Abstract:The synthesis of analogues of the potent acetylcholinesterase inhibitor, Territrem B, was carried out starting from the naturally occurring jujubogenin glycosides. Dihydrojujubogenin-2-en-1-one (1), a dammarane derivative that possesses a skeleton and pharmacophore partially similar to those of Tenitrem B, was synthesized via three different paths. The derivative 21, which contains two potential pharmacophores, was also synthesized. The anti-AChE activity of the analogues was measured. Abstract:A mimetic synthesis of a potent acetylcholinesterase inhibitor, Territrem B, was carried out from the naturally occurring jujubosides. The anti-AChE activity of the product, which possesses the 2-en-1-one pharmacophore and the aromatic ring, was measured. The aromatic ring moiety seemed to be less influencing when comparing with the 2-en-1-one moiety.

Abstract:AIM:To design and synthesize novel AchE inhibitors.METHODS:The condensation of 2-bromo-5, 6-dimethoxy-indan-1-one with various aminoalkyl phenols in the presence of K2CO3 and acetonitrile gave the corresponding title compounds, and the in vitro AchE and BchE inhibitory activities were evaluated by the modified Ellman method.RESULTS:Sixteen novel target compounds 8a - p were synthesized, their structures were confirmed by 1H NMR, MS, IR and elemental analysis. Preliminary pharmacological test demonstrated that most of these compounds displayed high AchE inhibitory activities, the IC50 of the most potent inhibitor 8h was 50.0 nmol x L(-1), similar to that of Huperzine A (IC50 = 53.0 nmol x L(-1)), while all the compounds were almost inactive against BchE.CONCLUSION:2-Phenoxy-indan-1-one derivatives exhibit high activities of AchE inhibition and are worthy of further investigation.

Abstract:Two series of territrem B analogs, i.e., 5 - 10, containing both the 2-en-1-one-A-ring and the aromatic-E-ring pharmacophores were designed and synthesized from jujubogenin (4a). The anti-acetylcholinesterase (anti-AChE), anti-caspase-3, and other biological activities of these territrem-B analogs and their intermediates were assessed. Compound 9b, 22a, and 24f were shown to be weak inhibitors of AChE. None of the synthesized compounds exhibited significant inhibitory activity on caspase-3. On the other hand, compounds 22e, 24a, 7b, and 8a showed mild cytotoxicity on cultured KB cells, with IC50, values of 2.0, 3.5, 6.5, and 14 mum, respectively. In addition, compounds 23b and 5f were active against injury arising from oxygen-glucose deprivation.

Abstract:Two series of territrem B analogues (6, 11) were designed and synthesized from jujubogenin 2. Compound 11c inhibited AChE with the ratio of 70% at 10-4 mol/L. Compounds5a, 5b, 6a and 11b showed moderate cytotoxicity on cultured KB cells at 10-6 mol/L.Compounds 5c and 6b alleviated injury arising from oxygen-glucose deprivation (OGD).

Abstract:Jatrorrhizine was considered as one of the active constituents of Coptis chinensis Franch. Herein, jatrorrhizine derivatives with substituted amino groups linked at the 3-position were designed, synthesized, and biologically evaluated as inhibitors of acetylcholinesterase. Jatrorrhizine derivatives inhibited the activity of acetylcholinesterase (AChE) to a greater extent than the lead compound jatrorrhizine. All these jatrorrhizine derivatives were proved to be potent inhibitors of acetylcholinesterase (AChE) with submicromolar IC50 values, but less sensitive to butyrylcholinesterase (BuChE), which suggests that these jatrorrhizine derivatives are selective for AChE/BuChE. Compound 3g gave the most potent inhibitor activity for AChE (IC50 = 0.301 μM), which is greater than the lead compound jatrorrhizine. All these results demonstrated that these jatrorrhizine derivatives are potential inhibitors for AChE.

Abstract:Some 23 analogues of the potent acetylcholinesterase (AChE) inhibitor territrem B (1) were designed, synthesized, and tested for their biological activities. Some of the new synthetic derivatives exhibited IC50 values for AChE inhibition in the upper micromolar range. Molecular-modeling studies indicated that a planar conformation seems to be crucial for AChE inhibition. The two N-atoms of the piperazine moieties in 5o, 5p, and 5r might further enhance the inhibitory effects. The cytotoxicities of selected compounds against six human tumor cell lines were also determined.

Abstract:A new series of flavonoid derivatives have been designed, synthesized and evaluated as potent AChE inhibitors. Most of them showed more potent inhibitory activities to AChE than rivastigmine. The most potent inhibitor isoflavone derivative 10d inhibit AChE with a IC(50) of 4 nM and showed high BChE/AChE inhibition ratio (4575-fold), superior to donepezil (IC(50)=12 nM, 389-fold). Molecular docking studies were also performed to explore the detailed interaction with AChE.

Abstract:A series of novel 2-methoxy-phenyl dimethyl-carbamate derivatives were designed, synthesized and evaluated as site-activated MTDLs based on rivastigmine and curcumin. Most of them exhibited good to excellent AChE and BuChE inhibitory activities with sub-micromolar IC50 values. Among all the compounds, 6a demonstrated the most potent AChE inhibition with IC50 value of 0.097μM, which is about 20-fold than that of rivastigmine. In addition, the three selected compounds 5a, 6a and 6e demonstrated inhibitory activity against Aβ self-aggregation similar to cucurmin in TEM assay, which is obviously different from the weak activity of rivastigmine. Moreover, the hydrolysate of 6a (compound 7) also showed potent ABTS(+) scavenging and moderate copper ion chelating activity in vitro.

