Abstract:OBJECTIVE: To observe the in vivo effect of combined iontophoresis and laurocapram pretreatment on transdermal delivery of diclofenac sodium gel. METHODS: Diclofenac sodium gel was prepared using polyvinyl alcohol, carboxymethylcellulose sodium and hydroxypropylmethyl cellulose. The diclofenac blood level in rabbits was measured in four groups: passive diffusion, laurocapram pretreatment, iontophoresis (current density controlled at 0.3 mA/cm(2)) and combined laurocapram pretreatment and iontophoresis. Rabbit stratum corneum of each of the four groups was examined using a scanning electron microscope. RESULTS: Diclofenac blood concentration in the passive diffusion group was undetectable. The diclofenac blood concentration area under the curve compared with time was 8.4 &mgr;l ml(-1) h(-1) in the laurocapram pretreatment group, 2.7 &mgr;l ml(-1) h(-1) in the iontophoresis group and 15.4 &mgr;l ml(-1) h(-1) in the combination group. There was no detectable damage observed by scanning electron microscopyto the stratum corneum after iontophoresis or laurocapram pretreatment. CONCLUSION: The combination of iontophoresis and laurocapram pretreatment appears to enhance transdermal delivery of diclofenac sodium gel wi thout significant skin damage.

Abstract:OBJECTIVE To study the ultrasound, intensity,effective time, and different couplant on drug transdermal delivery in vitro. METHODS Preparated with lappaconitine hydrobromide gels, the influence of different ultrasound,intensity,effective time, and different couplants on the transdermal parameters per unit cumulative absorbing quantity (Q_(8hr)) and steady satepenetration rate (J_s) were determined. RESULTS The transdermal per unit (O_(8hr)) and (J_s) were increased with the ultrasoundfrequencies, ultrasound intensity and effective time could affect the action in some degrees on drugs transdermal delivery. CONCLUSION Sonophoresis s, including ultrasound,intensity,effective time, affected its action on drugs transdermal delivery. We can change the parametres to accelerate the transdermal absorption of lappaconitine hydrobromide.
Optimized Preparation of Naloxone Microemulsion and Its Transdermal Delivery Effect

Abstract:OBJECTIVE To prepare W/O microemulsion formulation containing naloxone and to investigate its transdermal delivery effect in vitro.METHODS The microemulsion was prepared with the system contained of isopropyl myristate-water-lecithin-propanol according to the central composite design(CCD).The steady permeation rate of naloxone hydrochloride was determined in vitro using Franz diffusion cell with the skin of SD male rats.The concentration of naloxone hydrochloride was measured by HPLC.RESULTS The optimal formulation contained lecithin(29%),propano(l25%) and IPM(35%).The steady permeation rate of naloxone microemulsion was 4.33 μg·h-1·cm-2,much higher than that of the aqueous solution(0.20 μg·h-1·cm-2).CONCLUSION Microemulsion enhanced the permeation of naloxone hydrochloride across the skin of SD male rats.
The study of ultrasound and iontophoresis on oxaprozin transdermal penetration using surface-enhanced Raman spectroscopy

Abstract:The potential for physicochemical driving forces facilitating topical transport of the lipid-soluble drug oxaprozin (OXA) was investigated using surface-enhanced Raman spectroscopy (SERS) in this study. Azone, iontophoresis (IP), and sonophoresis (SP) were combined and performed on mouse skin for the OXA transdermal penetration, and the synergistic effect was analyzed using Raman spectroscopy. The data of characteristic peak intensity were processed with overlapping peak resolving and standard normalization. The results showed that Azone promoted the transdermal penetration of OXA (5.9-fold greater than the OXA concentration of normal penetration); SP enhanced OXA transdermal penetration (5.5-fold); IP enhanced OXA transdermal penetration (4.2-fold); the combined application of Azone and SP (Azone+SP) and SP+IP can improve the enhancement coefficient of OXA transdermal penetration (8.4-fold and 6.1-fold, > 5.9, > 5.5, > 4.2), and their combined application has a synergistic effect; Azone+IP does not have a synergistic effect while the enhancement coefficient of Azone+IP (5.3-fold, < 5.9) and Azone+SP+IP (7.2-fold, < 8.4) was slightly reduced. As for the drug OXA, Azone+SP is an effective method of transdermal penetration.
Study on preparation and percutaneous absorption in vitro of piroxicam microemulsion

