Abstract:C-Glycosides are a class of carbohydrates with a variety of biological activities, and the construction of their unique C-C glycosidic bond is a hot and challenging topic in carbohydrate chemistry. The formation of new C-C bond from carbon-carbon double bond is a common strategy in organic chemistry. With the rapid development of ene-chemistry, great Progress in the synthesis of C-glycosides from glycol donors containing 1,2-double bond has been made. In this review, the methods for the synthesis of C-glycosides based on glycals are summarized, including Ferrier I-type C-glycosylation, Fleck-type C-glycosylation, C-glycosylation of 1-substituted glycols using transition metal-catalyzed coupling reaction on, chael-type C-glycosylation of 2-substituted glycols, and radical addition type C-glycosylation of glycols.

Abstract:Aryl-glycosides represent a significant subclass of crucial glycosidic compounds, increasingly capturing the attention of pharmaceutical developers as bioelectronic motifs embedded within glycosides. Their outstanding resistance to enzymatic hydrolysis bestows a distinctive advantage in the field of drug development, particularly in therapeutic domains such as diabetes treatment, where pharmaceuticals based on the C -aryl-glycoside architecture manifest compelling therapeutic efficacy. As a result, researchers in the realm of synthetic chemistry have diligently explored and devised a plethora of streamlined and efficacious synthetic methodologies. Abstract:The [5+2] cycloaddition is one kind of fascinating and powerful reaction to access seven-membered ring systems existing in complex natural products and pharmaceuticals. In this review, recent developments on the [5+2] cycloadditions and their application in the synthesis of complex natural products are discussed. A focus is given to the way in which the [5+2] cycloadditions have influenced the logical process of the synthesis. Abstract:C-oligosaccharides are pharmacologically relevant because they are more hydrolysis-resistant than O-oligosaccharides. Despite indisputable advances, C-oligosaccharides continue to be underdeveloped, likely due to a lack of efficient and selective strategies for the assembly of the interglycosidic C-C linkages. In contrast, we, herein, report a versatile and robust strategy for the synthesis of structurally complex C-oligosaccharides via catalyzed C(sp3 )-H activations. Thus, a wealth of complex interglycosidic (2→1)- and (1→1)-C-oligosaccharides becomes readily available by palladium-catalyzed C(sp3 )-H glycoside glycosylation. The isolation of key palladacycle intermediates and experiments with isotopically-labeled compounds identified a trans-stereoselectivity for the C(sp3 )-H glycosylation. The glycoside C(sp3 )-H activation manifold was likewise exploited for the diversification of furanoses, pyranoses and disaccharides.

Abstract:C-Glycosides represent a privileged class of carbohydrates, which exhibit remarkable biological and pharmaceutical activities. Transition metal-catalyzed cross coupling reaction is a powerful tool for the efficient and rapid construction of C-C bonds. Among various transition-metal catalyzed methods, palladium-catalyzed C-glycosylation is an effective and reliable approach to synthesize C-glycosides, which involves the reaction of glycoyl donors and various reaction partners via C-C glycosidic bond formation under mild conditions with high regio/stero-selectivities. In this review, palladium-catalyzed C-glycosylation reactions are summarized and discussed in detail with the focus on the synthesis of C-aryl glycosides, C-alkyl glycosides, C-alkenyl glycosides, and other functionalized C-glycosides since 2020 to 2024. Some representative synthetic strategies and their transformation application along with reaction mechanisms are also highlighted.

Abstract:Amino sugars are a kind of carbohydrates with one or more hydroxyl groups replaced by an amino group. They play crucial roles in a broad range of biological activities. Over the past few decades, there have been continuing efforts on the stereoselective glycosylation of amino sugars. However, the introduction of glycoside bearing basic nitrogen is challenging using conventional Lewis acid-promoted pathways owing to competitive coordination of the amine to the Lewis acid promoter. Additionally, diastereomeric mixtures of O-glycoside are often produced if aminoglycoside lack a C2 substituent. This review focuses on the updated overview of the way to stereoselective synthesis of 1,2-cis-aminoglycoside. The scope, mechanism, and the applications in the synthesis of complex glycoconjugates for the representative methodologies were also included.

Abstract:Compounds containing a 2‐deoxy β‐glycoside linkage play important roles in carbohydrate chemistry, serving as potential therapies with significant biological properties. From the viewpoint of synthesis, many strategies developed in recent years targeted the stereoselective construction of 2‐deoxy glycosides. This review aims to summarise recent advances in the synthetic routes toward 2‐deoxy β‐glycosides developed over the last decade. Where associated, practical applications of these strategies in the total synthesis of natural products are highlighted.

