Abstract:We synthesized a duplex cyclodextrin with two aliphatic links and functionalized with eight pentyl ethers orientated towards the centre of the duplex. We found that this molecule self-assembled in water to form stable vesicles with a diameter within the range 1–10μm.

Abstract:A new class of crystalline inclusion complexes of a multiarm hyperbranched polyether combined with various cyclodextrins (CDs) was successfully prepared. Using self-condensing ring-opening polymerization, a kind of multiarm polyether with a hyperbranched poly(3-ethyl-3-oxetanemethanol) core and multiple linear poly(ethylene glycol) (PEG) arms was obtained. It has been found that this kind of hyperbranched polyether can be dissolved in water. Adding alpha-CDs to the multiarm hyperbranched polyether solution, molecular recognition results in the formation of crystalline inclusion complexes based on the noncovalent interactions between the linear PEG arms of the polyether particles and the a-CDs. These multiarm polyether inclusion complexes have been well characterized. Interestingly, quite different from inclusion complexes of CDs and linear polymeric guests, the complexes of the multiarm hyperbranched polyether with a-CDs show a novel lamellar morphology. The experimental results validate that the resultant lamellar crystals have a juxtaposed structure. In addition, the formation mechanism of these inclusion complexes of a multiarm polyether with a-CDs has also been well described. Besides the role of displacement of associated water molecules and the presence of hydrogen bonding between CDs in channel structure CD inclusion complexes, the noncovalent intermolecular forces between CDs and polymers also play an important role in the formation of complex architectures.

Abstract:A series of novel cyclodextrin derivatives, i.e., mono[6-O-(p-methylbenzoyl)]- β-cyclodextrin), mono[6-O-(p-bromobenzoyl)] -β-cyclodextrin and mono(6-O-cinnyl) β-cyclodextrin were synthesized from β-cyclodextrin to reveal the role of the hydrophilic group in the guest molecule. Their structures were confirmed by elementary analysis, IR, 1H NMR, 13C NMR, EI-MS and HR-MS.

Abstract:Two novel benzo-15-crown-5 tethered β-cyclodextrins 1 and 2have been synthesized by coupling substituted benzo-15-crown-5 with correspondingβ-cyclodextrin derivatives. Their inclusion complexation behavior withrepresentative guests, such as cyclohexanol, cyclohexane carboxylic acid, cyclohexaneacetic acid, sodium cyclohexane carboxylate, and potassium cyclohexane carboxylate,was investigated in aqueous solution by means of fluorescence spectrometry. As compared with parent β-cyclodextrin, benzo-15-crown-5 tethered β-cyclodextrins 1–2 display significantly enhanced molecular binding abilities and selectivities towards model substrates, especially towards substrates containing alkali-metal cations. These results indicate that, bearing two recognition sites in a single molecule, these supramolecular architectures can strongly enhance the molecular binding ability of parent β-cyclodextrin by the cooperative binding of the β-cyclodextrin cavity and the crown ether moiety. Possessing a shorter linker, crown ether-β-cyclodextrin 2 shows much higher binding affinity with guest molecules than crown ether-β-cyclodextrin 1, which may be attributed to the binding size and molecular multiple recognition behavior between host and guest.

Abstract:Cyclodextrins can encapsulate various molecules and have exciting applications in supramolecular, pharmaceutical, material, food and environmental chemistry. Typically, the encapsulation into the cyclodextrin cavity confers to the guest improved solubility, stability, bioavailability and organoleptic properties. In recent years, cyclodextrin multi-cavity systems have also been studied to encapsulate biomolecules or drugs. This context has inspired us to synthesize and characterize two new β and γ-cyclodextrin dimers and compare their complexation behavior. The two cavities in the same molecule significantly improve the stability of inclusion complexes. Particularly we investigated cholic acid, cholesterol and doxorubicin as guests. The extent of binding between a guest molecule and β and γ-cyclodextrin dimers was established by molecular docking. Cholic acid at physiological pH was chosen as a water-soluble cholesterol analog for the investigation in water by isothermal calorimetry. We found that dimers can include cholate species, cholesterol and doxorubicin better than native cyclodextrins, and the cyclodextrin type tunes the stability of inclusion complexes.