Abstract:Objective To prepare Piroxicam microemulsion and investigate its transdermal characteristics in vitro.Methods The o/w microemulsion was prepared with the system contained of Lauroglycol FCC as oil phase,Labrasol and Cremophor EL as surfactants,Transcutol P and Ethanol as cosurfactants according to the pseudo-ternary phase diagrams.The transdermal characteristics were evaluated by modified Franz diffusion cell method.Rat's skin was used as permeation barrier and the drug concentration was measured by HPLC.Results The optimum formulation consisted of 0.5%Piroxicam,10%Lauroglycol FCC,50%water,13.3%Labrasol,Cremophor El and Transcutol P.The permeation rate of Piroxicam was 10.04±1.73 μg/(cm2·h).Conclusion The results indicated that the optimum microemulsion system with high permeation rate may be a potential vehicle for the transdermal delivery of Piroxicam.
Synergistic Effect and Mechanism of Cineole and Terpineol on In-Vitro Transdermal Delivery of Huperzine A from Microemulsions

Abstract:The aim of the present study was to investigate the influence and the mechanisms of cineole and terpineol on the in-vitro transdermal delivery of huperzine A from microemulsions, and their potential synergistic effect on the permeation enhancement. The transdermal delivery of huperzine A from microemulsions with different concentrations of cineole and terpineol through the rat abdominal skin was determined by Franz-type diffusion cells. The partition coefficient of huperzine A between the full thickness skin and microemulsion was determined. Attenuated total reflection-Fourier transform infrared spectroscopy (ATR-FTIR) was carried out to analyze the effects of cineole and terpineol on the biophysical properties of the stratum corneum (SC) and the mechanisms of permeation enhancement. These results indicated that cineole and terpineol could synergistically increase the transdermal delivery of huperzine A from microemulsions through increasing the partition and diffusion coefficients of huperzine A. ATR-FTIR studies further validated the synergistic effect and revealed that the enhancing mechanisms were due to increasing the disorderliness and fluidity of SC lipid alkyl chains, disrupting the structure of keratin in SC, and extracting SC lipids. In conclusion, cineole and terpineol, acting synergistically to enhance the transdermal delivery of huperzine A from microemulsions, might provide an alternative permeation enhancer combination for the transdermal delivery of huperzine A.

Abstract:AIM To study the formulation of transdermal microemulsion of lappaconitine hydrobromide and its transdermal absorption behavior in vitro.METHODS Through the solubility determination and the drawing of microemulsion pseudo ternary phase,the microemulsion prescription was screened.Its transdermal absorption performance was tested through HFLC and TK - 20B skin diffusion test instruments.RESULTS Microemulsion with 5%oleic acid, labrasol,anhydrous alcohol and water had perfect transdermal absorption perfomiance.CONCLUSION Microemulsion for lappaconitine hydrobromide has good transdermal absorption performance.
The effect of ion-pair formation combined with penetration enhancers on the skin permeation of loxoprofen

Abstract:Context: Loxoprofen (LOXO) is a non-steroidal anti-inflammatory drug. Repeated oral administrations induce gastrointestinal side effects. Patches are a promising alternative. Objective: The aim of this study was to investigate the effects of organic amines on the skin permeation of LOXO and finally design a patch with a comparable permeation profile and pharmacodynamic effects to the commercial LOXONA® plaster. Materials and methods: The effects of organic amines were assessed by flux values of LOXO from isopropyl myristate (IPM), using horizontal diffusion cell and rabbit skin. FTIR spectroscopy was used to confirm ion-pair formation. Anti-inflammatory and analgesic activity assessments were performed in the adjuvant arthritis rat model and acetic acid-induced writhing syndrome in mouse, separately. Results and discussion: Results showed that triethylamine (TEA) was the most potential candidate in IPM, with the highest flux of 499.75 ± 32.40 µg/cm(2)/h. In patch, the highest flux of 369.37 ± 34.32 µg/cm(2)/h was still obtained by LOXO-TEA. Combined with penetration enhancers, the cumulative amounts were further increased in presence of 5% IPM, which exhibited a flux of 840.04 ± 66.38 µg/cm(2)/h as two times of the commercial one. Ultimately, anti-inflammatory and analgesic activity assessment presented that a comparable pharmacodynamic activity with the commercial one could be obtained by the patch we designed. Additionally, we also found that LOXO patch applied topically exerted a systemic effect, and the effect was dose-dependent. Conclusion: It was feasible for LOXO patch design by combination of ion-pair technology and chemical enhancers.
[Influence of Different Penetration Enhancers on Lappaconitine Transcutaneous Permeation].

