Abstract:The whole plants of Ajuga lupulina afforded five compounds, including three new clerodane diterpenes, lupulins A-C (1-3), whose structures were elucidated by spectral methods. Among these compounds, lupulins A (1) and B (2) as well as the acid hydrolysate (5) of lupulin D (4) showed antibacterial activities against Staphylococcus aureus, Pseudomonas aeruginosa, and Escherichia coli.

Abstract:Different neo-clerodane diterpenoids were isolated from a dichloromethane extract of Ajuga bracteosa depending on the isolation procedure used, owing to the labile nature of these tetrahydrofurofuran derivatives. Under "hydroxyl-free" purification conditions, both clerodin- and dihydroclerodin-type diterpenes were obtained [four new compounds, ajubractins A-D (1-4), along with clerodin (5), 3-epi-caryoptin (6), ajugapitin (7), 14,15-dihydroclerodin (8), 3-epi-14,15-dihydrocaryoptin (9), ivain II (10), and 14,15-dihydroajugapitin (11)]. When methanol-water mixtures were used for a C18 reversed-phase prepurification procedure and for semipreparative HPLC, the new ajubractin E (12) was also isolated along with 3 and 8-11, as previously, but 7 was the only tetrahydrofurofuran derivative obtained. Epimeric (15R and 15S) mixtures were obtained instead of 14-hydro-15-hydroxyclerodin derivatives [15-hydroxyajubractin C (13), 14-hydro-15-hydroxyajugachin A (14), and 14-hydro-15-hydroxyajugapitin (15)], along with 15-epi-lupulin B (16). The structures of the new compounds were elucidated by NMR and MS data analysis and by comparison with values previously reported. Antifeedant activity against Spodoptera littoralis larvae was evaluated for the compounds obtained.

Abstract:In order to explore the suitability of reversed-phase HPLC for the rapid isolation, identification and semi-preparative fractionation of neo-clerodane diterpenes from Ajuga remota, an extract from aerial parts of the plant was examined using a C18 column with UV detection and gradient elution with water:methanol. In addition to the known diterpenes ajugarins I, II, IV and V and clerodin, the presence and chromatographic behaviours of ajugapitin, dihydroajugapitin, dihydroclerodin and deacetylajugarin IV, not previously isolated from A. remota, were established. Presence of the epimeric mixture of 14-hydro-15-hydroxyclerodin (scutecyprol A) was also demonstrated together with that of 14-hydro-15-hydroxyajugapitin, the latter only in trace amounts. The structures of all isolated diterpenes were determined from NMR data. The anti-feedant activities of the isolated compounds against Spodoptera littoralis are also reported here.

Abstract:Five new compounds (1, 2, 7, 12, and 16), along with fifteen known ones, were isolated from Ajuga lupulina Maxim. Their structures were elucidated by spectroscopic data (MS, NMR) analysis, and the absolute configuration was determined by the experimental and calculated ECD spectra. Compound 16, with eight carbon atoms, was firstly isolated from the nature. All the isolates were assayed for their anti‐ferroptosis in RSL3‐induced HT22 mouse hippocampal neuronal cells. Among them, the abietane‐type diterpenoids (7‒11) significantly inhibited RSL3‐induced ferroptosis with EC50 values of 0.83 μM, 2.05 μM, 0.96 μM, 1.47 μM, and 1.19 μM, respectively.

Abstract:The phytochemical investigation of the leaves of Inula confertiflora, a medicinal plant endemic to Ethiopia, led to the isolation of 15 terpenoids; 1β‐hydroxy‐α‐costic acid (1), 3α‐hydroxycostic acid (2), isotelekin (3), asperilin (4), carabrone (5), carpesioline (6), graveolide (7), inuviscolide (8), 8‐epi‐inuviscolide (9), 1β,4β‐dihydroxy‐5α(H)‐guaia‐10(14),11(13)‐dien‐8α,12‐olide (10), isoinuviscolide (11), 4β,10β‐dihydroxy‐5α(H)‐1,11(13)‐guaidien‐8α,12‐olide (12), 4β,10β‐dihydroxy‐1β(H)‐5α(H)‐guai‐11(13)‐en‐8α,12‐olide (13), 4β,10α‐dihydroxy‐1β(H)‐5α(H)‐guai‐11(13)‐en‐8α,12‐olide (14), 4β,10α‐dihydroxy‐1α(H)‐5α(H)‐guai‐11(13)‐en‐8α,12‐olide (15). Herein, structural elucidation and full NMR data for compound 1 are presented for the first time. The structures were elucidated using NMR, HRESIMS, and by comparison with literature data. The relative configurations were defined by NOESY correlations and single‐crystal X‐ray crystallography. Herein, crystallography data of 6 and 7 were reported for the first time. The antibacterial efficacy of some of the isolated compounds was evaluated against two commonly dispersed environmental strains of Escherichia coli and Staphylococcus aureus. Compounds 1, 3, 6, 7, and 8 exhibited moderate antibacterial activities against the tested organisms. The chemotaxonomic significance of compounds is discussed.

