Abstract:The paper presents a novel strategy to identify analytical markers of traditional Chinese medicine preparation (TCMP) rapidly via direct analysis in real time mass spectrometry (DART-MS). A commonly used TCMP, Danshen injection, was employed as a model. The optimal analysis conditions were achieved by measuring the contribution of various experimental parameters to the mass spectra. Salvianolic acids and saccharides were simultaneously determined within a single 1-min DART-MS run. Furthermore, spectra of Danshen injections supplied by five manufacturers were processed with principal component analysis (PCA). Obvious clustering was observed in the PCA score plot, and candidate markers were recognized from the contribution plots of PCA. The suitability of potential markers was then confirmed by contrasting with the results of traditional analysis methods. Using this strategy, fructose, glucose, sucrose, protocatechuic aldehyde and salvianolic acid A were rapidly identified as the markers of Danshen injections. The combination of DART-MS with PCA provides a reliable approach to the identification of analytical markers for quality control of TCMP.

Abstract:Identification of molecular markers linked with gene can promote the development of MAS(molecular assistant selection),and it is also the basic of map-based cloning.BSA(Bulked Segregation Analysis)is a useful method for rapidly identifying markers linked to any specific gene or genomic region.At first,the samples with obviously difference in the target phenotypic character are selected and then crossed.Two bulks are constituted with chosen segregation progenies individuals that are identical for a particular trait or genomic region but arbitrary at all unlinked regions.The two pools or bulks are screened for differences using proper molecular marker,then we can find some special molecular markers showing the difference between the two gene pool.The special markers must be the closely linked markers with the target gene.Many molecular markers can be used in BSA,such as RFLP(Restriction Fragment Length Polymorphism),RAPD(Random Amplified Polymorphism DNA),AFLP(Amplified Fragment length polymorphism) and SSR(Simple Sequence Repeat).

Abstract:As a novel genetic marker, microhaplotype can be applied in the field of forensic genetics. Microhaplotype has the advantages of high polymorphism, low mutation rate, no stutter products and short amplification fragments. Microhaplotype can effectively detect mixture, and quantitatively analyze the contributors of mixture. DNA with severe fragmentation can be successfully genotyped by microhaplotype. It can be used as ancestry informative marker to effectively divide the global continental population according to genetic structure. Microhaplotype system can provide more information than traditional short tandem repeat and help to identify complex relationships. It can provide new ideas for tumor source identification, cell line identification and prenatal paternity testing. Here we review the applications of microhaplotype, intending to provide references for forensic practice.

Abstract:Dendrobium, as a valuable and expensive Chinese herb, has attracted great attention in China and abroad in recent years. This paper provides the basic principles and characteristics of some DNA molecular marker techniques, including RFLP, allele-specific diagnostic PCR, RAPD, AP-PCR, AFLP,ISSR, SRAP, and DNA sequencing analysis, genechip,and reviews their application progresses in Dendrobium. These techniques provide more effective and reliable methods for discrimination of true and false Dendrobium. Abstract:After more than 10 years of development,DNA molecular markers were widely used in loquat.Types and characteristics of molecular markers utilized in the study of loquat were summarized.The progresses and achievements have been reviewed on genetic diversity and phylogenetic relationship,genetic linkage mapping of molecular markers,fingerprinting and identification of germplasm.Meanwhile,the problems presented in Loquat were discussed.Besides,development and use of more types of molecular markers of loquat,germplasm collection and evaluation based on molecular markers,construction of high-density genetic linkage mapping of molecular markers and fingerprinting,were considered as the direction of development in the future.

Abstract:Codonopsis genus is comprised of species that are perennial plants primarily distributed across all east, southeast, and Central Asia. The most famous species of Codonopsis are C. tangshen, C. lanceolate, and C. pilosula. The records showed that they have a long story usage as traditional Chinese medicines, as they were alleged to be able to intensify the spleen and the lung as well as enriching blood and engendering liquid. Certain species have a culinary value in southern China and Southeast Asia, where they are considered as tea, wine, soup, plaster, and porridge. Codonopsis species were shown to be of great importance in medicine, due to their broad biological activity. Therefore, a clear understanding of their genetic diversity is needed. Adequate distinctions and descriptions of those species are necessary to preserve plant reservoir, investigations of genes associated with desirable traits, and understanding of evolutionary relationships. Subsequently, various molecular marker techniques such as Random Amplified Polymorphic DNA (RAPD), Amplified Fragment Length Polymorphism (AFLP), Simple Sequence Repeats (SSR), and Inter Simple Sequence Repeat (ISSR), Single Nucleotide Polymorphism (SNP), internal transcribed spacer (ITS), and Sequence-Characterized Amplified Region (SCAR) have been improved to provide detailed informations about genomes, that historically were not possible to obtain based on only phenotypic methods. This review represents the usage of DNA molecular markers for molecular diversity analysis of medically important species belonging to the genus Codonopsis.

Abstract:DNA barcoding is a taxonomic method that uses a short genetic marker in an organism's DNA to identify it as belonging to a certain species.DNA barcoding can be used to overcome the shortcomings of traditional species identification methods;in particular,it has the advantage of versatility,making the process of species identification more efficient.DNA barcoding has been well applied in the identification of animal species,and in recent years it has been rapidly developed and promoted in the identification of medicinal plants.This paper reviews the principles of DNA barcoding,and introduces and discusses the limitations of traditional identification methods for Chinese medicinal herbs.The significance,application,and prospects of DNA barcoding in the identification of Chinese medicinal herbs are also examined.

