Abstract:An improved process was reported to solve the problems such as low yield and high costs in the synthesis of 3-n-propyl-2, 4-dihydroxyacetophenone, which is a key intermediate of Leukotriene receptor antagonist. The target compound was synthesized starting from m-dihydroxybenzene, via acylation, allylation, Claisen rearrangement and hydrogenation reduction. The optimal reaction conditions were as follows. The acylation reaction which used acetic acid as acylation agent and ZnCl 2 as catalyst was carried out under reflux for 5 h, and 77.9% yield was obtained. The allylation reaction using allyl chloride as allylation reagent and NaBr, KI as catalyst was carried out at 40°C for 2 h and continued at 70°C for 14 h, to give 98.6% yield. In Claisen rearrangement carried out at 205°C for 4.5 h, diphenyl ether was used as reaction solvent and NaCl was used as catalyst, and the yield was improved to 67.4%. In the final step, the hydrogenated reduction with Pd/C (w(Pd) is 5%) as catalyst was carried out at 45°C for 6 h, and 88.6% yield was obtained. The process is much of industrial value because of high yield, cheap and available materials, and convenient operations. The structure of the main compounds was confirmed by 1H-NMR, GC-MS and IR.

Abstract:A parallel process for the new synthesis of 2,7-dimethyl-2,4,6-octatriene-1,8-dial was investigated.3-(5,5-Dimethyl-1,3-dioxan-2-yl)-2-butenal was obtained by a process involving aldehyde protection,ester exchange and catalytic oxidation from 4-acetoxy-2-methyl-2-butenal.A new catalytic oxidation process was proposed to get 3-(5,5-dimethyl-1,3-dioxan-2-yl)-2-butenal by oxidating 3-(5,5-dimethyl-1,3-dioxan-2-yl)-2-buten-1-ol with copper(Ⅰ) chloride、2,2,6,6-tetramethyl-1-piperidinyloxy and oxygen.3-(5,5-Dimethyl-1,3-dioxan-2-yl)-2-buten-1-ol was chlorizated with SOCl_2 in DMF to get 2-(3-chloro-1-methyl-1-propenyl)-5,5-dimethyl-1,3-dioxane.2,7-Dimethyl-2,4,6-octatriene-1,8-dial was acquired in an overall yield of 48% by one-pot synthesis method from 3-(5,5-dimethyl-1,3-dioxan-2-yl)-2-butenal,2-(3-chloro-1-methyl-1-propenyl)-5,5-dimethyl-1,3-dioxane and triethylphosphite.

Abstract:A novel class of chiral glycosyl-triazole-based P,N ligands were synthesized by 'click chemistry' and were applied to the Rh-catalyzed asymmetric hydrosilylation of a range of substituted acetophenones. Abstract:A new, practical and cost-effective route for scalable synthesis of 2-[(4-dodecyloxyphenyl)sulfonyl] butanoic acid, a key intermediate of a new cyan dye-forming coupler containing a sulfone group, was developed by adopting phenol as the starting material. The synthesis was accomplished in five steps with etherification, chlorosulfonation, reduction, nucleophilic reaction by C-S coupling and hydrolyzation. An important objective of the new synthetic route was the synthesis of 2-[4-(dodecyloxyphenyl)sulfonyl]butanoate. Overall yield obtained at optimized conditions increased to 66 %. The synthetic strategy was proven to be a process enabling rapid delivery of the target product with high purity.

Abstract:The synthesis of 2, 4-diisopropyl-5,5-dimethyl-1,3-dioxane through the acetalization of isobutyraldehyde with 2, 2, 4-trimethyl-1,3-pentanediol (TMPD) catalyzed by the novel carbon based acid was first carried out. High conversion (≥98%) and specific selectivity were obtained using the novel carbon based acid, which kept high activity after it was reused 5 times. Moreover, the catalyst could be used to catalyze the acetalization and ketalization of different aldehydes and ketones with superior yield. The yield of several products was over 90%. The novel heterogeneous catalyst has the distinct advantages of high activity, strikingly simple workup procedure, non-pollution, and reusability, which will contribute to the success of the green process greatly.

Abstract:(E)-4-bromo-3-methyl-2-butenoic acid was conveniently synthesized from 3-methyl-2-butenoic acid in 29% overall yield.In this procedure,the title compound was easily obtained by hydrolysis of (E)-trimethylsilyl 4-bromo-3-methyl-2-butenoate,which could be isolated from its mixture of Z-/E-isomer by distillation in vacuo.The isomeric mixture was prepared via the esterification of 3-methyl-2-butenoic acid with chlorotrimethylsilane,followed by bromination with NBS.

Abstract:A series of new chiral C-3-symmetric tris(beta-hydroxy amide) ligands have been synthesized via the reaction of 1,3,5-benzenetricarboxylic chloride and optically pure amino alcohols (up to 96% yield). The asymmetric catalytic alkynylation of aldehydes with these new C3-symmetric chiral tris(beta-hydroxy amide) ligands and Ti ((OPr)-Pr-/)4 Was investigated. Ligand 4c synthesized from (1R,2S)-(-)-2-amino-1,2-diphenylethanol is effective for the enantioselective alkynylation of various aldehydes, and high enantioselectivity was obtained with aromatic aldehydes and alpha,beta-unsaturated aldehyde (up to 92% ee).