Abstract:Two novel phosphoryl-bridged bis- and tris(beta-cyclodextrin)s of different tether lengths, i.e., bis[m-(N-(6-cyclodextryl)-2-aminoethylaminosulfonyl)phenyl]-m-(chlorosulfonyl)phenylphosphine oxide (5) and tris[m-(N-(6-cyclodextryl)-8-amino-3,6-diazaoctylaminosulfonyl)phenyl]phosphine oxide (6), have been synthesized by reactions of 6-oligo(ethylenediamino)-6-deoxy-beta-cyclodextrins with tris[m-(chlorosulfonyl)phenyl]phosphine oxide. The complex stability constants (K(S)), standard molar enthalpy (Delta H degrees ), and entropy changes (Delta S degrees ) were determined at 25 degrees C for the inclusion complexation of phosphoryl-modified bis- and tris-cyclodextrins (5 and 6, respectively), mono[6-O-(ethoxyhydroxyphosphoryl)]-beta-cyclodextrin (2), mono[6-O-(diethylamino-ethoxyphosphoryl)]-beta-cyclodextrin (3), and mono[6-O-(diphenoxyphosphoryl)]-beta-cyclodextrin (4) with representative alicyclic and N-Cbz-D/L-alanine guests in 0.1 M phosphate buffer solution at pH 7.2 by means of titration microcalorimetry. The thermodynamic parameters obtained indicate that the charge-dipole interaction between the phosphoryl moiety and the negatively charged guests, as well as the conformational difference of modified beta-cyclodextrins in aqueous solution, significantly contribute to the inclusion complexation and the enhanced chiral discrimination. The interactions and binding modes between the hosts and chiral guests were further studied by two-dimensional NMR spectroscopy to elucidate the influence of the structural features of hosts on their increased chiral recognition ability and to establish the correlation between the conformation of the resulting complexes and the thermodynamic parameters obtained.

Abstract:The supramolecular interaction between the host molecule β-cyclodextrin (β-CD) and the guest molecule adamantane is extensively applied in several disciplines. However, recent studies on the applications of this molecular recognition mainly focused on glass, silicon, and gold substrates. Few studies have explored the field of poly(dimethylsiloxane) (PDMS), an optically transparent elastomer widely used in biological microfluidic devices. In the present study, various functional group-modified adamantyl-terminated dendronized molecules were synthesized via an efficient and facile route. The binding of adamantyl-terminated dendronized molecules onto β-CD-conjugated PDMS surfaces and their reversible dissociation from PDMS surfaces through the competitive mechanism of β-CD-N3 were studied. The results showed that the dissociation is relatively slow compared with the binding, and the former process can be accelerated through exchanging the original β-CD-N3 solution with a fresh one. Afterward, more complex multilayer assemblies were constructed on PDMS surfaces using the host–guest interaction between adamantane and β-CD as well as between biotin and streptavidin. A further study on the association and dissociation of the established multilayer assemblies showed that the assembly constructed with a biotinylated monoadamantyl-terminated molecule has a reversible property, whereas that with a biotinylated triadamantyl-terminated molecule has an irreversible property. These results demonstrated that the reversible properties of the fabricated assembly can be easily modulated on the PDMS surfaces by regulating the valence of the supramolecular interaction between adamantane and β-CD.

Abstract:To investigate quantitatively the cooperative binding ability of beta-cyclodextrin dimers, a series of bridged bis(beta-cyclodextrin)s with 2,2'-diselenobis(benzoyl) spacer connected by different lengths of oligo(ethylenediamine)s (2-5) and their platinum(IV) complexes (6-9) have been synthesized and their inclusion complexation behavior with selected substrates, such as Acridine Red, Neutral Red, Brilliant Green, Rhodamine B, ammonium 8-anilino-1-naphthalenesulfonate, and 6-p-toluidino-2-naphthalenesulfonic acid, were investigated by means of ultraviolet, fluorescence, fluorescence lifetime, circular dichroism, and 2D-NMR spectroscopy. The spectral titrations have been performed in aqueous phosphate buffer solution (pH 7.20) at 25 degrees C to give the complex stability constants (K(S)) and Gibbs free energy changes (-DeltaG degrees ) for the inclusion complexation of hosts 2-9 with organic dyes and other thermodynamic parameters (DeltaH degrees and TDeltaS degrees ) for the inclusion complexation of 2-5with fluorescent dyes ANS and TNS. The results obtained indicate that beta-cyclodextrin dimers 2-5 can coordinate with one or two platinum(IV) ions to form 1:1 or 1:2 stoichiometry metallobridged bis(beta-cyclodextrin)s. As compared with parent beta-cyclodextrin (1) and bis(beta-cyclodextrin)s 2-5, metallobridged bis(beta-cyclodextrin)s 6-9 can further switch the original molecular binding ability through the coordinating metal to orientate two beta-cyclodextrin cavities and an additional binding site upon the inclusion complexation with model substrates, giving the enhanced binding constants K(S) for both ANS and TNS. The tether length between two cyclodextrin units plays a crucial role in the molecular recognition with guest dyes. The binding constants for TNS decrease linearly with an increase in the tether length of dimeric beta-cyclodextrins. The Gibbs free energy change (-DeltaG degrees ) for the unit increment per ethylene is 0.32 kJ.mol(-)(1) for TNS. Thermodynamically, the higher complex stabilities of both ANS and TNS upon the inclusion complexation with 2-5 are mainly contributed to the favorable enthalpic gain (-DeltaH degrees ) by the cooperative binding of one guest molecule in the closely located two beta-cyclodextrin cavities as compared with parent beta-cyclodextrin. The molecular binding ability and selectivity of organic dyes by hosts 1-9 are discussed from the viewpoints of the multiple recognition mechanism and the size/shape-fitting relationship between host and guest.