Abstract:Molecular pharmacognosy is based on molecular identification technologies and has provided an identification basis of crude drugs at the gene level. Molecular pharmacognosy includes the systematic assortment of varieties of traditional herbal medicine and Chinese herbs, and studies quality, standardization, conservation, and diversity of medicinal plants. The molecular pharmacognosy in twenty-first century embraces many items from the study of molecular taxonomy, phylogenetic evolution of medicinal plants and animals, conservation of endangered species, molecular identification of medicinal raw materials, controlling metabolic pathways and biosynthetic regulation of secondary metabolites in plants, conservation of biodiversity, genetic engineering, tissue culture technology, production of pollution-free medicinal plants, structure-activity relationships with a drug potential, etc. to genetic engineering technology based on molecular cloning and the related tissue culture technology, especially molecular marker technology based on PCR. Molecular pharmacognosy is a rising discipline combining molecular biology and pharmacognosy, and its development depends on systems biology with reference to genomics, proteomics, and metabolomics, and also involved methods of modern biotechnology. Molecular biology has become vital to drug discovery from medicinal plants, through the determination and accomplishment of appropriate screening assays directed toward physiologically relevant molecular targets. DNA markers based tools for molecular identification of traditional medicinal materials are considered more reliable for authentication of herbal materials.

Abstract:<p>DNA barcoding is an emerging molecular identification and classification technology that has been applied to medicinal plants since 2008. The application of this technique has greatly ensured the safety and effectiveness of medicinal materials. In this paper, we review the application of DNA barcoding and some related technologies over the past 10 years with respect to improving our knowledge of medicinal plant identification and authentication. From single locus-based DNA barcodes to combined markers to genome-scale levels, DNA barcodes contribute more and more genetic information. At the same time, other technologies, such as high-resolution melting (HRM), have been combined with DNA barcoding. With the development of next-generation sequencing (NGS), metabarcoding technology has also been shown to identify species in mixed samples successfully. As a widely used and effective tool, DNA barcoding will become more useful over time in the field of medicinal plants.</p>

Abstract:Molecular markers, due to their stability, cost-effectiveness and ease of use provide an immensely popular tool for a variety of applications including genome mapping, gene tagging, genetic diversity diversity, phylogenetic analysis and forensic investigations. In the last three decades, a number of molecular marker techniques have been developed and exploited worldwide in different systems. However, only a handful of these techniques, namely RFLPs, RAPDs, AFLPs, ISSRs, SSRs and SNPs have received global acceptance. A recent revolution in DNA sequencing techniques has taken the discovery and application of molecular markers to high-throughput and ultrahigh-throughput levels. Although, the choice of marker will obviously depend on the targeted use, microsatellites, SNPs and genotyping by sequencing (GBS) largely fulfill most of the user requirements. Further, modern transcriptomic and functional markers will lead the ventures onto high-density genetic map construction, identification of QTLs, breeding and conservation strategies in times to come in combination with other high throughput techniques. This review presents an overview of different marker technologies and their variants with a comparative account of their characteristic features and applications.

Abstract:The authors have reviewed one new kind of molecular marker in higher plants, functional markers (FMs), based on comparisons with random DNA markers (RDMs) and gene targeted markers (GTMs) that are usually applied in genetic analysis. FMs include direct functional marker (DFM) and indirect functional marker (IFM). FMs are derived from polymorphic sites within gene motifs causally involved in phenotypic trait variation, which exhibit dominant genetic characters. FMs are independent of molecular genetic mapping and may enhance the efficiency of marker-assisted selection in crops breeding.

Abstract:In this review, the authors summarized the new technologies and methods for identifying traditional Chinese medicinal materials, including molecular identification, chemical identification, morphological identification, microscopic identification and identification based on biological effects. The authors introduced the principle, characteristics, application and prospect on each new technology or method and compared their advantages and disadvantages. In general, new methods make the result more objective and accurate. DNA barcoding technique and spectroscopy identification have their owner obvious strongpoint in universality and digitalization. In the near future, the two techniques are promising to be the main trend for identifying traditional Chinese medicinal materials. The identification techniques based on microscopy, liquid chromatography, PCR, biological effects and DNA chip will be indispensable supplements. However, the bionic identification technology is just placed in the developing stage at present.

Abstract:There are several kinds of Chinese herbal medicines originating from diverse sources. However, the rapid taxonomic identification of large quantities of Chinese herbal medicines is difficult using traditional methods, and the process of identification itself is prone to error. Therefore, the traditional methods of Chinese herbal medicine identification must meet higher standards of accuracy. With the rapid development of bioinformatics, methods relying on bioinformatics strategies offer advantages with respect to the speed and accuracy of the identification of Chinese herbal medicine ingredients. This article reviews the applicability and limitations of biochip and DNA barcoding technology in the identification of Chinese herbal medicines. Furthermore, the future development of the two technologies of interest is discussed.

Abstract:Traditional species identification has gone through five stages -- morphology, cytology, biochemistry, immunology and molecular biology. At present, the use of DNA technology for species identification has become a research hotspot. In the use of DNA for species identification, the presentation and application of DNA barcode is of epoch-making significance. With the successful application of new technology in species identification, forensic species identification has also made corresponding development, and is expected to play an important role in forensic related fields. This paper briefly describes the general situation and principles of DNA barcode technology as well as its advantages and limitations when applied to biological classification, and discusses the future significance and feasibility of DNA barcode technology in forensic applications, in order to provide new ideas for future forensic identification.
