Abstract:The reductions of cytochrome c by a series of derivatives of Fe(EDTA)(2-) complex have been studied by a stopped-flow technique. The reactions of cytochrome c with Fe(EDTA)(2-), Fe(CDTA)(2-) and Fe(IDA)(2)(2-) present typical outer sphere mechanism, meanwhile the cytochrome c(III) and Fe(NTA)(-) system shows abnormal kinetic behavior, including the rate saturation, big negative entropy and lower overall charge and binding site charge calculated from the dependence of electron transfer rate on ionic strength. On the basis of these observations a semi-inner-sphere mechanism is proposed to illustrate the kinetics.
A Cu And Fe Dual-Atom Nanozyme Mimicking Cytochrome C Oxidase To Boost The Oxygen Reduction Reaction

Abstract:The intrinsically sluggish kinetics of the oxygen reduction reaction (ORR) and overuse of expensive and unstable Pt-based catalysts have severely hampered the development of clean energy technologies. Herein, a type of non-noble metal nanozyme as an ORR electrocatalyst, with Fe-Cu dual atomic sites embedded in three-dimensional porous N-doped carbon (FeCu-DA/NC), is fabricated by mimicking both the constituents of active centers and enzymatic microenvironment of Cytochrome c oxidase (CcO). The atomically dispersed Fe-Cu single atom sites are clearly identified by direct aberration-corrected transmission electron microscopy imaging, systematic X-ray absorption fine structure (XAFS) analyses, and density functional theory (DFT) calculations. FeCu-DA/NC exhibits a superior ORR activity, with about 30 mV more positive half-wave potential (E-1/2) than that of commercial Pt/C catalysts in a base, and moreover with only anE(1/2)gap of 20 mV to Pt/C in an acid. Importantly, FeCu-DA/NC displays an excellent durability under both acidic and basic conditions, which is much superior to that of Pt/C. DFT calculations further demonstrate that the enhanced performance can be attributed to the unique synergistic effect between Cu and Fe single atoms in which Cu-N(4)serves as the electron donor to increase the electron density of the active centers of Fe-N(4)and thus to facilitate O(2)activation.

Abstract:ABSTRACT Because of their cell surface locations, the outer membrane c-type cytochromes MtrC and OmcA of Shewanella oneidensis MR-1 have been suggested to be the terminal reductases for a range of redox-reactive metals that form poorly soluble solids or that do not readily cross the outer membrane. In this work, we determined the kinetics of reduction of a series of Fe(III) complexes with citrate, nitrilotriacetic acid (NTA), and EDTA by MtrC and OmcA using a stopped-flow technique in combination with theoretical computation methods. Stopped-flow kinetic data showed that the reaction proceeded in two stages, a fast stage that was completed in less than 1 s, followed by a second, relatively slower stage. For a given complex, electron transfer by MtrC was faster than that by OmcA. For a given cytochrome, the reaction was completed in the order Fe-EDTA > Fe-NTA > Fe-citrate. The kinetic data could be modeled by two parallel second-order bimolecular redox reactions with second-order rate constants ranging from 0.872 μM−1 s−1 for the reaction between MtrC and the Fe-EDTA complex to 0.012 μM−1 s−1 for the reaction between OmcA and Fe-citrate. The biphasic reaction kinetics was attributed to redox potential differences among the heme groups or redox site heterogeneity within the cytochromes. The results of redox potential and reorganization energy calculations showed that the reaction rate was influenced mostly by the relatively large reorganization energy. The results demonstrate that ligand complexation plays an important role in microbial dissimilatory reduction and mineral transformation of iron, as well as other redox-sensitive metal species in nature.
Reduction Kinetics of Cr(VI) in the Presence of Common Food Organics

