HEK293 cells as a useful model in investigating receptor-operated calcium movement

Abstract:AIM: To understand the properties of calcium-movement in HEK293 cells. METHODS: The intracellular calcium concentration ([Ca2+](i)) was measured with Fura-2 probe in native HEK293 cells and transfected HEK293 cells with α(1B)-adrenoceptor cDNA (HEKα(1B)). The effects of drugs which was capable of changing Ca2+ movement in cells on [Ca2+](i) were investigated. RESULTS: The [Ca2+](i) was not changed by KCl (50 mmol·L-1), Bay K8644 (10 μmol·L-1), caffeine (20 mmol·L-1) and ryanodine (1 μmol·L-1) in the presence or absence of 1.5 mmol·L-1 extracellular Ca2+. Cyclopiazonic acid (CPA) concentration-dependently caused increase in [Ca2+](i) in HEK293 cells. In the medium containing 1.5 mmol·L-1 CaCl2, CPA produced a biphasic Ca2+ signal response (initial transient induced by depletion of intracellular Ca2+ pools and sustaining phase resulting from Ca2+ influx). The CPA-induced Ca2+ influx phase was not changed by nifedipine (10 μmol·L-1) but inhibited by NiSO4 (1 mmol·L-1). In HEKα(1B) cells, adrenaline (10 μmol·L-1) induced increase in [Ca2+](i). Pretreated HEKα(1B) cells with maximum effective concentration of CPA (30 μmol·L-1) for 3~10 min to inducing full depletion of CPA- sensitive Ca2+ pool, subsequent administration of 10 μmol·L-1 Adr still stimulated Ca2+ release. CONCLUSION: The receptor operated Ca2+ channel exists but the voltage dependent Ca2+ channel not in HEK293 cells. There are two different kinds of releasable Ca2+ pools, one is CPA-sensitive and another is CPA-insensitive pool, in HEK293 cells. Release of Ca2+ from these Ca2+ pools are through IP3 receptor-operated Ca2+ release channel but ryanodine receptor-operated Ca2+ channel.