Abstract:A novel bridged P-CD dimer in which two beta-cyclodextrins were linked by a naphthalene at positions 2 and 7 has been synthesized. H-1 and (CNMR)-C-13 measurements showed that a large change in the conformation of the dimer occurred in aqueous solution. The dimer interacted with methyl and ethyl orange to form stable inclusion complexes via "induced fit" mechanism.

Abstract:A new soluble cyclodextrin derivative 6-O-(2-hydroxybutyl)-beta-cyclodextrin (6-HB-beta-CD) was prepared. Its molecular binding and recognition ability were investigated with the comparison of beta-cyclodextrin (beta-CD), 2-O-(2-hydroxypropyl)-beta-cyclodextrin (2-HP-beta-CD), 6-O-(2-hydroxypropyl)-beta-cyclodextrin (6-HP-beta-CD), and 2-O-(2-hydroxybutyl)-beta-cyclodextrin (2-HB-beta-CD). The relationship between the complex stability constants and the possible structures of inclusion compounds was discussed with the interaction of hosts and guests, including the weak hydrophobic interactions, the size/shape matching, the steric hindrance, and the hydrophilic property.

Abstract:Desolvation processes, though common in self-assembled biological structures, are rarely evidenced and utilized in the design of crystalline architectures. In this study, we introduce a novel approach using the [Mo8S8O8(OH)8(guest)]2- complex, formed by the self-condensation of four [MoV 2O2S2]2- fragments around a guest unit (MoVIO6H4 or oxalate), as a chaotropic scaffold for crystallizing hybrid organic-inorganic systems with natural cyclodextrins. Our findings reveal that β-cyclodextrin (β-CD) facilitates the formation of host-guest complexes, while α-cyclodextrin (α-CD) induces the formation of a Kagome-type structure with significant voids. These new compounds were thoroughly characterized using X-ray diffraction (both powder and single-crystal), N2 adsorption, elemental and thermogravimetric analysis. Additionally, solution studies using 1H NMR titration and small-angle X-ray scattering (SAXS) demonstrated pre-association of the building units in solution. These results enhance our understanding of the design principles for supramolecular structures composed of inorganic polyanions and cyclodextrins.

Abstract:Two triazinylanilino-bridged bis(beta-cyclodextrin)s were synthesized by the reactions of N,N-diethyl-4-(4,6-dichloro-1,3,5-triazin-2-yl)aniline with 6-ethylenediamino-6-deoxy-beta-cyclodextrin and 6-diethylenetriamino-6-deoxy-beta cyclodextrin. The complex stability constants for the inclusion complexation of these bis(beta-cyclodextrin)s with several dyes possessing different shapes and electron intensities were determined by means of fluorescence and UV-Vis titrations at 25 degrees C in phosphate buffer solution (pH 7.2). The results show that the triazinylanilino-bridged bis(beta-cyclodextrin)s exhibit not only the high complex stability constants, i.e. 6 similar to 26 times higher than that of native beta-cyclodextrin, for triangular guest crystal violent that possesses a larger electron intensity, but also the high molecular selectivity for crystal violent/neutral red pair (K-CV/K-NR = 23 similar to 31). Molecular dynamics studies show that the strong binding ability of triazinylanilino-bridged bis(beta-cyclodextrin)s can be mainly attributed to the cooperative binding of two cyclodextrin cavities and the bridge chain, especially the pi-pi interactions between the triazinylaniline group in the bridge chain and the guest dye.

Abstract:The monotosylates obtained by treatment of α- and β- cyclodextrin with p- methylbenzenesulfonyl chloride reacted with ammonia to give the title compounds. These amines are of unusually low basicity , with pKa values of 8.70 and 8.72, respectively. In water at 25°, the hydrochloride salt of the amine derived from β- cyclodextrin is approximately 40 times more soluble than β-cyclodextrin and, through complexation, the salt increases the solubility of Nabumetone over 800 times.