Abstract:The data of Cr(VI) reduction kinetics with 15 kinds of common organic compounds in food were determined by the diphenylcarbazide spectrophotometry at different pH values and temperatures. The results indicate that the L-Ascorbic acid, L-Cysteine hydrochloride and L-Methionine have relatively strong reduction activities for Cr(VI), while Cr(VI) hardly undergoes the reduction with other 12 kinds of organics within 5 hours. Cr(VI) is completely reduced by L-Ascorbic acid in a short period of time, and completely reduced by L-Cysteine hydrochloride within 60 minutes, but reduced for 23%by L-Methionine after 5 hours. Therefore, the sequence of the reduction activity of the three organic compounds is L-Ascorbic acid >L-Cysteine hydrochloride>L-Methionine. The values of pH have a great effect on the reduction kinetics. As the pH value increases, the reduction with L-Cysteine hydrochloride salt is enhanced, while the reduction with L-Methionine is decreased, and even decreased to none. The kinetics data of Cr(VI) reduction with L-Methionine were fitted by a zero-order kinetics equation at the pH of 3.0, and the apparent activation energy was obtained as 35.5 kJ×mol-1. In addition, the kinetics data of Cr(VI) reduction with L-Cysteine hydrochloride at the pH of 3.0, 5.0 and 7.0 were fitted by a first-order kinetics equation, and the apparent activation energies of the reduction were obtained as 34.4, 45.3, 34.9 kJ×mol-1, respectively.
Computational Study of CO 2 Reduction Catalyzed by Iron(I) Complex at Different Spin States: Cooperativity of Hydrogen Bonding and Auxiliary Group Effect

Abstract:To explore alternative approaches to the CO2 reduction to formate and provide an insight into the spin state effect on the CO2 reduction, we theoretically designed a kind of low-valence iron(I) model complex, whose doublet, quartet, and sextet states are denoted as 2 Fe(I), 4 Fe(I), and 6 Fe(I), respectively. This complex is featured with an iron(I) center, which bonds to a 1,2-ethanediamine (en) and a 2-hydroxy-biphenyl group. Reaction mechanisms for the CO2 reduction to formate catalyzed by this iron(I) model complex were explored using density functional theory (DFT) computations. Studies showed that the univalent iron(I) compound can efficiently fix and activate a CO2 molecule, whereas its oxidized forms with trivalent iron(III) or bivalent iron(II) cannot activate CO2. For the iron(I) compound, it was found that the lowest spin state 2 Fe(I) is the most favorable for the CO2 reduction as the reactions barriers involving 2 Fe(I), 4 Fe(I), and 6 Fe(I) are 25.6, 37.2, and 35.9 kcal/mol, respectively. Yet, a photosensitizer-free visible-light-mediated high-low spin shift from 4 Fe(I) and 6 Fe(I) to 2 Fe(I) is likely through the reverse intersystem crossing (RIC) because the 4 Fe(I) and 6 Fe(I) compounds have strong absorption in the visible-light range. Notably, the synergistic interaction between the hydrogen bonding from the auxiliary hydroxyl group in the 2-hydroxy-biphenyl moiety to CO2 and an intermediate five-membered ring promotes the proton transfer, leading to the formation of the -COOH moiety from CO2 and the Fe-O bond. With the addition of H2, one H2 molecule is split by the Fe-O bond and thus serves as H atom sources for both the CO2 reduction and the recovery of the auxiliary hydroxyl group. The present theoretical study provides a novel solution for the challenging CO2 reduction, which calls for further experimental verifications.

Reductive activation of the heme iron-nitrosyl intermediate in the reaction mechanism of cytochrome c nitrite reductase: a theoretical study