Abstract:Some 2-en-1-one-ebelactonyl-20-yl-benzoyl esters as Territrem B analogues were prepared with new method.Aldehyde 2 was treated with Li_2CO_3 in N,N-dimethylacetamide(DMAc) to give(2-en)-ebelactonyl-13-aldehyde which was oxidation with N-bromosuccinimide(NBS) under (irradiation)gave the corresponding 2-en-1-bromo-acyl bromide 4 in one step.Then 4 was esterified(directly) with substituted phenols to afford the corresponding esters 5 in one step.And compound 6 can be prepared from 5 through hydrolysis and oxidation in one step.During these process,the yield of 6 was(increased)5%～30% than the(original) method,and five new compounds 3 and 5b～5e were(prepared.) Among all of the compounds only 6a showed 17% inhibitory activity to AChE in(10~(-4) mol/L.)

Abstract:OBJECTIVE To study the design and synthesis of new territrem B analogues and their biological evaluation.METHODS New territrem B analogues 10a～10l with the common structure of 2H-pyran-2-one were designed and synthesized from ethyl acetoacetate.The cytotoxicities and inhibitory activities on acetylcholinesterase and butylcholinesterase of these synthetic analogues were examined by MTT and ELISA methods,respectively.RESULTS New analogues of territrem B were afforded and the structures were confirmed by()~(1)H-NMR,ESI-MS and IR spectra.Compound 10f showed IC_(50) value to acetylcholinesterase at 6.98×10~(-5) mol·L~(-1) and all target compounds showed no inhibition to butylcholinesterase at 4.0 g·L~(-1).CONCLUSION The bioassay results indicated that 10f exhibited potential AChE inhibitory effect and better selectivity of AChE.
Total Synthesis of Pulmonarin B and Design of Brominated Phenylacetic Acid/Tacrine Hybrids: Marine Pharmacophore Inspired Discovery of New ChE and Aβ Aggregation Inhibitors

Abstract:A marine natural product, pulmonarin B (1), and a series of related tacrine hybrid analogues were synthesized and evaluated as cholinesterase (ChE) inhibitors. The in vitro ChE assay results revealed that 1 showed moderate dual acetylcholinesterase (AChE)/ butyrylcholinesterase (BChE) inhibitory activity, while the hybrid 12j proved to be the most potent dual inhibitor among the designed derivatives, being almost as active as tacrine. Molecular modeling studies together with kinetic analysis suggested that 12j interacted with both the catalytic active site and peripheral anionic site of AChE. Compounds 1 and 12j could also inhibit self-induced and AChE-induced Aβ aggregation. In addition, the cell-based assay against the human hepatoma cell line (HepG2) revealed that 1 and 12j did not show significant hepatotoxicity compared with tacrine and donepezil. Taken together, the present study confirmed that compound 1 was a potential anti-Alzheimer's disease (AD) hit, and 12j could be highlighted as a multifunctional lead compound for anti-AD drug development.

Abstract:Spiroterreusnoids A-F, six undescribed spiro-dioxolane-containing adducts bearing 3,5-dimethylorsellinic acid-based meroterpenoid and 2,3-butanediol moieties were isolated from the endophytic fungus Aspergillus terreus Thom from Tripterygium wilfordii Hook. f. (Celastraceae). The structures of these adducts were established by spectroscopy, single-crystal X-ray diffraction, and experimental electronic circular dichroism (ECD) measurements. Spiroterreusnoids A-F represent the first examples of adducts composed of 3,5-dimethylorsellinic acidbased meroterpenoids. It is noteworthy that spiroterreusnoids A-F possessing a spiro-dioxolane moiety exhibited potential abilities in inhibiting BACE1 (IC50 values ranging from 5.86 to 27.16 mu M) and AchE (IC50 values ranging from 22.18 to 32.51 mu M), while the other analogues without this fragment displayed no such activities. Taken together, spiroterreusnoids A-F represent the first multitargeted natural adducts that could inhibit BACE1 and AchE, and might provide a new template for the development of new anti-Alzheimer's disease drugs.

Abstract:As a part of our project aimed at developing new agents of potential application in AD, a new series of 2-phenoxy-indan-1-one derivatives which possess alkylamine side chain were designed, synthesized, and evaluated for their inhibitory activity against AChE and BuChE. Most of the compounds were found to inhibit AChE in the nanomolar range. The optimum inhibitor 3g exhibited 34-fold increase in AChE inhibition than donepezil and displayed neuroprotective effect against H(2)O(2)-induced cell death.

Abstract:Euphorlactone A (1), a rare rearranged ent-atisane norditerpenoid with an undescribed 3-nor-2,4-olideent-atisane scaffold, and euphorlactone B (2), a new ent-atisane diterpenoid with an unprecedented seven-membered lactone ring C, were isolated from the roots of Euphorbia fischeriana. Their planar structures with absolute configurations were extensively elucidated by analysis of 1D and 2D NMR data, electronic circular dichroism (ECD) calculations, Rh-2(OCOCF3)(4)(-)induced ECD curves, and single-crystal X-ray diffraction. Euphorlactone A (ELA) showed a remarkable AChE (acetylcholinesterase) inhibitory activity (IC50 = 2.13 +/- 0.06 mu M and K-i = 0.058 mu M), which was five times stronger than that of the positive control (rivastigmine, IC50 = 12.46 +/- 0.82 mu M), and further in vitro enzyme inhibition kinetic analysis and molecular docking studies were performed to investigate the AChE inhibitory mechanism.