Abstract:The combination of zirconocene-mediated coupling of silylated alkynes with a protonation-desilylation or bromination-desilylation process afforded otherwise unavailable butadiene derivatives. When (E,E)-2,3-dialkyl-1,4-bis(trimethylsilyl)-1,3-butadienes were treated with 3 equiv of Br-2 in CH2Cl2, (E)-2,3-dialkyl-1,1,4,4-tetrabromo-2-butenes were obtained in excellent yields with perfect stereoselectivity.

Abstract:A nonracemic 3-silyl-3-borylhex-4-enoate reagent has been developed. Its asymmetric crotylboration of aldehydes provides Z-anti-homoallylic alcohols possessing a trisubstituted vinylsilane in high yields with excellent stereo- and enantioselectivity. Diverse decoration of vinylsilane and ester groups, as well as formation of functionalized THF rings, showcase the potential of the approach in the synthesis of polyketide natural products.

Abstract:A new way to synthesis of (S)-tert-butyl 6-benzyloxy-5-hydroxy-3-oxohexanoate was investigated. The target compound was successfully prepared via several mildly reaction processes by using R-epichlorohydrin, which is an inexpensive and easily available starting material. Through screening the amounts of BF3 center dot Et2O and comparing two different synthetic methods, this route becomes more suitable for large-scale preparation for its high yields, mildly reacting-conditions, easily available materials and facilely purification of products.

Abstract:Z(E)-ethyl 3-oxo-2-(4-phenyl-1,3-dithiolan-2-ylidene)butanoate (1) was prepared in moderate (40.3%) yield by the reaction of ethyl 3-oxobutanoate with carbon disulfide and 1-(1,2-dibromoethyl)benzene with potassium carbonate as the base in DMF.Subsequently,the condensation reaction of 1 with arylalde- hydes was performed in the presence of C_2H_5ONa in C_2H_5OH,in which the product of condensation and ester hydrolysis:(2Z(E),4E)-3-oxo-5-phenyl-2-(4-phenyl-1,3-dithiolan-2-ylidene)pent-4-enoic acid (2) were isolated in good yield.

Abstract:A highly efficient and enantioselective synthesis of axially chiral α-allenols was realized in practical yields with 96-99% ee or de from TBS-protected propargylic alcohols, aldehydes, and a commercially available, inexpensive, chiral, secondary amine (S)-α,α-diphenylprolinol or its enantiomer followed by desilylation. The easily removable TBS group not only acts as a protecting group, but also as a possible sterically directing group for the excellent enantioselectivity and in situ prevention of possible allene racemization.

Abstract:The Cu-catalyzed 1,4-protosilylation and protoborylation of trifluoromethyl-substituted conjugated enynes were developed to access functionalized homoallenylsilanes and homoallenylboronates. This protocol also provides a general method to synthesize optically active homoallenylsilanes and homoallenylboronates in moderate to excellent yields with high enantiomeric excess by using new designed chiral bisoxazoline ligands. Simultaneously, the transformations of homoallenylsilanes and homoallenylboronates were also explored to synthesize useful building blocks.

Abstract:A new synthetic method for ((4R,6S)-6-hydroxymethyl-2,2-dimethyl-1,3-dioxinan-4-yl)-acetic acid tert-butyl ester was accomplished by a chemoenzymatic approach. Racemic 4-(4-methylbenzyloxy)-3-hydroxybutyronitrile (3) was firstly synthesized via condensation and epoxy opening reactions. (S)-4-(4-Methylbenzyloxy)-3-hydroxybutyronitrile (4) was obtained from resolution of racemic 3 by the lipase from Artgribacter sp. with a high enantiomeric excess (98% ee) in the mixture solvent of acetonitrile/hexane (V : V= 1 : 4). Dihydroxy ester 7 was synthesized by a syn-selective triethylborane mediated reduction with a de value of 97%. After acid-catalyzed protection of 7 and hydrogenolysis of the benzyl protecting group, the objective 9 was obtained with a high optical purity.

Abstract:Dibenzocyclooctadiene lignans are an interesting class of molecules because of their unique structure based on an axially chiral biaryl moiety as well as their significant biological activity. Herein, we describe the development of a palladium-catalyzed atroposelective C-H alkynylation and its application in gram-scale, stereocontrolled formal syntheses of (+)-isoschizandrin and (+)-steganone. tert-Leucine was identified as an efficient, catalytic transient chiral auxiliary. A wide range of enantiomerically enriched biaryl compounds were prepared by this approach in good yields (up to 99 %) with excellent enantioselectivity (up to >99 % ee).

Abstract:A facile method for the preparation of tert-butyl (3R,5S)-6-hydroxy-3,5-O-isopropylidene-3,5-dihydroxyhexanoate is described by a chemoenzymatic approach. In this method, one hydroxyl stereocenter at C5 is obtained with a high ee value (up to 98.0%) via an enzymatic transesterification resolution of l-chloro-3-(4-methylbenzyloxy)-2-propanol. The other hydroxyl stereocenter at C3 was built with 98.0% de, by acid-hydrolysis of a 1,3-diol-acetonide syn/anti-10. It is noteworthy that the reduction of β-hydroxy ketone 8 with sodium borohydride can be carried out smoothly in aqueous isopropyl alcohol with a high diastereomeric ratio syn/anti (drs:a) of 4.0:1.

Abstract:Copper-catalyzed highly efficient hydrosilylation reaction of enynoates was developed. Under simple reaction conditions, various di-, tri-, and tetrasubstituted racemic allene products could be obtained in high yields. The asymmetric 1,6-addition of silyl group to the (Z)-2-alken-4-ynoates could be achieved under mild reaction conditions to afford the silyl-substituted enantioenriched chiral allene products in good yields and with high enantioselectivities.