Abstract:A new antimicrobial eudesmanolide, 1-oxo-8 alpha-hydroxy-11 alpha H-eudesm-4-en-12,6 alpha-olide (1), was isolated from a medicinal plant Artemisia giraldii and its structure was elucidated by spectroscopic methods (IR, MS, H-1 NMR, C-13 NMR, H-1-H-1 COSY and NOESY). Antimicrobial bioassay indicated that this compound inhibited the growth of human pathogenic bacteria Staphylococcus auerus, Bacillus subtilis and Escherichia coli as well as human opportunistic pathogenic fungi Candida tropicalis, Gecotrichum candidun, Aspergillus flavus and A. nigers.

Abstract:Four new diterpenoids including one cis clerodane-type (1) and three highly oxygenated labdane-type diterpenes (2–4) were isolated from the aerial parts of Leonurus macranthus. Their structures were elucidated on the basis of spectroscopic data (NMR, UV, IR, and MS). Compound 1 represents the first example of cis clerodane-type diterpene in the plants of Leonurus genus. Abstract:As part of our continuing efforts toward the discovery of the biologically active diterpenoids from medicinal plants, six new C‐ring aromatized abietane diterpenoids, clerodenoids A–F (1–6), were isolated from Clerodendrum chinense. These C‐ring aromatized compounds mainly belonged to the rearranged abietane skeletons of 17(15→16)‐abeo‐abietane (2–4) and 17(15→16),18(4→3)‐diabeo‐abietane (5 and 6). Their structures were elucidated by extensive spectroscopic data, X‐ray crystallography, and ECD calculation. Abstract:Nine clerodane diterpenes, solidagoic acids C-I (1-7), cleroda-3,13(14)-dien-16,15:18,19-diolide (8) and cleroda-3,13(14)-dien-15,16:18,19-diolide (9) were isolated and characterised from the ethanol-ethyl acetate (1:1) extract of Solidago virgaurea. Abstract:The Lamiaceae plant Ajuga forrestii Diels is a traditional Chinese herbal medicine with abundant glandular trichomes (GTs), but their chemistry and biological functions remain uninvestigated. Here, a panel of six highly functionalized neo-clerodane diterpenoids was localized to the peltate GTs of A. forrestii using laser microdissection coupled with HPLC analysis, indicating that the GTs of A. forrestii are an excellent material for the elucidation of the yet unclear biosynthetic pathway of natural neo-clerodane diterpenoids. In addition, four undescribed neo-clerodane diterpenoids with an acyclic C-9 side chain including two pairs of 1:1 mixture of inseparable diastereomers, ajuforrestins D-G, were isolated from the fresh leaves of A. forrestii together with six known compounds. The structures of the undescribed compounds were elucidated by spectroscopic (including 1D and 2D NMR and HR-ESI-MS) analyses. Biological assays indicated that the major GT compound ajugacumbin B and undescribed ajuforrestins D/E showed antifeedant activity against Helicoverpa armigera, suggesting that neo-clerodanes in A. forrestii should be involved in plant defence against insects. Moreover, the abietane diterpenoid ajuforrestin B exhibited significant anti-inflammatory activity on the secretion of interleukin-2 (IL-2) and cytotoxicity against three cancer cell lines, NCI-H1975, HepG2 and MCF-7, suggesting that ajuforrestin B could positively contribute to the therapeutic effects of this traditional Chinese medicine.

Abstract:Two new flavones, 4',6,7-trihydroxy-3',5'-dimethoxy-flavone (2) and 5',5- dihydroxy-3',4',8-trimethoxyflavone (3) were isolated from Artemisia giraldii and their structures were identified by spectroscopic methods. These two new flavones showed antibiotic activity against Staphylococcus aureus, Sarcina lutea, Escherichia coli, Pseudomonas aeruginosa, Proteus sp. Aspergillus flavus, and Trichoderma viride.

Their structures were elucidated on the basis of extensive spectroscopic analysis. Two new compounds were evaluated for their cytotoxic activities against four strains of cultured tumor cells.

Abstract:Six compounds including three new polyketide ones named eleubosas A-C (1–3) were isolated from the active frations of Eleutherine bulbosa. Their structures were elucidated by extensive spectroscopic methods, including NMR, MS and IR spectroscopic analyses data. All the isolates were evaluated against three pathogenic bacteria, Staphylococcus aureus, Escherichia coli, and Pseudomonas aeruginosa, and the results showed that compounds 1 and 2 displayed moderate inhibitory activities against E. coli with MIC values both 12.5 μg/mL, which are consistent with the clinical applications and need further studies.

Abstract:In this study, lupinifolin (1) and its natural analogues, mundulin (2), minimiorin (3), khonklonginol H (4), flemichin D (5), and eriosemaone A (27), were obtained by chemical synthesis for the first time. Key steps involved an electrocyclization to build the linear pyran rings and a Claisen/Cope rearrangement to install the 8-prenyl substituents. All compounds were assessed for their in vitro antimicrobial activities against clinically relevant human pathogens, including one Gram-negative bacterial strain (E. coli ATCC 25922) and four Gram-positive bacterial strains (S. aureus ATCC 29213, E. faecalis ATCC 29212, MRSA21-5, and VRE ATCC 51299). The result indicated that eriosemaone A (27) was the most potent one against Gram-positive bacteria, with minimum inhibitory concentrations in the range of 0.25-0.5 μg/mL. Mechanistic studies indicated that 27 has good membrane-targeting ability to bacterial inner membranes and can bind to phosphatidylglycerol and cardiolipin in bacterial membranes, thereby disrupting the bacterial cell membranes and causing bacterial death.
