Abstract:An improved process was reported to solve the problems such as low yield and high costs in the synthesis of 3-n-propyl-2, 4-dihydroxyacetophenone, which is a key intermediate of Leukotriene receptor antagonist. The target compound was synthesized starting from m-dihydroxybenzene, via acylation, allylation, Claisen rearrangement and hydrogenation reduction. The optimal reaction conditions were as follows. The acylation reaction which used acetic acid as acylation agent and ZnCl 2 as catalyst was carried out under reflux for 5 h, and 77.9% yield was obtained. The allylation reaction using allyl chloride as allylation reagent and NaBr, KI as catalyst was carried out at 40°C for 2 h and continued at 70°C for 14 h, to give 98.6% yield. In Claisen rearrangement carried out at 205°C for 4.5 h, diphenyl ether was used as reaction solvent and NaCl was used as catalyst, and the yield was improved to 67.4%. In the final step, the hydrogenated reduction with Pd/C (w(Pd) is 5%) as catalyst was carried out at 45°C for 6 h, and 88.6% yield was obtained. The process is much of industrial value because of high yield, cheap and available materials, and convenient operations. The structure of the main compounds was confirmed by 1H-NMR, GC-MS and IR.

Abstract:The practical synthesis of the representative nonsteroidal anti-inflammatory drug ketoprofen has been accomplished in 44% overall yield on the decagram scale. The key features in this synthetic work involve a TiCl4-Et3N mediated aldol/enollactonization annulation to affect dihydrobenzofuranone core formation and the use of a highly efficient pyrolytic aromatization condition to facilitate the construction of the vital 2-arylpropionic acid (2-APA) moiety. Furthermore, the avoidance of hazardous chemicals and crystallization of several intermediates would be greatly beneficial to the scalable synthesis of ketoprofen.

Abstract:As an intermediate for COX-2 inhibiters,4-(methylthio)benzeneacetic acid(Ⅳ) was prepared via Friedel-Crafts and Willgerodt-Kindler reaction from thioanisole(Ⅰ).When the molar ratio of Ⅰ to acetyl chloride and AlCl_3 was 1∶1.1∶1.3 and CS_2 was used as solvent,after reacting at(0°C) for 6 h the yield of 4-(methylthio) acetophenone(Ⅱ) was 75.9%.Ⅱ was refluxed 5 h with morpholine and sulfur to give thioacetmorpholide(Ⅲ),then Ⅲ was refluxed in 10% NaOH for 10 h to give Ⅳ with an overall yield of 63.5% and purity of 99.0%(by HPLC).The structure was confirmed by IR and()~1H NMR.

Abstract:An efficient ring-closing reaction was developed for the synthesis of spirocyclopropane quinolinediones from 2,2-disubstituted 2-benzoylacetamides in the presence of [bis-(trifluoroacetoxy)iodo]benzene (PIFA). This reaction proceeds at room temperature in the absence of a metal catalyst to provide spirocyclopropane quinolinediones in good to excellent yields. Furthermore, these compounds were readily converted to the corresponding pyrrolo[3,2-c] quinolinones to provide tricyclic ring structures similar to those found in the natural products martinellic acid and martinelline via an intermolecular amine ringopening cyclization reaction.

Abstract:In the past two decades, substantial advances have been made on the asymmetric alkyne functionalization by the activation of inert alkynes. However, these asymmetric transformations have so far been mostly limited to transition metal catalysis, and chiral Bronsted acid-catalyzed examples are rarely explored. Here, we report a chiral Bronsted acid-catalyzed dearomatization reaction of phenol- and indole-tethered homopropargyl amines, allowing the practical and atom-economical synthesis of a diverse array of valuable fused polycyclic enones and indolines bearing a chiral quaternary carbon stereocenter and two contiguous stereogenic centers in moderate to good yields with excellent diastereoselectivities and generally excellent enantioselectivities (up to >99% enantiomeric excess). This protocol demonstrates Bronsted acid-catalyzed asymmetric dearomatizations via vinylidene-quinone methides.