Abstract:Cytochrome c nitrite reductase catalyzes the six-electron, seven-proton reduction of nitrite to ammonia without release of any detectable reaction intermediate. This implies a unique flexibility of the active site combined with a finely tuned proton and electron delivery system. In the present work, we employed density functional theory to study the recharging of the active site with protons and electrons through the series of reaction intermediates based on nitrogen monoxide [Fe(II)-NO(+), Fe(II)-NO·, Fe(II)-NO(-), and Fe(II)-HNO]. The activation barriers for the various proton and electron transfer steps were estimated in the framework of Marcus theory. Using the barriers obtained, we simulated the kinetics of the reduction process. We found that the complex recharging process can be accomplished in two possible ways: either through two consecutive proton-coupled electron transfers (PCETs) or in the form of three consecutive elementary steps involving reduction, PCET, and protonation. Kinetic simulations revealed the recharging through two PCETs to be a means of overcoming the predicted deep energetic minimum that is calculated to occur at the stage of the Fe(II)-NO· intermediate. The radical transfer role for the active-site Tyr(218), as proposed in the literature, cannot be confirmed on the basis of our calculations. The role of the highly conserved calcium located in the direct proximity of the active site in proton delivery has also been studied. It was found to play an important role in the substrate conversion through the facilitation of the proton transfer steps.

Selective Four‐Electron Reduction of Oxygen by a Nonheme Heterobimetallic CuFe Complex

Abstract:We report herein the first nonheme CuFe oxygen reduction catalyst ([Cu II (bpbp)( μ ‐OAc) 2 Fe III ] 2+ , CuFe−OAc ), which serves as a functional model of cytochrome c oxidase and can catalyze oxygen reduction to water with a turnover frequency of 2.4×10 3 s −1 and selectivity of 96.0 % in the presence of Et 3 NH + . This performance significantly outcompetes its homobimetallic analogues (2.7 s −1 of CuCu−OAc with %H 2 O 2 selectivity of 98.9 %, and inactive of FeFe−OAc ) under the same conditions. Structure‐activity relationship studies, in combination with density functional theory calculation, show that the CuFe center efficiently mediates O−O bond cleavage via a Cu II ( μ ‐ η 1 : η 2 ‐O 2 )Fe III peroxo intermediate in which the peroxo ligand possesses distinctive coordinating and electronic character. Our work sheds light on the nature of Cu/Fe heterobimetallic cooperation in oxygen reduction catalysis and demonstrates the potential of this synergistic effect in the design of nonheme oxygen reduction catalysts.
Kinetic Studies on the Oxidation of Cytochrome B(5) Phe35 Mutants with Cytochrome C, Plastocyanin and Inorganic Complexes

Abstract:To illustrate the functions of the aromatic residue Phe35 of cytochrome b 5 and to give further insight into the roles of the Phe35-containing hydrophobic patch and/or aromatic channel of cytochrome b 5, we studied electron transfer reactions of cytochrome b 5 and its Phe35Tyr and Phe35Leu variants with cytochrome c, with the wild-type and Tyr83Phe and Tyr83Leu variants of plastocyanin, and with the inorganic complexes [Fe(EDTA)]–, [Fe(CDTA)]– and [Ru(NH3)6]3+. The changes at Phe35 of cytochrome b 5 and Tyr83 of plastocyanin do not affect the second-order rate constants for the electron transfer reactions. These results show that the invariant aromatic residues and aromatic patch/channel are not essential for electron transfer in these systems.
Atomistic determinants of co-enzyme Q reduction at the Qi-site of the cytochrome bc1 complex

Abstract:The cytochrome (cyt) bc1 complex is an integral component of the respiratory electron transfer chain sustaining the energy needs of organisms ranging from humans to bacteria. Due to its ubiquitous role in the energy metabolism, both the oxidation and reduction of the enzyme's substrate co-enzyme Q has been studied vigorously. Here, this vast amount of data is reassessed after probing the substrate reduction steps at the Qi-site of the cyt bc1 complex of Rhodobacter capsulatus using atomistic molecular dynamics simulations. The simulations suggest that the Lys251 side chain could rotate into the Qi-site to facilitate binding of half-protonated semiquinone - a reaction intermediate that is potentially formed during substrate reduction. At this bent pose, the Lys251 forms a salt bridge with the Asp252, thus making direct proton transfer possible. In the neutral state, the lysine side chain stays close to the conserved binding location of cardiolipin (CL). This back-and-forth motion between the CL and Asp252 indicates that Lys251 functions as a proton shuttle controlled by pH-dependent negative feedback. The CL/K/D switching, which represents a refinement to the previously described CL/K pathway, fine-tunes the proton transfer process. Lastly, the simulation data was used to formulate a mechanism for reducing the substrate at the Qi-site.
Ferrocene-Functionalized Metal.Organic Macrocycles Mimicking Hydrogenase for Photocatalytic Nitro Reduction