Abstract:2-Deoxy-α-C-Glycosides are a significant class of carbohydrates found in numerous bioactive molecules and medicines. Developing a concise strategy for the assembly of these α-configured C-glycosides is crucial in the field of carbohydrate chemistry. However, current methods are restricted to the utilization of glycosyl radical precursors, which are required for pre-syntheses. Herein, we present a novel approach for the synthesis of 2-deoxy-α-C-glycosides using a nickel-catalyzed stereoselective coupling reaction with commercially available glycals. Notably, this method circumvents the preparation for diverse glycosyl radical precursors. The developed protocol exhibits a broad substrate scope and remarkable stereoselectivity under mild reaction conditions. Furthermore, the raw materials required for this process are readily accessible, eliminating the necessity for pre-functionalization modifications of the glycosyl substrates and ensuring high atomic economy.

Naturally derived benzopyrans, benzopyrones, and flavonoids are an essentially prominent group of oxa-heterocycles, highly significant targets in medicinal chemistry owing to their extensive abundance in biologically active natural products and pharmaceuticals. The molecular complexity and stereoselectivity induced by heterocycles embedded with C -glycosides have attracted considerable interest and emerged as a fascinating area of research for synthetic organic chemists. This present article emphasizes the existing growths in the strategies involving the diastereoselective synthesis of C -glycosylated benzopyrans, benzopyrones, and flavonoids using naturally acquired glycones as chiral synthons.

Abstract:Polysaccharides are a class of bio-macromolecules with highly complex structures that are widely found in living organisms such as microorganisms, plants and animals. Polysaccharides serve not only as structural components and energy sources of cells, but also as important signaling molecules which are involved in many key biological processes. Studies on polysaccharide-mediated biological processes require access to structurally defined molecules, which approach the size and complexity of those found in nature, but naturally-occurring polysaccharides usually exist in microheterogeneous forms, making it difficult or even impossible to isolate pure polysaccharides from natural sources in most cases. Chemical synthesis represents a reliable solution to this problem, which can provide polysaccharide samples with defined chemical structures for functional studies and even a library of analogs of natural glycans for structure-activity relationship investigations. But unlike oligonucleotides and peptides, which can already be obtained by automated synthesizers in a very short of time, the chemical synthesis of glycans remains a great challenge for synthetic chemists. The major challenge for glycan synthesis lies in the need to handle both stereo-and regio-chemistry in the construction of each glycosyl linkage, and the extensive protecting-group manipulations as well as much intermediate separation make it a tedious and time-consuming process. Over the past decades, carbohydrate chemists have developed many glycosylation reactions. A series of strategies for glycan assembly have been also established. The advances in both synthetic methods and strategies have significantly increased the synthetic efficiency of carbohydrate molecules, and many great accomplishments in the field of polysaccharide synthesis have been witnessed in recent decades. Some representative methods and strategies, and their successful applications in the chemical synthesis of complex polysaccharides are summarized in this review.

Abstract:Multisubstituted guanidines have received much attention because of important applications in many fields, such as pharmaceutics, organometallic and coordination chemistry, and organic synthesis. Although classical methods are available for the preparation of guanidines, synthetic preparation approaches to guanidines are still in great demand. In this review, we summarize recent developments on synthetic methods via the C-N bond formation. Three aspects are included: transition-metal-catalyzed guanidine synthesis based on classical methods; catalytic guanylation reaction of amines with carbodiimides; and tandem catalytic guanylation/cyclization reactions.

Abstract:C -glycosides represent a large group of natural products with a C-C bond between the aglycone and the sugar moiety. They exhibit great structural diversity, wide natural distribution, and significant biological activities. By the end of 2021, at least 754 C -glycosides and their derivatives have been isolated and characterized from plants. Thus far, 66 functional C -glycosyltransferases (CGTs) have been discovered from plants, and provide green and efficient approaches to synthesize C -glycosides. Herein, advances in plant-derived C -glycosides are comprehensively summarized from aspects of structural diversity and identification, bioactivities, and biotechnological production. New strategies to discover novel C -glycosides and CGTs, as well as the applications of biotechnological methods to produce C -glycosides in the future are also discussed.

Abstract:C-Glycosides are critical motifs embedded in many bioactive natural products. The inert C-glycosides are privileged structures for developing therapeutic agents owing to their high chemical and metabolic stability. Despite the comprehensive strategies and tactics established in the past few decades, highly efficient C-glycoside syntheses via C-C coupling with excellent regio-, chemo-, and stereoselectivity are still needed. Here, we report the efficient Pd-catalyzed glycosylation of C-H bonds promoted by weak coordination with native carboxylic acids without external directing groups to install various glycals to the structurally diverse aglycon parts. Mechanistic evidence points to the participation of a glycal radical donor in the C-H coupling reaction. The method has been applied to a wide range of substrates (over 60 examples), including many marketed drug molecules. Natural product- or drug-like scaffolds with compelling bioactivities have been constructed using a late-stage diversification strategy. Remarkably, a new potent sodium-glucose cotransporter-2 inhibitor with antidiabetic potential has been discovered, and the pharmacokinetic/pharmacodynamic profiles of drug molecules have been changed using our C-H glycosylation approach. The method developed here provides a powerful tool for efficiently synthesizing C-glycosides to facilitate drug discovery.
