Abstract:Objective To prepare sinomenine hydrochloride(SIN-HCl) liposomes with high entrapment efficiency(EE) and to illustrate the effects of drug quantity and particle size on EE.Methods Centrifugation sedimentation-centrifugation ultrafiltration was employed to determine EE of liposomes.Thin film hydration(TFH),reverse phase evaporation(REV),and ether injection(EI) were screened based on EE and formability of liposomes.The effects of water type,pH value,ion concentration of hydration liquid,pH gradient active drug loading,lecithin-cholesterol ratio,and drug-lipid ratio on EE of liposomes were investigated.The relationship between EE and the factors affecting the drug quantity and particle size was probed with a comprehensive design experiment.The stability of typical liposomes was evaluated at 4 ℃.Results The optimal preparation technology was TFH for SIN-HCl liposomes and citrate buffer solution(CBS) was the best hydration liquid.The liposome EE increased with the increase of pH values of CBS.When the pH value of CBS was fixed,the EE increased as a result of decrease in the ion concentration of CBS.pH gradient active drug loading led to increase of EE.The preferable hydration liquid for liposomes was CBS with pH value of 2.5.The optimal ratio of soybean lecithin to cholesterol was 6∶1.Increasing ratios of SIN-HCl to soybean lecithin from 1∶6 to 6∶6 led to a slight decrease in EE of liposomes without probe signification.A quantitative relationship was established between the EE and drug quantity and liposome size.The EE of SIN-HCl liposomes prepared by certain particle size and drug quantity could reach over 80%.The typical liposomes showed a good stability.Conclusion The technology of pH gradient active drug loading is able to prepare SIN-HCl liposomes with high EE.

Abstract:Despite of great advances of phospholipids and liposomes in clinical therapy, very limited success has been achieved in the preparation of smart phospholipids and controlled-release liposomes for in vivo drug delivery and clinical trials. Here we report a supramolecular approach to synthesize novel supramolecularly engineered phospholipids based on complementary hydrogen bonding of nucleosides, which greatly reduces the need of tedious chemical synthesis, including reducing the strict requirements for multistep chemical reactions, and the purification of the intermediates and the amount of waste generated relative more traditional approaches. These upgraded phospholipids self-assemble into liposome-like bilayer structures in aqueous solution, exhibiting fast stimuli-responsive ability due to the hydrogen bonding connection. In vitro and in vivo evaluations show the resulted supramolecular liposomes from nucleoside phospholipids could effectively transport drug into tumor tissue, rapidly enter tumor cells, and controllably release their payload in response to an intracellular acidic environment, thus resulting in a much higher antitumor activity than conventional liposomes. The present supramolecularly engineered phospholipids represent an important evolution in comparison to conventional covalent-bonded phospholipid systems.
Preparation of lyophilized polyene-phosphatidylcholine lipid nanoparticles

Abstract:OBJECTIVE To prepare lyophilized polyene phosphatidylcholine lipid nanoparticles(PPC-LN)and investigate the main influencing factors.METHODS PPC-LN was prepared by the method of melt-emulsification-high pressure homogenization.The main factors affecting PPC-LN quality were investigated by single factor test.The formulations were optimized by the orthogonal design.The physicochemical properties of lyophilized LN were also investigated.RESULTS The results showed that the nanoparticles were spherical and well distributed.The mean particle size was 135±12 nm with the encapsulation efficiency of 97.4%±0.9% and PDI of 0.169±0.004,respectively.CONCLUSION The melt-emulsification-high pressure homogenization for the preparation of lipid nanoparticles containing PPC was highly feasible.Lyophilized PPC-LN with high encapsulation efficiency,narrow size distribution and good form could be obtained using this method.

Abstract:In the last decades, pharmaceutical interested researches aimed to develop novel and innovative drug delivery techniques in the medical and pharmaceutical fields. Recently, phospholipid vesicles (Liposomes) are the most known versatile assemblies in the drug delivery systems. The discovery of liposomes arises from self-forming enclosed phospholipid bilayer upon coming in contact with the aqueous solution. Liposomes are uni or multilamellar vesicles consisting of phospholipids produced naturally or synthetically, which are readily non-toxic, biodegradable, and are readily produced on a large scale. Various phospholipids, for instance, soybean, egg yolk, synthetic, and hydrogenated phosphatidylcholine consider the most popular types used in different kinds of formulations. This review summarises liposomes composition, characterisation, methods of preparation, and their applications in different medical fields including cancer therapy, vaccine, ocular delivery, wound healing, and some dermatological applications.

