Abstract:A stereoselective synthesis of (3S,5S,6S)-3,7-dimethyl-5,6-dihydroxy-7-en-octanal, a key intermediate, for the synthesis of the HIV-inhibitory didemnaketals analogue, was performed via a series of reactions from the natural(+)-pulegone. In this approach, an interesting and valuable diastereo-recognizable reaction between tertiary hydroxyepoxides and PCC was found. Therefore, the mixture of compounds 8 and 9 was treated with PCC to recover 8 in 93% yield and de 98% purity.

Abstract:The total synthesis of isomer of didemnaketal A was achieved in 26 steps. The position of esters is switched at C7 and C8 with respect to their proposed position in natural didemnaketal A, which shows potent anti-HIV activity but so far has not been synthesized in the laboratory. Structural analysis of synthetic isomer of didemnaketal A indicates that the esters at C7 and C8 are correctly assigned, suggesting that the problems for the structural reassignment of natural didemnaketal A lie elsewhere. Abstract:The marine natural product cyclodidemniserinol trisulfate displayed moderate HIV-1 integrase inhibitory activity. Its novel structure triggered our interest to synthesize it. In our total synthesis effort, the natural product was dissected into four fragments based on the rational retrosynthetic analysis. All four fragments were successfully prepared with orthogonal protection. And the assembly of fragment A and B furnished the C5–C30 key subunit by employing the I2-mediated deprotection and intramolecular ketal formation tandem reaction in the presence of NaHCO3 in MeCN. Our work provided flexible and practical approaches to synthesize and derive the 3,5,7-trisubstituted 6,8-dioxabicyclo [3.2.1] octane based analogs to search for new structure HIV-1 integrase inhibitors.

Abstract:A convergent diastereo- and enantioselective total synthesis of anti-HIV agent chloropeptin I is reported. Important features of the total synthesis include: (1) the use of Ti-catalyzed cyanide addition to imines to prepare a requisite amino acid moiety, (2) the discovery of the positive effect of MeOH in the Cu-mediated biaryl ether formation to afford one of the two macrocyclic peptide moieties, and (3) the discovery of the positive influence of collidine in the diastereoselective Pd-mediated cross-coupling to result in efficient formation of another macrocycle within this medicinally important molecule. This key step is performed in the presence of four unprotected phenols, two of which reside on dichlorophenylglycines.

Abstract:An efficient synthesis of the C3-C12 aldehyde fragment of 24-demethylbafilomycin C 1 was accomplished for assembling the 16-membered plecomacrolide skeleton according to a 1,3-diene-ene ring-closing metathesis (RCM) strategy. A boron-mediated ANTI-selective aldol condensation of Abiko’s chiral propionate was used to secure the C6 and C7 stereogenic centers while the C8 chirality was introduced from a chiral building block. The dithiane alkylation and the methyl ketone Horner-Wittig olefination using allyldiphenylphosphine oxide were employed for construction of the requisite ( E)-1,3-diene subunit.

Abstract:A highly stereocontrolled synthesis of the C18–C28 ketone fragment of the 16-membered plecomacrolide micromonospolide B has been accomplished. The C21–C23 syn–anti stereotriad is secured by the anti-selective aldol condensation of the ephedrine-derived chiral propionate with (E,E)-hexa-2,4-dienal and Sharpless asymmetric allylic epoxidation–regioselective reductive epoxide ring opening, respectively. The overall yield of this 14-step sequence is 18.4% and the target C18–C28 ketone was obtained in enantiomerically pure form.

Abstract:In consideration of the anti-HIV activities of delavirdine, resveratrol and Pidotimod, a novel kind of anti-AIDs drug(compounds 5 and 7) was synthesized. Compound 5 was synthesized by linking the delavirdine derivative to pidotimod, compound 7 was synthesized via linking the delavirdine derivative to resveratrol with terephthalic acid as the conjugated compound by two different routes. The structures were confirmed by means of H-1 NMR, C-13 NMR, IR, MS and elemental analysis. The results for HIV-1 reverse transcriptase assay, quantitative titration of HIV-1 by using MAGI assay and pharmacokinetics assay show that a novel kind of anti-AIDs drug has been synthesized.