Abstract:OBJECTIVE:To investigate the effects of different kinds and concentration of transdermal enhancers on Lappaconitine transcutaneous permeation when used individually or together.METHOD:Using modified Franz-type diffusion cell and excised human body skin as an in vitro transdermal model, the concentration of lappaconitine was determined by HPLC, then cumulative permeation quantity (Q) and stability rate (J) of progesterone were calculated.RESULT:Penetration enhancers such as propylene glycol, dodecanol, IPM, and particularly 3% OA and Azone, can significantly enhance the penetration rate of lappaconitine. Concentration effect of penetration enhancers concentration on lappaconitine transcutaneous permeation were found in experiments, the permeation effect of Azone was better than Azone + OA and Azone + propylene glycol.CONCLUSION:The transdermal rate of lappaconitine from batch which contains 3% OA or Azone is higher than others. Combination of Azone with other penetration enhancers is not recommended for Lappaconitine transcutaneous permeation.

Abstract:To promote transungual permeation of nystatin (NYST), molecule with high molecular weight, no water-soluble, amphoteric by iontophoresis. The synergic effect of the combination of cetylpyridinium chloride, CPC, or polyoxyethylene (20) sorbitan monooleate, TW80, and iontophoresis was investigated. In vitro permeation experiments were carried out through bovine hoof slices using vertical diffusion cells. A low current density (0.2 mA/cm2) was applied by introducing Ag/AgCl electrodes in the donor (anode) and receptor (cathode) chambers. The donor phase consisted of a solution, a suspension, or gel-type vehicles containing NYST and surfactants in pH 5.6 HEPES buffer. The addition of CPC to NYST suspension (SOSP) produced a fivefold increase on the permeability of the bovine hoof membrane to the drug. The application of anodal iontophoresis further improved NYST flux. Conversely, NYST transungual permeation was not influenced by TW80 either in the passive diffusion or iontophoretic flux. Furthermore, the iontophoretic treatment does not appear to induce irreversible alterations to the hoof bovine membranes. The present work demonstrated the efficacy of iontophoresis as a treatment for different nail pathologies with large molecules very slightly soluble in water without irreversibly affecting the nail structure. A synergistic effect between CPC and iontophoresis was observed.
Simultaneous Lc-Ms/Ms Determination of Danshensu and Paeoniflorin for Permeability Studies in Caco-2 Intestinal Absorption Model

Abstract:A high sensitive method based on liquid chromatography tandem mass spectrometry(LC-MS/MS) was developed and validated for the study of permeability of danshensu(DS) and paeoniflorin(PF) in Caco-2 intestinal absorption model. The DS and PF were extracted from cell culture by vacuum-lyophilizing and then separated on a Zorbax Stable Bond C-18 column with 0.1% acetic acid aqueous solution and methanol as mobile phase. Detection was carried out by negative electrospray ionization(ESI-) with selected reaction monitoring(SR-M) mode. The apparent permeability coefficients(P-app) of DS and PF in Caco-2 cell medium were calculated and the effects of verapamil on the coefficients Papp of the two test compounds were also illustrated. The permeability of PF was much better than that of DS when the two compounds were administrated individually. Co-administration of DS and PF led to the decrease of the transport from apical side to basolateral side for both the compounds. However, the transport in the contrary direction were accelerated. It was also observed that verapamil could accelerate the transport of the test compounds from apical side to basolateral side. However, the absorption-enhanced effect of verapamil was attenuated when DS and PF were co-administrated. These observations suggest that both passive diffusion and active efflux involved in P-gp would effect the passage of DS and PF across Caco-2 cell monolayer. At the same time, the co-administration of DS and PF to an alteration of transport behavior, which suggests that the interaction must be taken into account when 'n-in-one' samples were used in Caco-2 intestinal model.

Abstract:The purpose of this paper was to study the influence of drug physicochemical characteristics on in vitro transdermal absorption of hydrophobic drug nanosuspensions. Four drug nanosuspensions were produced by high-pressure homogenization technique, which were the same in stabilizer and similar in particle size. Differential scanning calorimetry and powder X-ray diffraction analysis showed that the crystalline state of the nanocrystals did not change. In vitro permeation study demonstrated that the drug nanosuspensions have a higher rate of permeation that ranged from 1.69- to 3.74-fold compared to drug microsuspensions. Correlation analysis between drug physicochemical properties and Jss revealed that log P and pKa were factors that influenced the in vitro transdermal absorption of hydrophobic drug nanosuspensions, and drugs with a log P value around 3 and a higher pKa value (when pKa < pH+2) would gain higher Jss in this paper.