Bile Salt/phospholipid Mixed Micelle Precursor Pellets Prepared by Fluid-Bed Coating

Abstract:Bile salt/phospholipid mixed micelles (MMs) are potent carriers used for oral absorption of drugs that are poorly soluble in water; however, there are many limitations associated with liquid formulations. In the current study, the feasibility of preparing bile salt/phospholipid MM precursor (preMM) pellets with high oral bioavailability, using fluid-bed coating technology, was examined. In this study, fenofibrate (FB) and sodium deoxycholate (SDC) were used as the model drug and the bile salt, respectively. To prepare the MMs and to serve as the micellular carrier, a weight ratio of 4:6 was selected for the sodium deoxycholate/phospholipids based on the ternary phase diagram. Polyethylene glycol (PEG) 6000 was selected as the dispersion matrix for precipitation of the MMs onto pellets, since it can enhance the solubilizing ability of the MMs. Coating of the MMs onto the pellets using the fluid-bed coating technology was efficient and the pellets were spherical and intact. MMs could be easily reconstituted from preMM pellets in water. Although they existed in a crystalline state in the preMM pellets, FB could be encapsulated into the reconstituted MMs, and the MMs were redispersed better than solid dispersion pellets (FB:PEG = 1:3) and Lipanthyl®. The redispersibility of the preMM pellets increased with the increase of the FB/PEG/micellar carrier. PreMM pellets with a FB:PEG:micellar carrier ratio of 1:1.5:1.5 showed 284% and 145% bioavailability relative to Lipanthyl® and solid dispersion pellets (FB:PEG = 1:3), respectively. Fluid-bed coating technology has considerable potential for use in preparing sodium deoxycholate/phospholipid preMM pellets, with enhanced oral bioavailability for poorly water-soluble drugs.

Abstract:The high physiology and low toxicity of therapeutic peptides and proteins have made them a hot spot for drug development in recent years. However, their poor oral bioavailability and unstable metabolism make their clinical application difficult. The bilayer membrane of liposomes provides protection for the drug within the compartment, and their high biocompatibility makes the drug more easily absorbed by the body. However, phospholipids—which form the membranes—are subjected to various digestive enzymes and mucosal adhesion in the digestive tract and disintegrate before absorption. Improvements in the composition of liposomes or modifying their surface can enhance the stability of the liposomes in the gastrointestinal tract. This article reviews the basic strategies for liposome preparation and surface modification that promote the oral administration of therapeutic polypeptides.

Preparation and Characterization of Liposomes Encapsulating Chitosan Nanoparticles

Abstract:Liposomes are an important colloidal carrier system for controlled drug delivery. However some highly hydrophilic small molecules are difficult to entrap into liposomes and store stably, resulting in poor encapsulation efficiency and fast leakage. In the present work, fluorescein sodium (FS) was used as a model drug that was loaded into chitosan nanoparticles and then encapsulated into liposomes by reverse-phase evaporation (RPV). The encapsulation efficiency, particle size, zeta potential, release in vitro and pharmacokinetics in rats were determined in order to characterize the novel drug delivery system. The entrapment efficiency was above 80% in nanoparticles (Np) and 95% in liposomes encapsulating the nanoparticles (Lip-Np). The Lip-Np was composed of soybean phospholipids, cholesterol and chitosan, which the average diameter was 202.6 nm and zeta potential was -34.8 mV The release rate of fluorescein sodium from Lip-Np was slower than from Np and liposomes. FS in Lip-Np administered to rats exhibited prolonged circulation and higher bioavailability than FS in Np. The results indicated that liposomal release kinetics can be controlled by encapsulating nanoparticles and thus solid-cored liposomes can be used as a potential drug delivery system.

Abstract:Objective: To prepare liposomes-containing polyelectrolyte microcapsules by layer-by-layer self-assemble technique,and study the structure,spontaneous deposition and release profile.Method: The polyelectrolyte microcapsules were fabricated by deposition of oppositely charged chitosan and alginate onto the liposomes-containing calcium carbonate colloidal particles in a layer-by-layer way followed by the removal of the template cores with disodium ethylenediaminetetraacetic acid.The structure,spontaneous deposition and release profile of the microcapsules were characterized by TEM and SEM.Result: TEM and SEM images revealed that liposomes-containing polyelectrolyte microcapsules were obtained successfully.The concentration of doxorubicin in the microcapsules was increased with the increase of doxorubicin concentration,and the highest concentration of doxorubicin in the microcapsules could reach 520.1 mg·ml-1 with the doxorubicin concentration of 1 mg·ml-1.The cumulative release amount of doxorubicin from the microcapsules in 48 h was 59.2%,54.3% and 44.8% in PBS with pH of 5.0,6.5 and 7.4,respectively.Conclusion: The liposomes-containing polyelectrolyte microcapsules show promising spontaneous deposition and release ability,suggesting good application prospects as novel drug carriers.

