Experimental study on the treatment of gastric cancer by TK gene combined with miL-2 gene

Abstract:AIM: To study the killing effect of suicide gene TK to treat mouse gastric cancer by the transgene methods, and combined with mIL-2 gene to study the immune response in the cancer treatment. METHODS: To transduce the suicide gene with the retrovirus, and to transduce the cytokine gene with liposome. In the in vitro experiment, we observed the killing effect of TK gene to treat mouse gastric cancer cell line MFC, and the bystander effect expressing in the experiment. In the in vivo experiment, we made the gastric cancer model on the 615 mouse and transduced the suicide gene by injecting the virus serum into the tumors. Liposome method was used to transduce the mIL-2 gene and the volume of the tumors was measured in each group. With the pathological and immunohistochemical methods to observe the tissue and immunological changes: in the tumors. RESULTS: TK gene expressed marked killing effect and bystander effect in the in vitro experiment. 20% gene transduced cells can give rise to 70% - 80% total cells to death. In vivo, TK/GCV can strongly cause tumor cells to death (P=0.046). When combined with mIL-2 gene, the killing effect can be enhanced (P=0.005), and many tumors disappeared. The immunohistochemical examination demonstrated the gathering of the CD4/+ and CD8/+ T lymphocytes. CONCLUSION: TK/GCV system yielded marked killing effect to mouse gastric cancer, through both the direct killing and bystander effect. The immune response takes part in the process. The cytokine gene mIL-2 could enhance the therapeutic effect.