Abstract:Two novel metal-organic macrocycles (M-Fe and M-Cu) were constructed using the flexible and redox-active ferrocene ligand. The macrocycles effectively quenched the fluorescence of organic dyes and exhibited the capability to trigger the photoinduced electron transfer. Due to relatively lower reduction potentials, M-Cu exhibited better catalytic features for proton reduction and could better catalyze the reduction of nitro compounds. The large size of the substrates leads to a decreased efficiency in the nitro reduction process. The Michaelis-Menten mechanism was used to verify the photocatalytic system, which revealed the importance of the interaction between substrate and catalyst. CCDC: 2166465, M-Cu; 2166466, M-Fe.

A Density Functional Theory Study on the Active Center of Fe-only Hydrogenase: Characterization and Electronic Structure of the Redox States.

Abstract:We have carried out extensive density functional theory (DFT) calculations for possible redox states of the active center in Fe-only hydrogenases. The active center is modeled by [(H(CH(3))S)(CO)(CN(-))Fe(p)(mu-DTN)(mu-CO)Fe(d)(CO)(CN(-))(L)](z)() (z is the net charge in the complex; Fe(p)= the proximal Fe, Fe(d) = the distal Fe, DTN = (-SCH(2)NHCH(2)S-), L is the ligand that bonds with the Fe(d) at the trans position to the bridging CO). Structures of possible redox states are optimized, and CO stretching frequencies are calculated. By a detailed comparison of all the calculated structures and the vibrational frequencies with the available experimental data, we find that (i) the fully oxidized, inactive state is an Fe(II)-Fe(II) state with a hydroxyl (OH(-)) group bonded at the Fe(d), (ii) the oxidized, active state is an Fe(II)-Fe(I) complex which is consistent with the assignment of Cao and Hall (J. Am. Chem. Soc. 2001, 123, 3734), and (iii) the fully reduced state is a mixture with the major component being a protonated Fe(I)-Fe(I) complex and the other component being its self-arranged form, Fe(II)-Fe(II) hydride. Our calculations also show that the exogenous CO can strongly bond with the Fe(II)-Fe(I) species, but cannot bond with the Fe(I)-Fe(I) complex. This result is consistent with experiments that CO tends to inhibit the oxidized, active state, but not the fully reduced state. The electronic structures of all the redox states have been analyzed. It is found that a frontier orbital which is a mixing state between the e(g) of Fe and the 2 pi of the bridging CO plays a key role concerning the reactivity of Fe-only hydrogenases: (i) it is unoccupied in the fully oxidized, inactive state, half-occupied in the oxidized, active state, and fully occupied in the fully reduced state; (ii) the e(g)-2 pi orbital is a bonding state, and this is the key reason for stability of the low oxidation states, such as Fe(I)-Fe(I) complexes; and (iii) in the e(g)-2 pi orbital more charge accumulates between the bridging CO and the Fe(d) than between the bridging CO and the Fe(p), and the occupation increase in this orbital will enhance the bonding between the bridging CO and the Fe(d), leading to the bridging-CO shift toward the Fe(d).

Abstract:An imidazole modified silver electrode is prepared by immersing the substrate silver electrode in a 2% imidazole solution of ethanol at 50°C for 10 min. The modified electrode is men swept in a cytochrome c solution and the modified layer takes off because the modified electrode is very unstable. Although the amount of the silver-imidazole complex is very small compared with the amount of cytochrome c in the protein solution, it greatly facilitates redo reactions involving the biomacromolecules.

