Abstract:OBJECTIVE:To analyze the genetic relationship of Dendrobium and design a specific primer to differentiate Dendrobium candidum effectively.METHOD:Random amplified polymorphic DNA (RAPD) technique was used to analyze the genetic relationships of 26 species of Dendrobium and one species of Ephemerantha. Dendrogram was constructed by UPGMA. According to the sequence of DNA fragment selected, Sangon 18 primer had been extended from 3' extreme to 20 bp in order to form a specific primer.RESULT AND CONCLUSION:This primer can be used to distinguish Dendrobium candidum from other Dendrobium effectively. Such a technique provides a new way for the identification of Chinese traditional medicines.

Abstract:Random amplified polymorphic DNA(RAPD)was used to study the differencial genes between the Lentinula edodes(Qingyuan) strain 135 with poor resistance and strain 9015(a new strain by natural variation).Several differencial genes were obtained,and then cloned and sequenced.The results in GenBank revealed that the partially possible Open Reading Frames(ORF) of these fragments had higher homology with some hydrolases and pheromone precursor receptor genes of several kinds of organisms.And we presume these genes fragments could be as the specific molecular markers for specifying Lentinula edodes 135 and 9015.

Abstract:The genetic polymorphisms in vegetable crops of Brassica campestris was investigated using random amplified polymorphic DNA markers (RAPDs), the detected materials included thirty-three cultivars. Thirty-seven primers selected from seventy primers amplified 353 RAPD fragments and 260 bands of them showed polymorphisms. The length of most amplified fragments ranged from 0.9 to 1.6 kb. Different primers exhibited different detective efficiency. There were ten RAPD characteristic markers, which were detected as a group of DNA fingerprints to identify all the seven cultivars of heading type Chinese cabbage.

Abstract:Object To indentify the crude drugs of Shegan Belamcanda chinensis (L.), DC.] and related Chinese materia medica of and related Chinese materia medica of Iris L., including Iris tectorum Maxim., I. dichotoma Pall., I. germanica L. and I. japonica Thunb. by RAPD technique. Methods General DNA was isolated from fresh leaves of B. chinensis and five plants of Iris L. by CTAB and DNeasy TM Plant Mini Kit. The random amplified polymorphic DNA (RAPD) was used to identify them. Results The polymorphic feather of DNA fingerprints of different species were compared by primer OPD 08. The studies showed that they have different relationships in heredity and the method of RAPD can be used to identify them easily. Conclusion Chinese materia medica of Iris L. can be idenlified by RAPD, and B. chinensis could be differentiated from I. tectorum, I. dichotoma, I. japonica, I. germanica.
The Use of RAPD Analysis to Identify 4 Main Medicinal Plants in Vitex

Abstract:Objective: To identify the plants in Vitex including Vitex negundo L. , V. negundo var. cannabifolia （Sieb. et Zucc. ） Hand. Mazz. , V. trifolia L. and V. trifolia L. var. simplicifolia Cham. . Methods: Both intra- and inter-species relationships among these plants were analyzed by RAPD marker. Twenty-one samples collected from different locations in China were tested using 25 RAPD arbitrary 10mer primers which were screened from 32 primers, Cluster analysis was conducted by Ward’s minimumvariance method of SAS software. Results: A total of 224 bands were produced and 128 bands showed polymorphism among 21 samples. The level of polymorphism within species was 51. 7%. The dendrogram constructed based on RAPD analysis showed that 21 samples can be placed in 2 groups at the level of SPRS value of 0. 3636. The first group included V. trifolia var. simplicifolia and V. trifolia. The other consisted of V. negundo and V. negundo var. cannabijolia. . At the level of SPRS value of 0. 1, 21 samples can be obviously divided into 3 groups. V. trifolia and V. trifolia var. simplicifolia were clustered into one group. V. negundo and V. negundo var. cannabifolia were separately clustered to 2 groups. Conclusion: RAPD analysis is in good agreement with the traditional plant taxonomic classification, and fundamentally identical with their origins and morphologic characteristics, which indicates that the genetic relationship a-mong samples is related to their center of origin. These medicinal species in Vitex have their specific bands which are available to identify. These specific bands can be used as species-specific molecular markers of Fructus Viticis for the application of germplasm identification and classification.