Abstract:Liposomes (or lipid vesicles) are a versatile platform as carriers for the delivery of the drugs and other macromolecules into human and animal bodies. Though the method of using liposomes has been known since 1960s, and major developments and commercialization of liposomal formulations took place in the late nineties (or early part of this century), newer methods of liposome synthesis and drug encapsulation continue to be an active area of research. With the developments in related fields, such as electrohydrodynamics and microfluidics, and a growing understanding of the mechanisms of lipid assembly from colloidal and intermolecular forces, liposome preparation techniques have been enriched and more predictable than before. This has led to better methods that can also scale at an industrial production level. In this review, we present several novel methods that were introduced over the last decade and compare their advantages over conventional methods. Researchers beginning to explore liposomal formulations will find this resource useful to give an overall direction for an appropriate choice of method. Where possible, we have also provided the known mechanisms behind the preparation methods.

Abstract:Overview of liposome preparation methods and applications. Liposomes are artificial microscopic vesicles composed of phospholipid bilayers with the ability to encapsulate both hydrophobic and hydrophilic molecules, owing to the amphipathic nature of lipids. Hydrophobic molecules can be stored within the bilayer membrane, while hydrophilic molecules can be embedded in the inner core of liposomes. Encapsulated compounds within liposomes are protected from environmental and chemical alterations, such as enzymatic and chemical modifications, as well as changes against extreme pH, temperature, and ionic strength. Liposomes protective nature highlights their importance as nanocarriers for a wide spectrum of hydrophobic and hydrophilic molecules. This review offers a concise introduction to the fundamental physicochemical properties of liposomes and the various production methods including the role of cholesterol and potential alternatives such as phytosterols. It also provides an up‐to‐date overview of liposomes applications as delivery vehicles in food, cosmetics, and pharmaceuticals.

Abstract:Liposome-based drugs have emerged as highly successful nanomedicines for clinical applications owing to their excellent biocompatibility, biodegradability, and targeting effects. However, the lipids that play a crucial role in most liposome formulations are often engineered rather than natural phospholipids. From this perspective, we outlined the classification of engineered lipids based on their material chemistry, including charged, polymer-conjugated, ligand-conjugated, and choline phosphate lipids. We also discussed their drug, gene, protein, and probe delivery applications. With the increasing requirements for the innovation of nano drug excipients by the Food and Drug Administration, attention to drug efficacy and side effects has increased in clinics, and the development of superior engineered lipids has become urgent. Therefore, we aim to provide a new idea for developing engineered lipids in which lipids can be engineered with minimal chemical structure changes to achieve maximum functions enhancement, with choline phosphate lipids being a favorable choice. In the future, novel engineered lipids and liposomes should be developed with more powerful functions and superior therapeutic safety, which will allow their utilization in the diagnosis and treatment of major diseases and be valuable for more clinical applications.

Abstract:Liposomes are essentially a subtype of nanoparticles comprising a hydrophobic tail and a hydrophilic head constituting a phospholipid membrane. The spherical or multilayered spherical structures of liposomes are highly rich in lipid contents with numerous criteria for their classification, including structural features, structural parameters, and size, synthesis methods, preparation, and drug loading. Despite various liposomal applications, such as drug, vaccine/gene delivery, biosensors fabrication, diagnosis, and food products applications, their use encounters many limitations due to physico-chemical instability as their stability is vigorously affected by the constituting ingredients wherein cholesterol performs a vital role in the stability of the liposomal membrane. It has well established that cholesterol exerts its impact by controlling fluidity, permeability, membrane strength, elasticity and stiffness, transition temperature (Tm), drug retention, phospholipid packing, and plasma stability. Although the undetermined optimum amount of cholesterol for preparing a stable and controlled release vehicle has been the downside, but researchers are still focused on cholesterol as a promising material for the stability of liposomes necessitating explanation for the stability promotion of liposomes. Herein, the prior art pertaining to the liposomal appliances, especially for drug delivery in cancer therapy, and their stability emphasizing the roles of cholesterol.

