Abstract:OBJECTIVE: The study attempted to develop extended release capsules of diazepam based on the principle of hydrodynamically balanced system. METHODS: The content of diazepam in HBS capsules was measured. The floating time, dissolution and stability of the capsules were studied. The gamma scintigraphy was used to monitor the transit of the HBS capsule in vivo. RESULTS: In vivo the HBS capsules floated for over 8 hours. Accelerating experiment showed both the content and total amount released after 12 hours of diazepam did not change apparently (P > 0.05), while the dissolution rate decreased slightly. The HBS capsules retained in stomach in fasting and non-fasting volumteers for longer time than the conventional ones, which was proved by gamma scintigraphy. CONCLUSION: HBS capsule of diazepam was extended release ones in vitro and could stay in stomach for longer time than conventional capsule.