Abstract:Many plants of the genus Isodon (Benth.) Kudo from Lamiaceae, such as Isodon serra (IS), Isodon lophanthoides (IL), etc., are widely used as the source of traditional Chinese herbal medicine “Xihuangcao” in Lingnan region of China. The origin of “Xihuangcao” herb is relatively chaotic and germplasm resources are unclear. In the present research, evaluation of genetic diversity and genetic variability in 358 accessions of ‘Xihuangcao’ herb from 33 populations were carried out based on ISSR and SCoT makers. Thirteen ISSR and 8 SCoT markers produced 491 and 272 fragments respectively, of which 99.80% (490) and 99.26% (270) were polymorphic, H=0.1719 I=0.1704 for ISSR and H=0.2057 I=0.3300 for SCoT revealed high genetic diversity. Both genetic differentiation analysis and AMOVA revealed higher variance among the populations than within populations. According to UPGMA and PCoA, the clustering patterns of populations were out of accordance with plant taxonomy or geographic distribution. This study showed that the germplasm resources of “Xihuangcao” had high genetic diversity, which could provide materials for excellent breed screening and core collections developing.

[A New Molecular Identification Method: Anchored Primer Amplification Polymorphism DNA].

Abstract:To build up a stable and easy doing method for molecular identification in traditional Chinese medicine, on basis of RAPD, the new method mainly changed the primer length and PCR annealing temperature. Panax ginseng, Panax quinquefolius and its nine adulterants were used to establish the method and test it using MARMS primers published in 2004. The new method also used to authenticate Chinese Materia Medica of Tian-hua-fen (Radix Trichosanthes) and Bai-zhi (Radix Angelica). Primer Pg-q36F obtained polymorphic bands of P. Ginseng, P. quinquefolius and its adulterants. The identification result is identical to that published before and more stable. Primer TkS1-64F obtained polymorphic bands of Tian-hua-fen and its nine adulterants. Primer AfS1-100F obtained polymorphic bands of Bai-zhi and its three adulterants. The method has good stability and reproducibility and can easily identify authertic medicines from their adulterants. It was a potential molecular method to identify other Chinese Materia Medica. The method was named as anchored primer amplification polymorphism DNA (APAPD).

Abstract:Aim To establish a simple method for molecular identification of original plants of D. chrysanthum and D. fimbriatum using molecular marker rDNA ITS region. Methods Restriction patterns of ITS fragments were obtained using PCR-RFLP method. The PCR products of D. chrysanthum and its morphologically allied species were digested at 37 degrees C by Cla I and Apa LI, those of D. fimbriatum and its morphologically allied species were digested by Sph I. Results D. chrysanthum, D. fimbriatum and their morphologically allied species could be identified by predicted restriction profiles of PCR-RFLP. The botanical origin of twenty-five fresh samples of Shihu collected in markets was identified by this method. Conclusion The results showed that PCR-RFLP analysis of the rDNA ITS region is a feasible, simple and inexpensive method for determining the botanical origin of the traditional Chinese medicine Shihu.
Authentification of Radix Cyathule and Its Adulterants Based on SCAR Markers

Abstract:Chuanniuxi (Radix Cyathule) is one of the most important geo-herb in Sichuan province, which adulterants are Hongniuxi (Cyathula capitata) and Huainiuxi (Achyranthes bidentata). In this paper Chuanniuxi and its adulterants were identified by SCAR markers. Nineteen populations from Tianquan, Baoxin, Huili and Jinkouhe were collected and their RAPD fingerprints were established. Based on the RAPD patterns, two polymorphic bands F300 and F500 were selected, recycled, cloned and sequenced. According to the sequences two pairs of sequence characterized amplified regions (SCAR) primers were designed and used to amplify all materials to prove the efficiency of identification of the different populations. Chuanniuxi and Huiniuxi could be distinguished by the primer SC-320, Chuanniuxi and Hongniuxi could be distinguished by the primer SC-495. Combining the two SCAR markers, Chuanniuxi, Hongniuxi and huainiuxi could be identified effectively and quickly.

