Abstract:An unexpectedly high yield of carbon monooxide as a deoxygenation product of carbonyl compound was obtained in the reaction of tetraphenyl-cyclopentadienone with dibromocarbene using phenyl tribromomethyl mercury as the carbene precursor. This unique phenomenon is rationalized in terms of the push-pull stabilization gained from the interaction between the partially aromatic cyclopentadienide moiety and the positively charged dibromomethylene moiety in the dihalocarbonyl Ylide-like transition state.

Abstract:We report a highly diastereo- and enantioselective synthesis of cyclopropanes with adjacent all-carbon quaternary stereocenters featuring a monofluoromethyl (CH2F) group. While the spiroketal bisphosphine (SKP) derived chiral digold complex was identified to be a powerful catalyst for the cyclopropanation of unprotected diazooxindoles with -CH2F styrenes, Rh-2(R-DOSP)(4) proved to be excellent in the corresponding reactions using aryl diazoacetate. In the cyclopropanation of diazooxindoles, dramatic rate acceleration (about twenty times) was observed when varying the reaction solvent from chlorobenzene (PhCl) to fluorobenzene (PhF), which cannot be explained by two factors that are widely used to explain the solvent effects on the olefin cyclopropanation (the dielectric constant and the coordinating ability of the solvent). Theoretical calculations suggest the formation of strong C-FH-N interaction between PhF and the N-H bond of the unprotected diazooxindole derived Au(i)-carbenoid intermediate, which effectively lowers the reaction barrier. This result first demonstrated that it is possible to harness the C-FH-X interaction between fluorinated solvents and the reactive intermediates to tune both the reactivity and stereoselectivity of a catalytic asymmetric reaction.

Abstract:A novel example of alcohol deoxycyclopropanation reaction for cyclopropane introduction with organophotocatalyst is described. This method demonstrates a broad substrate scope and high transformation efficiency. The mechanism involves the activation of alcohols in situ along with a photoredox‐catalyzed deoxygenative radical addition–polar cyclization. The direct introduction of cyclopropane to marketed drugs and natural products is also explored. Cyclopropane fragments, which widely exist in marketed drugs and natural products, can confer special pharmacological properties to small‐molecule drugs. Therefore, developing methods to construct cyclopropanes is of great significance. Nevertheless, the introduction of cyclopropane primarily relies on already‐formed cyclopropyl groups, which significantly restricts the diversity of cyclopropane skeletons. Late‐stage direct cyclopropanation is still a challenging task. Herein, a photo‐induced intermolecular deoxycyclopropanation reaction that employs alcohols as substrates, and 1 mol.% of 2,3,5,6‐tetrakis(carbazol‐9‐yl)‐1,4‐dicyanobenzene (4CzTPN) as organophotocatalyst is reported. This method proceeds with high transformation efficiency (up to 98% yield) and exhibits broad functional group tolerance, such as primary, secondary, and tertiary alcohols as well as various activated β‐halogenated alkenes. This process is mild, easy to operate, and has low equipment requirements. The power of this technology is demonstrated by the late‐stage functionalization of five marketed drugs and five natural products.

Abstract:A computational study using a hybrid DFT method (M06) on the cyclopropanation of endo-dicyclopentadiene with Simmons-Smith zinc carbenoids is reported. Each channel proceeds via the methylene-transfer mechanism with a reactant complex (RC) and subsquently a asynchronous transition state (TS). The channels with monomeric IZnCH2I attacking the double bonds from the exo-face have a much lower barrier (about 16.17-18.43 kcal/mol) in the gas phase, compared with the channels from the endo-face (21.80-31.13 kcal/mol). Thus, PI and P3 are the primary cyclopropanated compounds, and PS is the sole final product, representing remarkable stereospecificity. When considering the bulk solvent effect of diethyl ether, the barriers are decreased about 0.50-7.77 kcal/mol due to more destabilization of RC than TS. The solvated (ICH2)(2)Zn can further reduce the barriers about 0.18-2.30 kcal/mol. In addition, the solvated IZnCH2I and (ICH2)(2)Zn do not change the reaction pathways and retain the stereospecificity. Our computational results agree with the experimental observations quite well and suggest that both IZnCH2I and (ICH2)(2)Zn might be the active species in the real reaction system. Regarding the solvent effect, the polar continuum model is more realistic than the direct involvement of diethyl ether molecules.

