Abstract:The development of micro total analysis system (mu-TAS) technique is very prominent in recent years. More and more novel and practical methods for sample precondition, separation and detection have been established to solve real problems. Electrochromatography, as a new chromatography technique, is one of the promised separation techniques suitable for mu-TAS, because it has not only the high performance efficiency of conventional capillary electrophoresis and high selectivity of high performance liquid chromatography. Latest progress on chip electrochromatography is reviewed.

Abstract:Micro-total analysis systems (μTAS) integrate different analytical operations like sample preparation, separation and detection into a single microfabricated device. With the outstanding advantages of low cost, satisfactory analytical efficiency and flexibility in design, highly integrated and miniaturized devices from the concept of μTAS have gained widespread applications, especially in biochemical assays. Electrochemistry is shown to be quite compatible with microanalytical systems for biochemical assays, because of its attractive merits such as simplicity, rapidity, high sensitivity, reduced power consumption, and sample/reagent economy. This review presents recent developments in the integration of electrochemistry in microdevices for biochemical assays. Ingenious microelectrode design and fabrication methods, and versatility of electrochemical techniques are involved. Practical applications of such integrated microsystem in biochemical assays are focused on in situ analysis, point-of-care testing and portable devices. Electrochemical techniques are apparently suited to microsystems, since easy microfabrication of electrochemical elements and a high degree of integration with multi-analytical functions can be achieved at low cost. Such integrated microsystems will play an increasingly important role for analysis of small volume biochemical samples. Work is in progress toward new microdevice design and applications.

Abstract:Based on laser induced fluorescence detection a miniaturized and sensitive optical system is developed for micro total analysis systems (u-TAS) application. A micro optical system integrated with a micro fluid device is a convenient way to realize biochemical reaction and detection on one chip. Such a bi-functional micro system for DNA affinity assay is presented in this paper. The optical system composed of optical fiber coupling with planar waveguide for illuminating the labeled DNA samples, and micro lens arrays for collecting the induced fluorescence light. High signal-to-noise ratio can be achieved because of evanescent field excitation mode. A specially designed micro chamber, with chemical process, including small pipes for guiding sample agent is included in the system in order to enable DNA affinity reaction, and to wash the chamber after reaction. These components will be integrated within the encapsulation of a photoelectric array detector, so the size of this system can be greatly reduced. This bi-functional micro system has merits of integration of reaction and detection together, more compactness, multi-channel detection, high sensitivity and good compatibility with biological samples. This system is a promising candidate to be integrated into a micro total analysis system, such as a portable DNA diagnostics device.

Abstract:Accelerator mass spectrometry(AMS) is the most sensitive analytical technique for measuring long-lived radionuclides, which is widely used in the fields of environment, geology, archaeology and physics. In recent years, the miniaturization of AMS device has been greatly developed in the world. In order to develop miniaturized AMS devices and their analysis technology, China institute of atomic energy has independently developed a single-stage AMS device with an acceleration voltage of 0.2 MV and a tandem AMS device with an terminal voltage of 0.3 MV, respectively. Based on the single-stage AMS device, the high efficiency transport technique and background eliminating method have been developed for 14C measurement, and realized the high sensitivity measurement of 14C. The measurement sensitivity reaches 14C/12C (atomic number ratio)=2×10–15. Using the 0.3 MV tandem AMS device, the gas stripping conditions, background eliminating and detection methods of 129I at low ion energy were systematically studied, and the high efficient transmission and high sensitive measurement methods of 129I were established, the sensitivity is 129I/127I=1×10–14. This is the first time to develop miniaturized AMS devices in China, which lays the foundation for the localization of AMS.

Abstract:Miniature mass spectrometers are of an increasing interest for in-situ analyses, and their coupling with the ambient ionization sources is a valid path for direct analysis of complex samples. In this study, a miniature mass spectrometer using discontinuous atmospheric pressure interface was developed with a dual-LIT (linear ion trap) configuration. The comprehensive scan modes were enabled for tandem mass spectrometry analysis, which are critical for high-quality qualitative and quantitative analysis. A real-time pressure control was implemented to facilitate the ion transfer and collision induced dissociation (CID). Beam-type CID could be performed for tandem analysis at a high number of stages. In-trap CID at high q could also be performed with the fragment ions accumulated in a second trap. A precursor ion scan mode for analyzing target analytes has also been demonstrated.
A Compact and Low-Cost Miniaturized Analysis System Composed of Microchip Electrophoresis and Chemiluminescence Detection Manipulated by a Simple Subatmospheric Pressure Fluid-Driven Device