Abstract:OBJECTIVE To prepare compound asiaticoside cataplasm and study the effect of different percutaneous absorption enhancer on its in vitro percutaneous permeability.METHODS A modified Franz diffusion cell device was used to study the effect of 1% azone,0.5% menthol,1% 1,2-di-hydroxypropane and 1% azone combined with 1% 1,2-di-hydroxypropane on the in vitro percutaneous permeability of the compound asiaticoside cataplasm.The contents of aisaticoside and triamcinolone acetonide acetate in the modified Franz diffusion cell device were determined by HPLC.RESULTS Transdermal diffusion in vitro of asiaticoside complied with the Higuchi equation.Transdermal diffusion in vitro of triamcinolone acetonide acetate complied with zero order equation.The four penetration enhancers could increase the permeability of the drug.The order of enhancement for asiaticoside was 1% azone combined with 1% 1,2-di-hydroxypropane > 1% azone >0.5% menthol>1% 1,2-di-hydroxypropane.The order of enhancement for triamcinolone acetonide acetate was 1% azone combined with 1% 1,2-di-hydroxypropane > 1% azone >1% 1,2-di-hydroxypropane>0.5% menthol.CONCLUSION 1% azone combined with 1% 1,2-di-hydroxypropane could be used as penetration enhancer of compound asiaticoside cataplasm.

Abstract:OBJECTIVE: The purpose of this study was to research the mechanism of percutaneous penetration of Ibuprofen (IBU) nanoemulsion.METHOD: Transdermal penetration mechanism of IBU nanoemulsion was investigated by using Fourier transform infra-red spectral analysis (FTIR), differential scanning calorimeter thermogram (DSC), and activation energy (Ea) measurement. The in vivo skin penetration test of rats was carried out using Rhodamine B nanoemulsion to simulate the process of drug penetration into the skin, and the frozen section of the skin was observed by confocal laser scanning microscopy (CLSM).RESULT: FTIR spectra and DSC thermogram of rat skin treated with IBU nanoemulsion showed that infiltration occurred due to disruption of the stratum corneum (SC) protein-lipid structure and increasing of fluidity, hydration, and disruption of the lipid bilayer structure of the SC. The significant reduce in Ea (1.255 kcal/mol) for IBU permeating rat skin suggested crucial disruption of the SC lipid bilayers (P < 0.05), which is speculated that nanoemulsion may create new pathways to promote drug penetration. CLSM revealed that Rhodamine B penetrated into the SC in a shorter period of time and it accumulated around the sebaceous glands.CONCLUSION: The study of skin penetration mechanism indicated that nanoemulsion can be perfectly well used as the transdermal penetration of poorly soluble drugs.

Comparison of liposomal and NLC (nanostructured lipid carrier) formulations for improving the transdermal delivery of oxaprozin: Effect of cyclodextrin complexation

Abstract:The combined strategy of drug-cyclodextrin (CD) complexation and complex loading into nanocarriers (deformable liposomes or nanostructured lipid carriers (NLC)), was exploited to develop effective topical formulations for oxaprozin transdermal administration. Oxaprozin was loaded as ternary complex with randomly-methylated-ßCD and arginine, selected as the best system in improving drug solubility. The colloidal dispersions, characterized for particle size, zeta-potential and entrapment efficiency, were investigated for drug permeation properties in comparison with a plain drug aqueous suspension, a ternary complex aqueous solution and a plain drug liposomal or NLC dispersion. Experiments with artificial membranes showed that the joined use of CD and both liposomes or NLC enabled a marked increase of the drug permeability (16 and 8 times, respectively) and was significantly more effective (P<0.05) than the drug as ternary complex (3.2 times increase), and the corresponding liposomal or NLC dispersion of plain drug (5.6 and 4.3 times increase, respectively). Experiments with excised human skin confirmed the significantly (P<0.05) better performance of deformable liposomes than NLC in promoting drug permeation; moreover, they evidenced a more marked permeability increase compared to the plain drug (24 and 12 fold, respectively), attributed to a possible enhancer effect of the nanocarriers components and/or of the randomly-methylated-ßCD.