Abstract:A metal-free strategy has been developed for the α-arylation of carboxylic acids, secondary amides, and esters employing arenes as key reagents. This process entails the Lewis-acid catalyzed reductive Friedel-Crafts alkylation of arenes utilizing α-ketoacids, facilitated by silane in HFIP solvent. The transformation exhibits exceptional functional group tolerance, enabling late-stage functionalization of natural products. This one-step protocol has been successfully used to synthesize commercially available drugs, such as adiphenine, piperidolate, derivatives of ketoprofen, ibuprofen, flurbiprofen, and the pesticide bromopropylate.

Abstract:To discover an efficient and convenient method to synthesize C2-arylacylated benzothiazoles as potential drug scaffolds, a novel [bis(trifluoroacetoxy)iodo]benzene(PIFA)/KOH synergistically promoted direct ring-opening C2-arylacylation reaction of 2H-benzothiazoles with aryl methyl ketones has been developed. Various substrates were tolerated under optimized conditions affording the C2-arylacylation products in 70–95% yields for 38 examples. A plausible mechanism was also proposed based on a series of controlled experiments.

Abstract:A lipase-catalyzed irreversible resolution of 2-arylpropionic acids was developed using the corresponding racemic vinyl esters as activated substrates. The obtained products, (S)-ketoprofen vinyl ester, (S)-naproxen vinyl ester and (S)-ibuprofen vinyl ester would be useful as significant monomers for polymeric drug. The effect of enzyme sources, solvent, water amount and temperature on the hydrolysis resolution was investigated. Optically active, polymerizable ketoprofen prodrug can be obtained with excellent enantioselectivity (ee∼90%) after optimization of the reaction conditions. Lipased-catalyzed transesterification of (R,S)-ketoprofen vinyl esters with various alcohols was also studied. The use of vinyl ester effectively enhanced the enantioselectivity compared with other common ester, such as ethyl and butyl esters.

Abstract:A highly regioselective and efficient method has been developed for synthesizing novel β -amino alcohols fused with dihydropyrimidin-2-one. This method utilizes the enzyme Novozyme-435 to catalyze the reaction between epoxides and various aliphatic amines in acetonitrile. Novozyme-435 outperformed other catalysts, including Porcine Pancreatic Lipase (PPL), Pseudomonas aeruginosa lipase (PAL), and Candida rugosa lipase (CRL). This process yielded two series of β -amino alcohols (compounds 8a-h and 9a-h ), whose structures were confirmed through IR, NMR ( 1 H, Citation 13 C), and HRMS analyses. The anti-inflammatory and antioxidant properties of these compounds were evaluated, revealing mild to moderate inhibition of TNF- α -induced ICAM-1 expression in primary human endothelial cells, with compounds 9a and 9c showing approximately 60% inhibition. Antioxidant activity, assessed using the DPPH (2,2-diphenyl-1-picrylhydrazyl) method, indicated that compounds 9a , 9b , 9c , and 9 g had the superior activity than others. This study highlights the potential of these β -amino alcohols fused with dihydropyrimidin-2-one as anti-inflammatory and antioxidant agents.

Abstract:Ketoprofen is the non-steroidal anti-inflammatory drug which is a propionic acid derivative and is a non-selective COX-inhibitor. It is frequently used in case of osteo and rheumatoid arthritis and other inflammatory conditions. This drug having side effect of gastric ulcer which occur due to free carboxylic acid group present in its structure. Here, we synthesized the ester linked prodrugs of ketoprofen conjugated with different phytophenols. All the synthesized chemical entities were characterized by IR-spectroscopy, 1H-NMR, 13C-NMR and mass spectrometry. All the prodrugs showed significant analgesic and anti-inflammatory effect and less ulcerogenic potential.

The transformation is highly efficient and mild, providing significant advantages over existing protocols. Notably, the method exhibits exceptional tolerance towards various functional groups, enabling late-stage functionalization of pharmaceutical compounds and natural products such as Thymol, and Sesamol. The reaction mechanism has been studied through control experiments, providing valuable insight. This one-step reductive Friedel–Crafts type protocol has been successfully used in the synthesis of various commercially available drugs, such as Adiphenine, Piperidolate, derivatives of Ketoprofen, Ibuprofen, and Flurbiprofen, and Bromopropylate pesticide. Furthermore, after the gram-scale synthesis, the solvent (HFIP) was recovered, demonstrating the method's suitability for industrial applications.