Abstract:Objective To establish the DNA fingerprints for identification the three origin plants- Gentiana macrophylla,G.strarninea,and G.dahurica of Chinese traditional medicine"Qinjiao"(Radix Gentianae Macrophylla)from Gansu Province.Methods PCR was performed with the genomic DNA of Radix Gentianae Macrophyllae by RAPD marker,the genetic relationship among the Radix Gentianae Macrophylla was analyzed and the DNA molecular dendrogram was set up by the software NTSYSpc 2.1 and Popgene 3.2.Results The four effective primers for identification were selected from 80 RAPD primers.A total of 39 loci were amplified from the three species by four effective primers.In the total 39 amplified bands,37 were polymorphic with two common bands.Conclusion There was abundant genetic diversity among the three species from Gansu Province.The RAPD markers can be used for the identification them each other.

Abstract:Objective To seek after a new method for the identification of Fritillaria hupehensis and accurate distinguishment between Fritillaria hupehensis and Fritillaria unibracteata the certified product. Methods Random primers suitable for the distinguishment between Fritillaria hupehensis and Fritillaria unibracteata were screened out with the adoption of the random amplified polymorphic DNA(RAPD)technique. Results 20 random primers were screened out in total and one of which,the primer S1,was shown to accurately distinguish between Fritillaria hupehensis and Fritillaria unibracteata with very high specificity. Conclusion The results showed that RAPD marker technique could effectively distinguish between Fritillaria hupehensis and Fritillaria unibracteata

Abstract:This work aims to discriminate Huangqi(Radix Astragali seu Hedysari) from different producingareas using Raman spectroscopy combined with principal component analysis(PCA) and partial least squares discriminant analysis(PLS-DA).The Raman spectroscopy of 300 batches of Huangqi from five producing areas of Shanxi,Gansu,Nei Mongol,Heilongjiang and Shaanxi were measured and analyzed by PCA method to discriminate between samples from different producing areas.The discriminant model of producing areas based on PLS method was establish and verified.The results demonstrated that rapid identification of the producing areas of Huangqi could be achieved by Raman spectroscopy combined with PCA and PLS-DA methods.This method has a good application prospect and could be expanded to other medicinal herbs origin identification and authenticity identification.

Abstract:RAPD(random amplified polymorphic DNA) was used to appraise the genetic diversity of Lycium barbarum L.gathered from different areas of Ningxia.40 oligonucleotide primers from 400 oligonucleotide primers were selected for the RAPD analysis.The 10 DNA samples from diferent places were amplified by 9 selected primers.The result indicates that there are no significant genetic diferences among the 10 DNA samples,but the new bred material shows several different strips from the main variety Ningqi No.1.And DNA fingerprint of Ningqi No.1 was built by the RAPD techniques.

Abstract:RAPD technique was applied to evaluate genetic diversity of 45 Dayflower samples from six different locations,i.e.Wudalianchi,Tieli,Tongbei,Bawusi,Hongqiling and Jiamusi of Heilongjiang Province.The results showed that amplification with 60 random primers produced 349 clear and reproducible DNA fragments.The number of highly reproducible DNA fragments were 127,occupying 36.4 % of the total.The similarity coefficient ranged from 0.556 to 0.979.By using UPGMA clustering analysis the Dayflower samples were basically classified into four groups,indicating that RAPD technique was a valuable and reliable method to identify and classify Dayflower of different regions.

