Protective effect of heme oxygenase-1 on acetic acid induced gastric ulcer in rats

Abstract:AIM: To investigate the possible role of heme oxygenase-1 (HO-1) in acetic acid induced gastric ulcer in rats. METHODS: HO-1 and inducible nitric oxide synthase (iNOS) mRNA and protein were determined at different time points after acetic acid induced gastric ulcer in rats by RT-PCR and western blotting, respectively. The severity of ulcer was assessed by ulcer area. Tin mesoporphyrin(SnMP) was used to inhibit the activity of HO. RESULTS: HO-1 gene expression was markedly increased after gastric ulcer induction. The ulcer area in SnMP treatment group 1 day (71.75 ± 6.06mm2) and 3 days (50.75 ± 3.86mm2) after ulcer induction was significantly higher (P< 0.01) than the corresponding control (51.40 ± 3.61mm2 and 35.00 ± 3.7mm2 respectively). However, there was no significant difference between SnMP treatment group (27.20 ± 4.09mm2) and control (24.17 + 2.94mm2) in the ulcer area 7 days after ulceration. Along with the severity of mucosal lesion, the iNOS expression and activity increased markedly in SnMP group. CONCLUSION: HO-1 may play a protective role in acetic acid induced gastric mucosa damage by decreasing iNOS activity.
