated with leukemia (OR = 0.5874, Pc = 0.0235 and OR = 0.5874, Pc = 0.0329, respectively). In addition, we found that homozygotes for MICA A5 (OR = 14.0659, 95% CI: 3.1627-62.5574, Pc < 0.0001) and MICA*010 (OR = 10.1053, 95% CI: 2.2139-46.1260, Pc < 0.0004) were at an increased risk for leukemia, whereas heterozygotes for MICA*008 and MICA A5.1 were linked to a decreased risk for leukemia (OR = 0.4609, 95% CI: 0.2799-0.7590, Pc = 0.0027). MICA allelic variation is associated with leukemia in Hunan Han population; the data also suggest that MICA gene polymorphism affects susceptibility to different clinical subtypes of leukemia.

Abstract:In this study, we investigated the major histocompatibility complex (MHC) class I chain-related gene B (MICB) allelic variation by sequence-based typing (SBT) in 201 healthy, unrelated Han subjects from Hunan province, southern China. Eleven MICB alleles were observed, among which MICB(∗)005:02 predominated with a frequency of 64.93%. Significant linkage disequilibrium (LD) was observed for 5 HLA-B-MICB and 6 MICA-MICB haplotypes. Compared with a northern Chinese Han population, several MICB-containing haplotypes appeared to be highly specific to this southern Chinese Han population. Two new MICB alleles, MICB(∗)005:06 and MICB(∗)026, were identified. Aligned with MICB(∗)005:02, MICB(∗)005:06 has a synonymous T replacement at nucleotide 762 in exon 4; MICB(∗)026 has probably arisen from MICB(∗)004:01 through a single nucleotide substitution from G to A at position 826 in exon 4, leading to an amino acid change from glutamic acid to lysine at codon 253. HLA-A(∗)02-C(∗)01-B(∗)46-MICA(∗)010-MICB(∗)005:02-DRB1(∗)09 was the most prevalent six-locus haplotype with a frequency of 8.49%. HLA-A(∗)30-C(∗)06-B(∗)13:02-MICA(∗)008:01-MICB(∗)005:02-DRB1(∗)07 appeared to be a conserved extended haplotype. Our results provide new information about MICB genetic polymorphism in Chinese Han populations, and will inform future studies of the potential role of MICB in allogeneic organ transplantation and disease susceptibility in related ethnic groups.

Abstract:In this study, the 5′promoter region of MHC class I chain-related gene A (MICA) was investigated in 104 healthy, unrelated Han individuals recruited from northern China, using PCR-sequencing method. Twelve variable sites were detected, which were in very strong linkage disequilibrium with each other. Twelve different MICA 5′promoter haplotypes were identified, among which Promoter-7 predominated (0.5529). Twenty-six extended haplotypes incorporating MICA 5′promoter and MICA exons 2–5 were observed in this population, 9 of which were in significant linkage disequilibrium (LD). Phylogenetic analysis of 5′promoter refined MICA sub-lineage structure previously constructed according to MICA coding and 3′untranslated regions. Ewens–Watterson homozygosity statistics at MICA 5′promoter region were consistent with neutral expectations. None of the five variable sites detected within the minimal promoter of MICA gene was located in the putative binding sites for transcription factor. Our study provided for the first time the sequence information about 5′promoter of MICA gene at a human population level. The data will facilitate the understanding of regulation of MICA gene expression, which represents a promising pathway for immune intervention against cancer, autoimmune disorders and infections.