Abstract:Random amplified polymorphic DNA (RAPD) markers were applied to detect the genetic relationships and diversity among 33 accessions of Curcuma species in China, which involved six medicinal species. A total of 115 products were amplified by 21 primers, among which 106 products (92.17%) were found to be polymorphic. A total of 3 to 8 polymorphic bands were amplified by each polymorphic primer, with an average of 5.48 bands. The data of 115 RAPD bands were used to generate Jaccard's similarity coefficients and to construct a dendrogram by means of UPGMA. The results show that the genetic similarity coefficient of these six species is relatively large, from the cluster diagram, while the genetic relationships are not associated with their geographical distributions.
Chemical Pattern Recognition of Atractylodes Chinensis from Different Producing Areas and Establishment of Quantitative Analysis of Multi-Components by Single Marker(qams)method for Four Components

Abstract:This study established the fingerprint and combined it with chemical pattern recognition to evaluate the quality of Atractylodes chinensis samples from different producing areas and then employed the quantitative analysis of multi-components by single marker(QAMS) method to verify the feasibility and applicability of the established method in the quality evaluation of A. chinensis. The fingerprints of A. chinensis samples were constructed via high performance liquid chromatography(HPLC) to evaluate the inter-batch consistency. With the quality control component atractylodin as the internal reference, the relative correction factors(RCFs) were established for atractylenolide Ⅰ, atractylenolide Ⅲ, and β-eudesmol and the content of the four components was calculated. The external standard method was used to verify the accuracy of QAMS method. The quality of A. chinensis was further evaluated by similarity analysis, clustering analysis, and principal component analysis. The fingerprints of 13 batches of samples were calibrated with 21 common peaks, and 4 common peaks were identified with the similarities all above 0.9. The RCFs established with atractylodin as the internal reference represented good reproducibility under different experimental conditions. Specifically, the RCFs of atractylenolide Ⅰ, atractylenolide Ⅲ, and β-eudesmol in A. chinensis were 2.091, 4.253, and 6.010, respectively. QAMS and ESM showed no significant difference in the results, indicating that the QAMS method established in this study was stable and reliable. Thus, HPLC fingerprint combined with QAMS can be used for the quality evaluation of A. chinensis, providing a basis for comprehensive and rapid quality evaluation of A. chinensis.
A STUDY ON THE ORIGIN OF GERMPLASM MATERIALS OF ILEX KUDINGCHA C.J.TSENG BASED ON RAPD ANALYSIS

Abstract:Taking 15 wild germplasm materials of Ilex kudingcha C.J.Tseng as CK,the origins of 26 cultivated materials of I.kudingcha C.J.Tseng were studied via RAPD-PCR analysis.16 random primers with abundant polymorphism were selected from 40 primers tested.These 16 random primers were used for the amplification on the 41 germplasm mentioned above,and 238 bands were obtained.Of which 190 band were polymorphic bands,amounting to 79.8% of the total bands amplified.The Jaccard genetic similarity(GS) of the materials was calculated based on the amplification results and the dendrogram was constructed with UPGMA(unweighted pair group method arithmetic averages).The results showed that the 41 germplasm materials tested were divided into 2 populations,i.e.Population Hainan Island and Population Nanning,Guangxi.The 12 germplasm materials that their precise origins were unknown were clustered into Population Hainan Island,which showed that they originated in Hainan Island.The 12 germplasm materials that their precise origins were unknown were clustered into Population Guanxi-Nanning,which showed that they originated in Nanning region,the Guangxi Zhuang Autonomous Region,or in the neighboring region near Nanning region.The GS of all the 41 materials of I.kudingcha C.J.Tseng tested were distributed between 0.5654-0.9813,which showed that there was abundant considerable genetic diversity.The different origins of the various germplasm materials of I.kudingcha C.J.Tseng tested were clearly showed in the cluster analysis dendrogram.The analysis of RAPD molecular marker could be considered as an important basis for deciding the original region of I.kudingcha C.J.Tseng,and also,it could be used to study the genetic difference and relationship of the various germplasm materials of I.kudingcha C.J.Tseng.