Abstract:In the present study,the potential inhibition behaviors of notoginseng total saponins (NS),safflower total flavonoids (SF),and their combination (CNS) towards three major isoforms of UDP-glucuronosyltransferases (UGTs) in human liver microsomes (HLMs) were investigated to study the mechanism of the synergistic effect of CNS.Etoposide,trifluoperazine and azidothymidine were selected as the probe drugs to elucidate the activities of UGT1A1,1A4 and 2B7 by UPLC-MS/MS method,respectively.The results showed that CNS,NS and SF significantly inhibited the activities of UGT1A1,1A4 and 2B7 (P＜0.05) with the IC50 values less than 30 mg/mL.Furthermore,the inhibitory effects of CNS towards UGT1A1,1A4 and 2B7 were stronger than those of NS and SF (P＜0.05).In conclusion,the combination of NS and SF could increase their inhibitory effects on UGT1A1,1A4 and 2B7 activities in HLMs and might be conducive to reduce the phase Ⅱ metabolism of the effective constituents in CNS.The potential herb-drug interactions of CNS based on UGT enzymes provided a useful experimental basis for its further research and development.

Abstract:Objective:To study the effect of ultramicro-shatter technology on penetrating skin absorption of isorhamnetin-3-O-neohesperidin in Zhongtongxiao Cataplasm.Methods:To apply reformed Frans penetrating skin absorption cell marching extraorgan penetrating skin experiment.HPLC method was used to determine the content of isorhamnetin-3-Oneohesperidin in ultramicro-shatter Zhongtongxiao Cataplasm and in common Zhongtongxiao Cataplasm.Results:The Q-t equation of ultramicro-shatter Zhongtongxiao Cataplasm:Q=3.0382t+47.082,penetrating skin velocity:3.0382(μg.cm2/h);the Q-t equation of common Zhongtongxiao Cataplasm:Q=2.7967t+39.752,penetrating skin velocity:2.7967(μg.cm2/h);Extraction rate of dynamic extracting micro-powder,the ephedrina hydrochloridum,glycyrrhizic acid and glycyrrhizae glycoside were higher than the trdtional cut crude drug decocting.Conclusion:The accumulating osmolality and penetrating skin velocity of isorhamnetin-3-O-neohesperidin in ultramicro-shatter Zhongtongxiao Cataplasm were all better than those in common Zhongtongxiao Cataplasm,it explained that ultramicro-shatter technology accelerate the dissolution of medicine compsitions.

Abstract:Objectiae:To study the Lappaconitine hydrobromide two preparation for percutaneous administration of analgesic effect.Method: Prepare two formulations of Lappaconitine hydrobromide : microemulsion,gel.And determine the analgesic effect of two lappaconitine hydrobromide transdermal preparation by the mouse hot plate method and mouse writhing method Results: The analgesic effect of two preparations is better than that of the blank,the analgesic effect of Lappaconitine hydrobromide microemulsion is better than that of gel,the writhing method and hot plate has significant analgesic effect(P<0.01).Conclusion: Preparation of lappaconitine gel and microemulsion have analgesic effect,analgesic effect is better than that of microemulsion gel.
Enhancing effect of fumaric acid on transdermal penetration of loxoprofen sodium

Abstract:Topical administration is a promising clinical strategy to avoid serious gastrointestinal adverse reactions of loxoprofen sodium (LOX), a new non-steroidal anti-inflammatory drug. Small molecule organic acids had been reported with the ability of promoting transdermal rate of several drugs. In this article, the effect of small molecule organic acids on the transdermal delivery of LOX was studied, and the possible mechanism was also explored by Fourier infrared spectroscopy, differential scanning calorimetry, tape stripping, etc. The results showed that lactic acid and fumaric acid could significantly increase the penetration rate of LOX and reduce time lag even without the help of acidic environment. The preliminary mechanism investigation inferred that fumaric acid could increase LOX's distribution in stratum corneum and might change its complexation state, but had lithe effect on the drug structure and skin's lipids and proteins configuration. The topical LOX gel using fumaric acid as penetration enhancer had higher transdermal rate, significant anti-inflammatory effect and no obvious skin irritation. This study proved the promising application of small molecule organic acids in transdermal enhancing and provided a potential strategy for transdermal delivery of LOX combined with fumaric acid.
