Abstract:Aim To study the saponin constituents of the seeds of Aesculus chinensis Bunge var. chekiangensis (Hu et Fang) Fang. Methods Compounds were separated and purified by macroreticular resin column chromatography and high-performance liquid chromatography. Their structures were elucidated by spectroscopic and hydrolysis analysis. Results Six compounds were isolated from the 70% ethanolic extracts. They were identified as escins IVc, IVd, Ia, Ib, isoescins Ia and Ib, respectively. Conclusion The above compounds were obtained from the seeds of Aesculus chinensis Bunge var.chekiangensis (Hu et Fang) Fang for the first time.

Abstract:Eight bioactive triterpenoid saponins (1-8) were isolated from the seeds of Aesculus chinensis, four of which are novel compounds. The major saponins were identified as escin Ia (1), Ib (2), isoescin Ia (3) and Ib (4), while the new compounds were identified as 22alpha-tigloyl-28-acetylprotoaescigenin-3beta-O-¿beta -D-glucopyranos yl (1-2) ¿beta-D-glucopyranosyl (1-4)-beta-D-glucopyranosiduronic acid (escin IVc, 5), 22alpha-angeloyl-28-acetylprotoaescigenin-3beta-O-¿bet a-D-glucopyrano syl (1-2) ¿beta-D-glucopyranosyl (1-4)-beta-D-glucopyranosiduronic acid (escin IVd, 6), 28-tigloylprotoaescigenin-3beta-O-¿beta-D-glucopyranosyl (1-2) ¿beta-D-glucopyranosyl (1-4)-beta-D-glucopyranosiduronic acid (escin IVe, 7), and 28-angeloylprotoaescigenin-3beta-O-¿beta-D-glucopyranosyl (1-2) ¿beta-D-glucopyranosyl (1-4)-beta-D-glucopyranosiduronic acid (escin IVf, 8). The structures were determined by chemical and spectroscopic methods. All the above compounds were evaluated for their inhibitory activity against HIV-1 protease.

Abstract:Three triterpenoid saponins were isolated from 90％ ethanolic extract of the seeds of Aes-culus turbinata Bl.. Their structures were identified on the basis of spectral data as escin IVc:22α-O-tigloyl-28-O-acetylprotoaescigenin 33-O-[β-D-glucopyranosyl (1→2)][β-D-glucopyranosyl (1→4)]-β-D-glucopyranosiduronic acid (Ⅰ), isoescin Ia: 21β-O-tigloyl-28-O-acetylprotoaescigenin-3β-O-[β-D-glucopy-ranosyl (1→2)][β-D-glucopyranosyl (1→4)]-β-D-glucopyranosiduronic acid (Ⅳ), and isoescin Ib:21β-O-angeloyl-28-O-acetylprotoaescigenin-3β-O-[β-D-glucopyranosyl (1→2)][β-D-glucopyr anosyl (1→4)]-β-D-glucopyranosiduronic acid (Ⅴ). Escin IVc and isoescins Ia, Ib were obtained for the first time from thisplant. Escins Ia, Ib, IVc and isoescins Ia, Ib had inhibitory effect against recombinant HIV-1 protease atconcentration of 100 μmol/L in vitro.

Abstract:AIM: To study the saponins from the seeds of Aesculus assamica Griff,and explore a new source of escin-like saponins.METHOD: The ethyl acetate and n-BuOH fractions from EtOH ext.of Aesculus assamica were studied.Chromatography and spectral analysis were used to isolate and elucidate the constituents.RESULT: Two triterpenoid aglycons together with one triterpenoid saponin and one coumarin glycoside were identified as protoaescigenin(1),21-angeloyl-protoaescigenin(2),21β-angeloyl-protoaescigenin-3β-O-[β-D-glucopyranosyl(l-2)][β-D-glucopyranosyl(l-4)]β-D-glucopyranosiduronic acid(3),and isofraxoside(4).CONCLUSION: Compounds 1～4 were first identified from the seeds of Aesculus assamica.

Abstract:Phytochemical study of the ethanol extract of the seeds of Aesculus assamica led to the isolation of two new triterpenoid saponins. The structure of the new compounds were elucidated on the basis of spectral data to be 28-O-acetyl-21-O-(4-O-angeloyl)-6-deoxy-beta-glucopyranosyl-3-O-[beta-glueopyranosyl(1-2)-O-[beta-glucopyranosyl(1-4)]-beta-glucuronopyranosyl]protoaeseigenin (1), and 21-O-(4-O-angeloyl)-6-deoxy-beta-glueopyranosyl-3-O-[beta-glueopyranosyl(1-2)-O-[beta-glucopyranosyl(1-4)]-beta-glueuronopyranosyl]protoaescigenin (2). Their in vitro bioactivity against plant pathogenic fungus Pyricularia oryzae and cytotoxicity against K562 and HCT-15 cell lines were evaluated.

Abstract:The American Journal of Chinese Medicine, Ahead of Print. [math]-Escin is an oleanane-type pentacyclic triterpenoid saponin extracted from the seeds of Aesculus hippocastanum (AH), which is more widely distributed. [math]-Escin sodium has been approved by the American FDA for clinical usage. This paper is intended to summarize an updated and comprehensive review of the pharmacological activities, pharmacokinetic properties, toxicity, and analytical methods of [math]-escin. Studies have shown that [math]-escin has significant antitumor, antiviral, anti-inflammatory, and other activities alongside less adverse effects and higher safety than other compounds. The review shows that the pharmacological effects of [math]-escin involve mechanisms such as ATM/[math]H2AX, RhoA/Rock, GSK-3[math]/[math]-Catenin, HER2/HER3/Akt, and PI3K/Akt signaling pathways, and Cyclin A, p21[math], survivin, Bcl-2, Mcl-1, Caspases, TGF-[math], MMPs, and TNF-[math] among other inflammatory factors. [math]-Escin has significant cytotoxicity; the use of the chitosan/xanthan gum-based polyelectrolyte complexes PA1 and PC-11 to modify it not only to reduces its toxicity, but also improves its drug efficacy. Because of this, these compounds may become a new research hotspot. [math]-Escin in vivo metabolism can be converted by the CYP1A2 enzyme in the intestinal flora to produce [math]-escin, deacylated, deglycosylated, and 21[math]-[math]-crotonoyl-protoescin, and the binding rate of the plasma proteins is higher than 90%. These are mainly metabolized by the liver, kidneys, and other organs, and excreted in the form of urine and feces. The number of reports on the specific mediators of the metabolism of [math]-escin and their mechanisms and metabolites is relatively small; furthermore, the results are vague. Therefore, a complete and in-depth exploration of the pharmacokinetic characteristics of [math]-escin is needed to provide a more complete and effective theoretical reference for the study of its pharmacodynamic activity.

Compounds 1–3 possess a novel 12β,30-dihydroxy-olean-28,13β-lactone aglycone and a 12β-hydroxy substituent that is rarely found in this kind of triterpenoid saponin.

Their structures were determined on the basis of extensive spectroscopic analysis, and the absolute stereochemistries were established by single-crystal X-ray diffraction and CD experiments. A plausible biosynthetic pathway of 1-3 was also discussed.