Preparation and in vitro evaluation of liposomal chloroquine diphosphate loaded by a transmembrane pH-gradient method

Abstract:This study developed an active loading method for encapsulating chloroquine diphosphate (CQ) into liposomes. The effects of different formulation factors on the encapsulation efficiency (EE) and the size of CQ liposomes were investigated. These factors included the internal phase of liposomes, the external phase of liposomes, the ratio of drug to soybean phosphatidylcholine (drug/SPC), the ratio of cholesterol to soybean phosphatidylcholine (Chol/SPC), and the incubation temperature and time. The EE (93%) was obtained when using drug/SPC (1:50 mass ratio), SPC/Chol (1:5 mass ratio) at 0.10 M citrate-sodium citrate buffer (pH 3.6). As 5 mol% methoxypoly(ethylene glycol)(2,000) cholesteryl succinate (CHS-PEG(2000)) or distearoyl phosphatidylethanolamine-poly (ethylene glycol)(2,000) (DSPE-PEG(2000)) was added, the size of particle was reduced and the EE was improved. Freeze-drying with 5% trehalose as a cryoprotectant was carried out to achieve long-term stability. The drug release studies were performed in vitro simulating the desired application conditions, such as physiological fluids (pH 7.4), tumor tissues (pH 6.5) and endosomal compartments (pH 5.5). The release of CQ from the liposomes prepared via remote loading showed the significant pH-sensitivity and retention properties, which favored the application of liposomal CQ at tumor tissues and endosomal compartments.

Abstract:OBJECTIVE：To prepare the liposomes carrier materials lecithin.METHODS:The lecithin was extracted and prepared from fresh yolks.The purity,the content,the percentage of oxidation and the quality standard were studied.RESULTS:The lecithin we prepared was chromatographically homogeneous phospholipid,with no indication of any other components.The content of lecithin was more than 98%,the percentage of oxidation was very low,and the results of studies on the quality standard were fit for the USP.CONCLUSION:The results showed that,the lecithin we prepared could be used as the liposomes carrier materials.

Egg phosphatidylcholine liposomes incorporating hydrophobically modified chitosan: pH-sensitive release

Abstract:Egg phosphatidylcholine (EPC) liposomes incorporating hydrophobically modified chitosan (HmCh) were prepared by a sonication method. In order to immobilize chitosan on the surface of EPC liposomes, pentyl chains were covalently attached to chitosan. Following the proton NMR spectrum, the number of pentyl chains per chitosan molecule was calculated to be 2.5. Once the ratio of HmCh to EPC was kept below 1:5, the degrees of fluorescence quenching in the liposomal suspension were greater than 70%, indicating a high yield of liposome formation. No significant release from the liposome was observed in neutral and akali condition. The liposomes started to release their content at pH 6.5, and the release became more extensive as the pH of release medium decreased. The pH-sensitivity of the liposomes in terms of the release strongly depended on the amount of HmCh incorporated in liposomes.

Abstract:The physicochemical compatibility between amphiphilic polymers and hydrophobic drugs has been recognized as an important issue for improving the drug solubilisation in polymeric micelle formulations. In this work, poly-L-lysine (PLL) grafted by cholate pendants as the only hydrophobic moiety were synthesized in order to facilitate the solubilisation of sterol drugs. Results showed that micelles formed by cholate grafted PLL encapsulated significantly higher level of prednisolone and estradiol than palmitoylated PLL micelles, whereas the solubilisation capacity of non-sterol drug (griseofulvin) is inefficient for both polymers. This suggests that higher drug-polymer incorporation can be achieved by the inclusion of hydrophobic moieties with similar architecture as the drugs, i.e. ‘drug-like’ functional groups, which will be useful for the future design of colloidal systems for the encapsulation of specific drug.

Abstract:OBJECTIVE To prepare and characterize Propylgallate(PG) liposomes.METHODS PG liposome was prepared by film dispersion method and ultrafiltrate centrifugation method was used to determine encapsulation efficiency.RESULTS The optimized formulation parameters included:the concentration of soybean lecithin(SPC) 5 mg·mL-1,PG-SPC(1:5),SPC-cholesterol(5:1),ionic strength 20 mmol·L-1.The particle diameter was 181.3 nm and Zeta potential was-21.8 mV.The cumulative of release amount was up to 80% within 4 h.CONCLUSION PG liposomes are of high encapsulation efficiency with a small and uniform particle size.
