Abstract:A new βcyclodextrin derivative with the anion side-arm, mono[6-deoxy-6-(2-sodium thio-1,2-dicyane ethylenylthio)]-βcyclodextrin (6-mnt-βCD), and its inclusion complex with ferrocene (6-mnt-βCD/Fc), have been prepared and characterized by elemental analysis, IR spectroscopy, UV spectroscopy, mass spectrometry, 13 C-NMR spectroscopy, thermogravimetry, and cyclic voltammetry (CV). Thermogravimetry analysis of the compound shows that the thermal stabilities of both the host and the guest in the inclusion complex have been greatly improved due to an additional interaction between the side-arm of the host and the guest. The interplay between the guest and the host with side-arm in the complex resulted in smaller positive potential shifts in CV compared to that in the inclusion complex βCD/Fc.

Abstract:Inclusion compounds of ferrocene and its derivatives with α-, β-, and γ-cyclodextrins have been prepared in a crystalline state in high yields. β-Cyclodextrin and γ-cyclodextrin formed 1 : 1 stoicheiometric inclusion compounds. α-Cyclodextrin formed 2 : 1 (host:guest) complexes with ferrocene and its monosubstituted derivatives, but did not form complexes with 1,1′-disubstituted derivatives. The complexes of α- and β-cyclodextrin with ferrocene are thermally stable and do not liberate ferrocene on heating at 100 °C in vacuo. The inclusion compounds were characterized by 1H n.m.r., i.r., u.v., and circular dichroism spectroscopy. A large positive induced Cotton effect was observed in the case of complex of β-cyclodextrin with ferrocene, while the complex of γ-cyclodextrin showed a negative spectrum. The binding mode is discussed.

Abstract:The inclusion complexation behaviour of ferulic acid (FA) with -cyclodextrin (-CD) and hydroxypropyl--cyclodextrin (HP--CD) was investigated by UV-vis, fluorescence and 1H NMR spectroscopy. Since the guest may exist in either anionic or neutral form, the experiments were performed at different pH values. The stoichiometry and association constants of the complexes were determined by nonlinear regression analysis. The phase-solubility studies indicated that the water solubility of FA was improved through complexation with -CD and HP--CD. An increase in the antioxidant reactivity was observed when inclusion complexes that FA formed with CDs were studied. Based on the NMR data, the spatial configurations of FA/-CD and FA/HP--CD complexes were proposed, which suggested that FA entered into the cavity of -CD from the narrow side, with the lipophilic aromatic ring and ethylenic moieties inside the CD cavity, and the -COOH group was close to the wider rim and exposed outside the cavity. A theoretical study of the complexes using molecular modelling gives the results in good agreement with the NMR data.

Abstract:A new water-soluble inclusion complex of ferrocene (Fc) with β-cyclodextrin polymer (β-CDP) was prepared by a facile strategy and characterized by 1H NMR spectroscopy, elemental analysis, powder X-ray diffractometry, thermogravimetry, UV–vis spectroscopy and cyclic voltammetry. Compared with Fc and the inclusion complex of Fc with β-cyclodextrin (Fc-β-CD), the solubility of ferrocene-β-cyclodextrin polymer (Fc-β-CDP) was greatly enhanced due to the water-soluble β-CDP host. The ratio of β-cyclodextrin (β-CD) unit in β-CDP to Fc was determined as 1:1. At 25°C, the dissociated constant of Fc-β-CDP was measured as 3.65mM by UV–vis spectroscopy and cyclic voltammetry. The electrochemical properties of Fc-β-CDP in water were studied. The diffusion coefficients of oxidation state and reduction state were calculated as 3.52×10−7cm2s−1 and 3.93×10−7cm2s−1. The resulting value of standard rate constant was measured as 1.95×10−3cms−1. The diffusion activation energy was calculated as 21.8kJmol−1.

The formation constant of the inclusion complex was determined by fluorescence spectra. The results indicate that the stoichiometry of the inclusion complex of beta -cyclodextrin with 5, 10, 15, 20-tetrakis(4-proplpyidinium) porphyrin bromide is 2:1. The molecular modeling provides further evidence for the formation of the inclusion complex.

Abstract:Bromination of the inclusion complex of trans-ferrocenylchalcone derivatives with β-cyclodextrin was studied. In the absence of β-cyclodextrin, the analogous ferrocenylchalcone does not undergo bromination at room temperature, but its inclusion complex in β-cyclodextrin brominates smoothly and gives the erythro-dibromide derivatives. This complexation does not induce any variation in the bromination pattern and the ferrocenyl moiety is not attacked in the reaction. These observations have been accounted for in terms of guest–host interaction patterns. The formation of complex and dibromination product has been confirmed by elemental analyses, IR, 1H-NMR, 13C-NMR, UV–Vis and electrochemical methods.

Abstract:Calculations have been done on β-cyclodextrin 6-O-monobenzoate and its complexes using molecular mechanics MM2 force field. The guest molecules involved were n-hexane, n-heptane, n-octane, cyclohexane, methylcyclohexane, cyclooctane and methylcyclooctane. Owing to lack of dipole–dipole and hydrogen-bonding interactions in the inclusion complexation of the modified cyclodextrin, the calculations of the geometry of the complexes and the inclusion process indicated that the complex formation is stabilized by van der Waals interaction. The energy differences of the modified cyclodextrins by calculation show that the size, shape, conformation of the guest molecules play significant roles in the inclusion complexation.