Abstract:The efficient total syntheses of Nigrasin I and Kuwanon C, two biologically interesting natural flavonoids with two regioisomeric isoprenyl side chains, were realized for the first time starting from commercially available 1-(2,4,6-trihydroxyphenyl)ethanone via a linear reaction sequence of 11 steps with the overall yields of 22% and 21%, respectively, in which the selection of protective groups and the Claisen rearrangement of the allylated 16 featured the synthetic strategy. The use of acetic anhydride as the solvent and the employment of microwave irradiation are disclosed to be critically important in the efficient and selective Claisen rearrangement.

Abstract:A facile one-pot synthesis of functionalized (E)-β -arylvinyl bromides bearing various 4-alkoxycarbonyl and 4-aryloxycarbonyl groups was achieved by debrominative decarboxylation of anti-2,3- dibromo-3-(4-carboxyphenyl)propanoic acid in the presence of AgOAc and subsequent esterification in the presence of dicyclohexyl carbodiimide and dimethylaminopyridine. This method gives (E)-β - arylvinyl bromides in high stereoselectivities and high yields, and tolerates aldehyde, ester, and nitro functional groups.

Abstract:Prostaglandins have garnered significant attention from synthetic chemists due to their exceptional biological activities. In this report, we present a concise chemoenzymatic synthesis method for several representative prostaglandins, achieved in 5 to 7 steps. Notably, the common intermediate bromohydrin, a radical equivalent of Corey lactone, is chemoenzymatically synthesized in only two steps, which allows us to complete the synthesis of prostaglandin F 2α in five steps on a 10-gram scale. The chiral cyclopentane core is introduced with high enantioselectivity, while the lipid chains are sequentially incorporated through a cost-effective process involving bromohydrin formation, nickel-catalyzed cross-couplings, and Wittig reactions. This cost-efficient synthesis route for prostaglandins holds the potential to make prostaglandin-related drugs more affordable and facilitate easier access to their analogues.

Abstract:This paper investigated a rapid one-pot synthesis method of some substituted (9-carbazoly)-carboxylic acids(3a-3m). The reaction was carried out by mixing of the 9H-carbazoles derivatives with bromo-esters and potassium hydroxide using DMF as solvent under microwave irradiation without catalyst. The optimized experiment conditions were 4-8 min irradiation time and 375 W microwave power. The required range of molar ratio of KOH to substituted carbazole was 3-5. The biological activities of some compounds have been tested. They have medium to weak antimicrobial activities.
Synthesis, hydrolysis kinetics and pharmacological evaluation of aceclofenac prodrugs

Abstract:The mutual prodrugs of aceclofenac with various naturally available antioxidants; menthol, thymol, eugenol, guiacol and vanillin have been synthesized by the DCC coupling method, purified and characterized by spectral data, as well as, partition coefficient, solubility and hydrolytic studies. The title compounds have more lipophilic character as compared to the parent moieties and good stability in acidic environment, which is prerequisite for the oral absorption of the drug. Under gastric as well as intestinal pH conditions these prodrugs showed variable susceptibility towards hydrolysis. The synthesized derivatives were evaluated for antiinflammatory, analgesic activities and ulcerogenic potential. Prodrug showed improved solubility in organic solvents, which implies lipophilic character of ester prodrugs and were also found to be chemically stable in acidic environment. The aceclofenac mutual prodrugs showed improved analgesic and anti-inflammatory activities with reduced ulcerogenicity.

Abstract:We report the photo-mediated 1,2-aryl migration of 2‑chloro-1-arylpropanones to 2-arylpropionic acids using HCOONa as an acid scavenger. This pragmatically focused study obviates the multiple-step sequence in the industrially employed, ZnO-promoted rearrangement strategy, and offers rapid access to various 2-arylpropionic acids under environmentally friendly conditions. Furthermore, the successful transfer of this batch photochemistry to a continuous flow platform led to improved scalability and enabled the gram-scale synthesis of loxoprofen.