Abstract:Dimocarpus longan Lour. is an edible and traditional herb in China, commonly referred to as longon. An improved randomly amplified polymorphic DNA (RAPD) protocol was here developed in order to determine the geographical origins of D. longan samples collected from 5 provinces in the southern and southwestern areas of China, including Sichuan, Hainan, Fujian, Guangdong, and Guangxi. Generally, the improved RAPD method generated good fingerprinting of the 5 samples using the selected 17 primers. In particular, primers SBS-A5, SBS-A13, SBS-I9, SBS-I20, SBS-M1, and SBS-Q12 produced distinguishable bands that clearly separated all 5 cultivars, suggesting that there are variations in RAPD genetic sites among the samples. The similarity index ranged from 0.69 to 0.76. The Sichuan and Hainan clades clustered together with a 0.73 similarity index. The Guangxi and Fujian clades clustered together with a 0.76 similarity index, and they formed the sister clade to the Sichuan/Hainan clade with a 0.71 similarity index. The Guangdong clade was in a basal polytomy with a 0.70 similarity index. Based on the abundant DNA polymorphisms, these longan accessions are distinguishable using our improved RAPD technique. Therefore, RAPD analysis is an effective technique in distinguishing the geographical origins of D. longan. Moreover, the improved method could also be employed for a variety of applications including genetic diversity and fingerprinting analyses.

Abstract:Rhubarb (official Da-huang) is an important medicinal herb in Asia. Many adulterants of official Da-huang have been discovered in Chinese markets in recent years, which has resulted in adverse effects in medicinal treatment. Here, novel molecular markers based on a short maturase K (matK) gene were developed for authenticating official Da-huang. This study showed that all the species from official Da-huang were clustered together in one clade in the polygenetic trees based on short matK. Two highly conserved single nucleotide polymorphisms of short matK were mined in the species from official Da-huang. Based on these polymophisms, four improved specific primers of official Da-huang were successfully developed that generated reproducible specific bands. These results suggest that the short matK sequence can be considered as a favorable candidate for distinguishing official Da-huang from its adulterants. The established multiplex allele-specific PCR was determined to be simple and accurate and may serve as a preferable tool for authentication of official Da-huang. In addition, we suggest that short-sized specific bands be developed to authenticate materials used in traditional Chinese medicine.

Abstract:Background The rhizome of Atractylodes chinensis (RAC), also known as "Cangzhu", is a well-known Chinese herbal medicine (CHMs). Due to the scarcity of wild resources, cultivated RAC and counterfeit products derived from the rhizome of Atractylodes japonica (RAJ) are also prevalent on the market. Diverse origins lead to varying quality. At present, the Chinese Pharmacopoeia (2020 edition) uses atractylodin as the only quantitative index component for RAC, and there is a lack of research on quality markers (Q-markers) that are closely related to efficacy. Purpose This work aims to discover the Q-markers of RAC based on chemical and pharmacological strategies. Methods The chemical composition of RAC was analyzed and identified through gas chromatography-mass spectrometry (GC–MS). The ′′Chromatographic Fingerprint Similarity Evaluation System for Traditional Chinese Medicine′′ was utilized to assess the similarity of the samples and identify the characteristic peaks. Network pharmacology was employed to screen targets and pathways for characteristic components of the RAC, construct "active ingredients–targets–pathways" network diagrams, and predict the Q-markers. The Q-markers for RAC were identified through an integrated analysis of the chemical components, network pharmacology, pharmacological activities and clinical applications. In addition, a preliminary evaluation of 28 batches of "Cangzhu" from different origins were conducted by GC-MS and chemical pattern recognition. Results A total of 16 specific components of RAC were identified by GC–MS fingerprint. The network pharmacology study found that these 16 components primarily function through 29 core targets and 23 pathways. Three compounds, hinesol, atractylon, and β-eudesmol were identified as the potential Q-markers for RAC. The evaluation of 28 batches of "Cangzhu" showed that these three Q-markers could effectively distinguish RAC from RAJ, and RAJ could not be used as a substitute for RAC. There was no significant difference between cultivated RAC and wild RAC, and the quality of cultivated RAC was more uniform than wild RAC. Conclusion This study identified and verified three potential Q-markers for RAC by using a comprehensive strategy combing of "GC–MS fingerprinting – network pharmacology – chemical pattern recognition". It provides a basis for the establishment of a more reasonable RAC quality evaluation system. Furthermore, it provides a reference for the discovery of Q-markers in other CHMs.