Abstract:An inclusion system of embedding ferulic acid into β-cyclodextrin (FACD) with different host-guest stoichiometries was prepared by a co-precipitation method. Then, the physicochemical properties and release kinetics of the FACD were evaluated. The results of thermal analysis, X-ray diffraction, scanning electron microscopy, and Fourier transform infrared spectroscopy (FTIR) proved the successful embedding of FA into the β-cyclodextrin matrix. Four mathematical models were applied to adjust the ferulic acid release profile and identify preferential kinetics. The results of physicochemical properties confirmed the successful formation of the complexes. The loading capacity (LC) and encapsulation efficiency (EE) of the inclusion complex (1 : 0.5) were 41.0 ± 3.28 mg g-1 and 52.1 ± 2.31%, respectively, which were significantly higher than other molar ratios. The release behaviour revealed that loaded FA molecules under various host-guest stoichiometries obey different release models. While lower host-guest stoichiometry (1 : 0.5) provided desirable EE, the moderate host-guest stoichiometry (1 : 1) exhibited faster release behaviour. The FACD inclusion complex could be a promising bioactive material for food preservation.

Abstract:A single crystal of a unique 4:5 inclusion complex between beta-cyclodextrin and ferrocene was prepared and the structure was determined by X-ray crystallographic analysis, showing that four of the five ferrocenes were included in the cavities of four beta-cyclodextrins along the axial orientation of the tetramer, while another was co-included between two beta-cyclodextrins from the equatorial orientation.

Abstract:Abstract In our work, the inclusion ability and binding pattern of flavonoids (catechin, iso‐liquiritigenin, luteolin, puerarin, rutin, and curcumin) with β‐cyclodextrin (β‐CD) and curcumin (CUR) with CDs (α‐CD, β‐CD, γ‐CD, dimethyl‐β‐CD, Hydroxypropyl‐β‐CD, and glucosyl‐β‐CD) were studied by molecular docking. The results showed that CUR among the six flavonoids was more likely to form stable inclusion complexes with β‐CD, and the HP‐β‐CD is the most suitable for CUR inclusion among the 6 types of CD. Phase solubility studies showed that CUR with β‐CD or HP‐β‐CD, respectively, could form stable inclusion complexes in a stoichiometric ratio of 1 : 1. The CUR/β‐CD and CUR/HP‐β‐CD inclusion complexes (ICs) were prepared by freeze‐drying method. The successful preparation of IC was confirmed by SEM images, FT‐IR spectra, and TG‐DSC. The results of in vitro release showed that the release of CUR from IC prepared with β‐CD or HP‐β‐CD could be improved significantly, and the release effect of HP‐β‐CD was better than that of β‐CD. The inclusion with CD could be used to improve the solubility and bioavailability of CUR.

Abstract:In this study, β-carotene and β-cyclodextrin inclusion complex are prepared by encapsulation. Optimal condition are established by orthogonal test: mol ratio of β-CD and β-C4.5:1, temperature40℃, rotate speed600r/min, time7h, and inclusion rate reach 93.18%. The accelerative experiment of light stability carrying out at 40℃,5000LX shows that the stability of β-carotene are improved significantly by encapsulation.

Abstract:The formation of supramolecular inclusion complexes between luteolin and five cyclodextrins namely β-cyclodextrin (β-CD), methyl-β-cyclodextrin (M-β-CD), hydroxyethyl-β-cyclodextrin (HE-β-CD), hydroxypropyl-β-cyclodextrin (HP-β-CD) and glucosyl-β-cyclodextrin (G-β-CD) was investigated. Results from phase-solubility studies showed that luteolin formed 1:1 stoichiometric inclusion complexes with these cyclodextrins with the G-β-CD complex displaying the greatest stability constant. The supramolecular structure of the luteolin/G-β-CD complex was investigated by ultraviolet-visible spectroscopy (UV), Fourier transform infrared spectroscopy (FT-IR), scanning electron microscopy (SEM), differential scanning calorimetry (DSC) and X-ray diffractometry (XRD). Results showed clearly the formation of a supramolecular complex in which the guest molecule, luteolin, was entrapped inside the cavity of the host, G-β-CD. The close association between luteolin and G-β-CD resulted in changes in some of the characteristic spectral, phase transitional and morphological properties of luteolin. Furthermore, molecular docking study showed that the complex was formed with the B ring of luteolin inserted into the cavity of G-β-CD.

Abstract:In this study, ferulic acid (FA) was covalently bound to one of the primary hydroxyls of β-cyclodextrin (β-CD) via an ester linkage to improve its stability and biological activity. Experiments using UV, FTIR, FS, NMR, MS and FE-SEM were performed to characterize the structure of the FA-β-CD conjugate. Physicochemical properties such as solubility, crystallinity, thermal stability and photo-stability of the conjugate were also investigated, and the results revealed that the conjugate was in an amorphous form with improved thermal stability and photostability compared with the FA/β-CD inclusion complex. Furthermore, in vitro cytotoxicity of the FA-β-CD conjugate against HepG2 cell was enhanced with effective growth inhibition. Taken together, the present study indicates that the FA-β-CD conjugate might be a novel and potential functional composition for application in food and medicine.

Abstract:The inclusion process involving β-cyclodextrin (β-CD) and quercetin has been investigated by using the PM3 quantum-mechanical semi-empirical method. In the β-CD quercetin inclusion complex, a large portion of the flavonoid skeleton is included in the β-CD cavity and the bond connected ring B with ring C is inclined to the molecular axis of β-CD. The orientation in which the B ring of the guest molecule located near the secondary hydroxyls of the β-CD cavity is preferred in energy. One intermolecular hydrogen bond is formed. The molecular modeling results are in agreement with the NMR observations and molecular dynamics (MD) simulations. The statistical thermodynamic calculations at 1atm and 298.15K by PM3 demonstrate that 1:1 quercetin/β-CD complex is favored by a negative enthalpy change.