Abstract:A portable and low-cost miniaturized analysis system was proposed, in which microchip electrophoresis (MCE) and chemiluminescence (CL) were used as the separation and detection units, respectively. A porous monolithic plug was created in the separation channel of the microchip as a select valve, which prevented the pressure-driven CL reagents flowing back into the separation channel but allowed electrophoretic migration along the separation channel. The sensitivity was greatly enhanced by improving the mixing efficiency using a spiral detection channel with an increased length facing the photomultiplier tube (PMT). Peak width could be significantly reduced by increasing the flow rate of CL reagents. Putting grounding electrode before the detection channel significantly improved the reproducibility. A simple and compact subatmospheric pressure fluid-driven device was developed for manipulating the whole analytical process, including variable-volume sample loading, electrophoretic separation and CL reagents transportation. All miniaturized components for constructing a portable MCE-CL system are commercially available. This approach considerably simplified the operation and equipments for constructing an efficient MCE-CL system. Nine metal cations, such as Cr(III), Co(II), Cu(II), Ni(II), Au(III), Mn(II), Zn(II), Pt(II), Pb(II) were successfully separated within 200 s. Migration time precisions ranging from 0.39% for Cr(III) to 2.1% for Cu(II) were obtained for ten consecutive determinations with peak height precisions from 1.67% for Co(II) to 5.73% for Pb(II). Detection limits ranging from 7.5 x 10(-11) mol L(-1) for Co(II) to 8.3 x 10(-9) mol L(-1) for Pb(II) were achieved, which were about three orders lower than the present MCE-CL system. It has been applied for the determination of metals in tea. The results with the recoveries from 97.0% to 102.3% proved that the proposed MCE-CL system offers a number of benefits including miniaturization, high sensitivity and better resolution.

Abstract:A benchtop miniature mass spectrometer system, Mini 12, with ambient ionization source and tandem mass spectrometry capabilities has been developed and characterized. This instrument was developed as a self-contained system to produce quantitative results for unprocessed samples of small volumes including nonvolatile analytes. The ion processing system, vacuum system, and control system are detailed. An integrated sample loading system facilitates automated operation. A user interface has been developed to acquire and to interpret analytical results for personnel who have limited mass spectrometry knowledge. Peak widths of Δm/z 0.6 Th (full width at half-maximum) and a mass range of up to m/z 900 are demonstrated with the rectilinear ion trap mass analyzer. Multistage experiments up to MS(5) are accomplished. Consumable cartridges have been designed for use in ambient paper spray ionization, and the recently developed extraction spray ionization method has been employed to improve the quantitative performance. Monitoring of trace-levels of chemicals in therapeutic drugs, as well as in food safety and environmental protection operations is demonstrated. Dual MS/MS scans are implemented to obtain the intensities of the fragment ions from the analyte and its internal standard, and the ratio is used in quantitative analysis of complex samples. Limits of quantitation (LOQ) of 7.5 ng/mL, with relative standard deviations below 10%, have been obtained for selected therapeutic drugs in whole blood throughout their individual therapeutic ranges.

Abstract:Mass spectrometry (MS) is known for highly specific and sensitive analysis. The general applicability of this technique makes it a good candidate for biological applications over a much broader range than is now the case. The limiting factors preventing MS from being applied at the biologist's bench or in a physician's office are identified as the large size of the systems, as well as the complicated analytical procedures required. An approach for developing miniature MS analysis systems with simplified operational procedures is described and the associated technical developments are discussed.

Abstract:The key concepts and technologies developed in our laboratories in Purdue University for the miniaturization of mass spectrometry analysis systems are introduced. Mass analyzers of simple geometries with a novel atmospheric pressure interface were used to allow reduction in the size of the ion trap mass spectrometer. Ambient ionization methods were developed and coupled to miniature mass spectrometers to allow direct MS analysis of complex samples without sample preparation and chemical separation. The performance of desorption electrospray ionization, low temperature plasma probe, paper spray, and two handheld MS systems, Mini 10 and Mini 11, are described with demonstrations of capabilities for chemical analysis.

Abstract:Top‐down mass spectrometry (TD‐MS) provides unique information on compositions, structures, and functions of proteins or protein complexes and has been recognized as a powerful approach to complement conventional tools in protein analysis. Employing microfluidic chips in TD‐MS workflows offers unique advantages including flexible integration and automation of multiple sample treatment functions, lowered sample consumption, compatibility with high‐sensitive ionization and higher throughput. Here we reviewed the reported microchips developed for TD‐MS in aspects of design of ionization, separation and mixing modules, and fabrication approaches; we also attempted to summarize the design considerations that may inspire development of microchips with better performance and wider applications.