Abstract:The cyclopropanation reaction of alkenes with photolytically‐generated chlorocarbenes from chlorodiazirines is reported as an effective way to prepare substituted 3‐chloro‐3‐aryl‐cyclopropanes. This practical and efficient approach allows the synthesis of various 3‐chloro‐3‐aryl‐cyclopropanes (32 examples) in continuous flow in 5‐minute residence time under light‐emitting diode (LED) irradiation. The conditions using 380 nm LED irradiation were successfully extended to the synthesis of substituted 3‐bromo‐3‐aryl‐cyclopropanes (3 examples).

Abstract:DeMuynck and colleagues describe a new, iron-catalyzed method for cyclopropanation of a sterically and electronically diverse range of unactivated alkenes. The strategy enables synthesis of novel classes of cyclopropanes bearing alkyl, benzyl, allyl, halide, and heteroatom substituents, as well as spirocyclic and fused bicycles. By harnessing non-stabilized alkyl carbenes, novel substituted cyclopropanes can now be accessed to facilitate the discovery of new medicines containing these privileged motifs. Mechanistic insights are provided for this non-stabilized alkyl carbene reactivity.

Abstract:A detailed mechanism for the Kulinkovich hydroxycyclopropanation reaction has been explored with density functional theory calculations on the reactions between R(1)COOMe and Ti(OMe)(2)(CH(2)CHR(2)) (R(1) and R(2) are hydrogen and alkyl groups). Addition of ester to titanacyclopropane is found to be fast, exothermic, and irreversible. It has a preference for the alpha-addition manifold over the beta-addition manifold in which its cycloinsertion transition states suffer from the steric repulsion between the R(2) and ester. The following intramolecular methoxy migration step is also exothermic with reasonable activation energy. The cyclopropane-forming step is the rate-determining step, which affords the experimentally observed cis-R(1)/R(2) diastereoselectivity in the alpha-addition manifold by generating cis-R(1)/R(2) 1,2-disubstituted cyclopropanol when R(1) is primary alkyl groups. On the contrary, the unfavored beta-addition manifold offers the diastereoselectivity contradicting the experimental observations. The effects of R(1) and R(2) on the regio- and stereoselectivity are also discussed.

Abstract:The ring opening-coupling reaction of cyclopropanols with propargylic carbonates affording synthetically attractive allenyl ketones has been developed. The mechanism involves the ligand-exchange reaction of in situ formed allenyl palladium methoxide with cyclopropanols followed by carbon-carbon bond cleavage and reductive elimination. The reactions proceeded smoothly under mild reaction conditions with Pd(0)/XPhos catalysis in the absence of any external base and displayed a wide scope and application to a steroidal skeleton. The efficiency of chirality transfer and synthetic utility of the allene products have also been demonstrated.

Abstract:The arene cyclopropanation between diazo compounds and benzene is well known to produce a tautomeric mixture of norcaradiene and cycloheptatriene in favour of the latter species. Nevertheless, previous studies have suggested that the initially formed norcaradiene can be stabilized by a C-7 cyano group with prevention of its 6π-electrocyclic ring opening. According to this feature, a synthetic route to functionalized cyclohexadienes had been designed using α‐cyanodiazoacetates and α‐diazo-β-ketonitriles as the starting materials respectively. The Rh2(esp)2-catalyzed arene cyclopropanation of α‐cyanodiazoacetates in benzene afforded the expected 7-alkoxycarbonyl-7-cyanonorcaradienes as isolable compounds, which then served as template for the second cyclopropanation with ethyl diazoacetate or α-cyanodiazocarbonyls to give the formation of bis(cyclopropanated) adducts. Subsequent treatment of them with SmI2 triggered a double ring-opening process, allowing for the generation of 1,4- and/or 1,3-cyclohexadienes as either regio- or diastereomeric mixtures. On the other hand, the norcaradienes generated from phenyl- or methyl-substituted α‐diazo-β-ketonitriles were found to undergo an in-situ rearrangement to yield dihydrobenzofurans that could be converted to benzofuran derivatives by DDQ oxidation.

