Disequilibrium transmission of the Ser9Gly polymorpphism of the dopamine D3 receptor gene in 151 Chinese pedigrees

Abstract:Objective: To investigate the association between Ser9Gly polymorphism of dopamine D3 receptor (DRD3) gene and schizophrenia. Methods: The Ser9Gly polymorphism of DRD3 was tested in 151 Chinese Han's pedigrees each of which includes two or more affected schizophrenia siblings who fulfill ICD-10 criteria. At most only one of their alive parents has a history of suffering schizophrenia. Results: 1) The distributions of alleles or genotypes of the DRD3 Ser9Gly polymorphism are not significantly different between the groups of parents, unaffected -siblings and affected-siblings, and between the clinical sub-types of schizophrenia. 2) The transmission disequilibrium test (TDT) showed the transmission disequilibrium in affectedsiblings (2 = 4.73, P = 0.0297, OR = 4.78, 95%IC: 1.27-18.46). 3) The correlation analysis is taken between genotypes and clinical status, the results showed that schizophrenia siblings with Ser9 allele are apt to have negative symptoms. Conclusion The results show that there is significant association between DRD3 and schizophrenia. It suggests that DRD3 may play a role in the etiology of schizophrenia. This research support that DRD3 is one of the candidate genes of schizophrenia.