Abstract:Ambien ionization and miniaturization of mass spectrometers are two fields in mass spectrometry that have advanced significantly in the last decade. The integration of the techniques developed in these two fields is leading to the development of complete miniature analytical systems that can be used for on site or point-of-care analysis by non-expert users. In this review, we report the current status of development in ambient ionization and miniature mass spectrometers, with an emphasis on those techniques with potential impact on the point-of-care (POC) diagnostics. Chip-Based MEMS Platform for Thermogravimetric/Differential Thermal Analysis (TG/DTA) Joint Characterization of Materials

Abstract:Combined use of thermal analysis techniques can realize complementarity of different characterization methods. Comprehensive thermal analysis with both thermogravimetric analysis and differential thermal analysis (TG/DTA) can measure not only mass change of a sample but also its temperature change during programmed heating-induced reaction or phase transition processes, thereby obtaining multiaspect thermal information of the material such as dehydration, structural decomposition, phase change and thermal stability. This study proposes and develops a MEMS chip-based TG/DTA microsystem that integrates both programmed heating and detecting elements into a TG chip and a DTA chip to enable the microinstrument performing TG/DTA joint characterization under microscope observation. The TG chip contains a self-heating resonant microcantilever to measure heating-induced mass change of a sample and the DTA chip is with a microheater and a temperature-detecting thermopile integrated on a suspended thermal-insulating diaphragm. Only nanogram and microgram-level samples are needed for the TG and DTA chips, thereby achieving safe measurement to energetic materials such as strong oxidants. The chip-based microinstrument surpasses the state-of-the-art commercial TG/DTA instruments that have, in the long term, suffered from large sample-amount (milligram level) requirements and have been unable to measure energetic materials. Compared with commercial instruments, the chip-based microinstrument is advantageous given its more accurate analysis, much higher heating rate, much smaller instrument volume and much lower power consumption, etc. The microinstrument has been fabricated by using wafer-level MEMS techniques. Testing results show that the mass-detection sensitivity of the TG-chip is as high as 0.45 Hz/pg in air and the temperature sensitivity of the DTA chip achieves 2.9 mV/K under the high heating rate of 25 °C/s. The strong oxidant of KMnO4 is analyzed with the TG/DTA joint characterization under microscopic observation. At the same time as microscope observation of the thermal decomposition phenomena, two-step thermal decomposition process of KMnO4 is identified and the thermal decomposition temperatures are obtained. The TG/DTA microinstrument is promising to be applied for study of various materials.

Abstract:A miniature mass spectrometry system with a sampling probe has been developed for real-time analysis of chemicals from sample surfaces. The sampling probe is 1.5 m in length and is comprised of one channel for introducing the spray and the other channel for transferring the charged species back to the Mini MS. This system provides a solution to the problem of real-time mass spectrometry analysis of a three-dimensional object in the field and is successful with compounds including those in inks, agrochemicals, explosives, and animal tissues. This system can be implemented in the form of a backpack MS with a sampling probe for forensic analysis or in the form of a compact MS with an intrasurgical probe for tissue analysis.

Abstract:Current nano-scale liquid chromatography-tandem mass spectrometry (nanoLC-MS/MS) approaches in proteome research are reviewed from an analytical perspective. For comprehensive analysis of cellular proteins, analytical methods with higher resolution, sensitivity, and wider dynamic range are required. Miniaturized LC coupled with tandem mass spectrometry is currently one of the most versatile techniques. In this review, the current status of nanoLC-MS/MS systems as well as data management systems is addressed. In addition, the future prospects for complete proteomics are discussed.

Abstract:TTSPM is a new computer program system for quantitative thick target analysis by nuclear microprobe based on PIXE analysis. It is multifunctional and most adaptable for different incident projectiles and various matrices while every process of production and detection of PIXE signals is considered including matrix attenuation and secondary fluorescence enhancement. This paper describes the principles of quantitative micro-PIXE analysis and the computer calculation methods, demonstrates the structure and the functions of TTSPM system, shows several results of standards comparisons and applications of the system in geology, material and other fields. Some new methods for resolving the overlapping peaks in PIXE spectra and eliminating the artifacts in 2D elemental maps are presented.