Abstract:The selective reductionof carbon dioxide remains a significantchallenge due to the complex multielectron/proton transfer process,which results in a high kinetic barrier and the production of diverseproducts. Inspired by the electrostatic and H-bonding interactionsobserved in the second sphere of the [NiFe]-CODH enzyme, researchershave extensively explored these interactions to regulate proton transfer,stabilize intermediates, and ultimately improve the performance ofcatalytic CO2 reduction. In this work, a series of cobalt(II)tetraphenylporphyrins with varying numbers of redox-active nitro groupswere synthesized and evaluated as CO2 reduction electrocatalysts.Analyses of the redox properties of these complexes revealed a consistentrelationship between the number of nitro groups and the correspondingaccepted electron number of the ligand at -1.59 V vs. Fc(+/0). Among the catalysts tested, TNPPCo with four nitro groupsexhibited the most efficient catalytic activity with a turnover frequencyof 4.9 x 10(4) s(-1) and a catalyticonset potential 820 mV more positive than that of the parent TPPCo.Furthermore, the turnover frequencies of the catalysts increased witha higher number of nitro groups. These results demonstrate the promisingdesign strategy of incorporating multielectron redox-active ligandsinto CO2 reduction catalysts to enhance catalytic performance.
Facile Electrocatalytic Proton Reduction by a [Fe-Fe]-Hydrogenase Bio-Inspired Synthetic Model Bearing a Terminal CN - Ligand

Abstract:An azadithiolate bridged CN - bound pentacarbonyl bis-iron complex, mimicking the active site of [Fe-Fe] H 2 ase is synthesized. The geometric and electronic structure of this complex is elucidated using a combination of EXAFS analysis, infrared and Mössbauer spectroscopy and DFT calculations. The electrochemical investigations show that complex 1 effectively reduces H + to H 2 between pH 0-3 at diffusion-controlled rates (10 11 M -1 s -1 ) i.e. 10 8 s -1 at pH 3 with an overpotential of 140 mV. Electrochemical analysis and DFT calculations suggest that a CN - ligand increases the pK a of the cluster enabling hydrogen production from its Fe(I)-Fe(0) state at pHs much higher and overpotential much lower than its precursor bis-iron hexacarbonyl model which is active in its Fe(0)-Fe(0) state. The formation of a terminal Fe-H species, evidenced by spectro-electrochemistry in organic solvent, via a rate determining proton coupled electron transfer step and protonation of the adjacent azadithiolate, lowers the kinetic barrier leading to diffusion-controlled rates of H 2 evolution. The stereo-electronic factors enhance its catalytic rate by 3 order of magnitude relative to a bis-iron hexacarbonyl precursor at the same pH and potential.

Abstract:CO2 can be electrochemically reduced to different products depending on the nature of catalysts. In this work, we report comprehensive kinetic studies on catalytic selectivity and product distribution of the CO2 reduction reaction on various metal surfaces. The influences on reaction kinetics can be clearly analyzed from the variation of reaction driving force (binding energy difference) and reaction resistance (reorganization energy). Moreover, the CO2RR product distributions are further affected by external factors such as electrode potential and solution pH. A potential-mediated mechanism is found to determine the competing two-electron reduction products of CO2 that shifts from thermodynamics-controlled product formic acid at less negative electrode potentials to kinetic-controlled product CO at more negative electrode potentials. Based on detailed kinetic simulations, a three-parameter descriptor is applied to identify the catalytic selectivity of CO, formate, hydrocarbons/alcohols, as well as side product H2. The present kinetic study not only well explains the catalytic selectivity and product distribution of experimental results but also provides a fast way for catalyst screening.

