Delayed Type Hypersensitivity On Corneal Allograft Rejection After Murine Keratoepithelioplasty

Abstract:Purpose. We have established murine keratoepithelioplasty (KEP) model and reported that grafted comeal lenticules were capable of reepitheiializing the denuded comeal surface and causing alloepithelial graft rejection (IOVS. 1995; 36:2623). In the present study, we examined whether it correlated with the elicitation of delayed type hypersensitivity (DTH) responses directed to alloepithetium. Methods. After corneal epithelial and limbal conjunctiva! removal, C3H/He comeal lenticules were grafted around the BALB/c limbus (KEP). Allograft rejection arter reepithelialization was evaluated by microscopic observation. Corneal opacity (epithelial edema, stromal opacity, or both) and neovascularization were respectively graded to 0-4 (rejection score). DTH responses of the recipients to C3H/He antigen were assessed by calculating specific ear swelling at 1, 2, and 4 weeks postoperatively. Results. The induction of epithelial edema was observed 1 week postoperatively. Corneal opacity score was at plateau during 1 -2 postoperative weeks (1 w: 1. 0 ��0. 0, 2 w: 1. 0 ��1. 2) and decreased toward 4 weeks (4 w: 0. 2 + 0. 4). Corneal neovascularization was induced at 1 week (neovascularization score: 1. 0 ��0. 6) and enhanced until 4 weeks (neovascularization score: 2. 4 ��0. 9). DTH responses were provoked within 2 weeks and maintained untii 4 weeks. Spearman's correlation analysis showed significant correlation between rejection score (comeal opacity score + neovascularization score) and DTH responses (Spearman's correlation coefficient: rs=0. 59,p<0. 05). Conclusions. Taken together with our previous findings that the induction of ACAID can inhibit alloepithelial rejection, DTH responses are considered to be responsible for mediating the allograft rejection after KEP.