Abstract:Cyclopropanes with three chiral centers were afforded in good to high yields with dr ratio of 4–23:1 and ee of 83–99% in the reaction of acyclic amides with monosubstituted allyl carbonates as well as polyenyl carbonates under the Pd-catalysis in the presence of (Sphos,R)-SIOCPhox L1 as the ligand, while allylic alkylated products were provided only if (Rphos,R)-SIOCPhox L2 was the ligand. The amide group of product was easily reduced to hydroxymethyl group in high yield. The active reaction intermediate was determined and transition states for cyclopropanation and allylation were calculated. The origin of cyclopropane formation was investigated by experiments, NMR studies, X-ray analysis of allyl-Pd-ligand complexes, and DFT calculations.
Divergent ring-opening coupling between cyclopropanols and alkynes under cobalt catalysis

Abstract:Cobalt-diphosphine catalysts promote ring-opening coupling reactions between cyclopropanols and unactivated internal alkynes, affording either β-alkenyl ketones or multisubstituted cyclopentenol derivatives in good yields with good to excellent regioselectivities. The chemoselectivity between these β-alkenylation and [3 + 2] annulation reactions, which likely share a cobalt homoenolate as a key catalytic intermediate, is exquisitely controlled by the reaction conditions, with the solvent being a major controlling factor. The reactions are proposed to involve ring opening of cobalt cyclopropoxide into homoenolate, migratory insertion of the alkyne into the Co-C bond, and protodemetalation or intramolecular carbonyl addition of the resulting alkenylcobalt species. The feasibility of these reaction steps was supported by DFT calculations.

Abstract:The cyclopropane skeleton plays a significant role in bioactive molecules due to its distinctive structural properties. This has sparked keen interest and in-depth exploration in the field of stereoselective synthesis of cyclopropane derivatives. In the present study, the mechanism and the origin of stereoselectivity of diastereodivergent synthesis of cyclopropane derivatives via the catalyst-free [2 + 1]-cyclopropanation reactions of 3-diazo- N -methylindole ( R1 ) with two types of electron-deficient olefins ( R2 and R3 ) in both aqueous and toluene media have been studied using the DFT calculations. The findings indicate that these [2 + 1] cycloaddition reactions proceed in two stages, where the first step is not only the rate-determining step but also critically dictates the stereoselectivity of the product. The calculated diastereomeric ratios are in agreement with the experimental results. Furthermore, by utilizing non-covalent interaction (NCI) analysis and energy decomposition analysis based on molecular force fields (EDA-FF), we elucidated that the electrostatic interactions between reactant fragments in the transition state TS1s for the first step are the predominant factors determining the stereoselectivity, as opposed to the experimentally hypothesized steric hindrance and π-π stacking interactions.

Abstract:The ring-opening reaction of 1,2-disubstituted cyclopropanols, prepared from β-diketones, mediated by Cu(NO3)2, p-TsOH, and NaOH is reported. The Cu(II)-mediated ring-opening of cyclopropanols gave α-methylene-γ-diketones in good yields. The reaction took place at the less substituted carbon of the cyclopropanols, involving mild conditions and a simple procedure. The reaction induced by p-TsOH in CH2Cl2 afforded α-methyl-γ-diketones as the major product with minor amounts of δ-diketones. The 2,3,5-trisubstituted furans were obtained in high yields when the ring-opening reaction was mediated by p-TsOH in methanol under reflux conditions. In the presence of sodium hydroxide the reaction proceeded smoothly in preference to the formation of δ-diketones, particularly for substrates with an aromatic group on the cyclopropane.