Abstract:To characterize the cytochrome b(5) (Cyt b(5))-cytochrome c (Cyt c) interactions during electron transfer, variants of Cyt b(5) have been employed to assess the contributions of electrostatic interactions (substitution of surface charged residues Glu44, Glu48, Glu56, and Asp60 and heme propionate), hydrophobic interactions, and the thermodynamic driving forces (substitutions for hydrophobic residues in heme pocket residues Phe35, Pro40, Val45, Phe58, and Val61). The electrostatic interactions play an important role in maintaining the stability and specificity of the Cyt b(5)-Cyt c complex that is formed. There is no essential effect on the intraprotein complex electron transfer even if most of the involved negatively charged residues on the surface of Cyt b(5) have been removed. The results support a dynamic docking paradigm for Cyt b(5)-Cyt c interactions. The orientation that is optimal for binding may not be optimal form for electron transfer. Substitution of hydrophobic residues does not have a significant effect on the binding between Cyt b(5) and Cyt c; rather, it regulates the electron transfer rates via changes in the driving force. Combining the electron transfer studies of the Cyt b(5)-Cyt c system and the Cyt b(5)-Zn-Cyt c system, we obtain the reorganization energy (0.6 eV) at an ionic strength of 150 mM.
Selectivity and Activity Modulation of Electrocatalytic Carbon Dioxide Reduction by Atomically Dispersed Dual Iron Catalyst

Abstract:Through regulating the electronic environment of Fe active centers to modulate electrocatalytic CO2 reduction behavior, the advanced dual Fe2-site catalyst (Fe2 DAC) exhibiting CO current density (jCO) of 10 mA/cm2 at overpotential of 330 mV in CO2-saturated 0.5 M KHCO3 electrolyte was designed and characterized by HAADF-STEM microscopy and XAS/EPR spectroscopies. With regard to Fe2 DAC displaying the higher charge transfer coefficient ( = 0.53), turnover frequency (TOFCO = 2.03 s-1) and CO Faradaic efficiency (FECO = 98.6 %) at overpotential of 400 mV, compared to that of single iron atom catalyst (Fe SAC,  = 0.31, TOFCO = 0.25 s-1, FECO = 60.1 %), the kinetic mechanism was investigated/elucidated by cation effect and in operando spectroscopy. The higher DMPO-CO2 EPR intensity (g = 2.0065, aN = 15.6 G, aH = 18.9 G) and the smaller separation of ATR-SEIRAS stretching frequencies (1554 (asym), 1288 (sym) cm-1,  = 266 cm-1) suggests that the structural type of [*COOCs] ̇/[*COOCs]- intermediate is 2-3 CO2 coordination (class II) for Fe2 DAC-triggered electrocatalytic CO2 reduction in CO2-saturated CsHCO3 solution. The strong orbital interaction among dual Fe2 site, intermediate [CO2] ̇ˉ/[CO2]2- and Cs+ cation (6s orbital) is proposed to accelerate charge transfer kinetics and shift the rate-determining step from electron transfer step (Li+, Na+) to protonation step (K+, Cs+), as evidenced by Cs+-induced increase in proton reaction order (0.86) and Cs+-induced decrease in kinetic Tafel slope (57.2 mV/dec) and electrochemical activation energy (23.7 kJ/mol). In contrast, the structural transformation from dual FeII2 motif to single FeII site revealed by the disappearance of Fe-Fe distance (3.10 Å) in operando Fe K edge EXAFS lends support to the absence of stretching frequencies (1429 (asym), 1380 (asym), 1241 (asym) cm-1) ascribed to 2-2 CO2 coordination in CO2-saturated LiHCO3 aqueous medium, unraveling that the transformation of [*COOCs]-/[*COOK]- into the bridge [CO2]2- [Fe--C(=O)O-Fe] is vital for electrocatalytic CO2-to-CO conversion. In addition to identify dinuclear Fe2II site as catalytic center, this study demonstrates that the thermodynamic stabilization effect of both cation size (large s orbital/soft hydration shell) and dual FeII2 motif toward [CO2] ̇ˉ/[CO2]2- intermediate is pivotal to the superior CO2RR kinetics (activity/selectivity). The proposed pathways (Cs+/K+/Na+ vs Li+ and dual Fe2 site vs single Fe site) may provide insight into how the orbital interaction and the peculiar electronic structure of dinuclear Fe2 site impacts the molecular-level mechanism for efficient electrocatalytic CO2 reduction.
